Currently, ocean acidification is occurring at a faster rate than at any time in the last 300 million years, posing an ecological challenge to marine organisms globally. There is a critical need to understand the effects of acidification on the vulnerable larval stages of marine fishes, as there is potential for large ecological and economic impacts on fish populations and the human economies that rely on them. We expand upon the narrow taxonomic scope found in the literature today, which overlooks many life history characteristics of harvested species, by reporting on the larvae of Rachycentron canadum (cobia), a large, highly mobile, pelagic‐spawning, widely distributed species with a life history and fishery value contrasting other species studied to date. We raised larval cobia through the first 3 weeks of ontogeny under conditions of predicted future ocean acidification to determine effects on somatic growth, development, otolith formation, swimming ability, and swimming activity. Cobia exhibited resistance to treatment effects on growth, development, swimming ability, and swimming activity at 800 and 2100 μatm pCO2. However, these scenarios resulted in a significant increase in otolith size (up to 25% larger area) at the lowest pCO2 levels reported to date, as well as the first report of significantly wider daily otolith growth increments. When raised under more extreme scenarios of 3500 and 5400 μatm pCO2, cobia exhibited significantly reduced size‐at‐age (up to 25% smaller) and a 2–3 days developmental delay. The robust nature of cobia may be due to the naturally variable environmental conditions this species currently encounters throughout ontogeny in coastal environments, which may lead to an increased acclimatization ability even during long‐term exposure to stressors.  相似文献

Cell migration is a highly integrated, multistep process that plays an important role in physiological and pathological processes. The migrating cell is highly polarized, with complex regulatory pathways that integrate its component processes spatially and temporally.1 Ridley AJ, Schwartz MA, Burridge K, Firtel RA, Ginsberg MH, Borisy G, Parsons JT, Horwitz AR. Cell migration: integrating signals from front to back. Science 2003; 302:1704-9; PMID:14657486; http://dx.doi.org/10.1126/science.1092053[Crossref], [PubMed], [Web of Science ®] [Google Scholar] The Drosophila tumor suppressor, Lethal (2) giant larvae (Lgl), regulates apical-basal polarity in epithelia and asymmetric cell division.2 Etienne-Manneville S. Polarity proteins in migration and invasion. Oncogene 2008; 27:6970-80; PMID:19029938; http://dx.doi.org/10.1038/onc.2008.347[Crossref], [PubMed], [Web of Science ®] [Google Scholar] But little is known about the role of Lgl in establishing cell polarity in migrating cells. Recently, we showed that the mammalian Lgl1 interacts directly with non-muscle myosin IIA (NMIIA), inhibiting its ability to assemble into filaments in vitro.3 Dahan I, Yearim A, Touboul Y, Ravid S. The tumor suppressor Lgl1 regulates NMII-A cellular distribution and focal adhesion morphology to optimize cell migration. Mol Biol Cell 2012; 23:591-601; PMID:22219375; http://dx.doi.org/10.1091/mbc.E11-01-0015[Crossref], [PubMed], [Web of Science ®] [Google Scholar] Lgl1 also regulates the cellular localization of NMIIA, the maturation of focal adhesions, and cell migration.3 Dahan I, Yearim A, Touboul Y, Ravid S. The tumor suppressor Lgl1 regulates NMII-A cellular distribution and focal adhesion morphology to optimize cell migration. Mol Biol Cell 2012; 23:591-601; PMID:22219375; http://dx.doi.org/10.1091/mbc.E11-01-0015[Crossref], [PubMed], [Web of Science ®] [Google Scholar] We further showed that phosphorylation of Lgl1 by aPKCζ prevents its interaction with NMIIA and is important for Lgl1 and acto-NMII cytoskeleton cellular organization.4 Dahan I, Petrov D, Cohen-Kfir E, Ravid S. The tumor suppressor Lgl1 forms discrete complexes with NMII-A and Par6α-aPKCζ that are affected by Lgl1 phosphorylation. J Cell Sci 2014; 127:295-304; PMID:24213535; http://dx.doi.org/10.1242/jcs.127357[Crossref], [PubMed], [Web of Science ®] [Google Scholar] Lgl is a critical downstream target of the Par6-aPKC cell polarity complex; we showed that Lgl1 forms two distinct complexes in vivo, Lgl1-NMIIA and Lgl1-Par6-aPKCζ in different cellular compartments.4 Dahan I, Petrov D, Cohen-Kfir E, Ravid S. The tumor suppressor Lgl1 forms discrete complexes with NMII-A and Par6α-aPKCζ that are affected by Lgl1 phosphorylation. J Cell Sci 2014; 127:295-304; PMID:24213535; http://dx.doi.org/10.1242/jcs.127357[Crossref], [PubMed], [Web of Science ®] [Google Scholar] We further showed that aPKCζ and NMIIA compete to bind directly to Lgl1 through the same domain. These data provide new insights into the role of Lgl1, NMIIA, and Par6-aPKCζ in establishing front-rear polarity in migrating cells. In this commentary, I discuss the role of Lgl1 in the regulation of the acto-NMII cytoskeleton and its regulation by the Par6-aPKCζ polarity complex, and how Lgl1 activity may contribute to the establishment of front-rear polarity in migrating cells.  相似文献

Zebrafish larvae provide models of muscle development, muscle disease and muscle-related chemical toxicity, but related studies often lack functional measures of muscle health. In this video article, we demonstrate a method to measure force generation during contraction of zebrafish larval trunk muscle. Force measurements are accomplished by placing an anesthetized larva into a chamber filled with a salt solution. The anterior end of the larva is tied to a force transducer and the posterior end of the larva is tied to a length controller. An isometric twitch contraction is elicited by electric field stimulation and the force response is recorded for analysis. Force generation during contraction provides a measure of overall muscle health and specifically provides a measure of muscle function. Although we describe this technique for use with wild-type larvae, this method can be used with genetically modified larvae or with larvae treated with drugs or toxicants, to characterize muscle disease models and evaluate treatments, or to study muscle development, injury, or chemical toxicity.  相似文献

In order to evaluate the early tolerance of Mactra chinensis to salinity, the treatments of salinity gradients and salinity gradual changes were set in this study, and the post growth and development of juveniles were analyzed in recovery experiment, respectively. The result showed that the optimum hatching of zygotes was found at a salinity from 24 to 32, which is narrower than that of larvae (20–32); a slight of low salinity (16–32) will benefit the early growth and development of M. chinensis; at planktonic and creeping stages, low salinity stress (20) was conducive to promoting the growth of juvenile M. chinensis philippi; 4, 48 was the ultimate salinity of M. chinensis; The range of early tolerance of larvae M. chinensis philippi to salinity can be widened through a short period of salinity acclimation.  相似文献

The ontogenetic structure and longitudinal variability of ichthyoplankton in a confluence zone of two river systems in the Eastern Amazon were evaluated between December 2018 and April 2019, at the time 137 eggs and 7,687 larvae of fish were captured. Ichthyoplankton proved to be heterogeneous between sections of rivers with differences in the number of taxa and abundance. The assemblages were distinguished by a NMDS Analysis, a ANOSIM, a SIMPER and a CCA, they presented a spatial zonation pattern with strong distinction and variability in the composition of the species between the river systems, under strong influence of limnological variables. The assemblages were mainly represented by larvae of Clupeiformes and Perciformes in the clear waters of the Tapajós river and Characiformes and Siluriforms in the cloudy waters of the Amazon river, influenced by the existence of a limnological gradient. Larvae in more advanced development stages were predominant in the Tapajós river while early stages were dominant in the Amazon river. The confluence zone does not seem to be a spawning area, but it is an important transport site for the larvae to reach the nursery areas in the lower stretch of the Amazon river. The encounter of these two river systems seems to guarantee the survival and biological recruitment of several fish species.  相似文献

Atlantic bluefin tuna is an iconic scombrid species with a high commercial and ecological value. Despite their importance, many physiological aspects, especially during the larval stages, are still unknown. Metabolic rates are one of the understudied aspects in scombrid larvae, likely due to challenges associated to larval handling before and during respirometry trials. Gaining reliable estimates of metabolic rates is essential to understand how larvae balance their high growth needs and activity and other physiological functions, which can be very useful for fisheries ecology and aquaculture. This is the first study to (a) estimate the relationship between routine metabolic rate (RMR) and larval dry weight (DW) (mass scaling exponent) at a constant temperature of 26°C, (b) measure the RMR under light and darkness and (c) test whether the interindividual differences in the RMR are related to larval nutritional status (RNA/DNA and DNA/DW). The RMR scaled nearly isometrically with body size (b = 0.99, 0.60–31.56 mg DW) in contrast to the allometric relationship observed in most fish larvae (average b = 0.87). The results show no significant differences in larval RMR under light and darkness, suggesting similar larval activity levels in both conditions. The size explained most of the variability in RMR (97%), and nutritional condition was unrelated to the interindividual differences in routine metabolism. This is the first study to report the metabolic rates of Atlantic bluefin tuna larvae and discuss the challenges of performing bioenergetic studies with early life stages of scombrids.  相似文献

ABSTRACT

Larvae of the invasive mosquito Culex quinquefasciatus Say are morphologically similar to those of the native Culex pervigilans Bergroth, yet distinguishing these species can be hampered by morphological variations in Cx. quinquefasciatus. We present detail about the extent of these variations in an urban population of Cx. quinquefasciatus in Auckland. To aid in identification of this exotic species, we provide images of key diagnostic characters and some observed exceptions to these. Details regarding variation in diagnostic characters for < 3rd instar and 3rd/4th instar larvae are given. Of the nine characters used for identification, three were highly consistent (dorsal papillae, mantle plate, pecten teeth); each observed in > 90% of larvae, although these characters were not always visible. Other characters were less reliable, for instance, the expected position of seta 1a-S in relation to the pecten teeth was observed in < 10% of larvae. Further exploration of regional morphological variation in both Cx. quinquefasciatus and Cx. pervigilans is recommended, ideally with associated molecular characterisation.  相似文献

The shrimp Macrobrachium amazonicum (Heller 1862) has an extremely large geographic range (>4000?km across) in northern and central South America, comprising estuarine and fully limnic inland populations, which are hydrologically isolated from each other. Significant variations in ecology, physiology, reproduction, and larval development suggest an at least incipient allopatric speciation due to limited genetic exchange. In a comparative experimental investigation with shrimps from the Pantanal (upper Paraguay River basin) and the Amazon delta, respectively, we measured larval body size, dry weight (W), biochemical (total protein; lipid; fatty acids, FA), and elemental composition (carbon, hydrogen, nitrogen; collectively CHN) at hatching. All these early larval traits are relevant for the degree of developmental dependence on planktonic food sources. Various consistent differences were observed between the two populations: Newly hatched larvae produced by shrimps from the Amazon delta were significantly smaller and showed lower values of W, CHN, protein, and unsaturated FA compared to those from the Pantanal. On the other hand, they contained significantly higher quantities of total lipid and saturated FA and, in consequence, higher ratios of lipid:protein, C:N, and saturated:unsaturated FA. All these differences in biomass and chemical composition suggest that the larvae of the Amazon population are energetically better adapted to planktonic food limitation, which likely occurs during riverine downstream transport toward coastal marine waters, also explaining previous observations of much stronger initial starvation tolerance in larvae from the Amazon versus those from the Pantanal. The latter develop in highly productive lentic inland waters, where large body size, an early onset of feeding, and a strong musculature (indicated by a high protein content) should facilitate their role as planktonic predators and allow for fast growth. An initial independence from food (lecithotrophy in the zoea I stage) as well as a preference for oligohaline rather than fully limnic conditions observed in the Pantanal larvae are interpreted as traits that have persisted from an ancestral coastal marine clade. Altogether, consistent ontogenetic differences between shrimps from the Pantanal and the Amazon estuary support the hypothesis that the taxon M. amazonicum comprises a complex of closely related but separate species.  相似文献

The mangrove horseshoe crab, Carcinoscorpius rotundicauda, is an Asian species found in Malaysia. This ancient arthropod has a long incubational period during which it depends on various energy sources for both embryogenesis and organogenesis. This study describes the trend of energy utilization from the endogenous reserves by the developing embryos from 0 to 40 days of incubation (until the hatching of the larvae). The dry weight, insoluble protein, carbohydrate, glycogen and lipid showed a declining trend from 0 to 40 days of incubation, whereas the wet weight, water content, ash content and soluble protein showed an increasing trend. Selected micro-elements such as Cu2+, Fe2+ and Zn2+ also demonstrated an interesting trend in the developing eggs when the egg mass was subjected to inductively coupled plasma mass spectrometry analysis, where these elements showed a high correlation with the moulting stages and egg development. Maximum variations within the micro-elements were observed during the 1st (10 days after fertilization) and 2nd (35–36 days after fertilization) moulting stages within the developing eggs. This study clearly indicated that the moulting cycles of C. rotundicauda during embryonic development influence energy utilization in the form of carbohydrates, lipids, proteins, glycogen and other micro-elements.  相似文献
