Phylogenetic relationships in the Olea europaea complex and the phylogeography of 24 populations of the Macaronesian olive (O. europaea ssp. cerasiformis) were assessed by using three molecular markers: nuclear ribosomal internal transcribed spacer 1 (ITS-1) sequences, randomly amplified polymorphic DNAs (RAPD), and intersimple sequence repeats (ISSR). Parsimony analysis of the ITS-1 sequences and Neighbour-joining (NJ) analyses of RAPD and ISSR banding variation revealed four major lineages in the O. europaea complex: (1) ssp. cuspidata; (2) ssp. cerasiformis from Madeira; (3) ssp. laperrinei; and (4) ssp. cerasiformis from the Canary Islands plus ssp. europaea. These results provide unequivocal support for two independent dispersal events of Olea to the Madeira and Canary Islands. Molecular and morphological evidence led to recognition of two separate olive taxa in Macaronesia, to date included in ssp. cerasiformis. NJ analyses of the combined RAPD and ISSR data suggest that the colonization of the Canaries by O. europaea may have followed an east to west stepping-stone model. An interisland dispersal sequence can be recognized, starting from the continent to Fuerteventura, Gran Canaria, Tenerife, La Gomera, and finally La Palma. High dispersal activity of the lipid-rich Olea fruits by birds in the Mediterranean region is congruent with multiple dispersal of olives to Macaronesia and successive colonization of the archipelagos. The observation of strong genetic isolation between populations of different islands of the Canary Islands suggests, however, that subsequent interisland dispersal and establishment has been very rare or may not have occurred at all.  相似文献

Olive plantation systems occur in a wide variety throughout the Mediterranean, especially in sloping and mountainous areas. Recent drivers of change, including the widespread introduction of mechanisation, increased use of (chemical) inputs and (drip)irrigation have considerably added to this diversity. The various systems have very different resource use patterns and environmental and social performances. This article attempts to grasp these differences and to link systems characteristics to options for natural resources management in the spirit of contemporary agricultural policies that seek to promote a more sustainable agriculture. Cluster analysis was employed to classify 28 olive plantation systems distinguished by regional typologies developed for six study areas: Trás‐os‐Montes (Portugal), Córdoba and Granada/Jaén (both in Spain), Haffouz (Tunisia), Basilicata/Salerno (Italy) and West‐Crete (Greece). Six types of olive plantation systems resulted: (a) very extensive, (b) traditional extensive, (c) semi‐intensive low input, (d) semi‐intensive high input, (b) intensive and (e) organic. Natural resources management options to address soil erosion, low biodiversity, wildfire risk and excessive water use are explored for each of these systems. In the discussion, it becomes evident that an important quality for a typology lies in its capacity to differentiate likely future development pathways. If options are known, policy‐makers can make choices as to what the desired pathway is and what instruments to design to facilitate it.  相似文献

The olive tree (Olea europaea L., Oleaceae) is one of the most important fruit trees in Mediterranean basin and has been associated with numerous biological assets. These effects have been mainly attributed to certain phenolic compounds found in fruits, olive oil and by-products of olive oil production. However, other Olea organs such as stems, roots and drupe stones have received little attention leading to limited knowledge about their phytochemical content. Thus, the main goal of the current study was the investigation of the chemical composition of diverse organs from two O. europaea varieties (i.e. Koroneiki and Chetoui) using combinations of modern analytical techniques. A fast UHPLC-DAD-FLD method was developed and applied for the profiling of different extracts of O. europaea organs as well as for the quantification of oleuropein. In addition, a dereplication strategy was developed using an Orbitrap platform (UHPLC-ESI-HRMS/MS) aiming to further characterization of the contained secondary metabolites. In total, 86 molecules were identified including compounds described for the first time in O. europaea such as coumarins. Some compounds were found to be organ specific such as nuzhenide derivatives in stone, flavonoids in leaves and oleuropein which was mainly found in Olea roots, in both varieties. Overall, it is noticeable that except olive oil, diverse organs of olive tree might comprise an alternative and valuable source of biologically active compounds.  相似文献

Our study reports the triglyceride (triacylglycerol, TAG) composition of five new Tunisian virgin olive oil cultivars obtained through controlled crossings of the cultivar (cv.) Chemlali Sfax. These cultivars have been selected among a progeny of 500 olive descendants, based on an evaluation of the fatty‐acid (FA) composition of their oils. Among these samples, two were derived from the crossing with the cv. Sigoise as pollinator (SM634) or pollen acceptor (SM1110) and the others from the crossing with the cv. Meski as pollen acceptor (SM513, SM514, and SM517). The five descendants were characterized by a good fat value, a balanced FA composition, and a high content of triolein, varying between 26.9 (SM514) and 45.46% (SM1110). They had an improved FA composition as compared to that of the cv. Chemlali Sfax and their fruits were slightly bigger. The principal component analysis suggested that the TAG variables were more suitable than the total FAs for an optimum classification of the cultivar samples analyzed. The cultivars obtained through the crossing with the cv. Sigoise (as pollen acceptor or pollinator) had a more favorable composition of FAs and TAG than those obtained through the crossing with the cv. Meski, which indicated that genetic factors had the most important influence on the quality of the virgin olive oils.  相似文献

Metabolism of ammonia (NH3) and hydroxylamine (NH2OH) by wild-type and a nitrite reductase (nirK) deficient mutant of Nitrosomonas europaea was investigated to clarify the role of NirK in the NH3 oxidation pathway. NirK-deficient N. europaea grew more slowly, consumed less NH3, had a lower rate of nitrite (NO2 ) production, and a significantly higher rate of nitrous oxide (N2O) production than the wild-type when incubated with NH3 under high O2 tension. In incubations with NH3 under low O2 tension, NirK-deficient N. europaea grew more slowly, but had only modest differences in NH3 oxidation and product formation rates relative to the wild-type. In contrast, the nirK mutant oxidized NH2OH to NO2 at consistently slower rates than the wild-type, especially under low O2 tension, and lost a significant pool of NH2OH–N to products other than NO2 and N2O. The rate of N2O production by the nirK mutant was ca. three times higher than the wild-type during hydrazine-dependent NO2 reduction under both high and low O2 tension. Together, the results indicate that NirK activity supports growth of N. europaea by supporting the oxidation of NH3 to NO2 via NH2OH, and stimulation of hydrazine-dependent NO2 reduction by NirK-deficient N. europaea indicated the presence of an alternative, enzymatic pathway for N2O production.  相似文献

AIMS: To obtain ammonia-oxidizing bacterial (AOB) strains inhabiting low dissolved oxygen (DO) environments and to characterize them to better understand their function and ecology. METHODS AND RESULTS: Using a serial dilution method, two AOB strains (ML1 and NL7) were isolated from chemostat reactors operated with low DO concentrations (0.12-0.24 mg l(-1)). Phylogenetically, strains ML1 and NL7 are affiliated to AOB within the Nitrosomonas europaea and Nitrosomonas oligotropha lineages, respectively. Kinetically, strain ML1 had high affinity for oxygen (0.24 +/- 0.13 mg l(-1)) and low affinity for ammonia (1.62 +/- 0.97 mg N l(-1)), while strain NL7 had high affinity for ammonia (0.48 +/- 0.35 mg l(-1)), but a surprisingly low affinity for oxygen (1.22 +/- 0.43 mg l(-1)). A co-culture experiment was used to iteratively estimate decay constants for both strains. CONCLUSIONS: The results indicated that AOB without high affinity for oxygen may have other mechanisms to persist in low DO environments, with high affinity for ammonia being important. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides a method to determine AOB growth kinetic parameters without assuming or neglecting decay constant. And, this is the first report on oxygen affinity constant of a N. oligotropha strain.  相似文献

With the aim to select new olive cultivars with superior physical and chemical properties than the cultivar Chemlali Sfax, the present study focused on the comparison of the chemical composition and the sensory profile of the virgin olive oils (VOOs) of two wild olive trees (Oleasters K and M) with those of VOOs obtained from Chemlali Sfax and Neb Jmel olive cultivars, all growing in the coastal region of Tunisia. Despite the variability in the chemical composition (fatty acids, pigments, and phenolic and volatile compounds) and the organoleptic profile of the VOOs of the oleasters and the cultivars, the quality indices (free fatty acids, peroxide value, and spectrophotometric indices K232 and K270) as well as the fatty acid composition of all VOOs studied met the commercial standards. Both the α-tocopherol and phenol contents varied between the genotypes. The Neb Jmel and Oleaster K VOOs had more than two times higher total phenol levels than the Chemlali Sfax and Oleaster M VOOs. Also the contents of volatile compounds differed between the olive oils studied. Chemlali Sfax and Oleaster K oils were more abundant in aldehydes, whereas Oleaster M VOO had higher contents of alcohols. These results were confirmed by a sensorial analysis showing that the later oil was deprived for consumption despite its abundance in α-tocopherol. In conclusion, the oleasters studied revealed to be interesting, since they produced oils with good quality characteristics in terms of minor compounds (phenols and volatiles) compared to the Chemlali Sfax cultivar.  相似文献

Samples from three hot springs (Alla, Seya and Garga) located in the northeastern part of Baikal rift zone (Buryat Republic, Russia) were screened for the presence of thermophilic nitrifying bacteria. Enrichment cultures were obtained solely from the Garga spring characterized by slightly alkaline water (pH 7.9) and an outlet temperature of 75 degrees C. The enrichment cultures of the ammonia- and nitrite oxidizers grew at temperature ranges of 27-55 and 40-60 degrees C, respectively. The temperature optimum was approximately 50 degrees C for both groups and thus they can be designated as moderate thermophiles. Ammonia oxidizers were identified with classical and immunological techniques. Representatives of the genus Nitrosomonas and Nitrosospira-like bacteria with characteristic vibroid morphology were detected. The latter were characterized by an enlarged periplasmic space, which has not been previously observed in ammonia oxidizers. Electron microscopy, denaturing gradient gel electrophoresis analyses and partial 16S rRNA gene sequencing provided evidence that the nitrite oxidizers were members of the genus Nitrospira.  相似文献

The contents of Na+, K+, water, and dry matter were measured in leaves and roots of euhalophytes Salicornia europaea L. and Climacoptera lanata (Pall.) Botsch featuring succulent and xeromorphic cell structures, respectively, as well as in saltbush Atriplex micrantha C.A. Mey, a halophyte having bladder-like salt glands on their leaves. All three species were able to accumulate Na+ in their tissues. The Na+ content in organs increased with elevation of NaCl concentration in the substrate, the concentrations of Na+ being higher in leaves than in roots. When these halophytes were grown on a NaCl-free substrate, a trend toward K+ accumulation was observed and was better pronounced in leaves than in roots. Particularly high K+ concentrations were accumulated in Salicornia leaves. There were no principal differences in the partitioning of Na+ and K+ between organs of three halophyte species representing different ecological groups. At all substrate concentrations of NaCl, the total content of Na+ and K+ in leaves was higher than in roots. This distribution pattern persisted in Atriplex possessing salt glands, as well as in euhalophytes Salicornia and Climacoptera. The physiological significance of such universal pattern of ion accumulation and distribution among organs in halophytes is related to the necessity of water absorption by roots, its transport to shoots, and maintenance of sufficient cell water content in all organs under high soil salinity.  相似文献
