A mathematical model is presented for the emergence of perceptual-cognitive-behavioral modes in psychophysical experiments in which participants are confronted with two alternatives. The model is based on the theory of self-organization and, in particular, the order parameter concept such that the emergence of a mode is conceptualized as an instability leading to the emergence of an appropriately defined order parameter. The order parameter model is merged with a second model that describes adaptation in terms of a system parameter dynamics. It is shown that the two-component model predicts hysteretic mode-mode transitions when control parameters are increased or decreased beyond critical values. The two-component model can account for both positive and negative hysteresis effects due to the interaction between order parameter and system parameter dynamics. Moreover, the model-based analysis reveals that response time curves look rather flat when response times are relatively decoupled from the mode-mode transition phenomenon. In general, response time curves exhibit a peaked close to the mode-mode transition point. In this context, the possibility is discussed that such peaked response time curves belong to the class of critical phenomena of self-organizing systems. In order to illustrate the relevance of peaked response time curves for future research and research reported in the past, results from a perceptual judgment experiment are reported, in which participants judged their ability to stand on a tilted slope for various angles of inclination. Binomial group testing involves pooling individuals into groups and observing a binary response on each group. Results from the group tests can then be used to draw inference about population proportions. Its use as an experimental design has received much attention in recent years, especially in public‐health screening experiments and vector‐transfer designs in plant pathology. We investigate the benefits of group testing in situations wherein one desires to test whether or not probabilities are increasingly ordered across the levels of an observed qualitative covariate, i.e., across strata of a population or among treatment levels. We use a known likelihood ratio test for individual testing, but extend its use to group‐testing situations to show the increases in power conferred by using group testing when operating in this constrained parameter space. Most biological phospholipids contain at least one unsaturated alkyl chain. However, few order parameters of unsaturated lipids have been determined because of the difficulty associated with isotopic labeling of a double bond. Dipolar recoupling on axis with scaling and shape preservation (DROSS) is a solid-state nuclear magnetic resonance technique optimized for measuring 1H–13C dipolar couplings and order parameters in lipid membranes in the fluid phase. It has been used to determine the order profile of 1,2-dimyristoyl-sn-glycero-3-phosphocholine hydrated membranes. Here, we show an application for the measurement of local order parameters in multilamellar vesicles containing unsaturated lipids. Taking advantage of the very good 13C chemical shift dispersion, one can easily follow the segmental order along the acyl chains and, particularly, around the double bonds where we have been able to determine the previously misassigned order parameters of each acyl chain of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC). We have followed the variation of such order profiles with temperature, unsaturation content and cholesterol addition. We have found that the phase formed by DOPC with 30% cholesterol is analogous to the liquid-ordered (lo) phase. Saposin C (Sap C) is known to stimulate the catalytic activity of the lysosomal enzyme glucosylceramidase (GCase) that facilitates the hydrolysis of glucosylceramide to ceramide and glucose. Both Sap C and acidic phospholipids are required for full activity of GCase. In order to better understand this interaction, mixed bilayer samples prepared from dioleoylphosphatidylglycerol (DOPG) and dioleoylphosphatidylserine (DOPS) (5:3 ratio) and Sap C were investigated using 2H and 31P solid-state NMR spectroscopy at temperatures ranging from 25 to 50 °C at pH 4.7. The Sap C concentrations used to carry out these experiments were 0 mol%, 1 mol% and 3 mol% with respect to the phospholipids. The molecular order parameters (SCD) were calculated from the dePaked 2H solid-state NMR spectra of Distearoyl-d70-phosphatidylglycerol (DSPG-d70) incorporated with DOPG and DOPS binary mixed bilayers. The SCD profiles indicate that the addition of Sap C to the negatively charged phospholipids is concentration dependent. SCD profiles of 1 mol% of the Sap C protein show only a very slight decrease in the acyl chain order. However, the SCD profiles of the 3 mol% of Sap C protein indicate that the interaction is predominantly increasing the disorder in the first half of the acyl chain near the head group (C1-C8) indicating that the amino and the carboxyl termini of Sap C are not inserting deep into the DOPG and DOPS mixed bilayers. The 31P solid-state NMR spectra show that the chemical shift anisotropy (CSA) for both phospholipids decrease and the spectral broadening increases upon addition of Sap C to the mixed bilayers. The data indicate that Sap C interacts similarly with the head groups of both acidic phospholipids and that Sap C has no preference to DOPS over DOPG. Moreover, our solid-state NMR spectroscopic data agree with the structural model previously proposed in the literature [X. Qi, G.A. Grabowski, Differential membrane interactions of saposins A and C. In this paper, we report the results of a systematic attempt to relate the intrinsic plasma membrane fluidity of three different
cell lines to their electroporation behaviour, which consists of reversible and irreversible electroporation. Apart from electroporation
behaviour of given cell lines the time course required for membrane resealing was determined in order to distinguish the effect
of resealing time from the cell’s ability to survive given electric pulse parameters. Reversible, irreversible electroporation
and membrane resealing were then related to cell membrane fluidity as determined by electron paramagnetic resonance spectroscopy
and computer characterization of membrane domains. We found that cell membrane fluidity does not have significant effect on
reversible electroporation although there is a tendency for the voltage required for reversible electroporation to increase
with increased membrane fluidity. Cell membrane fluidity, however, may affect irreversible electroporation. Nevertheless,
this effect, if present, is masked with different time courses of membrane resealing found for the different cell lines studied.
The time course of cell membrane resealing itself could be related to the cell’s ability to survive. 相似文献

Recently, two independent (15)N NMR relaxation studies indicated that in contrast to the decreased flexibility expected for induced-fit interactions, the backbone flexibility of major urinary protein isoform I (MUP-I) slightly increased upon complex formation with its natural pheromone 2-sec-butyl-4,5-dihydrothiazol. We have investigated the subtle details of molecular interactions by molecular dynamics simulations in explicit solvent. The calculated order parameters S(2) for a free- and ligand-bound protein supply evidence that mobility in various regions of MUP-I can be directly related to small conformational changes of the free- and complexed protein resulting from modifications of the hydrogen bonding network. 相似文献

A statistical surveillance system gives a signal as soon as data give enough evidence of an important event. We consider on-line surveillance systems for detecting changes in influenza incidence. One important feature of the influenza cycle is the start of the influenza season, and another one is the change to a decline (the peak). In this report we discuss statistical methods for on-line peak detection. One motive for doing this is the need for health resource planning. Surveillance systems were adapted for Swedish data on laboratory verified diagnoses of influenza. In Sweden, the parameters of the influenza cycles vary too much from year to year for parametric methods to be useful. We suggest a non-parametric method based on the monotonicity properties of the increase and decline around a peak. A Monte Carlo study indicated that this method has useful stochastic properties. The method was applied to Swedish data on laboratory verified diagnoses of influenza for seven periods. 相似文献
