The impact of storm-flow on river biofilm architecture was investigated using transmission (TEM) and scanning (SEM) electron microscopy. TEM resin substrata were colonized under light-grown (LG) or dark-grown (DG) conditions for 33 weeks in the Clywedog River, North Wales, prior to exposure to ambient-flow (approx. 60 cm·s?1) or storm-flow (approx. 235 cm·s?1+ river sediment) in a laboratory flume. Line transect methodology was used to quantify information from TEM ultrathin sections of LG material. In the LG ambient-flow biofilm, bacteria were more abundant directly adjacent to the substratum and were noticeably denser directly under the adnate diatom Cocconeis. Higher in the biofilm, the bacteria were loosely dispersed in the matrix between other cells. Cyanobacteria occurred most frequently as single cells but were also found in large “palisade” formations adjacent to the substratum. Significant horizontal and vertical nearest-neighbor associations were noted for both bacteria and cyanobacteria. Cells of Cocconeis were common adjacent to the substratum, providing shelter to, and often elevated upon, an “organic pad” of bacteria, cyanobacteria, and densely staining exopolysaccharide. Cyanobacteria and Cocconeis were resistant to removal by storm-flow, but Cocconeis frustules were sometimes damaged. Bacteria in the LG storm-flow samples were less common adjacent to the substratum and were sometimes more dispersed higher in the biofilm than in ambient-flow samples. We suggest that storm-flow hydrodynamic forces may redistribute bacteria adjacent to the substratum into higher areas of the biofilm. In addition, bacteria and the exopolysaccharide matrix were sometimes removed down to the substratum by storm-flow, unless beneath Cocconeis. The DG biofilm consisted almost entirely of bacteria. Storm-flow only removed surface growth from DG biofilms, and SEM revealed peritrich stalk abrasion and “blow-down.” Pre-disturbance biofilm architecture appears to influence the form of destruction. We suggest that the “microcosms” of Cocconeis and their underlying cells not only serve as an inoculum to recolonize the surface when conditions permit but enhance immigration by interrupting flow patterns across the surface.  相似文献

The biomechanics of the sauropod dinosaur pes is poorly understood, particularly among the earliest members of the group. To date, reasonably complete and articulated pedes in Early Middle Jurassic sauropods are rare, limited to a handful of taxa. Of these, Rhoetosaurus brownei, from eastern Australia, is currently the only one from the Gondwanan Middle Jurassic that preserves an articulated pes. Using Rhoetosaurus brownei as a case exemplar, we assessed its paleobiomechanical capabilities and pedal posture. Physical and virtual manipulations of the pedal elements were undertaken to evaluate the range of motion between the pedal joints, under both bone-to-bone and cartilaginous scenarios. Using the results as constraints, virtual reconstructions of all possible pedal postures were generated. We show that Rhoetosaurus brownei was capable of significant digital mobility at the osteological metatarsophalangeal and distal interphalangeal joints. We assume these movements would have been restricted by soft tissue in life but that their presence would have helped in the support of the animal. Further insights based on anatomy and theoretical mechanical constraints restricted the skeletal postures to a range encompassing digitigrade to subunguligrade stances. The approach was extended to additional sauropodomorph pedes, and some validation was provided via the bone data of an African elephant pes. Based on the resulting pedal configurations, the in-life plantar surface of the sauropod pes is inferred to extend caudally from the digits, with a soft tissue pad supporting the elevated metatarsus. The plantar pad is inferred to play a role in the reduction of biomechanical stresses, and to aid in support and locomotion. A pedal pad may have been a key biomechanical innovation in early sauropods, ultimately resulting in a functionally plantigrade pes, which may have arisen during the Early to Middle Jurassic. Further mechanical studies are ultimately required to permit validation of this long-standing hypothesis.  相似文献

Summary Primary events during the establishment of the fungus-root symbiosis in ectomycorrhizas are still little understood. No attention has been paid so far to the adhesion of hyphae to the root cuticle and penetration of this barrier, although the importance of the cuticle has been shown for pathogen-plant interactions. Early developmental stages of in vitro mycorrhization ofLaccaria amethystea onPicea abies after short periods of incubation in growth chambers under elevated CO2 concentrations were studied by light and transmission electron microscopy. No structural changes in mycorrhization related to elevated CO2 were found, but fine roots and mycorrhizas developed faster. Adhesion pad formation was observed at hyphal tips in contact with the root cuticle. The adhesion pad was connected to the outer cell wall layer of the hypha and reacted positively to the Swift reaction for cysteine rich proteins. Although the reaction cannot be considered as totally specific, findings are discussed in respect to hydrophobins, which have recently been found to be expressed during early steps in ectomycorrhizal development. The root cuticle was dissolved and penetrated by fungal tips of the fingerlike branching mycelium attached to the root surface. The findings are compared with well documented pathogenic fungus-plant interactions at the cuticle. The possibility of restriction of hyphal attack to that part of the cuticle covering cell junctions is discussed.  相似文献

Secondary sexual structures in millipedes have evolved to improve the efficiency with which spermatophores are transferred from males to females. Here, we demonstrate the microstructural characteristics of the sexual dimorphism in the leg adhesive pads of the millipede Orthomorphella pekuensis, using field emission scanning electron microscopy. The adult male of this millipede has developed typical adhesive pads on the legs as a secondary sexual character: these help the male hold on to the female during mating. The adhesive pads are located on the ventral surface of the tarsus in the form of numerous filamentous bristles with horizontal striation. There are approximately 300 bristles, each with an average length of approximately 60 µm, on each tarsus. Each bristle has a round socket on the surface of the tarsus and an acuminate tip. The striation appears only on the distal half of each bristle, typically.  相似文献

Biosynthesis of Nylon 12 monomer using dodecanoic acid (DDA) or its esters as the renewable feedstock typically involves ω-hydroxylation, oxidation and ω-amination. The dependence of hydroxylation and oxidation-catalyzing enzymes on redox cofactors, and the requirement of L-alanine as the co-substrate and pyridoxal 5′-phosphate (PLP) as the coenzyme for transamination, raise the issue of redox imbalance and cofactor shortage, challenging the development of efficient biocatalysts. Simultaneous regeneration of the redox equivalents, PLP and L-alanine required in the artificial pathway was enabled by its interfacing with the native metabolism of the host using glucose dehydrogenase (GDH), L-alanine dehydrogenase (AlaDH) and an exogenous ribose 5-phosphate (R5P)-dependent PLP synthesis pathway as bridges. Further engineering of the host by blocking β-oxidation and enhancing substrate uptake improved the ω-aminododecanoic acid (ω-AmDDA) yield to 96.5%. This study offers a strategy to resolve the cofactor imbalance issue commonly encountered in whole-cell biocatalysis and meanwhile lays a solid foundation for Nylon 12 bioproduction.  相似文献

Salicylic acid (SA)-dependent signaling controls activation of a set of plant defense mechanisms that are important for resistance to a variety of microbial pathogens. Many Arabidopsis mutants that display altered SA-dependent signaling have been isolated. We used double mutant analysis to determine the relative positions of the pad4, cpr1, cpr5, cpr6, dnd1 and dnd2 mutations in the signal transduction network leading to SA-dependent activation of defense gene expression and disease resistance. The pad4 mutation causes failure of SA accumulation in response to infection by certain pathogens, while the other mutations cause constitutively high levels of SA, defense gene expression and resistance. The cpr1 pad4, cpr5 pad4, cpr6 pad4, dnd1 pad4 and dnd2 pad4 double mutants were constructed and assayed for stature, presence of spontaneous lesions, resistance to Pseudomonas syringae and Peronospora parasitica, SA levels, expression of PAD4, PR-1 and PDF1.2, and accumulation of camalexin. We found that the effects of the cpr1 and cpr6 mutations on SA-dependent gene expression are completely dependent on PAD4 function. In contrast, SA accumulation in the lesion-mimic mutant cpr5 is partially PAD4-independent, while in dnd1 and dnd2 mutants it is completely PAD4-independent. A model describing a possible arrangement of activities in the signal transduction network is presented.  相似文献

This article discusses the analysis of the hydrolysis products from one-step scouring of cotton using pectinase and two-step scouring of cotton using lipase then cellulase, protease then cellulase, or lipase/protease then cellulase, to improve water absorbency of cotton. UV spectrophotometric analysis indicated that the pectinase scouring process produced approximately 18-fold higher amounts of reducing sugars and galacturonic acid than any of the two-step scouring processes. The production rate of reducing sugars and galacturonic acid from most of the scouring processes showed a decrease with an increase in time. HPLC analysis revealed that the lipase/protease/cellulase scouring processes produced approximately 5-fold higher amounts of 17 amino acids than the pectinase scouring process. GC analysis for 18 fatty acids (C(8)-C(24)) revealed that three major fatty acids, palmitic acid, stearic acid, and behenic acid, were found on both the scoured and the unscoured fabrics. Scoured fabrics were tested for content of proteins, extractable components, waxes, and anionic components including pectins, and some differences among the fabric scoured with different enzyme combinations were found.  相似文献

利用石蜡切片的方法,观察并比较了褐飞虱Nilaparvata lugens (Stal)长翅型和短翅型纯系各龄若虫及成虫的翅芽和间接飞行肌的发育情况。发现在4龄的第8h以后间接飞行肌就开始分化,长翅型若虫的间接飞行肌正常发育而短翅型若虫间接飞行肌的发育则被抑制。在5龄初始时方可明显观察到翅芽分化,短翅型若虫前翅芽细胞增殖速度明显慢于长翅型,并且其后翅芽停止发育。本文还比较了长翅型雌、雄性个体之间间接飞行肌的发育情况。  相似文献
