Effective containment of gene flow in transgenic plants requires a promoter that is highly specific for male and female gametes or tissues. Here, we report the creation of a novel pollen-, stigma- and carpel-specific (PSC) promoter through the fusion of the pollen-specific LAT52 and carpel-specific AGL5 enhancers to a stigma-specific SLG promoter. Gene expression analysis showed that fusion of the LAT52 enhancer to the SLG promoter enables the latter to gain pollen-specific activity while the acquirement of carpel-specific activity requires the correct orientation of the inserted AGL5 enhancer in the PSC promoter, and only a forward- but not a reverse-oriented one is functional. The resulting fPSC promoter, when fused to DT-A, generated at least three aberrant gynoecium phenotypes. Type I plants exhibited shortened stigmatic tissues, resembling plants containing the DT-A gene controlled by the SLG promoter. However, type II and III plants displayed partial or complete ablation of gynoecia, and were unable to support the reproductive process. Type II and III plants also produced severely perturbed anthers and pollen in comparison to type I or SLG::DT-A plants, and transgenic pollen grains were unable, when out-crossed with control plants, to pass the transgene to the next generation in all plants examined, indicating that they are selectively eliminated. This tissue-specific ablation or perturbation is highly specific, and does not compromise vegetative growth. Evidently, the fPSC promoter faithfully acquires tissue specificity from the incorporated enhancers and promoter, and should have a practical application for transgene containment in non-fruit and -grain producing plant crops. 相似文献

由人羊水中分离羊水多能性干细胞,通过形成类胚体诱导其向心肌细胞分化.取人羊水标本进行体外培养,分离得到人羊水干细胞,已连续传代培养至42代,采用免疫细胞化学、RT-PCR和流式细胞仪技术对羊水干细胞的生物学特性进行检测.取10~15代羊水干细胞,悬浮培养使其形成类胚体,进而向心肌细胞诱导分化.培养的羊水干细胞呈成纤维样,表达部分胚胎干细胞特异标志基因,悬浮培养可形成类胚体.类胚体碱性磷酸酶(AP)检测呈阳性,表达三胚层特异标志基因fgf5、ζ-globin和α-fetoprotein.羊水干细胞形成类胚体后进行诱导,得到α-actin阳性细胞,表达心肌细胞特异标志基因Tbx5、Nkx2.5、GATA4和α-MHC.试验结果表明,从人羊水标本中可分离得到具有胚胎干细胞特性的细胞,经初步检测确定为羊水干细胞,并能通过形成类胚体诱导其向心肌细胞分化. 相似文献

Although theory predicts that dispersal has a pivotal influence on speciation and extinction rates, it can have contradictory effects on each, such that empirical quantification of its role is required. In many studies, dispersal reduction appears to promote diversification, although some comparisons of migratory and nonmigratory species suggest otherwise. We tested for a relationship between migratory status and diversification rate within the dominant radiation of temperate Southern Hemisphere freshwater fishes, the Galaxiidae. We reconstructed a molecular phylogeny comprising >95% of extant taxa, and applied State-dependent Speciation Extinction models to estimate speciation, extinction, and diversification rates. In contrast to some previous studies, we revealed higher diversification rates in nonmigratory lineages. The reduced gene flow experienced by nonmigratory galaxiids appears to have increased diversification under conditions of allopatry or local adaptation. Migratory galaxiid lineages, by contrast, are genetically homogeneous within landmasses, but may also be rarely able to diversify by colonizing other landmasses in the temperate Southern Hemisphere. Apparent contradictions among studies of dispersal-diversification relationships may be explained by the spatial context of study systems relative to species dispersal abilities, by means of the “intermediate dispersal” model; the accurate quantification of dispersal abilities will aid in the understanding of these proposed interactions. 相似文献

Exposure of microbial cells to sub-lethal stresses is known to increase cell robustness. In this work, a two-compartment bioreactor in which microbial cells are stochastically exposed to sub-lethal temperature stresses has been used in order to investigate the response of the stress sensitive Bifidobacterium bifidum THT 0101 to downstream processing operations. A stochastic model validated by residence time distribution experiments has shown that in the heat-shock configuration, a two-compartment bioreactor (TCB) allows the exposure of microbial cells to sub-lethal temperature of 42 °C for a duration comprised between 100 and 300 s. This exposure resulted in a significant increase of cell resistance to freeze–drying by comparison with cells cultivated in conventional bioreactors or in the TCB in the cold shock mode (CS-TCB). The mechanism behind this robustness seems to be related with the coating of microbial cells with exopolysaccharide (EPS), as assessed by the change of the zeta potential and the presence of higher EPS concentration after heat shock. Conditioning of Bifidobacteria on the basis of the heat shock technique is interesting from the practical and economical point of view since this strategy can be directly implemented in the bioreactor during stationary phase preceding cell recovery and freeze–drying. 相似文献

The California Floristic Province is home to more than 8000 species of beetles, yet their geographical patterns of supra- and infraspecific diversity remain largely unexplored. In this paper, we investigate the phylogeography and population demographics of a flightless ground beetle, Calathus ruficollis (Coleoptera: Carabidae), in southern California. We sampled 136 specimens from 25 localities divided into 10 populations using a fragment of the mitochondrial cytochrome oxidase I gene. We tested several hypotheses, including the association of geography with particular clades and populations, the degree of differentiation among regions, and the expansion of populations. Parsimony and Bayesian phylogenetic analyses along with nested clade analysis and amova indicate a deep split between the southern Sierra Nevada population and populations south and west. This split corresponds closely to the split between subspecies C. ruficollis ignicollis (southern Sierra Nevada) and C. ruficollis ruficollis . Populations otherwise exhibit limited geographical structure, though F st values indicate some local differentiation. Mismatch distributions and Fu's F s indicate range expansion of several populations, suggesting that some structure may have been obscured by recent exchange. The population of C. ruficollis on Santa Cruz Island, which might have been expected to be isolated, shares several haplotypes with mainland populations, appearing to represent multiple colonizations. 相似文献

- 1 The mammal fauna of the British Isles has seen major perturbations since the end of the last ice age, some natural and some anthropogenic. Today, 61 species of terrestrial mammals breed in the British Isles, but only 39 of them are native species, the rest have been introduced. Furthermore, 19 native species have disappeared from the fauna including all the large predators, lynx Lynx lynx, wolf Canis lupus and brown bear Ursus arctos.
- 2 Inevitably, these changes in species composition have been accompanied by major changes in community function including changes in patterns of energy flow through the mammalian community. For example, a high percentage of all the energy now flows through the introduced rabbit Oryctolagus cuniculus population and red deer Cervus elaphus, which, in the absence of natural predators, are living at extremely high densities.
- 3 Could the reintroduction of species help to reverse such changes in community structure and function? The successful return to the wild of species such as the Arabian oryx Oryx leucoryx and beaver Castor fiber has shown that reintroductions are certainly possible. However, the impact on community function of returned species is more difficult to evaluate.
- 4 This question is addressed in relation to the consequences for deer populations of any possible reintroduction of the wolf to Scotland. Based on what we know of wolves elsewhere, predictions are made about the likely demography and patterns of killing behaviour of introduced wolves. These values are then used to parameterize a Leslie matrix simulation of the impact of wolves on contemporary populations of red deer in Scotland. The simulations suggest very strongly that wolves are very unlikely to have any significant impact on the high‐density populations of deer now living in the Scottish landscape.

Background
Gd-DTPA-enhanced magnetic resonance imaging (MRI) is a conventional method for non-invasive investigation of blood-brain-barrier (BBB) permeability in animal models. It allows the visualization of serious injury to the BBB. We developed a novel approach for detecting very small disruptions in BBB permeability induced by dietary cholesterol by using carbamoyl-PROXYL (CMP) as an MRI contrast probe.Methods
Mice were separated into two groups: normal diet (ND-mice) and high cholesterol diet (CD-mice). MRI-signal dynamics, plasma cholesterol, matrix metalloproteinase (MMP-9, MMP-2), and the white blood cell profile were analyzed. For the MRI analysis, two regions-of-interest (ROI) were selected: brain (ROI-1) and surrounding area (ROI-2).Results
In the ROI-2 of ND-mice, CMP- or Gd-enhanced MRI-signal followed typical kinetics with a half-life of signal decay (τ1/2) ~ 8 or ~ 15 min, respectively. In CD-mice, the MRI-signal increased continuously without decay.In the ROI-1 of ND- and CD-mice, MRI-signal enhancement was not detected by Gd-DTPA. In the ROI-1 of ND-mice, CMP-induced MRI-signal enhancement was negligible, while in CD-mice, it was significant (τ1/2 > 15 min).Hypercholesterolemia increased the plasma levels of MMP-9 and neutrophils.Conclusions
Hypercholesterolemia increases vascular permeability, which is mediated by MMP-9 and neutrophils.General significance

A new chemiluminescence (CL) reaction was observed when chloramphenicol solution was injected into the mixture after the end of the reaction of alkaline luminol and sodium periodate or sodium periodate was injected into the reaction mixture of chloramphenicol and alkaline luminol. This reaction is described as an order‐transform second‐chemiluminescence (OTSCL) reaction. The OTSCL method combined with a flow‐injection technique was applied to the determination of chloramphenicol. The optimum conditions for the order‐transform second‐chemiluminescence emission were investigated. A mechanism for OTSCL has been proposed on the basis of the chemiluminescence kinetic characteristics, the UV‐visible spectra and the chemiluminescent spectra. Under optimal experimental conditions, the CL response is proportional to the concentration of chloramphenicol over the range 5.0 × 10?7–5.0 × 10?5 mol/L with a correlation coefficient of 0.9969 and a detection limit of 6.0 × 10?8 mol/L (3σ). The relative standard deviation (RSD) for 11 repeated determinations of 5.0 × 10?6 mol/L chloramphenicol is 1.7%. The contribution of sediment interstitial water and the water column to the transpiration stream of Myriophyllum aquaticum (Vellozo) Verdcourt was determined to estimate the significance of mass flow in supply of sediment nutrients for plant growth.
Sediment interstitial water accounted for about 2% of the water transpired over a 37 day period. Because of the small volume
of water that originated in the sediment we concluded that mass flow did not significantly enhance nutrient supply to the
roots of M. aquaticum. Relative growth rate (RGR) of adventitious, water roots was greater than whole plant RGR, and RGR of sediment roots was
not significantly different from zero, indicating a shift in the biomass allocation after emergence of the apical meristem
into the air. Water use, measured by the transpiration coefficient, averaged 260 ml H2O mg DW-1, which is similar to C-4 terrestrial plants. M. aquaticum has leaf characteristics commonly associated with xerophytic habitats. These characteristics may be necessary if a high transpiration
rate and a mechanical requirement for high cell turgor pressure, required by a reliance upon hydrostatic pressure for support
