Many marine Alphaproteobacteria of the Roseobacter group show a characteristic swim-or-stick lifestyle, for which motility is a crucial trait. Three phylogenetically distinct flagellar gene clusters (FGCs) have been identified in Rhodobacteraceae that have been named fla1, fla2 and fla3 according to their relative abundance. In addition to the flagellar-dependent swimming and swarming motility, pilus-dependent twitching mediates bacterial locomotion. Furthermore, filament independent modes of motility, namely gliding and sliding, have been described for various microorganisms. However, no mode of motility other than swimming has so far been described for roseobacters. In the present study, we investigated motility, distribution of flagellar systems and the phylogeny of 120 genome-sequenced Rhodobacteraceae. The phylogenetically broad taxon sampling that included 114 type strains revealed the presence of at least ten distinct clades that were statistically well supported. The investigation of the actual physiological capacity for swimming motility on soft agar plates showed that only about half of the 120 tested strains were motile under the tested conditions. Seven strains developed a conspicuous dendritic motility phenotype that was reminiscent of the swarming motility in Pseudomonas aeruginosa. The observed dendritic motility in two strains (i.e. Sulfitobacter pseudonitzschiae DSM 26842 and Roseovarius pacificus DSM 29589) was particularly surprising because they did not harbor any genes of the FGC. Accordingly, it was concluded that this form of dendritic motility was independent of a flagellum. A comparative genomics approach allowed a remarkable number of pilus-related candidate genes to be identified for this novel type of motility in Rhodobacteraceae. 相似文献

Various phosphatidylcholines differing either in the stereochemistry around their chiral center or in the position of a cis double bond along the acyl chains were synthesized in order to study critical contact regions in the phospholipid molecule with adjacent cholesterol in model membranes. Microviscosities calculated from fluorescence depolarization of diphenylhexatriene and chain order from spin label studies were measured to monitor physical membrane properties. The enhancing effect of cholesterol on the microviscosity of membranes containing phosphatidylcholines with comparable acyl chain length was largest when the two acyl chains were saturated and smallest when both were unsaturated. Membranes prepared from phosphatidylcholines having a single cis double bond at different positions along the sn-2 acyl chain showed roughly the same changes of microviscosity or chain order upon incorporation of cholesterol. No discrimination was evident in the interaction between cholesterol and enantiomeric phosphatidylcholines or between the enantiomeric phosphatidylcholine molecules themselves. We conclude that the rigidifying effect of cholesterol in membranes does not depend on specific sites of interaction and that with respect to physical membrane properties phosphatidylcholine behaves as an achiral molecule. 相似文献

Biosynthesis of the C-terminal carboxamide group of peptide hormones was studied using comparatively pGlu-His-Pro-Gly and Glu-His-Pro-Gly-Lys-Arg as putative precursors of the tripeptide, thyroliberin (TRH). Rat hypothalamus granules were found to contain an amide group forming activity which converts both peptide substrates into TRH. Comparison of the rate of conversion of the two substrates indicated that the C-terminal dibasic extension favored a 10-fold increase in the production of amidated peptide. It is suggested that this type of structure may be present in the putative biosynthetic precursor of TRH and that it may provide a better substrate for the enzyme(s) involved in C-terminal amidation. 相似文献

The fine structure of the organic matrix of the shore crab cuticle (Carcinus maenas L.), observed in transmission electron microscopy, reveals three different levels of organization of the chitin—protein complex. The highest level corresponds to the ‘twisted plywood’ organization described by Bouligand (1972). Horizontal microfibrils, parallel to the cuticle plane, rotate progressively from one level to another. When viewed in oblique section this structure gives superimposed series of nested arcs, visible in light microscopy or at the lowest magnifications of the electron microscope, in all the chitin-protein layers. At the highest magnifications of the electron microscope and with the best resolution, when the ultrathin sections are exactly transverse to the microfibril, a constant pattern can be observed which consists of rods transparent to electrons, which are embedded in an electron-opaque matrix. In cross-section, these rods often form more or less hexagonal arrays. We call a microfibril one rod and the adjacent opaque material, and question the usual interpretation of the microfibril molecular structure. Between these two levels of organization, there is an intermediate level, which corresponds to the grouping of microfibrils. Microfibrils form a dense structure, with few free spaces in the membranous layer, the deepest and non-calcified layer of the cuticle. In other parts of the cuticle, microfibrils are grouped into fibrils of various diameters or form a reticulate structure, the free spaces of the organic matrix being occupied by the mineral. 相似文献

A vinculin-like protein was identified in chicken as well as in bovine platelets by ELISA competitive binding assay using antibodies against vinculin from chicken gizzard. By a modified procedure (J. Biol. Chem. (1980) 255, 1194–1199) we succeeded in isolating bovine platelet vinculin to apparent homogeneity. The structural identity of platelet and chicken gizzard vinculin was demonstrated by circular dichroism analysis. It was also shown that platelet vinculin induces a significant decrease in the low shear viscosity of F-actin. Vinculin, in all probability, plays an important role in the organization of actin filaments in platelets, especially in the linkages of microfilaments to the membrane. 相似文献

All-trans retinoic acid increased the incorporation of D-[3H]galactose into particulate and soluble glycoproteins in the epidermis of cultured pig skin slices nearly two-fold. Increased incorporation of D-[3H]galactose was not blocked by tunicamycin. This effect was specific for D-[3H]galactose since the incorporation of D-[3H]glucosamine and L-[14C]leucine into epidermal glycoproteins was unaffected by all-trans retinoic acid. All-trans retinoic acid and 13-cis retinoic acid had quantitatively similar effects on D-[3H]galactose incorporation. All-trans retinyl acetate and an aromatic retinoic acid analogue (‘Etretinate’) were less effective. SDS polyacrylamide gel electrophoresis and fluorography showed increased incorporation of D-[3H]galactose into all epidermal glycoproteins in the presence of all-trans retinoic acid. 本文报道两种测定λ原噬菌体紫外诱导频率的新方法 - 菌落计数法和平板诱导法.将溶源菌液经紫外诱导暗培养稀释后直接涂布在平板上培养,根据平板上菌落形成单位数计算λ噬菌体紫外诱导频率.另一种是将溶源菌与指示菌混合制备的平板用紫外线诱导,根据平板上噬菌体形成单位确定λ噬菌体紫外诱导频率.这两种方法不仅能准确测定噬菌体紫外诱导频率,而且操作简便,节省时间和用具,重复性好.本研究还将冬虫夏草浸出汁与溶源菌混合后进行紫外辐射,通过几种方法进行比较,结果证明建立的新方法确实可行,易操作;同时也表明冬虫夏草具有较强的抗紫外辐射作用. 相似文献
