Abstract Nitrate reduction to ammonia by marine Vibrio species was studied in batch and continuous culture. In pH-controlled batch cultures (pH 7.4; 50 mM glucose, 20 mM KNO3), the nitrate consumed accumulated to more than 90% as nitrite. Under these conditions, the nitrite reductase (NO2→ NH3) was severely repressed. In pH-controlled continuous cultures of V. alginolyticus with glucose or glycerol as substrates ( D = 0.045 h−1) and limiting N-source (nitrate or nitrite), nitrite reductase was significantly derepressed with cellular activities in the range of 0.7–1.2 μmol min−1 (mg protein)−1. The enzyme was purified close to electrophoretic homogeneity with catalytic activity concentrations of about 1800 nkat/mg protein. It catalyzed the reduction of nitrite to ammonia with dithionite-reduced viologen dyes or flavins as electron donors, had an M r of about 50 000 (determined by gel filtration) and contained c-type heme groups (probably 4–6 per molecule).  相似文献

The assimilation of nitrate under dark-N2 and dark-O2 conditions in Zea mays leaf tissue was investigated using colourimetric and 15N techniques for the determination of organic and inorganic nitrogen. Studies using 15N indicated that nitrate was assimilated under dark conditions. However, the rate of nitrate assimilation in the dark was only 28% of the rate under non-saturating light conditions. No nitrite accumulated under dark aerobiosis, even though nitrate reduction occurred under these conditions. The pattern of nitrite accumulation in leaf tissue in response to dark-N2 conditions consisted of three phases: an initial lag phase, followed by a period of rapid nitrite accumulation and finally a phase during which the rate of nitrite accumulation declined. After a 1-h period of dark-anaerobiosis, both nitrate reduction and nitrite accumulation declined considerably. However, when O2 was supplied, nitrate reduction was stimulated and the accumulated nitrite was rapidly reduced. Anaerobic conditions stimulated nitrate reduction in leaf tissue after a period of dark-aerobic pretreatment.  相似文献

The rate of photosynthetic nitrate utilization in Anacystis nidulans is strongly influenced by the availability of carbon dioxide. This dependence can be relieved by inhibiting the metabolism of the ammonium derived from nitrate reduction. Nitrate uptake seems to be modulated through a sensitive regulatory system integrating the photosynthetic metabolism of carbon and nitrogen, with CO2 fixation products antagonizing the inhibitory effect of ammonium derivatives.  相似文献

Summary Two grass species, the annual Bromus sterilis and the perennial Bromus erectus, were grown from seeds for 28 days in a hydroponic culture system at 1 and 100 M NO3 - in the nutrient solution. At 100 M NO3 -, the relative growth rate (RGR) of the perennial was 30% lower than that of the annual. This was only the consequence of the higher specific mass of its leaves, since its leaf mass ratio was higher than that of the annual and the unit leaf rates (ULR), calculated on an area basis, were similar for the two species. At 1 M, the RGR of the annual was 50% lower than at 100 M, while that of the perennial was not significantly lower. This was due mainly to a lower ULR for the annual. while for the perennial ULR was the same in both treatments. These differences between the two species were all the more striking in that the differences in total nitrogen concentrations and nitrate reductase activities between the two treatments were very similar for both species. These different responses together with differences in the nitrogen productivity of the two species suggest that the level of nutrient availability may play an important role in the distribution of these Bromus species in natural habitats. Summary Effects of fire and forest harvesting on inorganic-N in the soil, on net N-mineralization, and on the leaching of NO inf3 sup- -N and metallic cations were measured in forests of Eucalyptus regnans following a severe wildfire in 1983. E. regnans regenerates profusely by seed after fire, and this study compared unburnt forest with forests burnt at varying intensities (surface fire and crown fire), and with logged and burnt forest (slash fire). Total inorganic-N in soil (0–5 cm) increased with increasing fire intensity to a maximum of 158 g g-1 in the slash fire plot (compared with 51 g g-1 in the unburnt forest) over the first 205 days after fire. Total inorganic-N returned to a concentration equal to that in the unburnt forest after 485 days at the slash fire plot, and after only 205 days at the surface fire plot. Studies of net mineralization in situ and of NO inf3 sup- -N in soil solution support the hypothesis that inorganic-N was immobilized in all of the burnt forests; microbial immobilization after fire is identified as a key process in N-conservation, limiting the substrate available for nitrification and thereby limiting the loss of N from the system by leaching. The concentrations of NO inf3 sup- -N and metallic cations in soil solution increased with increasing fire intensity. For the first 318 days after the fire, [NO inf3 sup- -N] in soil solution at 10 cm averaged 0.6 g ml-1 in the unburnt forest, 9.7 mg l-1 in the surface fire plot, 26 mg l-1 in the crown fire plot, and 70 mg l-1 in the slash fire plot. The concentration of metallic cations in soil solution was significantly correlated with [NO inf3 sup- -N], the observed order of mobility being Ca2+>Mg2+>K+>Na+. Processes which limit the production and persistence of NO inf3 sup- -N in soil solution following disturbance will significantly reduce nutrient losses or redistribution.  相似文献

The loading of amino acids and nitrate into the xylem was investigated by collection and analysis of root-pressure exudate from the cut hypocotyl stumps of seedlings of Ricinus communis L. Glutamine was found to be the dominant amino acid in the exudate and also to be the amino acid which is transferred to the xylem most rapidly and accumulated to the greatest extent. The comparison between uptake and xylem loading showed significant differences in specificity between these two transport reactions, indicating a different set of transport systems. Nitrate is transferred to the xylem at a higher relative rate than any amino acid despite the great nitrate-storage capacity of the root system. Thus the supply of nitrate to Ricinus plants leads to enhanced nitrogen allocation to the shoots.  相似文献

Nitrogen-starved sunflower plants (Helianthus annuus L. cv. Peredovic) cannot absorb NO 3 or NO 2 upon initial exposure to these anions. Ability of the plants to take up NO 3 and NO 2 at high rates from the beginning was induced by a pretreatment with NO 3 . Nitrite also acted as inducer of the NO 2 -uptake system. The presence of cycloheximide during NO 3 -pretreatment prevented the subsequent uptake of NO 3 and NO 2 , indicating that both uptake systems are synthesized de novo when plants are exposed to NO 3 . Cycloheximide also suppressed nitrate-reductase (EC 1.6.6.1) and nitrite-reductase (EC 1.7.7.1) activities in the roots. The sulfhydryl-group reagent N-ethylmaleimide greatly inhibited the uptake of NO 3 and NO 2 . Likewise, N-ethylmaleimide promoted in vivo the inactivation of nitrate reductase without affecting nitrite-reductase activity. Rates of NO 3 and NO 2 uptake as a function of external anion concentration exhibited saturation kinetics. The calculated Km values for NO 3 and NO 2 uptake were 45 and 23 M, respectively. Rates of NO 3 uptake were four to six times higher than NO 3 -reduction rates in roots. In contrast, NO 2 -uptake rates, found to be very similar to NO 3 -uptake rates, were much lower (about 30 times) than NO 2 -reduction rates. Removal of oxygen from the external solution drastically suppressed NO 3 and NO 2 uptake without affecting their reduction. Uptake and reduction were also differentially affected by pH. The pericarp of the dormant sugarbeet fruit acts as a storage reservoir for nitrate, ammonium and -amino-N. These N-reserves enable an autonomous development of the seedling for 8–10 d after imbibition. The nitrate content of the seed (1% of the whole fruit) probably induces nitrate-reductase activity in the embryo enclosed in the pericarp. Nitrate that leaks out of the pericarp is reabsorbed by the emerging radicle. Seedlings germinated from seeds (pericarp was removed) without external N-supply are able to take up nitrate immediately upon exposure via a low-capacity uptake system (vmax = 0.8 mol NO 3 - ·(g root FW)–1·h–1; Ks = 0.12 mM). We assume that this uptake system is induced by the seed nitrate (10 nmol/seed) during germination. Induction of a high-capacity nitrate-uptake system (vmax = 3.4 mol NO 3 - ·(g root FW)–1·h–1; Ks = 0.08 mM) by externally supplied nitrate occurs after a 20-min lag and requires protein synthesis. Seedlings germinated from whole fruits absorb nitrate via a highcapacity uptake mechanism induced by the pericarp nitrate (748 nmol/pericarp) during germination. The uptake rates of the high-capacity system depend only on the actual nitrate concentration of the uptake medium and not on prior nitrate pretreatments. Nitrate deprivation results in a decline of the nitrate-uptake capacity (t1/2 of vmax = 5 d) probably caused by the decay of carrier molecules. A series of amphiphilic polymethylenecarboxymaleimides has been synthesized for use as sulfhydryl reagents applicable to membrane proteins. Physical properties of the compounds which are relevant to their proposed mode of action have been determined. By comparing rates of reaction in aqueous and aprotic solvents, the compounds have been shown to react exclusively with the thiolate ion. The effects of the reagents on three membrane-associated proteins are reported, and in two cases a comparative study has been made of the effects on the proteins in the absence of membranes. A mechanism is proposed whereby the reagents are anchored at the lipid/water interface by the negatively charged carboxyl group, thus siting the reactive maleimide in a plane whose depth is defined by the length of the reagent. Supporting evidence for this model is provided by the inability of the reagents to traverse membranes, and variation of their inhibitory potency with chain length when the proteins are embedded in the membrane, but not when extracted into solution. As examples of general use of the reagents to probe sulfhydryl groups in membrane proteins, the reagents have been used to (a) determine the depths in the membrane at which two populations of sulfhydryl groups occur in the mitochondrial phosphate transporter; (b) locate a single sulfhydryl associated with the active site ofD--hydroxybutyrate dehydrogenase in the inner mitochondrial membrane; (c) examine sulfhydryl groups in theD-3-glyceraldehyde phosphate dehydrogenase associated with the human red blood cell membrane.  相似文献

The decrease in the electron flow of the aerobic respiratory chain of the bacterium Paracoccus denitrificans, owing to either the drop in the saturation of terminal oxidases by oxygen or to the inhibition of the rate of respiration by azide or nitrite, resulted in the synthesis of dissimilatory nitrate reductase and nitrite reductase. The dependence of the resulting activities of the two enzymes (after a three-hour adaptation) on the initial value of the parameter Vmax/kLa (oxidase activity of the volume unit of the culture divided by the volumetric oxygen transfer coefficient) or on the concentrations of the inhibitors had a similar form, characterized by the appearance of a maximum. The increasing parts of the obtained curves reflect the synthesis of enzymes, probably initiated by the increase in the intracellular degree of reduction, the subsequent drop being evidently in connection with the lack of metabolic energy for biosynthesis. The possible mechanisms of the effect of nitrogenous terminal acceptors (NO-3 and NO-2) on the formation of the denitrification pathway are discussed.  相似文献
