Short-term studies for comparing some primary metabolic and growth-responses to salt stress in seedlings of two maize genotypes differing in drought resistance were carried out under controlled conditions. Both genotypes revealed high yielding ability in favourable environments. Treatments: Control (Hoagland-Arnon No 1 solution) and salt stress (Hoagland-Arnon solution plus NaCl, s = –0.84MPa). It was found that in both genotypes the activity of the principal metabolic pathway supplying reduced nitrogen (15N) for the synthesis of amino acids and proteins as well as the assimulatory number (14CO2—assimilation relation rate per chlorophyll unit) were decreased under the effect of the stress. These effects were more marked in the resistant genotype. In this genotype the stress induced metabolic activity decline was accompanied by a corresonding reduction of the relative growth rate. Conversely, continuing growth, resulting probably from accumulation of solutes, was observed in the susceptible genotype.On the basis of these and other observations it is assumed that the resistant genotype manifests short-term energy saving stress reactions.  相似文献

Inorganic-N concentrations in soil solution of whole tree harvest (WTH) and conventional fell (CF) plots were monitored for two years before felling and four years after felling. Concentrations in the mineral soil after felling were higher than in standing forest for up to 14 months in both felling treatments. In the WTH plots inorganic-N concentrations then dropped steadily until four years after felling they approached zero. In contrast, inorganic-N concentrations of the CF plots remained comparatively large. Inorganic-N was dominated by nitrate throughout the period of the study, and especially in the mineral horizons.Felling debris was not a source of inorganic-N, unless indirectly through release and mineralisation of soluble organic-N. Vegetation cover, biomass and N content were substantially greater in the WTH plots two to three years after felling, compared with the CF. Vegetation cover and brash cover (slash cover in N. America) were negatively correlated. There was also a negative correlation between inorganic-N concentration in soil water samplers and the vegetation cover within the collection area of, or a 1 m square surrounding, these samplers.Two factors are probably responsible for the reduction in inorganic-N concentrations after felling in the WTH — the rapid re-establishment of vegetation and the lack of a N source in felling debris. In the CF plots, brash prevents re-establishment of vegetation over wide areas for at least four years. However, brash is not directly a source of inorganic-N at this stage.  相似文献

A newly-developed field-portable multi-flash kinetic fluorimeter for measuring the kinetics of the microsecond to millisecond reactions of the oxidizing and reducing sides of photosystem 2 in leaves of intact plants is described and demonstrated. The instrumental technique is a refinement of that employed in the double-flash kinetic fluorimeter (Joliot 1974 Biochim Biophys Acta 357: 439–448) where a low-intensity short-duration light pulse is used to measure the fluorescence yield changes following saturating single-turnover light pulses. The present instrument uses a rapid series of short-duration (2 s) pulses to resolve a complete microsecond to millisecond time-scale kinetic trace of fluorescence yield changes after each actinic flash. Differential optics, using a matrix of optical fibers, allow very high sensitivity (noise levels about 0.05% Fmax) thus eliminating the need for signal averaging, and greatly reducing the intensity of light required to make a measurement. Consequently, the measuring pulses have much less actinic effect and an entire multi-point trace (seven points) excites less than 1% of the reaction centers in a leaf. In addition, bu combining the actinic and measuring pulse light in the optical fiber network, the tail of the actinic flash can be compensated for, allowing measurements of events as rapidly as 20 s after the actinic flash. This resolution makes practical the routine measurement of the microsecond turnover kinetics of the oxygen evolving complex in leaves of intact plants in the field. The instrument is demonstrated by observing flash number dependency and inhibitor sensitivity of the induction and decay kinetics of flash-induced fluorescence transients in leaves of intact plants. From these traces the period-two oscillations associated with the turnover of the two-electron gate and the period-four oscillations associated with the turnover of the oxygen evolving complex can be observed. This work was aimed to investigate whether shoot Sr concentrations of plant species are related to respective Ca concentrations and to soil properties and to compare the Sr-Ca observed ratios (OR), defined as the quotient of the ratios Sr/Ca in shoots and in the soil solution or in the extractable form, among species and soils. Ten pasture plant species were grown in pots (1-L volume) filled with eight soils differing in the various physicochemical characteristics. Each pot received 50 mg Sr except those of the soil with the highest cation exchange capacity (C.E.C.) that received 100 mg Sr per pot. For each soil, shoot Sr concentrations of species were linearly and positively related with the respective Ca concentrations. C.E.C, organic matter content and Ca in the soil solution or in the extractable form were the only soil properties that were related, all negatively, with shoot Sr concentrations. The ratio of extractable Sr and Ca was positively and linearly related with the ratio of Sr and Ca. in the soil solution. OR was affected by both species and soils. Most of OR values of all species in all soils ranged between 0.8 and 1.5, except for the grass Agrostis capillaris which had the highest values for most of soils. This indicates that Agrostis capillaris compared to other species, takes up proportionally more Sr than Ca.  相似文献

Five strawberry (Fragaria sp.) and five raspberry (Rubus ideaus L.) cultivars were evaluated for resistance to two spotted spider mite (Tetranychus urticae Koch.). Two methods of assessing the development of two spotted mite populations using detached leaves were compared. The number of eggs laid and mites which developed were compared. The strawberry cvs Hapil and Pegasus had significantly greater development of two spotted mite populations than the cvs Rhapsody, Symphony and Elsanta. The raspberry cv. Joan Squires had higher populations of two spotted mite whilst the raspberry cv. Leo the least, when compared with cvs Glen Clova, Glen Moy and Glen Prosen. Differences were observed in oviposition sites and mite distribution when comparing raspberries with strawberries. The method of assessing the populations development of two spotted mite which involved maintaining the cut leaf stem in water may be of potential use for studying population dynamics of both two spotted mite and possible predators over extended periods of time.  相似文献

In this paper we analyze a model for the HIV-infection transmission in a male homosexual population. In the model we consider two types of infected individuals. Those that are infected but do not know their serological status and/or are not under any sort of clinical /therapeutical treatment, and those who are. The two groups of infectives differ in their incubation time, contact rate with susceptible individuals, and probability of disease transmission. The aim of this article is to study the roles played by detection and changes in sexual behavior in the incidence and prevalence of HIV. The analytical results show that there exists a unique endemic equilibrium which is globally asymptotically stable under a range of parameter values whenever a detection /treatment rate and an indirect measure of the level of infection risk are sufficiently large. However, any level of detection/ treatment rate coupled with a decrease of the transmission probability lowers the incidence rate and prevalence level in the population. In general, only significant reductions in the transmission probability (achieved through, for example, the adoption of safe sexual practices) can contain effectively the spread of the disease.  相似文献

Lolium perenne growing with high root density on a fine nylon mesh (Kuchenbuch and Jungk, 1982) caused the development of element gradients in the rhizosphere below the mesh. Micro-liter soil solutions from 2-mg soil samples were sprayed onto Formvar-coated grids and analyzed by X-ray microanalysis in a transmission electron microscope. The results were comparable to those obtained by flame photometry and atomic absorption spectrometry (AAS) of conventional soil solutions from 1 g soil. X-ray microanalysis of micro-soil solutions allows the application of different extraction procedures to even small amounts of soil usually available from rhizosphere experiments. Information about soil buffering characteristics in the rhizosphere can thus be obtained. Aluminum accumulation in the rhizosphere of small segments of single Picea abies fine roots grown in undisturbed natural forest soil could be detected with this technique.  相似文献

The influence of base cation concentrations on pH and aluminium sensitivity of the woodland grasses Bromus benekenii and Hordelymus europaeus was studied in flowing solution culture experiments. Plants were exposed to low pH (3.9, experiment 1) and Al concentrations of 19 and 37 M (experiment 2) at two base cation (Ca+Mg+K) levels, all within the ranges measured in natural forest soil solutions. Elevated base cation concentrations ameliorated both H and Al toxicity, as indicated by increased root and shoot growth. In the third experiment, interactions between pH (4.3 and 4.0) and Al (0 and 19 M) were investigated. It was shown that the combined toxicity effects of H and Al were not greater than the separate H or Al effects. Tissue concentrations of base cations and Al increased with increasing concentrations in the solution, but were also influenced by the base cation : Al ratio. Relating the experimental evidence with the composition of forest soil solutions suggests an important role of soil pH and Al in controlling the distribution of the two species. Growth conditions also differ at various soil depths. Concentrations of free cationic Al were higher and base cation concentrations lower at 5–10 cm than at 0–5 cm soil depth. Increasing base cation concentrations may protect roots from both H and Al injury during periods of drought when concentrations of most elements increase in the soil solution, whereas molar ratios between base cations, H and Al remain unchanged.  相似文献

The pH of the nutrient solution bathing the roots of four-month-oldPinus contorta var.latifolia Englm. seedlings was monitored continuously between additions of nutrients. Nitrogen was supplied in the form of NH4NO3, and was added three times per week in amounts relative to seedling fresh weight. No pH change was associated with the nutrient addition cycle; however, extinguishing of the lights at night resulted in a decrease in pH of almost half a pH unit in the first hour. The pH reverted to normal within a few hours. Re-illumination resulted in a pH increase of a smaller magnitude, but over a similar time span. Estimation of the proton extrusion rate gave values of about 17 µmol (g FW root)–1 h–1.  相似文献

The role of polyamines (PA) synthesis in NMDA receptor-mediated45Ca2+ fluxes and norepinephrine release was studied in rat hippocampal synaptosomes. NMDA (50M) caused a sharp (>2-fold) transient increase in PA synthesis regulating enzyme, ornithine decarboxylase (ODC) activity with concomitant elevation in PA levels in the order putrescine>spermidine>spermine. ODC inhibitor, -difluoromethylornithine (DFMO), and NMDA antagonist, 2-amino-5-phosphonovaleric acid (D-AP5), both blocked increases in ODC activity and PA levels. Activation of NMDA receptors induced a sharp (3 to 4-fold) and quick (15 seconds) increase in45Ca2+ uptake by synaptosomes within 15 seconds of exposure at 37°C. The efflux of45Ca2+ and3H-norepinephrine (NE) release at 22°C from pre-loaded synaptosomes was also significantly (2 to 4-fold) enhanced by NMDA within 15 seconds. These NMDA receptor-mediated effects on calcium fluxes and NE release were blocked by NMDA receptor-antagonists (DAP-5 and MK-801) and PA synthesis inhibitor, DFMO and the DFMO inhibition nullified by exogenous putrescine. These observations establish that ODC/PA cascade play an important role in transduction of excitatory amino acid mediated signals at NMDA receptors.Special issue dedicated to Dr. Sidney Ochs.  相似文献
