Invasive species pose a major threat to aquatic ecosystems. Their impact can be particularly severe in tropical regions, like those in northern Australia, where >20 invasive fish species are recorded. In temperate regions, environmental DNA (eDNA) technology is gaining momentum as a tool to detect aquatic pests, but the technology's effectiveness has not been fully explored in tropical systems with their unique climatic challenges (i.e. high turbidity, temperatures and ultraviolet light). In this study, we modified conventional eDNA protocols for use in tropical environments using the invasive fish, Mozambique tilapia (Oreochromis mossambicus) as a detection model. We evaluated the effects of high water temperatures and fish density on the detection of tilapia eDNA, using filters with larger pores to facilitate filtration. Large‐pore filters (20 μm) were effective in filtering turbid waters and retaining sufficient eDNA, whilst achieving filtration times of 2–3 min per 2‐L sample. High water temperatures, often experienced in the tropics (23, 29, 35 °C), did not affect eDNA degradation rates, although high temperatures (35 °C) did significantly increase fish eDNA shedding rates. We established a minimum detection limit for tilapia (1 fish/0.4 megalitres/after 4 days) and found that low water flow (3.17 L/s) into ponds with high fish density (>16 fish/0.4 megalitres) did not affect eDNA detection. These results demonstrate that eDNA technology can be effectively used in tropical ecosystems to detect invasive fish species. 相似文献

SUMMARY The behaviour of a single specimen of Tilapia rendalli Boulenger was observed June through October 1986 in Lake Ngezi, Zimbabwe, by the use of a surgically-implanted radiotransmitter. The individual displayed a distinct homing response on two occasions, returning to the cove of capture (1) 1.5 km away within 48 hours of release (95Z of the distance was covered in 24 hours), and (2) after a lengthy migration some 6.0 km up the lake with other tilapia. During the only observed migration in late June, the fish travelled a conservative distance of 6.7 km over a 5-day period, sometimes at rates > 150 m/hr. The fish was also characterized by strong territoriality (i.e., preference to the cove of capture) and avoidance to the approach of the researcher by boat. 相似文献

硬骨鱼胚后发育中,边缘生长区及胚胎裂缝代表神经形成的活跃位点,同时对新的神经胶质细胞和光感器(视锥和视杆细胞)的形成有积极作用。此项工作首次研究红腹罗非鱼(硬骨鱼类辐鳍亚纲)视网膜中视网膜色素上皮和光感器的胚后生长及分化。采用光学和透射电子显微镜术分析了四个大小不同的鱼群组。正如至目前为止,在已研究过的大多数其它硬骨鱼中所观察到的结果一样,我们研究的所有样本中,成年红腹罗非鱼的视网膜表现为气球样扩张。此外,尚具有一弯曲的、有开口的胚胎裂缝,表明眼的不对称性胚后生长。在胚后分化过程中,视网膜色素上皮表现为自外周向中央的梯度变化,即边缘生长区周围,该上皮仅含有球形的黑色素体,而在中央部位发育为长形的黑色素体及顶端突起。同样,发育中的光感受器亦表现为由侧面向中央逐步发展变化的程序。光感觉器先形成1内节及其附属的突起,随后内节分化为一椭圆体加一肌样结构,这两种成分与其它已研究过的硬骨鱼视网膜中观察到的胚胎发育程序大致相符。因此提供了另一线索:硬骨鱼胚后视网膜的生长极大地刺激(促进)了胚胎视网膜的发育。本研究表明:双体视锥细胞可能是发育中的单体视锥细胞裂开的结果[动物学报49(6):813~818,2003]。 相似文献

In order to test whether histopathological changes of gills demonstrated a good dose–response relationship with water copper levels, juvenile Nile tilapia Oreochromis niloticus, of both sexes and similar mass (36·3 + 7·7 g), were kept in dechlorinated tap water (temperature 25° C, range ±1° C; pH 6·5–7·5; hardness 74·5 mg l?1 CaCO3) and exposed to 40 and 400 μg l?1 of copper. Gill samples were collected after 3, 7, 14 and 21 days. Six major histopathological changes (oedema, lifting, changes in filament epithelium thickness, lamellar fusion, vasodilatation and aneurisms) and three minor ones (proliferation of the lamellar epithelium, necrosis and adjacent lamellar fusion) were found and their prevalence estimated. The extent and severity of each histopathological change were used to develop a severity gradation scale (SGS). Semi‐quantitative analysis of the histopathological changes and measurements of gill copper deposition levels revealed a good dose– and time–response relationship. Oedemas and aneurisms were significantly correlated with acute exposure periods and lamellar fusion with chronic exposure. Epithelial lifting and changes in filament epithelial thickness were seen at lower and higher metal concentrations, respectively. We surveyed the watersheds covering more than 80% of the surface area of Nicaragua, and review the history of deliberate introductions
and unintentional invasion of tilapias, Oreochromis spp., into the freshwater of Nicaragua. The species have become widely established, with a range of negative consequences
for the rich natural fish fauna of this Central American country. Tilapias compete directly with native cichlids in a number
of ways, and have also supplanted native species as food fish in local markets. We suggest that introduced tilapias may have
been responsible for the outbreak of blindness in native cichlids. We make recommendations on the management of these exotic
species and on further introductions.

Arsenic is an environmental contaminant and potential carcinogen. Toxicological assessment of As, which causes hematological alterations and chromosomal aberrations, was studied in freshwater fish Oreochromis mossambicus. Fish were exposed to 3 ppm, 28 ppm, and 56 ppm concentrations of sodium arsenite (NaAsO2) and blood samples were collected after 48 h, 96 h, and 192 h of exposure. Hematological assay of exposed fish revealed abnormal mature and immature erythrocytes, deformed erythrocytes (spindle-shaped and triangular erythrocytes) and erythrocytes with segmented nuclei in all treatments. Arsenic exposure induced chromosomal aberration in a concentration-dependent manner, whereas, a decreasing trend was found after 192 h exposure. Observations on blood cells of exposed fish revealed chromosome breaks, chromatid breaks, and chromatid gaps. The alterations and aberrations of these parameters can be effectively used to assess toxicological effects of As on fish in the aquatic environment and at the same time this study elucidates the potential risks to humans who live in arsenic-contaminated areas. 相似文献

In terms of number of species, perciform (perch-like) fishes are one of the most diversified groups of modern vertebrates.
Within this group, the family Cichlidae is best known for its spectacular adaptive radiation in the great lakes of East Africa.
The molecular tool kit used in the study of this radiation includes the major histocompatibility complex (Mhc) genes. To refine this tool, information about the organization of the Mhc regions is badly needed. In this study, the first step was taken toward providing such information for the Mhc class one regions of Oreochromis niloticus, a representative species of the tilapiine branch of the Cichlidae, for which good bacterial artificial chromosome library
is available. Screening of the library with class I gene probes led to the identification and isolation of 31 class-I-positive
clones. Sequencing of one of these clones and partial characterization of the remaining clones for the presence of class I
exons resulted in the construction of two contigs representing the class I region of this species as well as identification
of seven additional class-I-positive singleton clones. The O. niloticus genome was shown to contain at least 28 class I genes or gene fragments. The shorter of the two contigs was approximately
330 kb long and contained eight class I genes/gene fragments; the longer contig encompassed 1,200 kb of sequence and contained
minimally 17 class I genes/gene fragments; three additional class I genes were found to be borne by a clone that might be
part of the shorter contig.
