Phytochrome (120 kdalton or 60 kdalton) was isolated from etiolated seedlings of Avena sativa L. cv. Pirol (Baywa München). Irradiation with red light of the Pr form at −23°C in aqueous medium or at −40°C in 66% glycerol leads to the intermediate meta-Rb. Acidification of the glycerol solution at −40°C leads to the absorption of the 15(E) phytochrome chromophore (= Pfr chromophore). Subsequent irradiation transforms this into the 15(Z) chromophore (= Pr chromophore). The presence of the 15(E) chromophore was demonstrated by the same methods also in phytochrome bleached either as Pfr in the dark by 4 M urea, methanol, acetone, ethylene glycol, 8-anilinonaphthalene-1-sulfonate, or as Pr by irradiation with red light in the presence of the same agents. Phytochrome bleached by sodium dodecylsulfate or by dehydration was also investigated. It was concluded that bleached phytochrome contains the Pfr chromophore without specific interaction with the protein.  相似文献

Light inhibits while carbon dioxide enhances the conversion of 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene in oat ( Avena sativa L. cv. Victory) leaf segments. The possibility that the light inhibition is mediated through changes of carbon dioxide has been investigated. The level of CO2 increases or decreases in the sealed incubation vial in darkness or in light, respectively, which can apparently account for the differences in ACC-dependent ethylene production between the dark and light treatments. However, although the evolution of ethylene from ACC in the dark is reduced upon depletion of CO2, the difference between light and dark is still very noticeable. Moreover, the production of the ethylene in CO2-free air in the dark was still higher than in the light, where the concentration of CO2 was 0.01%. It is proposed that the light effect on the conversion of ACC to ethylene is composed of two distinguishable components: one CO2-mediated and the other CO2-independent.  相似文献

It has long been assumed that Al3+ is an important rhizotoxic ion in acid soils around the world, but the toxicity of Al3+ relative to mononuclear hydroxy-Al [AlOH2+ and Al(OH)+2] has been examined in detail only for an Al-sensitive wheat variety ( Triticum aestivum L. cv. Tyler). That plant appears to be sensitive to Al3+ but not to AlOH2+ and Al(OH)+2. New experiments, and reanalyses of previously published experiments, provide evidence that dicotyledonous species may be sensitive to mononuclear hydroxy-Al and that Al3+ may be nontoxic, or less toxic, to those plants. Despite these consistently measured differences between wheat and the dicotyledons, the determination of relative toxicities (Al3+ vs mononuclear hydroxy-Al) may be an intractable problem. Because of hydrolysis equilibria, (AlOH2+) and (Al(OH)+2) are equivalent to (Al3+)k1(H+)−1 and (l3+)k2(H+)−2, respectively, in which k1 and k2 are the first and second hydrolysis constants (braces denote activities). Thus, any expression of root elongation as a function of mononuclear hydroxy-Al can be alternatively expressed as a function of (Al3+) and (H+). Toxicity attributed to mononuclear hydroxy-Al may actually be Al3+ toxicity that increases as pH rises (i.e. Al3+ toxicity ameliorated by H+).  相似文献

Gravitropism of maize and rice coleoptiles was investigated with respect to its dependence on the angle of displacement or the initial stimulation angle (ISA). Close examination of curvature kinetics and the response to a drop in stimulation angle (SA) indicated that the gravtropic response during an early but substantial part of the curvature development is directly related to the ISA, there being no effect of the reduction of SA resulting from the curvature response itself. On the basis of this finding, the relationship between the steady SA and the curvature rate was determined. In maize, the curvature rate increased linearly with the sines of SAs up to an SA of 90 degrees. Rice coleoptiles, however, showed a saturation curve in the same range of SAs. The saturation profile was nearly identical between coleoptiles grown in air and those submerged in water, although the latter elongated much faster. Rice coleoptiles appeared to be far more sensitive to gravity than maize coleoptiles. It is concluded that the sensitivity to gravity, assessed through dependence on ISA, is a property inherent to a given gravitropic organ. Long-term measurements of curvature indicated that the coleoptiles bend back past the vertical. This overshooting was marked in submerged rice coleoptiles.  相似文献

Biosynthesis of polyamines in plants is controlled primarily by the enzymes ornithine decarboxylase (EC 4.1.1.17) and arginine decarboxylase (ADC: EC 4.1.1.19), which are responsible for the production of putrescine, and S -adenosyl-L-methionine (SAM) decarboxylase (EC 4.1.1.50) that is necessary for the formation of spermidine and spermine (Spm). Little is known about the metabolic or molecular mechanisms regulating the synthesis of these enzymes. We have studied the regulation of ADC synthesis by Spm in osmotically-stressed oat ( Avena sativa L. ev. Victory) leaves, using a polyclonal antibody to oat ADC and a cDNA clone encoding oat ADC. Treatment with Spm in combination with osmotic stress resulted in increased steady-state levels of ADC mRNA, yet the levels of ADC activity decreased. This absence of correlation is explained by the fact that Spm inhibits processing of the ADC proenzyme, which results in increased levels of this inactive ADC form and a consequent decrease in the ADC-processed form. Spermine treatment leads to delayed loss of chlorophyll in dark-incubated and osmotically-treated oat leaves. Thus, post-translational regulation of ADC synthesis by Spm may be important in explaining its anti-senescence properties.  相似文献

Enhanced production and accumulation of free and conjugated polyamines as well as increased activities of their biosynthetic enzymes in plants have been associated with heat stress. Perchloric acid-soluble free, as well as conjugated polyamines, and their metabolic enzymes were studied under 45°C heat stress in callus raised from heat-tolerant and -sensitive rice cultivars. The levels of free and conjugated polyamines, as well as arginine decarboxylase (EC 4.1.1.19) and polyamine oxidase (EC 1.4.34) activities were higher in tolerant than in sensitive callus under non-stressed conditions. Heat stress caused greater accumulation of free and conjugated polyamines in callus of the heat-tolerant cultivar N22 than in that of the heat-sensitive cultivar IR8. In particular, the uncommon polyamines norspermidine and norspermine were detected in cv. N22, which increased appreciably during stress, but they were not detected in callus of cv. IR8. Arginine decarboxylase and polyamine oxidase activities increased to a larger extent in N22 than in IR8 callus during stress, activities that were well correlated with the increased levels of common and uncommon polyamines. Increased levels of transglutaminase activity indicated the high titre of conjugated polyamines.  相似文献

Screening for mutants deficient in the high affinity system of nitrate uptake was performed using mutagenized M2 population of rice ( Oryza sativa , cv. Nipponbare or Kinmaze). For selecting mutants, M2 seedlings were transferred individually to 10 ml solution containing 250 μ M potassium nitrate and 500 μ M calcium sulphate at 20 or 28°C. After 6 or 24 h, nitrate concentration of the solution was determined with a nitrate selective electrode and the seedlings showing impaired nitrate uptake were selected as nitrate uptake deficient variants. Of 74 variants, three were confirmed to be mutants with low nitrate uptake ability in the M3 generation. Potassium uptake ability also decreased in the mutants. Three mutants were divided into two groups based on the analysis of nitrate reductase (NR, EC 1.6.6.1) activity and chlorate resistance. Two, NUE13 and NUE36 , had a lower level of NR activity than the original cultivar and were not resistant to chlorate, while the seedlings of NUE50 had the same level of NR activity as the original cultivar and were more resistant to chlorate than the original cultivar. All mutants were resistant to cesium, a toxic ion analogue for potassium, suggesting that the decreased levels of both nitrate and potassium uptake were coupled to the change of plasma membrane H+-ATPase activity.  相似文献
