Summary Interference microscopy was used to measure the dry mass of nucleoli in unfixed nuclei isolated from neurones of the paraventricular (PV) and supraoptic (SO) nuclei of female rats. Changes in nucleolar dry mass during pregnancy and lactation have been interpreted as reflecting changes in rates of synthesis of ribosomes and protein in these neurones. Measurements were made on a total of 6580 nucleoli from 135 rats. At the end of pregnancy nucleolar dry mass of both PV and SO neurones was increased compared with virgin female rats. Nucleolar dry mass of PV neurones but not SO neurones increased further during lactation. This change was biphasic, with a nadir at 2 weeks post partum. After day 5 post partum, nucleolar dry mass of PV and SO neurones was increased only in rats suckling pups. Adjustment of litter size to 10 or 22 to 24 pups on the first day post partum did not affect nucleolar changes in PV and SO neurones. Nucleolar changes were less when only one pup was nursed. The results are discussed in relation to oxytocin secretion induced by the suckling stimulus and the synthetic response of PV and SO neurones to increased secretion.  相似文献

Obesity is a major public health problem. The prevalence of obesity has significantly increased in developed countries, particularly in France with an overall increase of 76% over the last 15 years. In pregnant women, obesity is associated with alterations in the quality of labor, such as delayed onset of labor, a higher rate of prolonged pregnancies, prolonged labor, and higher oxytocin requirements. There is also an increased prevalence of Cesarean sections, particularly during the active phase of labor, and perinatal complications (postpartum hemorrhage).It seems that some of these functional changes and their consequences can be attributed to a disruption of hormonal balance encountered in obese women and involving adipokines (apelin, ghrelin, visfatin, leptin), but also to the interactions between adipose tissue and the “oxytocin (OT) – oxytocin receptor (OTR)”.In this review, we detailed mechanisms to understand the impact of specific metabolic alterations in obesity on uterine contractility.Better knowledge of the impact of obesity on labor and delivery pathophysiology should strengthen the prevention of obesity in women of childbearing age and provide a suitable and effective management. The beneficial effect of weight loss and exercise in non-pregnant women on the correction of metabolic disorders secondary to obesity should be studied in populations of overweight women to demonstrate its effectiveness.  相似文献

We have characterized the interaction between the Zn2+ ions and the histidine analogues of oxytocin and arginine-vasopressin. Potentiometric methods were used for the determination of the stoichiometry of the complexes formed and the calculation of their stability constants. The NMR measurements revealed detailed structures of the complexes and confirmed the binding mode at physiological pH.  相似文献

In order to investigate the effects of the nonapeptide hormone arginine vasotocin (AVT) on the maturation of the brain, the following developmental data were tabulated between 5 and 21 days of postnatal life, in kittens, after the daily intraperitoneal administration of 10?6 mg synthetic AVT: sleep, daily increase of body weight and locomotor, and investigative activities (LIA). Likewise, the day of the eye opening was noted and the brain weight as well as the total lipid levels within the brain in the day of sacrifice (21 days of age) were measured. The daily administration of AVT induced: (1) an increase of the total amount as well as of the intensity of active sleep (AS); (2) a decrease of the LIA; (3) a decrease of the total lipid levels within the brain and (4) a retardation of the eye opening. These effects appeared to be specific because neither arginine vasopressin, nor oxytocin, in the same doses (10?6 mg), were able to reproduce the effects of AVT. The present results demonstrate that chronic administration of AVT is associated with a retardation of brain maturation. Whether AVT induces this effect by an unique mechanism or there are different mechanisms for the reported developmental data that were affected by AVT, is unknown. However, the present results suggest that the pineal gland, by its effector within the brain, AVT, is involved by an inhibitory pathway in the brain maturation and the hypothesis is advanced that the decrease of AVT content of fetal and neonatal brain could represent a hormonal signal for triggering the beginning of the brain maturation phenomena.  相似文献
