为明确拟水狼蛛对食物中镉的吸收和排泄及生物学响应,采用原子吸收光谱法检测了连续3代拟水狼蛛对食物中Cd2+的吸收和排泄情况,并测定了Cd2+对其生长发育、繁殖力和耐饥力的影响。结果表明:食物中过量的Cd2+能够通过食物链进行传递并在拟水狼蛛体内积累,积累量随拟水狼蛛代数的增加而增加,第2代和第3代拟水狼蛛体内Cd2+积累量浓度显著高于第1代中Cd2+积累量,第2代和第3代间差异不显著,积累量与消耗的黑腹果蝇数量极显著正相关,P<0.01;与消耗食物中Cd含量显著正相关,P<0.05。连续3代拟水狼蛛分别吸收了食物中65.4%、68.5%和69.1%的Cd,生物营养级放大因子分别为1.71、2.12和2.17。连续3代拟水狼蛛Cd排泄量极低。Cd处理能显著改变拟水狼蛛背胸甲宽、幼蛛存活率、生长历期和繁殖力。随拟水狼蛛受胁迫代数的增加,幼蛛存活率显著减少,生长历期显著延长。卵袋重量显著减轻,卵数目显著减少,卵体积显著升高。Cd处理还能显著降低拟水狼耐饥力。研究结果可为进一步研究环境中Cd沿土壤-昆虫-天敌传递、放大和生理耐受提供更为充分的理论依据。  相似文献

Glycosylation is a widespread PTM of proteins; the carbohydrate moieties provide various functional, immunological and structural aspects of both eukaryotic and prokaryotic glycoproteins. Traditional strategies used to analyse glycoprotein O-glycans involve glycoprotein isolation, followed by glycan release using solution-phase base-catalysed beta-elimination. However, in a proteomics context, mixtures of proteins and glycoproteins are routinely separated using SDS-PAGE. We have therefore developed a method to enable the profiling of O-linked glycans directly from glycoproteins on gels. This is achieved using in-gel reductive beta-elimination followed by mass spectrometric analysis of the released glycans. Here we describe our demonstration of the feasibility of this approach, our development and optimisation of the procedure using bovine submaxillary gland glycoproteins as a standard, and then show its usefulness by applying the developed procedure to the analysis of the O-glycans from a glycoprotein band from a Coomassie-stained SDS-PAGE separation of a mixture of Mycobacterium avium capsular proteins and glycoproteins. The procedure has been shown to be applicable to both CBB- and silver-stained gels. The method offers a quick and easy way to identify the O-glycans from gel-separated glycoproteins within gel-based proteomics workflows.  相似文献

The 1,3-dinitrobenzene-degrading Rhodococcus strain QT-1 was isolated under nitrogen limiting conditions from contaminated soil samples. Experimental data indicate that 1,3-dinitrobenzene is metabolized via 4-nitrocatechol. Both compounds were oxidized by resting cells and nitro groups were completely eliminated as nitrite. Strain QT-1 utilizes both 1,3-dinitrobenzene and 4-nitrocatechol as source of nitrogen in the absence as well as in the presence of high amounts of ammonia. 探讨中药五倍子提取液对铜绿假单胞菌R质粒的消除作用。对老年呼吸系统感染患者痰中分离的铜绿假单胞菌R质粒进行了检测 ,并选用中药五倍子提取液对该质粒进行了体外体内消除试验。结果可见 ,体外药物作用 2 4 ,4 8,72hR质粒消除率分别为 0 ,10 .2 % ,3.4 % ,SDS对照组分别为 0 ,0 .8% ,1.6 %。体内药物作用 2 4 ,4 8,72h ,R质粒消除率分别为 0 ,2 % ,13.6 % ,对照组均为 0。可见 ,中药五倍子提取液对铜绿假单胞菌R质粒在体外、体内均具有消除作用 ,体内消除作用强于体外。  相似文献

目的:通过检测人膝骨关节炎裸露软骨下骨中OPN的表达,探讨OPN在OA发病及病情进展中的意义。方法:选取接受膝关节置换手术的膝关节骨关节炎患者软骨下骨标本50例,采用综合评分法对OA患者进行严重程度分级,分为轻、中、重度三组,取正常膝关节软骨下骨(股骨髁关节面)10例作为正常软骨下骨对照;对标本进行免疫组织化学染色,用SPSS 17.0统计软件包分析各组间OPN表达的差异及OA患者OPN表达与综合评分、K-L分期的相关性。结果:人膝骨关节炎裸露软骨下骨OPN表达明显高于正常软骨下骨组,差异有统计学意义(P<0.01);OPN在轻、中、重度膝骨关节炎裸露软骨下骨的表达差异有统计学意义(P<0.05);膝骨关节炎裸露软骨下骨OPN的表达与骨关节炎的综合评分、K-L分期呈正相关。结论:OA患者膝关节软骨下骨OPN表达与疾病严重程度呈正相关,提示OPN在骨关节炎发病及病情进展中可能起作用。  相似文献

采用十二烷基硫酸钠(SDS)和提高生长温度结合法消除T1828菌株质粒后,筛选到无质粒突变株ZWL-15。以原始亲株T1828和无质粒突变株ZWL-15为出发菌株,考察了两者部分生物学特性。结果表明,菌株ZWL.15生长速率快于T1828,进入对数期和稳定期的时间分别提前1h和4h。在菌株ZWL-15发酵过程中添加0.01%的SDS有利于冠菌素的合成,最适发酵温度比T1828升高5℃,达到35℃,发酵周期提前4h,COR相对值是T1828的两倍左右。ZWL-15传代实验表明该突变株很稳定,且对氨苄青霉素敏感,氨苄青霉素基因初步定位于该菌属质粒上。  相似文献

Rheumatoid arthritis (RA), osteoarthritis (OA), and periodontal disease (PD) are chronic inflammatory diseases that are globally prevalent, and pose a public health concern. The search for a potential mechanism linking PD to RA and OA continues, as it could play a significant role in disease prevention and treatment. Recent studies have linked RA, OA, and PD to Porphyromonas gingivalis (PG), a periodontal bacterium, through a similar dysregulation in an inflammatory mechanism. This study aimed to identify potential gene signatures that could assist in early diagnosis as well as gain insight into the molecular mechanisms of these diseases. The expression data sets with the series IDs GSE97779, GSE123492, and GSE24897 for macrophages of RA, OA synovium, and PG stimulated macrophages (PG-SM), respectively, were retrieved and screened for differentially expressed genes (DEGs). The 72 common DEGs among RA, OA, and PG-SM were further subjected to gene–gene correlation analysis. A GeneMANIA interaction network of the 47 highly correlated DEGs comprises 53 nodes and 271 edges. Network centrality analysis identified 15 hub genes, 6 of which are DEGs (API5, ATE1, CCNG1, EHD1, RIN2, and STK39). Additionally, two significantly up-regulated non-hub genes (IER3 and RGS16) showed interactions with hub genes. Functional enrichment analysis of the genes showed that “apoptotic regulation” and “inflammasomes” were among the major pathways. These eight genes can serve as important signatures/targets, and provide new insights into the molecular mechanism of PG-induced RA, OA, and PD.  相似文献

Ants (Formicinae, Hymenoptera) are considered efficient accumulators of Zn and Cd. In this study the relationship between Cd concentrations in soil and in Lasius niger workers was assessed for 20 colonies located along a gradient of Cd pollution. As soil Cd concentration increased from 1 to 8 mg/kg, ant body Cd concentration increased rapidly to about 40 mg/kg, and remained stable at this level, through a progressive increase of soil Cd concentration from 8 to 21 mg/kg. The phase of rapid increase confirms the high ability of L. niger for Cd accumulation, while the stabilization of Cd body concentration indicates either increasing elimination rate or decreasing rate of uptake. Therefore it can be concluded that Lasius niger has a high ability to accumulate Cd, but simultaneously an ability for Cd regulation was detected.  相似文献
