Summary Immunocytochemical methods were used to investigate the occurrence and distribution of sauvagine, corticotropin-releasing factor-, or urotensin I-like immunoreactivities (SVG-ir, CRF-ir, UI-ir, respectively) in the bullfrog (Rana catesbeiana) brain, using specific antisera raised against non-conjugated SVG, ovine CRF, rat/human CRF, and UI. In the hypothalamus, SVG-ir was found in the magnocellular perikarya, in the dorsal and ventral regions of the preoptic nucleus, and in the hypothalamo-hypophyseal projections to the external zone as well as the internal zone of the median eminence, to pars nervosa, and in fibres running from the pars nervosa to the pars intermedia of the pituitary. In contrast, CRF-ir was found only in parvocellular perikarya, mainly localized in the rostro-ventral part of the preoptic nucleus, with fine processes protruding through the ependyma of the third ventricle, fibre projections terminating in the anterior preoptic area and in the neuropil of the periventricular gray, and a caudal projection to the external zone of the median eminence. No CRF-ir staining was seen in the pars nervosa and pars intermedia. The use of UI-specific antisera failed to give a positive response in the frog brain. It is concluded that, in the frog brain, two anatomically different CRF-like (or SVG-like) systems co-exist, comparable to the reported co-existence of UI-ir and CRF-ir neuronal systems in fish brain.  相似文献

In this study, we hypothesize that hydroxyurea could provide an additional benefit as a free radical scavenger and/or iron chelator in β-thalassemia patients with iron overload. Twenty-one β-thalassemia intermedia patients who presented between 3 and 17 years but later required regular blood transfusions were enrolled for hydroxyurea therapy for a year. Fourteen patients responded to the therapy with hemoglobin levels maintained above 7.5?g/dl without transfusions. Hydroxyurea was discontinued after 6 months in seven patients who did not respond to the therapy and had to be continued on regular blood transfusions. We observed a statistically significant decrease in serum ferritin levels from 4194?±?4850?ng/ml to 2129?±?2380?ng/ml among the responders and from 2955?±?2909?ng/ml to 2040?±?2432?ng/ml among the non-responders and statistically significant decrease in labile iron pool from 18678.7?±?10067.4 mean fluorescence intensity (MFI) to 14888.5?±?5284.0?MFI among responders and from 17986.3?±?9079.8?MFI to 15634.8?±?8976.9?MFI among the non-responders after therapy. Phosphatidylserine externalization also showed a statistically significant decrease from 44.2?±?22.2?MFI to 16.6?±?6.7?MFI among the responders and from 46.9?±?33.1?MFI to 39.8?±?7.4?MFI among the non-responders along with a statistically significant decrease in the levels of reactive oxygen species from 72.8?±?35.5?MFI to 29.0?±?8.3?MFI among the responders and from 80.9?±?41.4?MFI to 40.5?±?15.8?MFI among the non-responders after therapy. A statistically significant increase in reduced glutathione levels was also observed from 430.8?±?201.1?MFI to 715.5?±?292.4?MFI among the responders and from 359.6?±?165.6?MFI to 450.3?±?279.5?MFI among the non-responders after therapy. This suggests the possible additional role of hydroxyurea as a free radical scavenger and/or iron chelator but requires a larger study for substantiation.  相似文献

Summary Quantified ultrastructural observations of the pars intermedia (PI) of the murine hypophysis enable evaluation and kinetic study of relatively fine secretory changes in the gland. Changes in volume of rER and newly formed dense secretory granules (Golgi granules) appear to best translate functional variations in the PI, as shown by the morphological effects of drugs affecting the dopaminergic control of the gland. Our morphometric results show that the PI is stimulated, but only briefly (no longer than 8–12 days), by both salt-loading and Na deprivation. However, the PI displays different secretory patterns in salt-loaded and Na-deprived mice; moreover, bromocriptine, which abolishes PI stimulation in Na-deprived mice, has only a slight inhibitory effect in salt-treated animals. Thus, it appears that the stimulation of the PI under both experimental conditions is triggered by different mechanisms. These results underline the plurifactorial control of the PI and show that the gland may have complex effects on hydromineral regulation.  相似文献

Abstract: Wild turkey (Meleagris gallopavo) broods spend the first several days of life on the ground until poult flight capabilities are attained. This is a critical period of wild turkey life history, with poult survival ranging from 12% to 52%. We measured vegetation in plots used by Rio Grande wild turkey (M. g. intermedia) preflight broods at 4 sites in southwest Kansas and the Texas Panhandle, USA, to determine microhabitat selection for ground roosting and to determine if microhabitat was related to poult survival. Hens selected ground-roost locations with more visual obstruction from multiple observation heights than random sites. Plots surrounding ground roosts had 1) greater visual obstruction; 2) increased tree decay; 3) higher percent grass, shrub, litter, and forb cover; and 4) lower percent bare ground cover than random sites. Grass, shrubs, and downed trees appeared to provide desired cover for ground-roosting broods. Poult survival increased with age of poult, size of brood, and density of shrubs 1–2 m tall. Plots used by broods <10 days old with above average survival contained more visual obstruction and shrubs than plots used by broods 10–16 days old with above average survival, signifying a shift in habitat use by successful broods as poults attain flight abilities. Density of shrubs 1–2 m tall in brood-use areas appears to be important for poult survival to 16 days of age on southern Great Plains rangeland habitats. Ground-level vegetative cover appears to be a significant factor in preflight poult survival. Provisions of ground-level vegetative cover should be considered during wild turkey brooding periods where increased poult survival is desired.  相似文献

This study was designed to clarify whether the pars flaccida (PF) as a flexible part of the tympanic membrane is capable of reacting to pressure oscillations (PO) with amplitudes and frequencies typical for natural atmospheric pressure fluctuations in the far infrasound frequency range (APF). If so, the PF mechanical reactions to APF might be involved in the overall physiologic regulation processes, which make organisms susceptible to APF. The displacements of the PF in response to PO were measured in vitro in ears of gerbils and rabbits by means of laser Doppler vibrometry. The index of the PF reactivity (Ra) was determined as the ratio of the amplitude of the PF oscillations (PFO) to the amplitude of the PO. All kinds of PO applied caused PFO. The amplitude of the PFO increased when the amplitude of the PO was increased. In gerbils, a decrease in Ra with the increase in amplitude of the PO was observed. In the range of PO lowest amplitudes (4–20 Pa) Ra proved to be 1.4 times higher than in the range of highest amplitudes (90–105 Pa). Considering that the natural APF are usually within the range of ±20 Pa, this fact points to an important contribution of the PF to the pressure dynamics in the middle ear (ME) of gerbils. In rabbit ears, Ra was lower and recovery from plastic deformation was slower than in gerbils. Our findings are in line with the suggestion that the PF might play an important role in respect of adaptation to natural APF.  相似文献

Antisera against a variety of vertebrate and invertebrate neuropeptides were used to characterize neurons with somata in the pars intercerebralis (PI), pars lateralis (PL), and subesophageal ganglion (SEG), designated as PI neurons, PL neurons, and SEG neurons, respectively, all of which project to the retrocerebral complex in the blow fly, Protophormia terraenovae. Immunocytochemistry combined with backfills through the cardiac-recurrent nerve revealed that at least two pairs of PI and SEG neurons for each were FMRFamide-immunoreactive. Immunoreactivity against [Arg7]-corazonin, beta-pigment-dispersing hormone (beta-PDH), cholecystokinin8, or FMRFamide was observed in PL neurons. Immunoreactive colocalization of [Arg7]-corazonin with beta-PDH, [Arg7]-corazonin with cholecystokinin8, or beta-PDH with FMRFamide was found in two to three somata in the PL of a hemisphere. Based on their anatomical and immunocytochemical characteristics, PI neurons were classified into two types, PL neurons into six types, and SEG neurons into two types. Fibers in the retrocerebral complex showed [Arg7]-corazonin, beta-PDH, cholecystokinin8, and FMRFamide immunoreactivity. Cholecystokinin8 immunoreactivity was also detected in intrinsic cells of the corpus cardiacum. The corpus allatum was densely innervated by FMRFamide-immunoreactive varicose fibers. These results suggest that PI, PL, and SEG neurons release [Arg7]-corazonin, beta-PDH, cholecystokinin8, or FMRFamide-like peptides from the corpus cardiacum or corpus allatum into the hemolymph, and that some PL neurons may simultaneously release several neuropeptides.  相似文献

BACKGROUND AND AIMS: Natural hybridization was investigated between two predominantly allohexaploid wheatgrasses, weedy Elytrigia repens and steppic E. intermedia, with respect to habitats characterized by different degrees of anthropogenic disturbance. METHODS: Using flow cytometry (relative DNA content), 269 plants from three localities were analysed. Hybrids were further analysed using nuclear ribosomal (ITS1-5.8S-ITS2 region) and chloroplast (trnT-F region) DNA markers in addition to absolute DNA content and chromosome numbers. KEY RESULTS: Weedy E. repens was rare in a steppic locality whereas E. intermedia was almost absent at two sites of agricultural land-use. Nevertheless, hybrids were common there whereas none were found at the steppic locality, underlining the importance of different ecological conditions for hybrid formation or establishment. At one highly disturbed site, > 16 % of randomly collected plants were hybrids. Hexaploid hybrids showed intermediate genome size compared with the parents and additive patterns of parental ITS copies. Some evidence of backcrosses was found. The direction of hybridization was highly asymmetric as cpDNA identified E. intermedia as the maternal parent in 61 out of 63 cases. Out of nine nonaploid cytotypes (2n = 9x = 63) which likely originated by fusion of unreduced and reduced gametes of hexaploids, eight were hybrids whereas one was a nonaploid cytotype of E. repens. The progeny of one nonaploid hybrid demonstrated gene flow between hexaploid and nonaploid cytotypes. CONCLUSIONS: The results show that E. repens and E. intermedia frequently cross at places where they co-occur. Hybrid frequency is likely influenced by habitat type; sites disturbed by human influence sustain hybrid formation and/or establishment. Hexaploid and nonaploid hybrid fertility is not negligible, backcrossing is possible, and the progeny is variable. The frequent production of new at least partially fertile cyto- and genotypes provides ample raw material for evolution and adaptation.  相似文献

SIFamide receptor (SIFR) is a Drosophila G protein-coupled receptor for the neuropeptide SIFamide (SIFa). Although the sequence and spatial expression of SIFa are evolutionarily conserved among insect species, the physiological function of SIFa/SIFR signaling remains elusive. Here, we provide genetic evidence that SIFa and SIFR promote sleep in Drosophila. Either genetic ablation of SIFa-expressing neurons in the pars intercerebralis (PI) or pan-neuronal depletion of SIFa expression shortened baseline sleep and reduced sleep-bout length, suggesting that it caused sleep fragmentation. Consistently, RNA interference-mediated knockdown of SIFR expression caused short sleep phenotypes as observed in SIFa-ablated or depleted flies. Using a panel of neuron-specific Gal4 drivers, we further mapped SIFR effects to subsets of PI neurons. Taken together, these results reveal a novel physiological role of the neuropeptide SIFa/SIFR pathway to regulate sleep through sleep-promoting neural circuits in the PI of adult fly brains.  相似文献

EVIDENCE FOR INCREASES IN ANTARCTIC BLUE WHALES BASED ON BAYESIAN MODELLING   总被引:2,自引:1,他引:1  
Antarctic blue whales ( Balaenoptera musculus intermedia ) are the largest and formerly most abundant blue whale subspecies, but were hunted to near extinction last century. Estimated whaling mortality was unsustainable from 1928 to 1972 (except during 1942–1944), depleting them from 239,000 (95% interval 202,000–311,000) to a low of 360 (150–840) in 1973. Obtaining statistical evidence for subsequent increases has proved difficult due to their scarcity. We fitted Bayesian models to three sighting series (1968–2001), constraining maximum rates of increase to 12% per annum. These models indicated that Antarctic blue whales are increasing at a mean rate of 7.3% per annum (1.4%–11.6%). Informative priors based on blue whale biology (4.3%, SD = 1.9%) and a Bayesian hierarchical meta-analysis of increase rates in other blue whale populations (−3%, SD = 11.6%), suggest plausible increase rates are lower (although the latter has wide intervals), but a meta-analysis of other mysticetes obtains similar rates of increase (6.7%, SD = 4.0%). Possible biases affecting the input abundance estimates are discussed. Although Antarctic blue whales appear to have been increasing since Sovier illegal whaling ended in 1972, they still need to be protected-their estimated 1996 population size, 1,700 (860–2,900), was just 0.7% (0.3%–1.3%) of the pre-exploitation level.  相似文献
