DELTA系统是针对分类学研究开发的综合性多功能软件包。新版DELTA系统在运行环境、程序、指令、界面、操作方式及中文字符处理等重要功能上比旧版本均有较大改进。作为国际分类学数据库数据交换标准,应用该系统可以规范分类学描述,对研究中所有常用性状进行识别和处理,自动生成各个研究对象分类学特征描述并对其进行详尽的相似性和差异性比较,对性状特征和分类单元进行注释和图像示例,将数据转换为数量分类学或分支分类学所需格式以生成树状图,自动生成检索表,建成基于本地和网络的交互式专家辅助鉴定系统等。DELTA系统作为植物分类学研究的常用手段和工具是非常适合及大有帮助的。  相似文献

All crocodilians are under varying degrees of threat due to over exploitation and these species have been listed in Appendix I or II of CITES. The lack of molecular techniques for the identification of confiscated samples makes it difficult to enforce the law. Conclusive forensic identification of species requires a complete gene sequence which is difficult in case of degraded samples. We have developed two novel sets of primers to amplify two partial cytochrome b gene sequences of six crocodile species i.e. Crocodylus palustris, Crocodylus porosus, Crocodylus siamensis, Crocodylus niloticus, Gavialis gangeticus and Caiman crocodilus. These partial sequences were edited to give a complete cyt b gene sequence, which can be used as an effective tool for forensic authentication of crocodile species. A phylogeny of crocodile species was reconstructed using these sequences. The described primers hold great promise in forensic identification of crocodile species, which can aid in the effective enforcement of law and conservation of these ancient species.  相似文献

Considerable effort has been put into detecting and identifying parasitic nematodes in live ruminants, but to date most studies are limited to a small group of nematodes and/or to experimentally infected sheep. In this study, a PCR-based assay using species-specific primer pairs, located in the second internal transcribed spacer ribosomal DNA, was developed to identify nine different species from six different families of parasitic nematodes in a wild, unmanaged and naturally infected population of sheep. Each primer pair was tested for its specificity and sensitivity and it exclusively amplified the species it was designed for and exhibited a high degree of sensitivity. The method was applied to eggs and cultured larvae to identify the parasitic nematodes present in a pooled faecal sample from several host individuals with unknown parasite burden. To test detection reliability, a faecal sample from an individual with known parasite burden (through post-mortem analysis) was also examined. All species present could be correctly identified by PCR, but detecting very low levels and/or early stages of infection proved to be difficult. The method was also tested for its applicability to high through-put screening of faecal samples.  相似文献

榆中贝母(Fritillaria yuzhongensis)生长于马啣山的高山地带,全生育期约100 d左右,是一种生育期较短的类短命植物。在果实成熟时,种子的胚发育不全,具有后熟作用的特点,包括形态后熟和生理后热两个阶段;因此,需要一定时期的低温处理,才能完成后熟过程,以解除休眠,促使种子发芽。本试验表明:榆中贝母完成形态后熟过程的最佳温度是7℃左右,经过100d以上的低温处理才能使原胚发育成熟,继续在7℃低温下处理70d以上,才能完成生理后熟过程。榆中贝母种子休眠时间长,需要经过170d以上的低温处理,种子才能萌发。本试验结果为榆中贝母种子的萌发提出了适宜的温度范围和低温处理时间,为提高种子的出苗率和缩短栽培年限提供了科学依据。  相似文献

对新疆昌吉和石河子两地种植的甜菜内生菌进行了分离、鉴定和分析,结果表明甜菜内生菌多属于细菌,其中假单胞菌 (Pseudomonas sp. )和芽孢菌类(Bacillus sp.)的分离频率分别在33.2%~59.2%和12.7%~28.1%,是甜菜植株中的优势内生菌群.16S rDNA 和 ITS 序列同源性比较和系统发育分析表明内生菌具有丰富的多样性.根中内生菌的多样性高于茎、叶,昌吉地区种植的甜菜中分离出的内生菌种类较多.从感病品种及生长不良甜菜植株中分离出的内生菌种类比较丰富.通过回接分离及利用扫描电镜观察内生菌在植物体内分布发现,内生菌能够定殖于甜菜块根.  相似文献

运用紫外光谱技术结合化学计量学,建立快速鉴别不同基原黄精的方法。通过单因素实验确定黄精最佳提取溶剂、时间和用量,制备测试液,采用紫外光谱技术建立3种基原黄精的紫外指纹图谱,光谱数据转化后进行主成分(PCA)和系统聚类分析(HCA)。该方法重现性、精密度、稳定性较好,结果表明不同种类黄精紫外指纹图谱具有指纹特性,3种基原植物黄精紫外光谱图在210 nm、220 nm、280 nm附近差异明显;聚类分析和主成分分析三维投影图反映出不同种类黄精的化学成分积累具有差异,能较好地区分滇黄精(Polygonatumkingianum)、黄精(P.sibiricum)与多花黄精(P.cyrtonema)。紫外光谱结合化学计量学能快速鉴别不同种类黄精,可作为黄精的鉴别和质量控制新方法,为黄精临床应用、资源开发及黄精属植物分类提供辅助方法。  相似文献

The effect of 16,16-dimethyl-PGE2-methyl ester (di-M-PGE2) on humoral and cellular immunoresponsiveness has been compared in normal mice and in mice immunosuppressed by splenectomy and thymectomy plus antithymocyte serum (ATS). Splenectomy resulted in immunosuppression manifested by augmentation of B-16 melanoma growth; this stimulatory effect was reversed by di-M-PGE2. In animals immunosuppressed by thymectomy plus ATS, di-M-PGE2 augmented the humoral and cellular immune responses; this was manifested by slowing of the growth of B-16 melanoma and by stimulating the number of plaque-forming cells, hemagglutinin titers, and delayed-hypersensitivity reactions to sheep erythrocytes. In contrast, in normal (nonthymectomized) mice, di-M-PGE2 was mildly immunosuppressive. Finally, adriamycin-immunosuppressed normal mice and this suppression were reversed by the addition of di-M-PGE2 to the treatment regimen.  相似文献

描述了在中国贵州发现的楼梯草属一新种:都匀楼梯草Elatostema duyunense W.T.Wang&Y.G.Wei,本种在亲缘关系上与上林楼梯草E.shanglinense W.T.Wang甚为相近,但根状茎细长,明显,茎较高,叶上面近牙齿顶端处被极短糙伏毛,托叶较大,条形或宽条形,有1条褐色脉,雄花序有短梗,花序托近圆形,明显,苞片6,其中2枚较大,卵状三角形,4枚较小,船状条形,小苞片无毛而与后者不同。  相似文献

Macroinvertebrates have been recognized as key ecological indicators of aquatic environment and are the most commonly used approaches for water quality assessment. However, species identification of macroinvertebrates (especially of aquatic insects) proves to be very difficult due to the lack of taxonomic expertise in some regions and can become time‐consuming. In this study, we evaluated the feasibility of DNA barcoding for the classification of benthic macroinvertebrates and investigated the genetic differentiation in seven orders (Insecta: Ephemeroptera, Plecoptera, Trichoptera, Diptera, Hemiptera, Coleoptera, and Odonata) from four large transboundary rivers of northwest China and further explored its potential application to biodiversity assessment. A total of 1,144 COI sequences, belonging to 176 species, 112 genera, and 53 families were obtained and analyzed. The barcoding gap analysis showed that COI gene fragment yielded significant intra‐ and interspecific divergences and obvious barcoding gaps. NJ phylogenetic trees showed that all species group into monophyletic species clusters whether from the same population or not, except two species (Polypedilum. laetum and Polypedilum. bullum). The distance‐based (ABGD) and tree‐based (PTP and MPTP) methods were utilized for grouping specimens into Operational Taxonomic Units (OTUs) and delimiting species. The ABGD, PTP, and MPTP analysis were divided into 177 (p = .0599), 197, and 195 OTUs, respectively. The BIN analysis generated 186 different BINs. Overall, our study showed that DNA barcoding offers an effective framework for macroinvertebrate species identification and sheds new light on the biodiversity assessment of local macroinvertebrates. Also, the construction of DNA barcode reference library of benthic macroinvertebrates in Eurasian transboundary rivers provides a solid backup for bioassessment studies of freshwater habitats using modern high‐throughput technologies in the near future.  相似文献

铅锌尾矿中耐重金属镉的丝状真菌的分离鉴定   总被引:2,自引:0,他引:2  
【目的】旨在从重金属污染地分离出耐重金属镉真菌, 获得耐受重金属镉污染的高效菌株, 为重金属污染微生物修复提供菌种资源。【方法】利用稀释平板涂布法, 采用4种培养基对粤东北梅州市梅县铅锌尾矿废弃地9个样品进行分离, 并结合形态学和ITS rDNA基因序列分析, 鉴定分离到的耐镉真菌, 最后对分离到的耐镉真菌进行最小抑制浓度 (MIC) 检测。【结果】从粤东北梅州市梅县铅锌尾矿废弃地分离出72株丝状真菌, 经形态学和分子技术鉴定, 它们主要属于曲霉属(Aspergillus)、青霉属(Penicillium)、枝孢属(Cladosporium)、油瓶霉属(Lecythophora)、拟青霉属(Paecilomyces)、镰刀孢属(Fusarium)等。MIC检测发现有4株丝状真菌耐镉浓度较高, Paecilomyces lilacinus (Thom) Samson (6?20 p), Penicillium pinophilum Hedgcock (6?16 p), Penicillium rolfsii Thom (6?16 m) 和Fusarium oxysporum Schlecht. (8?11 p) 分别为200、40、25和15 mmol/L。【结论】从粤东北梅州市梅县铅锌尾矿废弃地分离到的72株丝状真菌, 不同程度耐受重金属镉, 在重金属污染的治理中有可能发挥作用。本研究为镉污染环境的微生物修复提供了重要菌株。  相似文献
