Although recent progress in cardiovascular tissue engineering has generated great expectations for the exploitation of stem cells to restore cardiac form and function, the prospects of a common mass-produced cell resource for clinically viable engineered tissues and organs remain problematic. The refinement of stem cell culture protocols to increase induction of the cardiomyocyte phenotype and the assembly of transplantable vascularized tissue are areas of intense current research, but the problem of immune rejection of heterologous cell type poses perhaps the most significant hurdle to overcome. This article focuses on the potential advantages and problems encountered with various stem cell sources for reconstruction of the damaged or failing myocardium or heart valves and also discusses the need for integrating advances in developmental and stem cell biology, immunology and tissue engineering to achieve the full potential of cardiac tissue engineering. The ultimate goal is to produce 'off-the-shelf' cells and tissues capable of inducing specific immune tolerance. 相似文献

乏氧诱导因子-1α (HIF-1α)是肿瘤细胞适应乏氧微环境的关键调控因子,具有作为治疗靶基因的潜力,以克服乏氧诱导的治疗抗拒等效应.下调其表达可能影响肿瘤细胞内一系列乏氧应答相关基因的表达.本研究采用已构建的HIF-1α RNAi慢病毒载体转导肺腺癌A549细胞,经杀稻瘟素(blasticidin)筛选建立HIF-1α基因稳定沉默的A549细胞株.应用cDNA微阵列技术检测并比较HIF-1α基因沉默A549细胞株和其亲本细胞株在常氧和乏氧状态下的基因表达谱改变. 应用定量RT PCR方法验证部分cDNA芯片差异表达基因的表达改变.HIF-1α基因稳定沉默细胞株A549/HIF-1α,在常氧和乏氧条件下HIF-1αmRNA水平分别较A549细胞下降89.2%和88.1%,HIF-1α蛋白水平分别下降97.2%和88.4%. 在乏氧条件下,cDNA微阵列检测的1 280个基因中,52个基因表达上调,15个基因表达下调. HIF-1α基因沉默显著影响其中27个基因的乏氧诱导效应.定量RT-PCR验证其中ENO2、BCL-2、CXCR4和MMP11的表达水平,与cDNA芯片结果相符合.结果提示,HIF-1α基因沉默能够在一定程度上阻断肺癌细胞的乏氧应答,在克服乏氧导致的肺癌治疗抗拒方面具有潜力. 相似文献

Question: Chronic stress events are defined as disturbance events that exceed the lifespan of the dominant plant species, fluctuate in intensity and lack abruptness or physical destruction of biomass. Can the effects of chronic stress events be measured on vegetation communities? Did two chronic stress events, the removal of a tide gate and a four year drought, cause a temporary or permanent shift in the vegetation communities of a tidal marsh? Location: Tidal marsh in southeastern United States. Methods: Change in species composition and dominance and community change on a landscape level salinity gradient were measured between time periods ranging from four months to seven years to construct a statistical baseline reference community at freshwater, oligohaline, and mesohaline sections of a tidal marsh. Statistical shifts in the plant community were defined as changes in the plant community that fell outside of the defined baseline reference community. Results: Plant community changes outside of the reference community occurred in 13 out of 378 community comparisons. Removal of the tide gate had a greater effect on interstitial salinity levels than the drought and was most intense in the oligohaline marsh, where between 20 to 45% of the freshwa‐ter/oligohaline community types permanently converted to oligohaline community types. However, community shifts in the freshwater and oligohaline marsh induced by the drought were temporary, lasting from 1 to 3+ years. Neither chronic stress event permanently altered the mesohaline plant communities. Conclusion: The effects of chronic stress events could be detected; an extended historical record of vegetation change (18 years) was necessary to identify community shifts outside of a reference condition of the community and to determine if those shifts were permanent or temporary. 相似文献

In the Loess Plateau, China, arable cultivation of slope lands is common and associated with serious soil erosion. Planting trees or grass may control erosion, but planted species may consume more soil water and can threaten long‐term ecosystem sustainability. Natural vegetation succession is an alternative ecological solution to restore degraded land, but there is a time cost, given that the establishment of natural vegetation, adequate to prevent soil erosion, is a longer process than planting. The aims of this study were to identify the environmental factors controlling the type of vegetation established on abandoned cropland and to identify candidate species that might be sown soon after abandonment to accelerate vegetation succession and establishment of natural vegetation to prevent soil erosion. A field survey of thirty‐three 2 × 2–m plots was carried out in July 2003, recording age since abandonment, vegetation cover, and frequency of species together with major environmental and soil variables. Data were analyzed using correspondence analysis, classification tree analysis, and species response curves. Four vegetation types were identified and the data analysis confirmed the importance of time since abandonment, total P, and soil water in controlling the type of vegetation established. Among the dominant species in the three late‐successional vegetation types, the most appropriate candidates for accelerating and directing vegetation succession were King Ranch bluestem (Bothriochloa ischaemum) and Lespedeza davurica (Leguminosae). These species possess combinations of the following characteristics: tolerance of low water and nutrient availability, fibrous root system and strong lateral vegetative spread, and a persistent seed bank. 相似文献

Group A Streptococcus (GAS) infections may trigger autoimmune sequelae thought to involve streptococcal antibodies cross-reactive to human antigens. Here, the antigenicity of the streptococcal collagen-like (CL) proteins, Scl1 and Scl2, that exhibit structural similarity to human collagen, was analyzed. Antibodies to Scl1.1 protein were detected in human sera from healthy individuals previously infected with M1 GAS and from patients with various GAS infections, as well as in sera from mice infected with M1 GAS. Linear B-cell epitope mapping identified immunoreactive peptides corresponding to the CL region of Scl1.1. Humoral responses to Scl1.28 and Scl2.28 were also detected in pediatric patients and mice infected with M28 GAS. 相似文献

Sensory receptors often receive strongly dynamic, or time varying, inputs in their natural environments. Characterizing their dynamic properties requires control and measurement of the stimulus over a frequency range that equals or exceeds the receptor response. Techniques for dynamic stimulation of olfactory receptors have lagged behind other major sensory modalities because of difficulties in controlling and measuring the concentration of odorants at the receptor. We present a new method for delivering olfactory stimulation that gives linear, low-noise, wide frequency range control of odorant concentration. A servo-controlled moving bead of silicone elastomer occludes the tip of a Pasteur pipette that releases odorant plus tracer gas into a flow tube. Tracer gas serves as a surrogate indicator of odorant concentration and is measured by a photoionization detector. The system has well-defined time-dependent behavior (frequency response and impulse response functions) and gives predictable control of odorant over a significant volume surrounding the animal. The frequency range of the system is about 0-100 Hz. System characterization was based on random (white noise) stimulation, which allows more rapid and accurate estimation of dynamic behavior than deterministic signals such as sinusoids or step functions. Frequency response functions of Drosophila electroantennograms stimulated by fruit odors were used to demonstrate a typical application of the system. 相似文献
