Aims: To evaluate the diversity and antimicrobial activity of bacteria from the marine sponges Suberites carnosus and Leucosolenia sp. Methods and Results: Two hundred and thirty‐seven bacteria were isolated from the sponges S. carnosus (Demospongiae) and Leucosolenia sp. (Calcarea). Isolates from the phyla Actinobacteria, Bacteroidetes, Firmicutes and Proteobacteria were obtained. Isolates of the genus Pseudovibrio were dominant among the bacteria from S. carnosus, whereas Pseudoalteromonas and Vibrio were the dominant genera isolated from Leucosolenia sp. Approximately 50% of the isolates from S. carnosus displayed antibacterial activity, and c. 15% of the isolates from Leucosolenia sp. demonstrated activity against the test fungal strains. The antibacterial activity observed was mostly from Pseudovibrio and Spongiobacter isolates, while the majority of the antifungal activity was observed from the Pseudoalteromonas, Bacillus and Vibrio isolates. Conclusions: Both sponges possess a diverse range of bioactive and potentially novel bacteria. Differences observed from the sponge‐derived groups of isolates in terms of bioactivity suggest that S. carnosus isolates may be a better source of antibacterial compounds, while Leucosolenia sp. isolates appear to be a better source of antifungal compounds. Significance and Impact of the Study: This is the first study in which cultured bacterial isolates from the marine sponges S. carnosus and a Leucosolenia sp. have been evaluated for their antibacterial activity. The high percentage of antibacterial isolates from S. carnosus and of antifungal isolates from Leucosolenia sp. suggests that these two sponges may be good sources for potentially novel marine natural products.  相似文献

目的:对采自海南、湛江等海域的海绵样品进行放线菌选择性分离,采用其发酵液进行抗肿瘤活性筛选,并对活性较好的菌株进行鉴定。方法:用含50μg/mL重铬酸钾为抑制剂的海水高氏一号合成培养基分离培养海绵放线菌;以MTT法进行菌株的抗肿瘤活性筛选;通过培养特征、形态特征、生理生化特征、16S rDNA序列测定及系统发育分析,对菌株HA01184进行鉴定。结果与结论:海水高氏一号合成培养基用于海绵放线菌分离培养具有很好的选择性,从海绵样品中共分离得到放线菌165株,细胞毒活性达80%以上的阳性菌株有10株,其中菌株HA01184的发酵液细胞毒活性为90%。结合形态观察、生理生化特征和16S rDNA序列比对分析,将HA01184归于链霉菌属,可能是来自海洋环境的一个潜在新种。  相似文献

This study presents the application of the porous poly(D,L-lactic-co-glycolic acid) (PLGA) sponges fabricated from an organic solvent free supercritical gas foaming technique. Two formulations of PLGA sponges with different co-polymer compositions (85:15 and 50:50) were fabricated as novel scaffolds to guide human hepatoma cell line, Hep3B cell growth in vitro. The PLGA sponges showed desirable biodegradability and exhibited uniform pore size distribution with moderate interconnectivity. It was observed in this study that cells cultured on PLGA sponges showed lower proliferation rate as compared to the control during 14 days of culture as measured by using total DNA and methylthiazol tetrazolium (MTT) assays. However, the cells cultured on the sponges tended to aggregate to form cell islets which were able to express better hepatic functions. The enzyme-linked immunosorbent assay (ELISA) results showed that the cell-sponge constructs secreted 1.5-3.0 times more albumin than the control when normalized to cellular content. In a similar fashion, its detoxification ability was also predominantly higher than that of the control as indicated by the ethoxyresorufin-O-deethylase (EROD) results. By comparing the cells growing on the two formulations of PLGA sponges, it was found that the PLGA 85:15 sponge exhibited better conductive and desirable environment for hep3B cells as justified by better cell infiltration, higher proliferation and hepatic function than the PLGA 50:50 sponge.  相似文献

从辽宁海域的繁茂膜海绵(Hymeniacidonperleve)中分离到1株解淀粉类芽孢杆菌(Paenibacillusamyloliquefaciens),定名为海绵细菌B25W。它产生的抗生素用白色念珠菌(Candidaalbicans)为指示菌,通过单因子和均匀设计实验,发现菌株最佳产抗摇瓶发酵培养基:玉米面0.34%,豆饼粉1.66%,葡萄糖0.1%,ZnSO40.04%,MgSO40.02%,KH2PO40.04%;发酵条件:种龄12h,接种量10%,发酵时间20h,初始pH6.0,250mL三角瓶内含25mL培养基,往复式摇床,116次/min,振幅11cm,培养温度28℃。应用二剂量法(以制霉菌素为标样)测定B25W抗生素的相对效价为13680u/mL,比优化前效价(8444u/mL)提高了62%。  相似文献

Abstract. It is widely accepted that multicellular animals (metazoans) constitute a monophyletic unit, deriving from ancestral choanoflagellate‐like protists that gave rise to simple choanocyte‐bearing metazoans. However, a re‐assessment of molecular and histological evidence on choanoflagellates, sponge choanocytes, and other metazoan cells reveals that the status of choanocytes as a fundamental cell type in metazoan evolution is unrealistic. Rather, choanocytes are specialized cells that develop from non‐collared ciliated cells during sponge embryogenesis. Although choanocytes of adult sponges have no obvious homologue among metazoans, larval cells transdifferentiating into choanocytes at metamorphosis do have such homologues. The evidence reviewed here also indicates that sponge larvae are architecturally closer than adult sponges to the remaining metazoans. This may mean that the basic multicellular organismal architecture from which diploblasts evolved, that is, the putative planktonic archimetazoan, was more similar to a modern poriferan larva lacking choanocytes than to an adult sponge. Alternatively, it may mean that other metazoans evolved from a neotenous larva of ancient sponges. Indeed, the Porifera possess some features of intriguing evolutionary significance: (1) widespread occurrence of internal fertilization and a notable diversity of gastrulation modes, (2) dispersal through architecturally complex lecithotrophic larvae, in which an ephemeral archenteron (in dispherula larvae) and multiciliated and syncytial cells (in trichimella larvae) occur, (3) acquisition of direct development by some groups, and (4) replacement of choanocyte‐based filter‐feeding by carnivory in some sponges. Together, these features strongly suggest that the Porifera may have a longer and more complicated evolutionary history than traditionally assumed, and also that the simple anatomy of modern adult sponges may have resulted from a secondary simplification. This makes the idea of a neotenous evolution less likely than that of a larva‐like choanocyte‐lacking archimetazoan. From this perspective, the view that choanoflagellates may be simplified sponge‐derived metazoans, rather than protists, emerges as a viable alternative hypothesis. This idea neither conflicts with the available evidence nor can be disproved by it, and must be specifically re‐examined by further approaches combining morphological and molecular information. Interestingly, several microbial lin°Cages lacking choanocyte‐like morphology, such as Corallochytrea, Cristidiscoidea, Ministeriida, and Mesomycetozoea, have recently been placed at the boundary between fungi and animals, becoming a promising source of information in addition to the choanoflagellates in the search for the unicellular origin of animal multicellularity.  相似文献

Perfusion culture of fetal human hepatocytes in microfluidic environments   总被引:1,自引:0,他引:1  
Various types of bioreactors composed of microstructured PDMS (Polydimethylsiloxane) layers have recently been fabricated for perfusion culture of mammalian cells such as adult rat hepatocytes. As a new feature of those bioreactors, in this study, cultivation of fetal human hepatocytes (FHHs) was attempted, because they have high possibility to mature in vitro with preserving their normality, which is suitable for inplantation of liver tissue equivalents reconstituted in vitro. During the perfusion culture in the PDMS bioreactors for 1 week, cells showed good attachment, spreading and reached their confluence over the channels. In addition, their albumin production was significantly enhanced in the perfusion culture using the PDMS bioreactors up to about four times during the FHH perfusion culture when compared in dish-level static culture. Hep G2 cell cultures were also performed and have also shown under perfusion conditions an enhanced cell activity multiplied by 2 compared to static conditions. Although, the cellular activities of FHH cells are still low even compared to those of the Hep G2 cells, the conclusions of this work is encouraging toward future liver tissue engineering based on in vitro propagation and maturation of hepatocyte progenitors combined with microfabrication technologies.  相似文献

The alkaloid ageladine A, a pyrrole-imidazole alkaloid isolated from marine Agelas sponges shows fluorescence in the blue-green range during excitation with UV light with the highest absorption at 370 nm. The fluorescence of this alkaloid is pH dependent. Highest fluorescence is observed at pH 4, lowest at pH 9 with the largest fluorescence changes between pH 6 and 7. Ageladine A is brominated, which facilitates membrane permeation and therefore allows for easy staining of living cells and even whole transparent animal staining. To calculate the exact pH in solutions, cells, and tissues, the actual concentration of the alkaloid has to be known. A ratiometric measurement at the commonly used excitation wavelengths at 340/380 nm allows pH measurements in living tissues with an attenuated influence of the ageladine A concentration on calculated values. The fluorescence changes report small intracellular pH changes induced by extracellular acidification and alkalization as well as intracellular alkalization induced by ammonium chloride.  相似文献

Sponges assemblages were sampled in four coastal study regions (Malindi, Kenya; Quirimba Archipelago, northern Mozambique; Inhaca Island, Southern Mozambique and Anakao, Madagascar) in the west Indian Ocean. Sponge species were counted in multiple 0.5 m2 quadrats at depths of between 0 and 20 m at a number of sites within localities within each region. Despite the relatively small areas sampled, sponge samples comprised a total of 130 species and 70 genera of the classes Demospongiae and Calcarea. Sponges are clearly a major taxon in these regions in terms of numbers of species, percentage cover or biomass, although their ecology in the west Indian Ocean is virtually unknown. Nearly half of the genera, e.g. Iotrochota, found were species with a so‐called Tethyan distribution. Most of the other genera were cosmopolitan, e.g. Clathria, but some were cold water (Coelosphaera), Indo‐Australian (Ianthella) or circum‐African (Crambe). Many of the species encountered in the present study occurred in at least two study regions, many in more and could occupy large areas of substratum. Some of these, e.g. Xestospongia exigua, are commonly found throughout the Indo‐west Pacific region where they also occupy much space. The endemicity of the shallow water sponge faunas in East Africa (20–25%) seem to be high within the Indo‐Pacific realm but are lower than northern Papua New Guinea. The tropical regions (Kenya and Northern Mozambique) were more speciose than subtropical regions (southern Mozambique and Madagascar) but not significantly more diverse (Shannon H′). Although latitude was not a major influence on sponge community patterns, hard substratum assemblages did form a cline from the tropics to Southern Mozambique, linked by Madagascar. Substratum nature (habitat) was most important in influencing the suite and number of species present. Sponge assemblages of soft substrata were much more dissimilar, both within and between habitats, than those on hard substrata. There was a predictable variability in species richness between hard substratum habitats: coral reefs being speciose and caves being less so. Our findings showed that both patterns and influences on species richness may be decoupled from those influencing diversity. In our data species richness, but not diversity, showed striking regional and bathymetric trends. In addition, sponge species richness mainly split at coral reef vs. non‐reef habitats, whilst diversity divided principally into assemblages on hard and soft substrata. We consider this dichotomy of findings between species richness and diversity values to be important, as these are two principal measures used for the interpretation of biodiversity.  相似文献
