Ginsenoside Rh(4) (1), a saponin isolated from the roots of Panax notoginseng (Burk.) F. H. Chen, was evaluated for its haemolytic activity and adjuvant potential on specific antibody and cellular response to ovalbumin (OVA) in mice. Compound 1 showed a slight haemolytic effect, its concentration inducing 50% of the maximum haemolysis (HD(50) value) being 407+/-12 microg/ml using a 0.5% suspension of red blood cells. Compound 1 significantly increased the concanavalin A (Con A)-, lipopolysaccharide (LPS)-, and OVA-induced splenocyte proliferation in OVA-immunized mice especially at a dose of 25 microg (P<0.05, P<0.01, or P<0.001). The OVA-specific serum IgG, IgG1, and IgG2b antibody levels were also significantly enhanced by 1 at a dose of 25 microg compared to the OVA control group (P<0.05 or P<0.01). Moreover, the enhancing effect of 1 on the OVA-specific IgG2b antibody responses to OVA in mice was more significant than that of Alum (Al(OH)(3) gel; P<0.01). These results suggest that 1 could be safely used as adjuvant with low or non-haemolytic effect.  相似文献

Previous studies have revealed the inhibitory effects of an acidic polysaccharide purified from the root of Panax ginseng against the adhesion of Helicobacter pylori to gastric epithelial cells and the ability of Porphyromonas gingivalis to agglutinate erythrocytes. In this study, this acidic polysaccharide from P. ginseng, PG-F2, was investigated further, in order to characterize its antiadhesive effects against Actinobacillus actinomycetemcomitans, Propionibacterium acnes, and Staphylococcus aureus. The minimum inhibitory concentrations (MIC) were found to be in a range of 0.25-0.5mg/mL. However, results showed no inhibitory effects of PG-F2 against Lactobacillus acidophilus, Escherichia coli, or Staphylococcus epidermidis. PG-F2 is a pectin-type polysaccharide with a mean MW of 1.2 x 10(4) Da, and consists primarily of galacturonic and glucuronic acids along with rhamnose, arabinose, and galactose as minor components. The complete hydrolysis of PG-F2 via chemical or carbohydrolase enzyme treatment resulted in the abrogation of its antiadhesive activity, but limited hydrolysis via treatment with pectinase (EC. 3.2.1.15) yielded an oligosaccharide fraction, with activity comparable to the precursor PG-F2 (the MIC of ca. 0.01 mg/mL against H. pylori and P. gingivalis). Our results suggest that PG-F2 may exert a selective antiadhesive effect against pathogenic bacteria, while having no effects on beneficial and commensal bacteria.  相似文献

Roots of Panax ginseng exposed to various concentrations of Cu (0.0, 5, 10.0, 25.0, and 50.0 μM) accumulated high amounts of Cu in a concentration-dependent and duration-dependent manner. Roots treated with 50 μM Cu resulted in 52% and 89% growth inhibition after 20 and 40 days, respectively. Saponin synthesis was stimulated at a Cu concentration between 5 and 25 μM but decreased at 50 μM Cu. Malondialdehyde content (MDA), lipoxygenase activity (LOX), superoxide ion (O2 •−) accumulation, and H2O2 content at 5 and 10 μM Cu-treated roots were not increased but strongly increased at 50 μM Cu resulting in the oxidation of ascorbate (ASC) and glutathione (GSH) to dehydroascorbate (DHA) and glutathione disulfide (GSSG), respectively indicating a clear oxidative stress. Seven well-resolved bands of superoxide dismutase (SOD) were detected in the gel and an increase in SOD activity seemed to be mainly due to the induction of Fe-SOD 3. Five to 10 μM Cu slightly induced activity of ascorbate peroxidase (APX) and dehydroascorbate reductase (DHAR), guaiacol peroxidase (G-POD) but inhibited monodehydroascorbate reductase (MDHAR) and glutathione reductase (GR) enzyme activities. No changes in catalase (CAT) activity and in activity gel were found up to 25 μM Cu, but both G-POD and CAT activities were inhibited at 50 μM Cu. Glutathione metabolism enzymes such as γ-glutamylcysteine synthetase (γ-GCS), glutathione-S-transferase (GST), and glutathione peroxidase activities (GPx) were activated at 5 and 10 μM Cu but were strongly inhibited at 50 μM Cu due to the Cu accumulation in root tissues. The strong depletion of GSH at 50 μM Cu was associated to the strong induction of γ-glutamyltranspeptidase (γ-GGT) activity. These results indicate that plant could grow under Cu stress (5–25 μM) by modulating the antioxidant defense mechanism for combating Cu induced oxidative stress.  相似文献

The effect of the root-inoculum size and axuin concentration on growth of adventitious roots and accumulation of ginsenosides were studied during suspension cultures of ginseng (Panax ginseng C.A. Meyer). Of the various concentrations of indole-3-butyric acid (IBA) and γ-naphthaleneacetic acid (NAA) used as supplementary growth regulators along with Murashige and Skoog medium, 25 μM IBA was found suitable for lateral root induction and growth, as well as accumulation of ginsenosides. Inoculum size of 5 g L−1 was found suitable for optimal biomass (10.5 g L−1 dry biomass) and ginsenosides (5.4 mg g−1 DW) accumulation. Of the various length of root inocula tested (chopped to 1–3, 4–6, 7–10 mm and un-chopped), root inocula of 7–10 mm was found suitable for biomass and ginsenoside accumulation.  相似文献

目的:研究西洋参(Panax quinguefolium L)抗突变作用和对细胞增殖的影响.方法:采用小鼠骨髓淋巴细胞有丝分裂指数试验、染色体畸变试验和微核试验,观察西洋参对小鼠骨髓淋巴细胞遗传损伤效应的影响.结果:丝裂霉素(Mitomycin-c MMC)单独作用时,能抑制小鼠骨髓淋巴细胞的增殖(P<0.01),提高小鼠骨髓淋巴细胞的染色体突变(P<0.01)和微核率(P<0.01).西洋参高、中、低剂量组与MMC联合作用时,有丝分裂指数明显比模型组提高(P<0.01),并且均能显著地抑制由于MMC引起的染色体突变,其抑制率分别为68.25%、34.97%和14.29%,同时对MMC诱发的微核率也有明显的抑制效应.结论:西洋参能抑制MMC诱发的染色体畸变率和微核率,促进细胞的增殖能力.  相似文献

建立西洋参药材分类方法;采用电感耦合等离子体质谱(ICP-MS)法对12个西洋参样品中的15种无机元素的含量进行测定,用高效液相色谱(HPLC)法测定上述样品中的7种人参皂苷的含量,用蒽酮-硫酸法测定其中多糖的含量;进而采用主成分分析法(PCA)对所测得的西洋参样品的23个变量进行分类研究;12个西洋参样品能得到合理的分类,而各人参皂苷的含量是决定西洋参样品分类的第1关键因素,元素Mn、Cu、As、Ni、Mo以及多糖的含量是第2关键因素;主成分分析法是西洋参分析分类的有效方法。  相似文献

挖掘云南文山三七内生放线菌菌种资源以及对分离到的放线菌菌株进行三七根腐病病原菌的抗菌活性评价,为生物防治三七根腐病的发生、传播、有效治理及解决三七连作障碍和后续研究三七内生放线菌的代谢产物提供菌种资源。采用三种分离方法、六种分离培养基对文山两个产地三七全株植物进行内生放线菌分离,分离到的菌株通过16S rRNA基因扩增测序鉴定到属,通过琼脂扩散法对菌株发酵液进行抗菌活性初筛。共分离到56株内生放线菌,经形态特征初步排重后,选22株代表菌株进行16S rRNA基因扩增测序,并鉴定到属,其分属于链霉菌属(Streptomyces)、放线产孢菌属(Actinomycetospora)、拟诺卡氏菌属(Nocardiopsis)、黄英菌属(Yinghuangia)四个属。对23株菌株发酵液进行抗菌活性初筛,14株菌株(60.87%)具有不同程度的抗菌活性,其中菌株S004、S006、S014、S016、S022、S042、S047、S053对三七根腐病病原菌有较好的抗菌活性,具备进一步深入研究的价值。本研究初步探索出适合三七内生放线菌分离的分离方法,获得一批三七内生放线菌菌种资源,并筛选出8株对...  相似文献

人参(PanaxginsengC.A.Mey.)培养细胞经细胞平板克隆,获得近300个克隆系。克隆系在细胞生长速率和寡糖素含量及产率上均存在显著差异,且寡糖素产率和细胞生长之间有明显的相关性。经11代连续继代培养观察及过氧化物酶同工酶谱特征分析,筛选到一株寡糖素产率高且稳定的克隆系PG-180,其平均生长速率是0.495g于重/L·天,为原始株系的1.39倍,平均寡糖素含量为14.69%干重,是亲本的1.65倍,平均寡糖素产率是2.183g/L,为原始株系的2.32倍。比较克隆系PG-180和原始株系细胞悬浮培养时间进程发现,由人参培养细胞生产寡糖素的最佳细胞收获期为3周左右。  相似文献
