The specific binding protein for substance P (SP) was solubilized in an active form from the crude mitochondrial (P2) fraction of bovine brainstem. After incubation with 3-[(3-cholamidopropyl)dimethylammonio]-1-propane sulfonate (CHAPS) and 0.1 M NaCl at 0 degrees C for 30 min, the SP binding to the supernatant fraction (100,000 g, 60 min) was determined by the glass fiber filtration method reported by Bruns et al. (1983). The specific [3H]SP binding to the solubilized fraction was highly specific for SP and was displaced by nanomolar concentrations of SP and physalaemin, but only by micromolar concentrations of eledoisin. In addition, the binding was inhibited by GTP (approximately 40% of the specific binding decreased by 10 microM GTP) in both preparations. These results were virtually identical to those of P2 membrane preparations and suggested that this high-affinity SP binding site belongs to the SP-P type. Scatchard analyses of SP binding to the solubilized fraction revealed a single saturable component with a Bmax of 22.0 +/- 5.10 fmol/mg protein and a KD of 0.79 nM, and these values are almost the same as those obtained in the P2 fraction (Bmax = 31.3 +/- 3.56 fmol/mg protein, KD = 0.82 nM). Gel filtration analysis showed that the detergent-SP binding protein complex has two calculated molecular weights of greater than 1,000,000 and 55,000-60,000 (a corresponding Stokes radius of 35.5 nm). 相似文献

Phosphorus deficiency is one of the most growth-limiting factors in acid soils in various parts of the world. The objective of this study was to screen 25 rice cultivars (Oryza sativa L.) at low, medium, and high levels of soil P. Number of tillers, root length, plant height, root dry weight and shoot dry weight were related to tissue P concentrations, P uptake and P-use efficiency. Shoot weight was found to be the plant parameter most sensitive to P deficiency. Significant cultivar differences in P use efficiency were found. Phosphorus use efficiency was higher in roots than shoots and decreased with increasing levels of soil P. Positive correlations were found among growth parameters such as plant height, tillers, root and shoot weight, and P content of roots and shoots. These results indicate selection of rice cultivars for satisfactory performance under low P availability can be carried out using shoot and root dry weight as criteria. 相似文献

Sodium acetate extractable P and P sorption were measured in a high P-sorbing Andept soil (Mission series) following incubation with Winter wheat (Triticum aestivum (L.)Nugaines) and red clover (Trifolium pratense (L.)Florex) plant residue. The effects of plant residue P content, moisture regime, and quantity added were evaluated over a twelve week incubation period. Increased extractable P and decreased P sorption occured when the P content of residue exceeded 0.1% P for both winter wheat and red clover. The extractable P decreased and the P sorbed increased for all treatments and the check as the incubation time progressed. Extractable P increased and P sorption decreased more under air dry moisture conditions than at –0.03 MPa or saturation. Extractable P levels increased and P sorption decreased with increasing amounts of added residue. The effects of the residue additions decreased with increasing incubation time. 相似文献

Summary The calcium sensitivity of exocytosis from electroper-meabilized chromaffin cells is increased by activators of protein kinase C, such as TPA and certain phorbol esters, diacylglycerols, and mezerein. A range of putative inhibitors of protein kinase C block both the phorbol ester-sensitive component of secretion and also the underlying insensitive component. These inhibitors are also shown to inhibit medulla protein kinase C activity in vitro. The extent of secretion is reduced when electropermeabilized cells are exposed to Ca2+ levels much in excess of 50 m. The onset of inhibition is faster than the relatively slow rate of Ca-dependent exocytosis and is insensitive to inhibitors of proteolysis. Adrenal medulla protein kinase C activity is also irreversibly inhibited by high Ca2+ concentrations. Both the secretory response and the protein kinase C activity in vitro have similar nucleotide and cation specificities. Although these data do not definitely establish an involvement of protein kinase C in exocytosis, none argue against it.Deceased 相似文献

Neurotransmitter receptors and ion channels play a critical role in the transduction of signals at chemical synapses. The modulation of neurotransmitter receptor and ion channel function by protein phosphorylation is one of the major regulatory mechanisms in the control of synaptic transmission. The nicotinic acetylcholine receptor (nAcChR) has provided an excellent model system in which to study the modulation of neurotransmitter receptors and ion channels by protein phosphorylation since the structure and function of this receptor have been so extensively characterized. In this article, the structure of the nAcChR from the electric organ of electric fish, skeletal muscle, and the central and peripheral nervous system will be briefly reviewed. Emphasis will be placed on the regulation of the phosphorylation of nAcChR by second messengers and by neurotransmitters and hormones. Summary The effects of the antibiotics methotrexate and chloramphenicol on somatic embryogenesis inCitrus were evaluated. Relatively low levels (0.1 to 1.0 μg/ml) of these antibiotics did not inhibit embryo production from undeveloped
ovules of ‘Key’ lime [C. aurantifolia) (Christm.) Swing.]. Surprisingly, both antibiotics induced the formation of embryogenic callus in these cultures. This is
usually a rare event in cultures of undevelopedCitrus ovules, and ‘Key’ lime is especially recalcitrant. The effects of these antibiotics on embryogenic callus appeared to be
limited to the induction stage, because there was no consistent effect, either stimulatory or inhibitory, on established,
lines of embryogenic callus.

Influence of the food plants ofHeliothis armigera (Hb.) on the degree of parasitism by exotic parasiteCotesia kazak Telenga (Hymenoptera: Braconidae) was studied in cages in the laboratory on 7 food plants such as cotton (Gossypium hirsutum L.), tomato (Lycopersicon esculentum Mill), okra [Abelmoschus esculentus (L.) Moench], Dolichos (dolichos lablab L.), pigeonpea [Cajanus cajan (L.) Millsp.], Cowpea (Vigna unquiculata (L.) and chickpea (Cicer arietium L.). To determine the preference of the parasite 2 test methods were employed. In single plant choice test cotton was most
preferred. Next in order of preference were tomato and okra. Dolichos, pigeonpea, cowpea and chickpea were least preferred.
In multiple choice test, however, cotton and okra were preferred followed by tomato. Parasites were seen visiting these plants
very frequently and high parasitism was recorded on these plants. Chick pea, pigeon pea, cowpea and Dolichos were the least
preferred food plants. There appears to be some difference in fecundity as affected by some food plants. Exposure on okra,
cotton and tomato resulted in higher cocoon production as compared to pigeonpea, Dolichos, cowpea and chickpea. There was,
however, no difference in sex-ratio and longevity of the progeny as affected by food plants. This exotic parasite should be
released first in crops such as cotton, okra and tomato on whichH. armigera is a very serious pest in India and elsewhere.

Chronic treatment of hamsters with estradiol for several months has previously been shown to decrease the specific content
of cytochrome P450 in the kidney, a target of hormonal carcinogenesis, but not in liver. The reason for this decrease in metabolic
enzyme activity is unknown and has been examined in this investigation. We now report that the decrease in specific content
of renal cytochrome P450 by 73% in response to estradiol was not affected by co-treatment with tamoxifen for 1 month. The
subcutaneous infusion of 250 μg/day estradiol for 7 days lowered renal cytochrome P450 by 71% from control values and was
therefore used for further mechanistic studies. This treatment decreased renal activities of estradiol 2- or 4-hydroxylase
by 77 to 80%, of 7-ethoxycoumarin-O-deethylase by 66% of control values, respectively, and completely eliminated aryl hydrocarbon
hydroxylase activities, whereas liver enzymes remained unaffected. After 7 days of infusion of estradiol, fluorescent products
of lipid peroxidation were more than doubled in hamster kidney but remained unchanged in liver. The possibility of enzyme
destruction by binding of estradiol 2,3-quinone to metabolizing enzymes was investigatedin vitro. In the presence of 2-hydroxyestradiol, cumene hydroperoxide, and microsomes, conditions known to favor the oxidation of
the steroid to quinone, the binding of catechol estrogen metabolite to microsomal protein increased 60 fold over control values
in the absence of cofactor. Purified rat liver cytochrome P450c also oxidized 2-hydroxyestradiol to 2,3-estradiol quinone.
The rate of oxidation was linear for the first 2–3 min, but thereafter decreased with time. Under these incubation conditions,
irreversible binding of catechol estrogen metabolite to cytochrome P450c increased for the first 2–3 min and then remained
at this plateau level. It was concluded that enzyme destruction by a reactive estrogen metabolite or by lipid peroxides may
be a major reason for the organ-specific decrease in cytochrome P450 enzymes in kidneys of estrogen-treated hamsters. 相似文献

The present study examines the time dependent effects of n-6 and n-3 polyunsaturated fatty acids on liver microsomal lipid metabolism in FVB mice fed a diet supplemented with a mixture of free fatty acids (mainly 18:3n-6 and 20:5n-3) at 25 mg/g diet. Significant changes in the fatty acid composition of total liver and microsomal lipids were observed after 7 days on the diets. Thereafter, some animals remained on the same diet while others were fed a diet supplemented with hydrogenated coconut oil (HCO). With the exception of 20:5n-3 which showed a slower recovery, establishment of the HCO pattern was rapid indicating that the diet-induced changes could be easily reversed. The unsaturation index, the cholesterol/phospholipid ratio and the microviscosity of the microsomal membranes were not affected by these dietary manipulations. Unsaturated fatty acid supplementation reduced the activity of 9 desaturase by 50%. Feeding the HCO diet to mice previously fed the EPA/GLA diet led to a progressive increase in 9 desaturase activity, reaching 80% of the day zero values after 14 days. The monoene content of hepatic total lipids reflected, in most cases, the changes in enzyme activity. This study shows that a low dose of a n-3 and n-6 free fatty acid mixture increases the quantities of members of the n-3 family, without loss of n-6 fatty acids in microsomal membranes and modifies the activity of 9 desaturase without altering the microsome physicochemical parameters. 相似文献
