Citrus greening (Huanglongbing, HLB) is a destructive disease associated with the uncultivable, phloem-limited, gram-negative bacteria Candidatus Liberibacter spp., which affects citrus crops and other Rutaceae species. HLB is rapidly transmitted by the Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae). We developed an artificial medium on which D. citri eggs can hatch and first-instar nymphs can feed. The medium could be used to study insect physiology and screen molecules that may interfere with egg hatching and nymph development. 相似文献

Citrus decline has become a devastating problem in citrus-growing regions of southern Iran. The affected trees show progressive yellowing of leaves, thinning of the canopy, dieback and total collapse. Despite a number of attempts, the aetiology of the decline has remained uncertain. In the present study, we evaluated the potential association of various biotic and abiotic factors with the decline in the Fars Province of Iran. Biotic agents surveyed included Candidatus Liberibacter asiaticus, Spiroplasma citri, phytoplasmas, Tylenchulus semipenetrans and root infecting fungi and oomycetes. Abiotic factors studied were soil and water salinity, changes in prevailing temperature and other environmental conditions. In our surveys, Ca. L. asiaticus, either alone or in combination with other factors, had the highest frequency of association (92%) with the decline, followed by S. citri (75%). Ca. L. asiaticus was not detected in any of the non-decline trees examined. Pythium or Phytophthora species and a fungus of theFusarium solani species complex were also isolated from roots of many declining trees. Phytoplasmas were found only in few cases, and populations of citrus nematode were often below the threshold of economic loss. Soil/water salinity were in the suitable range for citrus cultivation in most cases. It is suggested that the decline is initiated by Ca. L. asiaticus infection, which is known to weaken the root system and make it vulnerable to infection by opportunistic soil fungi and oomycetes. S. citri, summer temperatures, low air humidity and overbearing of the trees seem to be other potential factors contributing to the intensity of the disease. 相似文献

Asiatic citrus psylla, Diaphorina citri, Kuwayama (Hemiptera: Liviidae) is an economic pest of citrus groves and a vector of the bacterium, Candidatus Liberibacter spp., one of the causative agents of citrus greening. In order to estimate the infectivity of six different isolates of Beauveria bassiana, Metarhizium anisopliae and Isaria fumosorosea, fungal bioassay was performed on the adults of D. citri. Adults of D. citri were treated individually with 1 × 105, 1 × 106, 1 × 107, 1 × 108, 2 × 108 spores/mL fungal concentrations by the immersion method. Subsequent to fungal bioassay, treated D. citri were used to determine the levels of esterase and glutathione S‐transferases (GST) enzymes over a period of 3–7 days. The mortality results suggest that I. fumosorosea isolates (If‐02) caused 82.2% mortality on the seventh day of treatment. However, B. bassiana isolate (Bb‐08) with lowest LC50 (1.4 × 107 spores/mL) proved to be highest potential isolate against D. citri. Biochemical determination of esterase and GST activity assay showed significant differences in activities after infection of fungi. Significantly high activity of esterase was observed by Bb‐01 (27.0 unit per mg protein) on the seventh day, while Ma‐11.1 and If‐2.3 (16.9 and 36.3 unit per mg protein) on the third day post treatment. However, maximum GST's activity was showen by isolates Bb‐08,Ma‐M2 and If‐2.3 (37.6, 1.40 and 10.9 unit per mg protein) on the third day. The current investigation will help to explore the relations between the insect defense system and entomopathogenic fungi. The citrus mealybug, Planococcus citri (Risso) (Hemiptera: Pseudococcidae) is a serious pest of economically important crops worldwide. The apefly, Spalgis epius (Westwood) (Lepidoptera: Lycaenidae) is a potential predator of various species of mealybugs. Earlier investigation on its daily preying capacity and preference for prey stages on P. citri is incomplete. Hence, a study was conducted to find out the daily prey consumption ability and preference for prey stages by different larval instars of S. epius reared on P. citri in the laboratory. Through the 8-day developmental period with four larval instars, the daily prey consumption of S. epius increased from the first to the seventh day and decreased on the eighth day prior to the prepupal stage. Generally, there was a significant difference in the prey consumption on different days. When the prey stages were offered separately, the first to fourth instar larva of S. epius consumed, respectively, a mean of 199.6, 722.6, 1908.8, and 4625.6 eggs or 21.5, 77.0, 168.5, and 670.5 nymphs or 3.2, 7.2, 16.0, and 35.1 adults of P. citri. When an S. epius larva was fed on P. citri eggs, nymphs and adults separately, it consumed a mean of 7456.7 eggs, 937.6 nymphs, or 62.3 adults during its entire development. When the prey stages were offered all together, a single S. epius larva consumed 2618.4 eggs, 170.4 nymphs, and 39.7 adults of P. citri throughout its entire development. The study revealed that S. epius is a voracious predator of P. citri and thus could be utilized as a major biological control agent. 相似文献

Abstract The population of the citrus red mite, Panonychus citri (McGregor), does not increase on pear from spring to mid-summer but thereafter increases abruptly. To elucidate this phenomenon, we compared the performance of the mites on pear leaves with that on citrus leaves, at different time throughout the pear-growing season. No significant difference was detected between the oviposition rate on pear and that on citrus throughout the season. However, the survival rate of ovipositing females that had fed on pear and the hatch rate of eggs laid by those females were significantly lower than those for females that had fed on citrus, until August. However, no significant difference was observed thereafter. The results showed that the decline of the population of citrus red mite before autumn is due to the high mortality of adult females that had fed on pear leaves and the low hatch rate of the eggs produced by those females. 相似文献

The YaeQ family of proteins are found in many Gram-negative and a few Gram-positive bacteria. We have determined the first structure of a member of the YaeQ family by X-ray crystallography. Comparisons with other structures indicate that YaeQ represents a new compact protein fold built around a variation of the PD-(D/E)XK nuclease motif found in type II endonucleases and enzymes involved in DNA replication, repair, and recombination. We show that catalytically important residues in the PD-(D/E)XK nuclease superfamily are spatially conserved in YaeQ and other highly conserved YaeQ residues may be poised to interact with nucleic acid structures. 相似文献

The Asian citrus psyllid (ACP), Diaphorina citri Kuwayama (Hemiptera:Psyllidae), is one of the most important pests of citrus plants worldwide, due to be the dominant vector of the devastating citrus disease Huanglongbing (HLB). Selection of suitable oviposition site is a crucial event of phytophagous insect life. Usually, the chemical and physical characteristics of a host plant were recognized by sensilla on the ovipositor and tarsi of a phytophagous insect after settling on a plant surface. In this study, the morphology, number, and distribution pattern of the ovipositorial and tarsal sensilla of adult female psyllids, were observed by scanning electron microscopy (SEM) techniques. The effect of these sensilla on oviposition was investigated by sensilla deactivation using hydrochloric acid (HCl). The results showed that sensilla trichoid (ST), sensilla chaetica (ChS) and sensilla basiconica (SB) were distributed on ovipositor, while sensilla trichoid (ST) were distributed on the external genitalia. Sensilla chaetica (ChS) and basiconica (SB) were mainly distributed on the end of the ovipositor back plate, and on the base of the sternum. On tarsus, sensilla trichoid (ST), sensilla chaetica (ChS), sensilla triangular (TeS) and sensilla ear ball (SE) were observed. However, whole tarsal segments were covered with Böhm's mane, which was arranged in a circular line. Oviposition assay revealed that a significantly lower number of eggs were laid on the Murraya paniculata seedlings confined by the psyllids with deactivated ovipositorial sensilla followed by protarsal sensilla, mesotarsal sensilla and hindtarsal sensilla.
