Summary Miscibility among phospholipids with different lipid chain-lengths or with different head groups has attracted a number of research efforts because of its significance in biological membrane structure and function. The general consensus about the miscibility of phosphatidylcholines with varying lipid chainlengths appears to be that binary mixtures of phospholipids with a difference of two carbon atoms in the lipid chain mix well at the main phase transition. Miscibility between phosphatidylcholines with differences of four carbon atoms appears to be inconclusive. Previous reports on the phase transition of binary phospholipid mixtures are concerned mainly with multilamellar vesicles and are mostly limited to the main transition. In the present study, unilamellar vesicles were used and miscibility in binary systems between dimyristoyl-, dipalmitoyl- and distearoyl-phosphatidylcholines at pretransition, as well as main transition temperatures was evaluated by constructing phase diagrams. Two methods were used to monitor the phase transitions: differential scanning microcalorimetry and optical absorbance methods. The optical method has the advantage that unilamellar vesicles of dilute phospholipid concentrations can be used. The liquidus and solidus phase boundaries were determined by the onset temperature of heating and cooling scans, respectively, because the completion temperature of a phase transition has no meaning in binary solutions. Natural forest communities are made up of different overlapping elementary subpopulations consisting of individuals of different
species and ages. To stratify the individuals of a forest stand into elementary subpopulations by using tree height records,
a graphical method was empirically proposed employing a derivative of Pearson's type VII distribution as a basis. Arranging
all of the individual trees in a stand in the descending order of their height (x), and introducing the rank (N) of any individual in the ordered ranking ofx, finite difference (n) ofN and three values ofx, i.e.,x(N), x(N−n), andx(N+n) labelled byN, N−n, andN+n, respectively, the proposed method used the linear relation betweenx(N) and [x(N−n)+x(N+n)]. Malaria parasite cloning is traditionally carried out mainly by using the limiting dilution method, which is laborious, imprecise, and unable to distinguish multiply-infected RBCs. In this study, we used a parasite engineered to express green fluorescent protein (GFP) to evaluate a single-cell sorting method for rapidly cloning Plasmodium falciparum. By dividing a two-dimensional scattergram from a cell sorter into 17 gates, we determined the parameters for isolating singly-infected erythrocytes and sorted them into individual cultures. Pre-gating of the engineered parasites for GFP allowed the isolation of almost 100% GFP-positive clones. Compared with the limiting dilution method, the number of parasite clones obtained by single-cell sorting was much higher. Molecular analyses showed that parasite isolates obtained by single-cell sorting were highly homogenous. This highly efficient single-cell sorting method should prove very useful for cloning both P. falciparum laboratory populations from genetic manipulation experiments and clinical samples. 相似文献

The X-ray powder diffractograms of racemic citalopram oxalate and (S)-citalopram oxalate are very similar, but the melting point of the racemate is higher than that of the pure enantiomer. The higher melting point indicates that the racemate is a racemic compound, rather than a conglomerate. The crystal structure of the enantiomer contains two molecules of (S)-citalopram in the asymmetric unit. The conformation of the two molecules is different but they approximate mirror images of each other if the aromatic groups are interchanged. The crystal structure of the racemate is essentially isostructural with that of the enantiomer, having almost the same cell parameters but containing a crystallographic inversion centre that is not retained in the enantiomer structure. The closely-comparable crystal structures permit solid solutions to be formed between racemic and enantiomeric citalopram oxalate. Phase diagrams of the (R)-citalopram and (S)-citalopram oxalate system are constructed, and they show that solid solutions are formed at all ratios of the two enantiomers. 相似文献

The structural basis of ligand-induced dimerization of the receptor tyrosine kinase MET by its natural ligand hepatocyte growth factor/scatter factor (HGF/SF) is not well understood. However, interesting insight into the molecular mechanism of MET dimerization has emerged from crystal structures of MET in complex with a bacterial agonist, the invasion protein internalin B (InlB) from pathogenic Listeria monocytogenes. MET activation by InlB promotes uptake of bacteria into host cells. Structural and biophysical data suggest that InlB is monomeric on its own but dimerizes upon binding to the membrane-anchored MET receptor promoting the formation of a signaling active 2:2 complex. The dimerization interface is small and unusually located on the convex side of the curved InlB leucine-rich repeat (LRR) domain. As InlB does not dimerize in solution, the dimerization site could only be identified by studying packing contacts of InlB in various crystal forms and had to be proven by scrutinizing its biological relevance in cellular assays. InlB dimerization is thus an example of a low-affinity contact that appears irrelevant in solution but becomes physiologically significant in the context of 2-dimensional diffusion restricted to the membrane plane. The resulting 2:2 InlB:MET complex has an InlB dimer at its center with one MET molecule bound peripherally to each InlB. This model of ligand-mediated MET dimerization may serve as a blue-print to understand MET activation by NK1, a naturally occurring HGF/SF splice variant and MET agonist. Crystal structures of NK1 repeatedly show a NK1 dimer, in which residues implicated in MET-binding are located on the outside. Thus, MET dimerization by NK1 may also be ligand-mediated with a NK1 dimer at the center of the 2:2 complex with one MET molecule bound peripherally to each NK1. Process diagrams are important tools in biology for explaining processes such as protein synthesis, compound cycles and the like. The aim of the present study was to measure the ability to solve process-diagram problems in biology and its relationship with prior knowledge, spatial ability and working memory. For this purpose, we developed a test that represents process diagrams and adjacent tasks used in secondary education biology. Results show that the ability to solve process-diagram problems is correlated to prior knowledge, spatial abilities and visuospatial working memory capacity. A difference in impact of spatial skills was demonstrated for the level of cognitive demand when solving process-diagram problems. 相似文献

Material flow analysis is a key tool to quantify and monitor the use of natural resources. A very visual way to undertake such analyses representing the mineral trade is through Sankey diagrams, in which the mineral resources that are extracted, imported, exported, recycled and consumed within the given boundaries are represented with the arrows proportional to their respective quantities. Yet Sankey diagrams alone are not sensitive to the quality of the resources as they only reflect tonnage. This issue can lead to misleading conclusions and thereby not effective resource policies. A way to overcome this deficiency is representing the flows in exergy replacement cost (ERC) terms instead of tonnage. Exergy replacement cost is a concept derived from the second law of thermodynamics and assesses the exergy cost required to return with available technologies a given mineral to its initial conditions of composition and concentration in the mines where it was found, once it has been dispersed after use. Using this methodology, minerals are physically valued in terms of their respective scarcities and the effort (in exergy cost terms) required to produce them. Accordingly, in this paper the so-called exergoecology method is used to evaluate mineral trade and foreign mineral dependency in the EU-28 for 1995 to 2012. Using the year 2011 as a case study, it can be seen using this novel approach that 45.8% of the total input of minerals are imported resulting in lower values of self-sufficiency than if a traditional MFA were applied (0.45 for minerals and 0.41 for fossil fuels, in contrast to 0.79 and 0.52 obtained respectively when using tonnes). Analyzing 10 of the 20 minerals deemed critical by the European Commission, of the total internal production, 0.88% corresponded to critical minerals when data were expressed in tonnes and 3.19% when expressed in exergy replacement costs, highlighting their relevance respect to other minerals. This external dependency leaves Europe in a delicate situation regarding fossil fuels and non-fuel minerals supply highlighting the importance of recycling especially scarce minerals and searching for alternative sources. 相似文献
