The fate of15N labeled nitrogen applied to mature citrus trees 总被引:1,自引:0,他引:1
Summary The efficiency and balance of nitrogen from one year's application was studied in a long-term fertigation experiment. Enriched
nitrogen fertilizer, K15NO3, was applied to a 22-year-old Shamouti orange tree with a history of high N applications (N3) and to an N-starved tree (N1). The distribution of N in the different parts of the trees and in the soil was determined after the experimental trees were
excavated.
Similar total recovery of the labeled fertilizer N was found in the trees and soil in both treatments (N1−61.7% N3−56%). However, the distribution between tree and soil was different. The amount of recovered residual fertilizer in the soil
was much larger in the N3 treatment than in N1. The highest percentage of fertilizer N was found in the new organs,i.e. fruits, twigs and leaves. The roots and branches took up only 6–14% from the labeled fertilizer.
Only 20.9% of the leaf N and 23.4% of the fruit N in the N3 tree originated in the labeled fertilizer, indicating translocation of N from older parts of the tree to new growth.
Evidence was found of storage of N in the wooded branches, while the roots contained a surprisingly small part of labeled
fertilizer.

Cells of obligate methylotrophic Gram-negative bacterium Methylobacillus flagellatum KT which can only grow on methanol and methylamine media possess three different carriers mediating uptake of methylamin depending on growth conditions. All three uptake systems are energy-dependent, the methylamine uptake was inhibited by oxidative phosphorylation uncoupler and respiratory inhibitors. The first active transport system for methanol in the cells of obligate methylotroph was also demonstrated. Background
Insertion sequences (ISs) are approximately 1 kbp long “jumping” genes found in prokaryotes. ISs encode the protein Transposase, which facilitates the excision and reinsertion of ISs in genomes, making these sequences a type of class I (“cut-and-paste”) Mobile Genetic Elements. ISs are proposed to be involved in the reductive evolution of symbiotic prokaryotes. Our previous sequencing of the genome of the cyanobacterium ‘Nostoc azollae’ 0708, living in a tight perpetual symbiotic association with a plant (the water fern Azolla), revealed the presence of an eroding genome, with a high number of insertion sequences (ISs) together with an unprecedented large proportion of pseudogenes. To investigate the role of ISs in the reductive evolution of ‘Nostoc azollae’ 0708, and potentially in the formation of pseudogenes, a bioinformatic investigation of the IS identities and positions in 47 cyanobacterial genomes was conducted. To widen the scope, the IS contents were analysed qualitatively and quantitatively in 20 other genomes representing both free-living and symbiotic bacteria.Results
Insertion Sequences were not randomly distributed in the bacterial genomes and were found to transpose short distances from their original location (“local hopping”) and pseudogenes were enriched in the vicinity of IS elements. In general, symbiotic organisms showed higher densities of IS elements and pseudogenes than non-symbiotic bacteria. A total of 1108 distinct repeated sequences over 500 bp were identified in the 67 genomes investigated. In the genome of ‘Nostoc azollae’ 0708, IS elements were apparent at 970 locations (14.3%), with 428 being full-length. Morphologically complex cyanobacteria with large genomes showed higher frequencies of IS elements, irrespective of life style.Conclusions
The apparent co-location of IS elements and pseudogenes found in prokaryotic genomes implies earlier IS transpositions into genes. As transpositions tend to be local rather than genome wide this likely explains the proximity between IS elements and pseudogenes. These findings suggest that ISs facilitate the reductive evolution in for instance in the symbiotic cyanobacterium ‘Nostoc azollae’ 0708 and in other obligate prokaryotic symbionts.Electronic supplementary material

Net carbon flux from agriculture: Carbon emissions, carbon sequestration, crop yield, and land-use change 总被引:10,自引:0,他引:10
There is a potential to sequester carbon in soil by changing agricultural management practices. These changes in agricultural management can also result in changes in fossil-fuel use, agricultural inputs, and the carbon emissions associated with fossil fuels and other inputs. Management practices that alter crop yields and land productivity can affect the amount of land used for crop production with further significant implications for both emissions and sequestration potential. Data from a 20-year agricultural experiment were used to analyze carbon sequestration, carbon emissions, crop yield, and land-use change and to estimate the impact that carbon sequestration strategies might have on the net flux of carbon to the atmosphere. Results indicate that if changes in management result in decreased crop yields, the net carbon flux can be greater under the new system, assuming that crop demand remains the same and additional lands are brought into production. Conversely, if increasing crop yields lead to land abandonment, the overall carbon savings from changes in management will be greater than when soil carbon sequestration alone is considered. 相似文献

Formate metabolism supported nitrogen-fixation activity in free-living cultures of Rhizobium japonicum. However, formate0dependent nitrogense activity was observed only in the presence of carbon sources such as glutamate, ribose or aspartate which by themselves were unable to support nitrogenase activity. Formate-dependent nitrogenase activity was not detected in the presence of carbon sources such as malate, gluconate or glycerol which by themselves supported nitrogenase activity. A mutant strain of R. japonicum was isolated that was unable to utilise formate and was shown to lack formate dehydrogenase activity. This mutant strain exhibited no formate-dependent nitrogenase activity. Both the wild-type and mutant strains nodulated soybean plants effectively and there were no significant differences in the plant dry weight or total nitrogen content of the respective plants. Furthermore pea bacteroids lacked formate dehydrogenase activity and exogenously added formate had no stimulatory effect on the endogenous oxygen uptake rate. 在内蒙古冷蒿小禾草草原1989年围封禁牧地,研究了天然羊尿尿斑土壤化学特性的变化规律,结果表明,土壤pH值于施尿后第2天达到最大值,以后呈下降趋势,58天后pH值基本稳定在6.5左右且低于对照区水平,施尿区土壤可溶性有机碳含量的变化趋势与pH值的变化基本一致,两者存在显著的正相关关系(P<0.01).施尿后土壤总可溶性磷含量显著提高(P<0.05),第2天达到最大值(35.1mg·L^-1)。总可溶性磷各组分钼酸反应磷、可溶性有机磷和可溶性缩合磷百分比均存在两个明显的变化阶段,钼酸反应磷和可溶性有机磷与可溶性缩合磷存在显著的负相关关系(P<0.05).施尿区土壤NH4^+-N在第2天达到最大值。以后呈下降趋势;NO3-N含量14天后开始显著增加,并于第21天达到最大值,其变化呈现明显的“双峰型”特征,并较NH4^+-N存在显著的时滞现象;施尿后土壤可溶性有机氮含量显著增加,并表现为“双峰型”变化特征。 相似文献

Variation in the stable N isotope ratio (δ15N) of plants and soils often reflects the influence of environment on the N cycle. We measured leaf δ15N and N concentration ([N]) on all individuals of Prosopis glandulosa (deciduous tree legume), Condalia hookeri (evergreen shrub), and Zanthoxylum fagara (evergreen shrub) present within a belt transect 308 m long × 12 m wide in a subtropical savanna ecosystem in southern Texas,
USA in April and August 2005. Soil texture, gravimetric water content (GWC), total N and δ15N were also measured along the transect. At the landscape scale, leaf δ15N was negatively related to elevation for all the three species along this topoedaphic sequence. Changes in soil δ15N, total N, and GWC appeared to contribute to this spatial pattern of leaf δ15N. In lower portions of the landscape, greater soil N availability and GWC are associated with relatively high rates of both
N mineralization and nitrification. Both soil δ15N and leaf [N] were positively correlated with leaf δ15N of non-N2 fixing plants. Leaf δ15N of P. glandulosa, an N2-fixing legume, did not correlate with leaf [N]; the δ15N of P. glandulosa’s leaves were closer to atmospheric N2 and significantly lower than those of C. hookeri and Z. fagara. Additionally, at smaller spatial scales, a proximity index (which reflected the density and distance of surrounding P. glandulosa trees) was negatively correlated with leaf δ15N of C. hookeri and Z. fagara, indicating the N2-fixing P. glandulosa may be important to the N nutrition of nearby non-N2-fixing species. Our results indicate plant 15N natural abundance can reflect the extent of N retention and help us better understand N dynamics and plant-soil interactions

Nitrogen assimilation in Rhodobacter capsulatus has been shown to proceed via the coupled action of glutamine synthetase (GS) and glutamate synthase (GOGAT) with no measurable glutamate dehydrogenase (GDH) present. We have recently isolated a novel class of mutants of R. capsulatus strain B100 that lacks a detectable GOGAT activity but is able to grow at wild type rates under nitrogen-fixing conditions. -supported growth in the mutants was normal under anaerobic/photosynthetic conditions, the growth rate was decreased under aerobic conditions. Ammonium and methylammonium uptake experiments indicated that there was a clear difference in the ammonium assimilatory capabilities in these mutants under aerobic versus anaerobic growth. Summary Millet plants (Pennisetum glaucum) were grown at three levels of nitrogen fertilization with and without an inoculum of live nitrogen-fixing Azospirillum cells. The highest average rate of nitrogen fixation as estimated from acetylene reduction by excised preincubated roots was only 23g N2 fixed per ha per day and occurred after treatment with low levels of nitrogen amendment. The average rates of acetylene reduction for intact plants at all treatments were also low. The lack of significant nitrogen fixation due to an Azospirillum-millet association in this study was substantiated by plant dry weight analysis, and determination of the nitrogen content of plants, pot leachate, and soil. There was significant correlation between the total nitrogen content of the plants per pot at the termination of the experiment and the amount of nitrogen fertilizer added initially, but there was no effect of inoculum on final total nitrogen content. 相似文献
