利用途径分析方法对黄色短杆菌(Brevibacterium flavum)TC-21 生产L-异亮氨酸的途径进行了分析,确定了黄色短杆菌TC-21生产L-异亮氨酸的最佳途径的通量分布,根据途径分析的结果,TCA循环的代谢流量对L-异亮氨酸产量有明显影响,而TCA循环与发酵过程中的溶氧密切相关,因此可以通过控制溶氧来提高L-异亮氨酸产量。在发酵过程的不同阶段,根据菌体生长和产酸的需求,改变TCA代谢流量,可以有效提高产酸率。实验证明,通过溶氧分阶段控制发酵生产L-异亮氨酸,比溶氧恒定控制方式发酵产率提高了15.77%。实验结果说明,用途径分析的结果指导发酵过程中的溶氧可以大幅度提高L-异亮氨酸的产量。  相似文献

Question: How are the effects of mineral soil properties on understory plant species richness propagated through a network of processes involving the forest overstory, soil organic matter, soil nitrogen, and understory plant abundance? Location: North‐central Arizona, USA. Methods: We sampled 75 0.05‐ha plots across a broad soil gradient in a Pinus ponderosa (ponderosa pine) forest ecosystem. We evaluated multivariate models of plant species richness using structural equation modeling. Results: Richness was highest at intermediate levels of understory plant cover, suggesting that both colonization success and competitive exclusion can limit richness in this system. We did not detect a reciprocal positive effect of richness on plant cover. Richness was strongly related to soil nitrogen in the model, with evidence for both a direct negative effect and an indirect non‐linear relationship mediated through understory plant cover. Soil organic matter appeared to have a positive influence on understory richness that was independent of soil nitrogen. Richness was lowest where the forest overstory was densest, which can be explained through indirect effects on soil organic matter, soil nitrogen and understory cover. Finally, model results suggest a variety of direct and indirect processes whereby mineral soil properties can influence richness. Conclusions: Understory plant species richness and plant cover in P. ponderosa forests appear to be significantly influenced by soil organic matter and nitrogen, which are, in turn, related to overstory density and composition and mineral soil properties. Thus, soil properties can impose direct and indirect constraints on local species diversity in ponderosa pine forests.  相似文献

The plant functional group approach has the potential to clarify ecological patterns and is of particular importance in simplifying the application of ecological models in high biodiversity ecosystems. Six functional groups (pasture grass, pasture sapling, top-canopy tree, top-canopy liana, mid canopy tree, and understory tree) were established a priori based on ecosystem inhabited, life form, and position within the forest canopy profile on eastern Amazonian region. Ecophysiological traits related to photosynthetic gas exchange were then used to characterize such groups. The ecophysiological traits evaluated showed considerable variations among groups. The pasture grass functional group (a C4 photosynthetic pathway species) showed high instantaneous water use efficiency (A max/g s@A max), high photosynthetic nitrogen use efficiency (A max/N area), and high ratio of A max to dark respiration (A max/R d). Among the species with the C3 photosynthetic pathway, the top-canopy liana group showed the highest mean of A max/g s@A max, statistically distinct from the lowest average presented by the understory tree group. Furthermore, the pasture sapling group showed the lowest average of A max/R d, statistically distinct from the high average observed for the understory tree group. Welch-ANOVAs followed by Games–Howell post hoc tests applied to ecophysiological traits produced reasonable distinctions among functional groups, although no significant distinction was detected between the groups top-canopy tree and pasture sapling. Species distribution within the functional groups was accurately reproduced by discriminant analyses based on species averages of ecophysiological traits. The present work convincingly shows that the functional groups identified have distinct ecophysiological characteristics, with the potential to respond differently to environmental factors. Such information is of great importance in modeling efforts that evaluate the effects of dynamic changes in tropical plant communities over ecosystem primary productivity.  相似文献

We examined how water and nitrogen addition and water–nitrogen interactions affect root and shoot competition intensity and competition–productivity relationships in a native rough fescue grassland in central Alberta, Canada. Water and nitrogen were added in a factorial design to plots and root exclusion tubes and netting were used to isolate root and shoot competition on two focal species (Artemisia frigida and Chenopodium leptophyllum). Both water and nitrogen were limiting to plant growth, and focal plant survival rates increased with nitrogen but not water addition. Relative allocation to root biomass increased with water addition. Competition was almost entirely belowground, with focal plants larger when released from root but not shoot competition. There were no significant relationships between productivity and root, shoot, or total competition intensity, likely because in this system shoot biomass was too low to cause strong shoot competition and root biomass was above the levels at which root competition saturates. Water addition had few effects on the intensity of root competition suggesting that root competition intensity is invariant along soil moisture gradients. Contrary to general expectation, the strength of root competition increased with nitrogen addition demonstrating that the relationship between root competition intensity and nitrogen is more complex than a simple monotonic decline as nitrogen increases. Finally, there were few interactions between nitrogen and water affecting competition. Together these results indicate that the mechanisms of competition for water and nitrogen likely differ.  相似文献

The spatial patterns of photosynthetic characteristics and leaf physical traits of 171 plants belonging to nine life-forms or functional groups (trees, shrubs, herbs, evergreen trees, deciduous trees, C3 and C4 herbaceous plants, leguminous and non-leguminous species) and their relationships with environmental factors in seven sites, Yangling, Yongshou, Tongchuan, Fuxian, Ansai, Mizhi and Shenmu, ranging from south to north in the Loess Plateau of China were studied. The results showed that the leaf light-saturated photosynthetic rate (Pmax), photosynthetic nitrogen use efficiency (PNUE), chlorophyll content (Chl), and leaf mass per area (LMA) of all the plants in the Loess Plateau varied significantly among three life-form groups, i.e., trees, shrubs and herbs, and two groups, i.e., evergreen trees and deciduous trees, but leaf nitrogen content differed little among different life-form groups. For the 171 plants in the Loess Plateau, leaf Pmax was positively correlated with PNUE. The leaf nitrogen content per unit area (Narea) was positively correlated but Chl was negatively correlated with the LMA. When controlling the LMA, the Narea was positively correlated with the Chl (partial r = 0.20, P < 0.05). With regard to relationships between photosynthetic characteristics and leaf physical traits, the Pmax was positively correlated with N area, while the PNUE was positively correlated with the Chl and negatively correlated with the Narea and LMA. For all the species in the Loess Plateau, the PNUE was negatively correlated with the latitude and annual solar radiation (ASR), but positively correlated with the mean annual rainfall (MAR) and mean annual temperature (MAT). With regard to the leaf physical traits, the leaf Chl was negatively correlated with the latitude and ASR, but positively correlated with the MAR and MAT. However, the Narea and LMA were positively correlated with the latitude and ASR, but negatively correlated with the MAR and MAT. In general, leaf Narea and LMA increased, while PNUE and Chl decreased with increases in the latitude and ASR and decreases in MAR and MAT. Vegetative and chemical responses to simulated leaf browsing during the growth season, and their subsequent effect on herbivory, were studied on Combretum apiculatum Sonder (Combretaceae) in Botswana. Treatments (50% and 100% leaf and shoot apex removal) were performed just before the shoot growth curve levelled out, and responses recorded 3 months later, just before leaf fall. Compared to controls, defoliation treatments, removing apical dominance, reduced growth in tree height and increased shoot mortality, although the production of lateral shoots increased. At the end of the trial, there was no difference in total length of annual shoots between treatment groups. Significant refoliation occurred only after 100% defoliation. Refoliated leaves were smaller and the 100% defoliated trees had a lower final leaf biomass. Total leaf biomass production was, however, equal for all treatment groups. Refoliated leaves contained higher levels of N, lower levels of acid-detergent fibre (ADF) and total phenolics, and showed a trend towards lower levels of condensed tannins, compared to leaves on control trees. Such chemical changes may be due to either carbon stress or to younger physiological age of new leaves. In spite of the observed potential increase in food quality, we found no evidence of increased levels of insect or ungulate herbivory on refoliated leaves, which, at least for insect herbivory, may be explained by the reduction in temporal availability of leaves. We conclude that the single severe defoliation was not detrimental to C. apiculatum in the short-term, although the resource loss and induced compensatory growth may produce negative effects during subsequent growth seasons.  相似文献

Physiology, regulation and biochemical aspects of the nitrogen assimilation are well known in Prokarya or Eukarya but they are poorly described in Archaea domain. The haloarchaeon Haloferax mediterranei can use different nitrogen inorganic sources (NO3, NO2 or NH4+) for growth. Different approaches were considered to study the effect of NH4+ on nitrogen assimilation in Hfx. mediterranei cells grown in KNO3 medium. The NH4+ addition to KNO3 medium caused a decrease of assimilatory nitrate (Nas) and nitrite reductases (NiR) activities. Similar effects were observed when nitrate-growing cells were transferred to NH4+ media. Both activities increased when NH4+ was removed from culture, showing that the negative effect of NH4+ on this pathway is reversible. These results suggest that ammonium causes the inhibition of the assimilatory nitrate pathway, while nitrate exerts a positive effect. This pattern has been confirmed by RT-PCR. In the presence of both NO3 and NH4+, NH4+ was preferentially consumed, but NO3 uptake was not completely inhibited by NH4+ at prolonged time scale. The addition of MSX to NH4+ or NO3 cultures results in an increase of Nas and NiR activities, suggesting that NH4+ assimilation, rather than NH4+ per se, has a negative effect on assimilatory nitrate reduction in Hfx. mediterranei. The Dynamic Energy Budget (DEB) theory quantifies the metabolic organisation of organisms on the basis of mechanistically inspired assumptions. We here sketch a scenario for how its various modules, such as maintenance, storage dynamics, development, differentiation and life stages could have evolved since the beginning of life. We argue that the combination of homeostasis and maintenance induced the development of reserves and that subsequent increases in the maintenance costs came with increases of the reserve capacity. Life evolved from a multiple reserves - single structure system (prokaryotes, many protoctists) to systems with multiple reserves and two structures (plants) or single reserve and single structure (animals). This had profound consequences for the possible effects of temperature on rates. We present an alternative explanation for what became known as the down-regulation of maintenance at high growth rates in microorganisms; the density of the limiting reserve increases with the growth rate, and reserves do not require maintenance while structure-specific maintenance costs are independent of the growth rate. This is also the mechanism behind the variation of the respiration rate with body size among species. The DEB theory specifies reserve dynamics on the basis of the requirements of weak homeostasis and partitionability. We here present a new and simple mechanism for this dynamics which accounts for the rejection of mobilised reserve by busy maintenance/growth machinery. This module, like quite a few other modules of DEB theory, uses the theory of Synthesising Units; we review recent progress in this field. The plasticity of membranes that evolved in early eukaryotes is a major step forward in metabolic evolution; we discuss quantitative aspects of the efficiency of phagocytosis relative to the excretion of digestive enzymes to illustrate its importance. Some processes of adaptation and gene expression can be understood in terms of allocation linked to the relative workload of metabolic modules in (unicellular) prokaryotes and organs in (multicellular) eukaryotes. We argue that the evolution of demand systems can only be understood in the light of that of supply systems. We illustrate some important points with data from the literature.  相似文献

Increasingly, organizations are working to reduce the environmental footprint of their supply chains. The use of environmentally preferable purchasing criteria is one strategy organizations use to address this goal. However, evaluating the environmental performance of these criteria (e.g., recycled content, biodegradable, renewable, and so on) has remained elusive. Life cycle assessment (LCA) can measure the impact reduction potential of sourcing strategies. However, full process‐based LCAs are time‐consuming and costly across multiple criteria of thousands of products and inputs purchased in an organizational setting. A streamlined “hotspot” methodology is presented using a combination of environmentally extended economic input‐output (EEIO) approaches and extant literature to identify hotspots in which to constrain a parameterized process‐based LCA. A case study of breakfast cereal manufacturing is developed to (1) assess the efficiencies associated with the hotspotting approach and (2) demonstrate its applicability in generating comparable decision signals of environmentally preferable sourcing criteria for procurement and supply‐chain managers along the dimensions of global warming potential and water use.  相似文献
