研究确定Nocardia orientalis NRRL18098发酵生产eremomycin的最佳工艺条件,以及对发酵产物进行分离纯化并得到eremomycin的纯品。通过正交设计方法优化发酵培养基的组成。采用树脂吸附、中压液相色谱技术相结合的方法对发酵产物进行分离纯化。在优化条件下,eremomycin的摇瓶发酵单位达115 mg/L,提高了63.5%,并采用树脂吸附和中压液相色谱相结合的方法能有效地将eremomycin从发酵液中分离出来,制备获得eremomycin精制品。  相似文献

目的:通过构建H22荷瘤小鼠模型,观察红色诺卡氏菌细胞壁骨架(Nocardia Rubra Cell Wall Skeleton,N-CWS)对H22荷瘤小鼠肝癌细胞生长的抑制效果。方法:复苏并培养H22细胞,将H22细胞以皮下方式接种到小鼠右侧腋部,建立H22荷瘤小鼠肝癌模型,随机分为对照组(生理盐水组)、低浓度N-CWS组(400μg/m L)与高浓度N-CWS组(800μg/m L),分别灌胃1次/d,共灌胃14 d,最长接种肿瘤15 d后处死小鼠,观察肝癌细胞的大小和重量。结果:治疗组小鼠肿瘤质量明显低于对照组(1.46±0.16)g,且高浓度组小鼠肿瘤质量(0.52±0.13)g明显低于低浓度组(0.79±0.13)g,组间对比差异有统计学意义(P0.05)。在治疗第3 d起直至结束,治疗组小鼠肿瘤体积均明显低于对照组,从第5 d起直至结束,高浓度组小鼠肿瘤体积明显低于低浓度组(P0.05)。第5 d起至结束,治疗组小鼠肿瘤体积增长的变化明显缓于对照组(P0.05)。高浓度组小鼠肿瘤体积的增长在第5 d后明显缓于低浓度组(P0.05)。对照组在第9 d时,增长变化最为显著,较第7 d增加约400 mm3。低浓度组变化较稳定。高浓度组肿瘤增长明显变缓,第7 d后最为明显,后逐渐趋于缓和。结论:N-CWS可以明显抑制肿瘤生长,且高浓度N-CWS抑制肿瘤生长效果更佳。  相似文献

酰胺酶是一种重要的工业酶。利用生物信息学手段,在和已知酰胺酶基因序列分析比对的基础上,首次从Ncordiasp.YS-2002中成功地克隆得到酰胺酶基因ami,并对其基因序列及氨基酸序列的性质进行了分析。结果表明,所得酰胺酶基因ami片段大小共为1446bp,由启动子区、阅读框和回文结构终止区三部分构成。序列分析和进化树分析表明,Ncordiasp.YS-2002酰胺酶是一种比较特殊的酰胺酶,不含大多数酰胺酶共同具有的保守区序列。进一步将酰胺酶基因连接到pET-28a( )上,转入大肠杆菌BL21(DE3)中筛选获得重组菌株PEAB。酶活测定结果表明重组菌具有酰胺酶酶活,但较低,其原因可能是因为大量表达的产物主要以包涵体的形式存在。  相似文献

Nocardia farcinica is a Gram positive, filamentous bacterium, and is considered an opportunistic pathogen. In this study, the highly expressed genes in N. farcinica were predicted using the codon adaptation index (CAI) as a numerical estimator of gene expressivity. Using ribosomal protein (RP) genes as references, the top ∼ ∼10% of the genes were predicted to be the predicted highly expressed (PHX) genes in N. farcinica using a CAI cutoff of greater than 0.73. Consistent with earlier analysis of Streptomyces genomes, most of the PHX genes in N. farcinica were involved in various ‘house-keeping’ functions important for cell growth. However, 15 genes putatively involved in nocardial virulence were predicted as PHX genes in N. farcinica, which included genes encoding four Mce proteins, cyclopropane fatty acid synthase which is involved in the modification of cell wall which may be important for nocardia virulence, polyketide synthase PKS13 for mycolic acid synthesis and a non-ribosomal peptide synthetase involved in biosynthesis of a mycobactin-related siderophore. In addition, multiple genes involved in defense against reactive oxygen species (ROS) produced by the phagocyte were predicted with high expressivity, which included alkylhydroperoxide reductase (ahpC), catalase (katG), superoxide dismutase (sodF), thioredoxin, thioredoxin reductase, glutathione peroxidase, and peptide methionine sulfoxide reductase, suggesting that combating against ROS is essential for survival of N. farcinica in host cells. The study also showed that the distribution of PHX genes in the N. farcinica circular chromosome was uneven, with more PHX genes located in the regions close to replication initiation site. The results provided the first estimates of global gene expression patterns in N.␣farcinica, which will be useful in guiding experimental design for further investigations. 为从基因水平上改造腈水合酶,进行了诺卡氏菌腈水合酶基因的外源表达研究。在重组大肠杆菌表达系统内,腈水合酶的α亚基几乎不能正常表达,在重组E. coli BL21(DE3) (pET32aNHBAX)中,腈水合酶活性仅为0.04U/mg。构建重组毕赤酵母表达质粒pPIC3.5kNHBAX,采用电穿孔转化法将其转入宿主菌P. pastoris GS115中,经过菌株培养和腈水合酶的诱导表达,筛选获得了优选菌株P. pastoris NH4。对P. pastoris NH4的细胞培养和腈水合酶的诱导表达条件进行优化,结果表明,重组腈水合酶在毕赤酵母中的表达水平可以达到0.52U/mg,但不能稳定积累。  相似文献

Aims:  To investigate the petroleum hydrocarbon (HC)-degrading potential of indigenous micro-organisms in a sandy Mediterranean coast, accidentally contaminated with petroleum-derived HCs.
Methods and Results:  Using culturable methods, a population of Gram-positive n -alkane degraders was detected in the contaminated soil. Five isolates, identified as one Nocardia , two Rhodococcus and two Gordonia strains, were able to degrade medium- and long-chain n -alkanes up to C36 as assessed by growth assays and gas chromatography-mass spectrometry analysis. Diverging alkane hydroxylase-encoding genes ( alkB ) were detected by PCR, using degenerated primers, in all the strains; multiple sequences were obtained from the Nocardia strain, while only one alkB gene was detected in the Rhodococcus and Gordonia strains. The majority of the alkB sequences were related to Rhodococcus alkB2 , but none was identical to it.
Conclusions:  Actinomycetes might have a key role in bioremediation of n -alkane-contaminated sites under dry, resource-limited conditions, such as those found in the Mediterranean shorelines.
Significance and Impact of the Study:  To our knowledge, this is the first study on the bioremediation potential in Mediterranean contaminated beaches.  相似文献

Aims:  The metabolism of phenanthrene and anthracene by a moderate thermophilic Nocardia otitidiscaviarum strain TSH1 was examined.
Methods and Results:  When strain TSH1 was grown in the presence of anthracene, four metabolites were identified as 1,2-dihydroxy-1,2-dihydroanthracene, 3-(2-carboxyvinyl)naphthalene-2-carboxylic acid, 2,3-dihydroxynaphthalene and benzoic acid using gas chromatography-mass spectrometry (GC-MS), reverse phase-high performance liquid chromatography (RP-HPLC) and thin-layer chromatography (TLC). Degradation studies with phenanthrene revealed 2,2'-diphenic acid, phthalic acid, 4-hydroxyphenylacetic acid, o -hydroxyphenylacetic acid, benzoic acid, a phenanthrene dihydrodiol, 4-[1-hydroxy(2-naphthyl)]-2-oxobut-3-enoic acid and 1-hydroxy-2-naphthoic acid (1H2NA), as detectable metabolites.
Conclusions:  Strain TSH1 initiates phenanthrene degradation via dioxygenation at the C-3 and C-4 or at C-9 and C-10 ring positions. Ortho -cleavage of the 9,10-diol leads to formation of 2,2'-diphenic acid. The 3,4-diol ring is cleaved to form 1H2NA which can subsequently be degraded through o -phthalic acid pathway. Benzoate does not fit in the previously published pathways from mesophiles. Anthracene metabolism seems to start with a dioxygenation at the 1 and 2 positions and ortho -cleavage of the resulting diol. The pathway proceeds probably through 2,3-dicarboxynaphthalene and 2,3-dihydroxynaphthalene. Degradation of 2,3-dihydroxynaphthalene to benzoate and transformation of the later to catechol is a possible route for the further degradation of anthracene.
Significance and Impact of the Study:  For the first time, metabolism of phenanthrene and anthracene in a thermophilic Nocardia strain was investigated.  相似文献

Nocardia beijingensis, a recently established new species, is an isolate from soil in China. During our taxonomic studies on 450 nocardial clinical isolates in Thailand and Japan, 17 strains from Thailand and 1 strain from Japan were found to have a similar physiological characteristic to those of N. beijingensis, such as a drug susceptibility pattern to three antimicrobial agents. Our phylogenetic studies on these 18 strains by 16S rRNA gene sequence analysis confirmed that these strains belong to N. beijingensis species. Phylogenetically, these newly isolated N. beijingensis strains were found to be classified into two distinct clades: one is a Japanese clade and other is a Chinese clade, including a reference strain and 17 Thai strains. This is the first report of human infection due to N. beijingensis strains, and we propose that the bacterium be categorized as an opportunistic infectious group regardless of its original isolation from soil.  相似文献

This paper deals with nocardiosis with brain abscess in a cynomolgus monkey imported from Indonesia. At necropsy, typical abscesses were found in the mandible, lung, kidney, heart, liver, and brain. Nocardia asteroides was isolated as the causative agent. The lesions in the kidney, heart, and brain were considered to be secondary metastases.  相似文献
