The twin-arginine transport (Tat) system is a prokaryotic protein transport system. Escherichia coli mutants in this pathway show a defect in cell separation during cell division, resulting in destabilization and permeability
of the outer membrane. Maltose uptake is catalysed by a membrane-bound transporter of the ATP binding cassette (ABC) superfamily,
where MalE is the essential periplasmic binding protein component. Here, we report that tat mutants are unexpectedly able to transport maltose in the absence of malE. This observation is specific to the MalE component since co-inactivation of malF, which encodes one of the channel components of the transporter, completely abolishes maltose transport even when the Tat
system is inactivated. Genetic repair of the outer membrane leaky phenotype of the tat mutant strain re-established the absolute requirement for MalE in maltose uptake. In addition, we demonstrate that phenotypic

OMP85 is a highly conserved outer membrane protein in all Gram-negative bacteria. We studied an uncharacterized OMP85 homolog of Porphyromonas gingivalis, a primary periodontal pathogen forming subgingival plaque biofilms. Using an outer-loop peptide antibody specific for the OMP85 of P. gingivalis, loop-3 Ab, we found a difference in the mobility of OMP85 on SDS-PAGE gel between the P. gingivalis wild-type and the isogenic galE mutant, a deglycosylated strain, suggesting that OMP85 naturally exists in a glycosylated form. This was also supported by a shift in OMP85 PAGE mobility after chemical deglycosylation treatment. Further, loop-3 Ab cross-reacted with the galE mutant stronger than the wild-type strain; and could inhibit biofilm formation in the galE mutant more than in the wild-type strain. In conclusion, this is the first report providing the evidence of OMP85 glycosylation and the involvement of OMP85 in biofilm formation. 相似文献

Serum glycans are promising markers for early-stage cancer detection, but the research remains challenging because low concentrations of serum glycoproteins are secreted from early-stage tumors. We have established an N-glycan profiling method using liquid chromatography electrospray ionization-mass spectrometry with high sensitive derivative, trimethyl(4-aminophenyl)ammonium chloride (TMAPA). The mass sensitivity of TMAPA-labeled oligosaccharides was enhanced more than 50 times compared with 2-aminopyridine (PA) labeled oligosaccharides, and the analytical period was significantly shortened compared with traditional HPLC 2D-mapping. Using this method, we found about 28 major N-linked oligosaccharides in human sera, and we investigated their alterations in patients who developed hepatocellular carcinoma (HCC). We found that outer arm fucosylation (attached GlcNAc via an alpha 1-3/4 linkage) in highly branched oligosaccharides increased significantly in sera of HCC patients. Normalizing the level of outer arm fucosylation by taking into account platelet concentration allowed us to distinguish more clearly between HCC and LC patients. 相似文献

为了评价国产钩端螺旋体外膜疫苗的流行病学效果。于1998年6-10月,用队列研究和病例对照研究等方法在钩体病流行区5-60岁农业人口 ,观察比较接种组和对照组钩体发病情况。队列研究显示,钩体外膜疫苗对同血清群钩体的保护率为75.17%,效果指烽为4.03。1:2配对调查的疫苗保护率为81.25%,1:3配对结果为73.33%。用筛选法估计的疫苗效果为755。不同研究方法的结果相近,均一致说明,该疫苗的流行病学效果较理想。研究结果还显示,该疫苗对异血清群钩体也有一定的保护作用。 相似文献

The dissected outer mantle epithelium (OME) of Anodonta cygnea, when mounted in Ussing type chambers, generated a spontaneous potential difference of 22.0±12.6 mV and, when short-circuited, a positive current (Isc) of 30.0±11 μA/cm2 towards the shell side and a conductance of 1.1±0.4 mS/cm2. When in contact with the shell side, Bafilomycin A1 and Concanamycin A, specific inhibitors of V-proton pumps, induced 90% inhibition of Isc in 5 and 35 min, respectively. They had no effect in the current from the hemolymph side. Both drugs induced a dramatic fall in conductance from the shell side. Tributyltin oxide (TBTO) inhibited 90% of the Isc in 3 min and induced a fall in conductance only from the shell side. Two observations suggest a direct effect on the proton pump: it was effective only from the shell side and the time course of the effect was identical to that of Bafilomicyn A1. Dicyclohexycarbodiimide (DCCD) and N-ethylmaleimide (NEM) inhibited Isc from both sides but very slowly and there was a delay of the effect from the hemolymph side in relation to the shell side. Taken together, the results suggest the presence of a V-type proton pump located at the apical (shell side) barrier of the OME. 相似文献

A noticeable characteristic of nervous systems is the arrangement of synapses into distinct layers. Such laminae are fundamental for the spatial organisation of synaptic connections transmitting different kinds of information. A major example of this is the inner plexiform layer (IPL) of the vertebrate retina, which is subdivided into at least ten sublayers. Another noticeable characteristic of these retina layers is that neurons are displayed in the horizontal plane in a non-random array termed as mosaic patterning.
