The impact of dimethoate on the spatial distribution of beneficial arthropods in winter wheat 总被引:3,自引:0,他引:3
The within‐field spatial distribution of beneficial arthropods was assessed using two‐dimensional grids of pitfall traps and suction sampling across two winter wheat fields of 4 and 16 ha, before and after an application of dimethoate. An unsprayed 6 m wide buffer zone was left around half the edge of the larger field. Arthropod numbers fluctuated to varying extents prior to spraying. Two species of Carabidae (Pterostichus madidus and P. melanarius), Linyphiidae, Lycosidae and Aphidius spp. (Braconidae) all showed their greatest reduction after spraying. For five carabid taxa, Tachyporus spp. (Staphylinidae) and Collembola the decline in numbers following spraying was no greater than any reduction found during the pre‐spraying period. Within field spatial distributions of three arthropod groups were analysed using SADIE. P. madidus, present in patches across the centre of both fields prior to spraying, was removed by dimethoate and by 34 days after spraying had recovered most at the field edges. Linyphiidae were evenly distributed across both fields prior to spraying. Their numbers were reduced considerably by dimethoate and they did not recover to pre‐spray levels. However, where recovery occurred this was across the centre of both fields indicating their potential to reinvade whole fields. Aphidius species were also evenly distributed across both fields prior to spraying, but did not recover after spraying. Some, but not all arthropods survived within the unsprayed buffer zone and there was some indication that reinvasion of the mid‐field was more extensive where this was present. The importance of field margins with respect to insecticide treatments is discussed. 相似文献

The comparative aspects of toxicity of phosphororganic and pyrethroid insecticides and their combinations for arthropods and warm‐blooded organisms were studied. The quantitative comparison of the coefficients of the selectivity, specific sensitivity and combined action were worked out. The most perspective mixtures were determined and the optimum ratios of their ingredients were stated. The management options for grape phylloxera, Daktulosphaira vitifoliae, a monophagous insect pest of Vitis species are reviewed. Although in a worldwide context, grape phylloxera is managed predominantly by the use of resistant rootstocks developed through conventional breeding of hybrid crosses of American Vitis species, this management aspect is largely excluded from the review so that emerging technologies in the field of detection, quarantine and alternative management are discussed. In some viticulture regions of the world, where grape phylloxera's geographic distribution is limited (e.g. Australia), the pest is managed through a combination of surveillance, detection and quarantine. Although some alternative management options for grape phylloxera exist they have received relatively limited research attention because of the relative success of resistant rootstocks. The resilience of resistant rootstocks as the primary management option could also be challenged in the future by host‐plant interactions with diverse grape phylloxera clonal lineages and by potential impacts of climate change on both grapevine and grape phylloxera distribution. A range of control options exist which could be integrated into an improved management system for grape phylloxera. Priority areas for future evaluation and further development include early detection techniques, investigation into the use of biological control agents and development of an integrated approach to grapevine phylloxera management. 相似文献

Purified P‐glycoprotein ATPase from Helicoverpa armigera (Ha‐Pgp), reconstituted in proteoliposomes composed of phospholipids and cholesterol, shows higher ATPase activity in the presence of cholesterol than in its absence. The Ha‐Pgp ATPase activity was increased 30–40% with cholesterol. The KM for ATP was found to be 1 and 0.8 mM in the absence and presence of cholesterol, respectively. The insecticide‐stimulated Ha‐Pgp ATPase activity was increased by 10–20% for all the insecticides in the reconstituted proteoliposomes containing cholesterol compared to those with no cholesterol. The effects of cholesterol on KM and Vmax values of insecticide‐stimulated Ha‐Pgp ATPase activity were unrelated to the size of the insecticide. Ha‐Pgp tryptophan fluorescence displayed a red shift of 3 and 8 nm in emission spectra upon binding of insecticides. Cholesterol enhances the interaction of insecticides with Ha‐Pgp. Kd values of different insecticides for binding to Ha‐Pgp were found to be lower in the presence of cholesterol in the proteoliposomes compared to its absence. Results suggest that cholesterol plays a role in the recognition and interaction of insecticides by modulating Ha‐Pgp ATPase and may be involved in efflux of insecticides from cells by the transporter. 相似文献

In a mushroom crop (Agaricus bisporus) affected by a very low level of sciarid fly (Lycoriella auripila) infestation, the effects of an indigenous isolate of insect-parasitic nematode (Steinernema feltiae) and of two commonly used insecticides (diazinon and diflubenzuron) were studied. When compared with untreated plots, nematodes applied to the casing had no adverse effects on mushroom yields whereas insecticides decreased yields. At a rate of 3 × 106 infective juveniles per tray (surface area = 0.56 m2), S. feltiae elicited increases of 28.5% and 19% in the mean total numbers and weights of mushrooms respectively. Treatment only with diflubenzuron resulted in 14.6% and 6% reductions in mean total numbers and weights of mushrooms, respectively; treatment with both diazinon and diflubenzuron caused 18.5% and 9.4% losses. Application of nematodes generally reduced the mean weight per mushroom whereas insecticides increased it; nematodes delayed the onset of mushroom production (first flush) whereas diflubenzuron delayed the third and fourth flushes. Nematode contamination of sporophores was minimal when S. feltiae was applied at casing. Although their numbers declined with time, the nematodes persisted, in the casing layer, throughout the cropping period of seven weeks. It is concluded that yield benefits associated with nematode application can result mainly from nematode effects on A. bisporus and not solely from suppression of a damaging pest population. 相似文献

We have developed a gas chromatography–high resolution mass spectrometry method for measuring pyrethroid, organophosphorus, carbamate and fipronil pesticides and the synergist piperonyl butoxide in human plasma. Plasma samples were extracted using solid phase extraction and were then concentrated for injection and analysis using isotope dilution gas chromatography–high resolution mass spectrometry. The limits of detection ranged from 10 to 158 pg/mL with relative recoveries at concentrations near the LODs (e.g., 25 or 250 pg/mL) ranging from 87% to 156% (9 of the 16 compounds were within ±15% of 100%). The extraction recoveries ranged from 20% to 98% and the overall method relative standard deviations were typically less than 20% with some exceptions. Our aim was to investigate the non-target effects on soil micro-organisms in agricultural environments caused by chemical control agents (CCAs) and microbial biocontrol agents (mBCAs), including the recovery from these effects. This was a desk study in which quantifiable endpoints, such as numbers of colony forming units (CFU), were derived from a series of studies and the combined data then analysed with a meta-regression analysis. Three analyses of the same dataset were performed. The first analysis, which was performed at the level of the CCAs and mBCAs in general, revealed that the effects of CCAs differed significantly from those of mBCAs. The second analysis, which included the type of non-target group (bacteria, fungi, actinomycetes, and protozoa) as additional input, revealed that mBCAs have greater effects than CCAs on fungi at study initiation, that CCAs had greater effects than mBCAs on bacteria and protozoa and that when effects were measured, recovery occurred within 100 days post-treatment initiation. The final analysis, which included the type of CCA (fungicide, insecticide, herbicide) or mBCA (antagonist) as additional input, revealed that (1) antagonists had a greater effect on fungi than insecticides and fungicides, (2) insecticides and to a lesser extent fungicides had a larger effect on bacteria than fungicides and antagonists, and (3) recovery of the CFU occurred within 100 days for all types of pesticides, mBCAs as well as CCAs, and for all non-target groups. Organophosphorus pesticides (OP) are highly toxic and are widely used as insecticides. Bacterial organophosphohydrolases which hydrolyze a variety of OPs have been considered for the clean-up of polluted environments. This study describes the engineering of Escherichia coli towards the overproduction of the organophosphohydrolase (OpdA) from Agrobacterium radiobacter at the surface of polyester inclusions. The OpdA was N-terminally fused via a designed linker region to the C-terminus of polyester inclusion-forming enzyme PhaC of Ralstonia eutropha. The PhaC-L-OpdA fusion protein was overproduced by using the strong T7 promoter and when coexpressed with genes phaA (encoding β-ketothiolase) and phaB (encoding acetoacetyl-CoA reductase) from R. eutropha this led to formation of polyester inclusions abundantly displaying OpdA. These OpdA beads showed organophosphohydrolase activity of 1,840 U/g wet polyester beads or 4,412 U/g protein. Steady state kinetics revealed that when compared with free OpdA the k(cat) (s(-1)) of 139 of immobilized OpdA was reduced by about 16.5-fold while the K(M) (M) of 2.5 × 10(-4) was increased by 1.6-fold. The immobilized OpdA showed increased temperature stability. Moreover, the stability of OpdA immobilized to polyester beads was assessed by incubating OpdA beads at 25°C for up to 11 days and no significant loss in enzyme activity was detected. The application performance of the OpdA beads with respect to hydrolysis of OPs in contaminated environments was demonstrated in wool scour spiked with fluorescent coumaphos. This study demonstrated a new strategy toward the efficient recombinant production of immobilized organophosphohydrolase, the OpdA, suitable for bioremediation applications. 相似文献
