Objective: To validate GE PIXImus2 DXA fat mass (FM) estimates by chemical analysis, to compare previously published correction equations with an equation from our machine, and to determine intermachine variation. Research Methods and Procedures: C57BL/6J (n = 16) and Aston (n = 14) mice (including ob/ob), Siberian hamsters (Phodopus sungorus) (n = 15), and bank voles (Clethrionomys glareolus) (n = 37) were DXA scanned postmortem, dried, then fat extracted using a Soxhlet apparatus. We compared extracted FM with DXA‐predicted FM corrected using an equation designed using wild‐type animals from split‐sample validation and multiple regression and two previously published equations. Sixteen animals were scanned on both a GE PIXImus2 DXA in France and a second machine in the United Kingdom. Results: DXA underestimated FM of obese C57BL/6J by 1.4 ± 0.19 grams but overestimated FM for wild‐type C57BL/6J (2.0 ± 0.11 grams), bank voles (1.1 ± 0.09 grams), and hamsters (1.1 ± 0.13 grams). DXA‐predicted FM corrected using our equation accurately predicted extracted FM (accuracy 0.02 grams), but the other equations did not (accuracy, ?1.3 and ?1.8 grams; paired Student's t test, p < 0.001). Two similar DXA instruments gave the same FM for obese mutant but not lean wild‐type animals. Discussion: DXA using the same software could use the same correction equation to accurately predict FM for obese mutant but not lean wild‐type animals. PIXImus machines purchased with new software need validating to accurately predict FM.  相似文献

Background

Effective therapy of Acute Lung Injury (ALI) is still a major scientific and clinical problem. To define novel therapeutic strategies for sequelae of blunt chest trauma (TxT) like ALI/Acute Respiratory Distress Syndrome, we have investigated the immunomodulatory and regenerative effects of a single dose of ex vivo expanded human or rat mesenchymal stromal cells (hMSCs/rMSCs) with or without priming, immediately after the induction of TxT in Wistar rats.

Methods

We analyzed the histological score of lung injury, the cell count of the broncho alveolar lavage fluid (BAL), the change in local and systemic cytokine level and the recovery of the administered cells 24?h and 5 days post trauma.

Results

The treatment with hMSCs reduced the injury score 24?h after trauma by at least 50% compared with TxT rats without MSCs. In general, TxT rats treated with hMSCs exhibited a lower level of pro-inflammatory cytokines (interleukin [IL]-1B, IL-6) and chemokines (C-X-C motif chemokine ligand 1 [CXCL1], C-C motif chemokine ligand 2 [CCL2]), but a higher tumor necrosis factor alpha induced protein 6 (TNFAIP6) level in the BAL compared with TxT rats after 24?h. Five days after trauma, cytokine levels and the distribution of inflammatory cells were similar to sham rats. In contrast, the treatment with rMSCs did not reveal such therapeutic effects on the injury score and cytokine levels, except for TNFAIP6 level.

Conclusion

TxT represents a suitable model to study effects of MSCs as an acute treatment strategy after trauma. However, the source of MSCs has to be carefully considered in the design of future studies.  相似文献

2007年9月至10月,以贵州省遵义市绥阳县宽阔水国家级自然保护区亚热带森林系统内食果(或食种子)啮齿类动物与肉果植物香港四照花Dendrobenthamia hongkongensis和干果植物西南山茶Camellia pitardii为研究对象,初步研究鼠类捕食对两种植物种子扩散的影响.结果表明啮齿动物对两种植物果实及种子的搬运存在较大差异:鼠类对于肉果植物主要表现为种子的散布者,香港四照花90%的种子被成功散布,但距离较靠近母树;鼠类对于干果植物则主要表现为种子的掠食者,超过90%的西南山茶种子被鼠类采食,仅极少数能够偶然幸存下来进入土壤种子库,鼠类对干果的平均搬运距离大于肉果.  相似文献

Using non-destructive sampling we provide further genetic characterisations for spiny mice (Acomys dimidiatus/cahirinus) and gerbils (Gerbillus sp.) in three regions in Saudi Arabia. All individuals were sequenced for a fragment of the cytochrome b gene, and compared against available conspecifics and closely related taxa. We confirm the existence of a second Acomys dimidiatus/cahirinus lineage specific to the Arabian Peninsula as seen previously. The Arabian Gerbillus nanus is shown to group with Middle Eastern rather than African conspecifics. A second cryptic Gerbillus lineage was also sampled across multiple locations, which may be an uncharacterised G. dasyurus.  相似文献

A series of four experiments was performed to determine whether acute exposure to a range of 50 Hz magnetic fields had any effect on a learning task in adult male CD1 mice. A radial-arm maze placed within the bore of an electromagnet was used to assess spatial discrimination learning for food reward. Subjects were reduced to 85% of their free-feeding weight and were placed in the maze for up to 15 minutes each day for 10 days. Performance of the task was measured by using maximum likelihood techniques to calculate the probability that an animal would not reenter any given arm of the maze. Experimental subjects were exposed to a vertical, 50 Hz sinusoidal magnetic field at 5 μT, 50 μT, 0.5 mT, or 5.0 mT (rms). Control subjects were exposed only to a background time-varying field of less than 50 nT and the ambient static field of about 40 μT. The variation in the applied magnetic field was less than 5% except at the ends of the arms, where it approached 10%. It was found that all eight groups of subjects (n = 10 in all cases) showed similar increases in performance with testing, and the acquisition curve for each group of experimental subjects was not significantly different from that of their control group (P > 0.05 in all cases). It was concluded that exposure had no effect on learning at any flux density. This result is contrary to the findings of a number of preliminary studies, although other studies have reported that magnetic fields do not affect spatial learning in adult male rodents. It is possible that differences between experimental conditions might explain some of this apparent discrepancy. © 1996 Wiley-Liss, Inc.  相似文献

Parasites can play an important role in the dynamics of host populations, but empirical evidence remains sparse. We investigated the role of bot fly (Cuterebra spp.) parasitism in red-backed voles (Myodes gapperi) by first assessing the impacts of the parasite on the probability of vole survival under stressful conditions as well as on the reproductive activity of females. We then identified the main factors driving both the individual risk of infection and the abundance of bot flies inside red-backed voles. Finally, we evaluated the impacts of bot fly prevalence on the growth rate of vole populations between mid-July and mid-August. Thirty-six populations of red-backed voles were sampled in the boreal forest of Québec, Canada. The presence and the abundance of parasites in voles, two host life history traits (sex and body condition), three indices of habitat complexity (tree basal area, sapling basal area, coarse woody debris volume), and vole abundance were considered in models evaluating the effects of bot flies on host populations. We found that the probability of survival of red-backed voles in live traps decreased with bot fly infection. Both the individual risk of infection and the abundance of bot flies in red-backed voles were driven mainly by vole abundance rather than by the two host life history traits or the three variables of habitat complexity. Parasitism had population consequences: bot fly prevalence was linked to a decrease in short-term growth rate of vole populations over the summer. We found that bot flies have the potential to reduce survival of red-backed voles, an effect that may apply to large portions of populations. Analysis of DNA sequences of 132 introns and 140 exons from 42 pairs of orthologous genes of mouse and rat was used to compare patterns of evolutionary change between introns and exons. The mean of the absolute difference in length (measured in base pairs) between the two species was nearly five times as high in the case of introns as in the case of exons. The average rate of nucleotide substitution in introns was very similar to the rate of synonymous substitution in exons, and both were about three times the rate of substitution at nonsynonymous sites in exons. G+C content of introns and exons of the same gene were correlated; but mean G+C content at the third positions of exons was significantly higher than that of introns or positions 1–2 of exons from the same gene. G+C content was conserved over evolutionary time, as indicated by strong correlations between mouse and rat; but the change in G+C content was greatest at position 3 of exons, intermediate in introns, and lowest at positions 1–2 in introns. The brush-tailed porcupine (Atherurus africanus) is a hystricomorph rodent, which frequents the forests of West and Central Africa. With an average weight of 3 kg, it is a favourite source of meat for urban and rural populations of Gabon, Nigeria, Cameroon or Congo. Hunted in large quantities, its price is often higher than that of other game or domestic animals. Although its current productivity in captivity is limited to a single young per birth and two to three births per year per female, this species could be a good candidate for minilivestock programmes in African forest areas if its current reproductive potential in captivity could be improved. Further research should be encouraged on its biology and reproduction since the current level of hunting for this species is probably not sustainable. Captive breeding programmes could play a role in assessing a better knowledge of the species' biology, and in reducing the effects of intensive hunting in areas where this activi ty is no longer sustainable.  相似文献

Bionomics of fleas (Siphonaptera) parasitizing rodent hosts, mostly the gerbil Gerbillus dasyurus (Wagner) and the jird Meriones crassus Sundevall (Gerbillidae), were investigated in the central Negev desert of Israel. Populations were sampled weekly (by Sherman trapping of hosts) from August 2000 to July 2001. Among 1055 fleas of nine species captured, four species predominated (94%). Two species of Pulicidae, Xenopsylla dipodilli Smit and X. ramesis (Rothschild), reproduced perennially, whereas adults of Nosopsyllus iranus theodori Smit (Ceratophyllidae) and Stenoponia tripectinata medialis Jordan (Hystrichopsyllidae) occurred only in cool months (October-March). During their main activity season on the most infested host species (estimated from > 300 trap-nights/month), prevalence of these four flea species reached 40-70%, 20-30%, 100% and 50-70%, respectively, with infestation intensities of 2-2.7, 7-12, 2-3.5 and 2.5-7 fleas per infested rodent, respectively. Xenopsylla dipodilli oviposition peaked during autumn with parous rate > 80% by September-October. During December-April, the majority of X. dipodilli females were immature and/or nulliparous (defined as having mature ovaries but no follicular relics). In contrast, X. ramesis had two reproductive peaks, in mid-spring and autumn, evidenced by the influx of immature females in late spring and summer (30-40%) and in winter (20-30%) after maximal parous rates: 80-100% in March-April and 95-100% in October-November. Nosopsyllus iranus theodori and Stenoponia tripectinata medialis adults occurred only during cool months. At the beginning of activity, during October-November, the sex ratio of N. i. theodori was strongly biased to females (86%) that were immature and/or nulliparous. In winter, adult females were 52-65% parous and 10-32% immature. In March, as the adult population of N. i. theodori declined, 78% of females were parous and 12% immature. Seasonal activity of S. t. medialis (November-March) was shorter than for the other three species; females were predominantly nulliparous in November (80%), after which the proportion of parous females increased gradually to 84% in February. Two females of S. t. medialis collected in March were mature but nulliparous, suggesting that this species of flea might 'oversummer' (as pupae or teneral adults) in the cocooned stage.  相似文献
