Identification of the predominant constituents produced by the plus and the minus strains of Blakeslea trispora is described. The occurrence of xylenes in the volatiles produced by the plus strain is reported. Additionally, production of 3-hydroxy-3-methylbutanal by the plus strain and dimethyl allyl alcohol by the minus strains were confirmed. The investigation of stems and leaves of Catha cassinoides afforded, in addition to sitosterol, β-amyrin, ursolic acid, lup-20(29)-en-3β,30-diol and friedelin, three new pentacyclic triterpenes: 30-hydroxyfriedelan-3-one, 29-hydroxyfriedelan-3-one and 3-oxo-friedelan-29-oic acid. The BALB/3T3 clone A31 mouse embryo cell line has been used by many investigators as a model “normal” “fibroblast” line for a variety of in vitro studies. It has been shown, however, that these cells are not “normal” because they will produce tumors within 2–4 months if 3 × 104 cells are implanted subcutaneously in BALB/c mice attached to 0.2 × 5 × 10-mm plastic plates. Previous studies also suggested that these cells were not fibroblasts because they gave rise to tumors with the characteristics of vascular endothelium not fibroblasts. We now report that BALB/3T3 (clone A31), BALB/3T3-T, a proadipocyte subclone of clone A31 cells, and six recent subclones of BALB/3T3-T cells show additional differentiation patterns when tumors derived by implantation of these cells attached to plastic plates are examined. Differentiation into pericytes, chondrocytes, and fibroblasts was observed. We conclude that the BALB/3T3 clone A31 cell line and related lines are multipotent mesenchymal cells which are capable of differentiation into a variety of cell types. 相似文献

Summary Tritiated -amanitin has been used as a specific and sensitive probe to estimate the number of RNA polymerase B molecules in isolated nuclei, chromatin and nucleoids, obtained from macroplasmodia ofPhysarum polycephalum. During mitosis (metaphase±10 min) there is at least 10-fold less RNA polymerase B than at all phases of the cell cycle, even if DNA replication has been blocked in vivo. It is concluded that many of the RNA polymerase B molecules leave the chromatin during decondensation of the chromosomes in telophase of the synchronous nuclear division ofPhysarum. 相似文献

Summary Simian virus 40-transformed 3T3 cells are dependent on serum for survival and growth. This growth activity can be separated
on a pH 2 Sephadex G100 column into two fractions: a high molecular weight activity and a low molecular weight substance that
has recently been characterized as containing as its major agent, biotin.
To replace the remainder of the serum requirement, hormones and other growth factors were tested. Both insulin at high, nonphysiological
concentrations (200 to 500 ng/ml) and transferrin (5×10−8
M) stimulate the growth rate in low serum medium (0.3% v/v bovine calf serum DME) individually and, when added together, are
nearly as growth enhancing as 10% serum.
The need for the residual serum in this medium can be eliminated by the use of crystalline trypsin during trypsinization.
Under these serum-free conditions, biotin and transferrin supplementation provide for moderately good growth (20 to 30 hr
population doubling time, 1×106 cells/3.2-cm dish final cell density). Insulin addition further stimulates the growth rate (16 to 20 hr) and the final density
(1.5×106 cells). Although the protein growth factors, EGF (0.5 to 1.0 ng/ml) and FGF (4 to 10 ng/ml), also appear to enhance growth
individually and additively, their effects are slight and very variable. Nevertheless, the complete serum-free medium (DME
supplemented with biotin, transferrin, insulin, EGF and FGF) yields growth comparable but still inferior to 10% serum supplementation
(14-versus 12-hr population doubling time, 1 to 2×106 versus 2 to 3×106 cells final cell density).

Effects of accumulation of 3-O-methylglucose on levels of endogenous osmotic solutes in Chlorella emersonii 总被引:2,自引:2,他引:0
Abstract. Regulation of the concentration of osmotic solutes was studied in Chlorella emersonii grown at external osmotic pressures (II) ranging between 0.08 and 1.64MPa. NaCl was used as osmoticum. The total solute content of the cells was manipulated by applying 2 mol m−3 3- O -methylglucose (MG), which was not metabolized, and accumulated at concentrations ranging between 60 and 230 mol m−3 within 4 h after its addition to the medium. Methylglucose uptake resulted in decreases in concentrations of proline and sucrose, the two solutes mainly responsible for osmotic adaptation of C. emersonii to high external II. The responses were consistent with the hypothesis that proline and sucrose concentrations are controlled by a system of osmotic regulation, with turgor and/or volume as a primary signal. Short-term experiments showed that even very small increases in turgor and/or volume, due to accumulation of methylglucose, resulted in large decreases in proline and sucrose. Over the first 30-60 min the total solute concentration in the cells increased by at most 15 osmol m−3 which would represent an increase in turgor pressure of at most 0.04 M Pa. Yet, the decreases in proline and sucrose were as fast as those in cells exposed to a sudden decrease of 0.25 MPa in external II, when the turgor pressure would have increased by at least 0.15 MPa. High concentrations of methylglucose in cells grown at high II did not affect the rapid synthesis of proline and sucrose which started when the cells were transferred to yet higher II. Thus, methylglucose had no direct effects on proline and sucrose metabolism, and it has been assumed that it acted solely as an inert osmotic solute within the cell. 相似文献
