A quantitative endonuclease assay, which relies on the introduction of single and double strand breaks into supercoiled plasmid DNA, was used to study the activity of the extracellular nuclease of Serratia marcescens SM6 in buffer and in groundwater. The parallel enzyme concentration-dependent production of relaxed and linear plasmid molecules suggests that the nuclease produces single and double strand breaks in duplex DNA. Bovine serum albumin stimulated the nuclease activity towards DNA and RNA and increased the stability of the enzyme against thermal inactivation. The DNase activity at 4 °C and 50 °C was almost half of that at the optimum temperature (37 °C). The nuclease was active in groundwater, although the specific activity was lower than in buffer. In a groundwater aquifer microcosm, mineral-adsorbed transforming DNA was substantially less accessible to the nuclease than was dissolved DNA. The data suggest that the extracellular nuclease of Serratia marcescens may contribute to DNA turnover in the environment and that adsorption of DNA to minerals provides protection against the nuclease.Abbreviations GW groundwater

The development of resistance to anticancer drugs urges the search for different treatment modalities. Several investigators have reported the concomitant development of drug resistance and resistance to natural killer (NK), lymphokine-activated killer (LAK) or monocyte/macrophage cell lysis, while others described unchanged or even increased susceptibility. We investigated this subject in the rat colon carcinoma cell line, CC531-PAR, which is intrinsically multidrug-resistant (MDR), and in three sublines derived from this parental cell line: a cell line with an increased MDR phenotype (CC531-COL), a revertant line from CC531-COL (CC531-REV), which demonstrates enhanced sensitivity to anticancer drugs of the MDR phenotype, and an independently developed cisplatin-resistant line (CC531-CIS). In a 4-h51Cr-release assay we found no difference in susceptibility to NK cell lysis. No significant differences in lysability by adherent LAK (aLAK) cells were observed in a 4-h assay. In a prolonged 20-h51Cr-release assay an enhanced sensitivity to aLAK-cell-mediated lysis was observed in the revertant, P-glycoprotein-negative cell line and in the cisplatin-resistant cell line (CC531-CIS). None of the cell lines was completely resistant to lysis by aLAK cells. Therefore, a role for immunotherapy in the treatment of drug-resistant tumors remains a realistic option. 相似文献

We investigated which of the following environmental factors: the number of years since the windthrow of the tree (the age of dead wood), the phytocenosis (the type of forest community), altitude, exposure, wood hardness and the spatial scale of forest disturbances (small gaps with a few fallen spruces vs large-area windthrows) contributed to the diversity and abundance of lichens inhabiting the exposed wood of windthrown spruce trees in Polish Western Carpathian forests. Both Shannon H index and sum of coverage coefficients rose with increasing age of the wood, levelling off after 11–14 y (diversity) and 14–17 y (abundance). This factor appeared to be the most important for this group of lichens, but the significant positive impact of large-area windthrows on the lichen abundance was also demonstrated by using a GLM model. The age of the wood we precisely determined on the basis of data on Norway spruce mortality collected annually in permanent plots of the Gorce National Park since 2000. Using the Shore durometer we linked the course of the wood-inhabiting lichen succession with wood decay more precisely than before. The largest number of species was associated with medium hard wood, i.e., 51 < x ≤ 80 on the Shore scale. Based on the NMDS analysis, we distinguished four age groups of logs, differing in lichen abundance and defined by the dominance of distinctive species. A large number of usually corticolous lichen species used the wood of windthrown spruce logs as an optional habitat to survive large-scale, post-hurricane forest disturbances. 相似文献

Recent rescue excavations at Bercy (Paris), a site on the alluvial plain of the Seine valley, yielded plant remains which
are associated with the recent occupation phase dating from the middle Neolithic II (Chasséen), when a village was established
on the former channel of the river Seine. Various contexts (the channel, the flat lower part of the bank, and several archaeological
features) have been studied and 84 taxa have been identified. Cultivated plants are represented by Triticum aestivum/durum, T. dicoccum, T. monococcum and Hordeum vulgare. Among the wild plants with potentially edible fruits or seeds, only very few satisfy the various criteria for association
with human activities: Corylus avellana, Vitis sylvestris, Cornus sanguinea, Quercus sp. and Prunus spinosa of which carbonized fruits were also present, were found in archaeological features and were very abundant. Though not found
in archaeological features, we consider that Crataegus monogyna (carbonized and well represented) and Rubus spp. (especially abundant) were not deposited there naturally and had also been intentionally collected. Finally, it is suggested
that the allochthonous (varied) origin of these taxa is the reason why there are no concentrations of their fruits in the
channel. The exploitation of wild seeds and fruits appears to have been very selective. All other wild taxa can be attributed

A large proportion of spontaneous mutations inDrosophila melanogaster strains of laboratory origin are associated with insertions of mobile DNA elements. As a first step toward determining whether spontaneous laboratory mutations are predictive for mutational events occurring in the wild, recessivebrown (bw) eye color mutants were isolated. By inbreeding the progeny of wild-caughtDrosophila melanogaster females,bw mutations were isolated from seven separate geographic sites distributed among Japan, California, Siberia and Hungary. Among a total of 14 mutations studied, no case of transposon mutagenesis was found. At least 4 mutations are associated with small deletions in thebw gene. The remainder are inseparable from wild-typebw by Southern analysis and are presumed to be basepair changes or very small indels. Summary On account, notably, of a competition between different component functions for individual sites in polypeptide chains, each protein molecule represents a functional compromise, with some functions optimized, but the overall state of the molecule –suboptimal–. The proposal is made that the selection coefficient relating to a protein molecule under given conditions can in principle be broken down into partial selection coefficients relevant to the different functions that the molecule carries out. At generalfunction sites, each fixation improves some function, while others deteriorate, at first nonsignificantly, and the overall adaptive state of the molecule fluctuates around its maximum. A selective mechanism is described whereby kaleidoscopic changes in primary structure at variable sites are indefinitely promoted, independently of any environmental changes and with the molecule remaining close to a state of maximal overall adaptation. The paradoxical aspect of this proposal is analyzed. The implication of specific functions in substitutions at general-function sites is noted. Further, it is shown that a certain category of changes in the internal environment of the organism can be integrated into the constantenvironment model for selection. Genetic sufficiency is considered a notion more adequate than genetic optimality for describing biological fitness and for providing a basis for the present model. On this basis selection occurs without genetic load. Multipolymorphism is one of the consequences. Several lines of evidence, in particular observations on polymorphism in deep sea organisms, seem to support the model. It is pointed out that it provides a theoretical foundation for a molecular evolutionary clock. The theoretical constancy of the clock depends on the constancy of functional density. The question of the evolution of functional density is examined. Comparisons of observed substitution frequencies with values expected on a random basis are rejected as a measure of the contribution to evolution of nondetermination. They are considered to reflect a hierarchy in the resistance of the molecules to different amino acid residues as substituents. A limited component of –true– randomness, again accompanied by selection, is on the other hand provided by the model. Most amino acid substitutions are considered evolutionary noise, even though noise compatible with selection. The analysis of spatial patterns is one of the ways to estimate the role of competition among trees in forest dynamics. Three hypotheses concerning distribution patterns in old-growth stands were tested: (1) fine-scale spatial patterns of trees are regular; (2) patterns do not differ significantly from a random distribution, and (3) spatial patterns at larger scales are clumped because of site heterogeneity. Old-growth forest stands in Poland and the Czech Republic were analysed with a modified Ripley K function, using distribution maps of tree stems. Fine-scale spatial patterns (with distances among trees not exceeding 15 m) were usually intermediate between random and regular. Trends towards a regular distribution occurred more often among dead than among live individuals. No significant relationships between tree species were found at smaller scales; however, at larger scales (distances from 15–25 m) negative associations between some species were found. This reflects site heterogeneity rather than any direct influence of one tree species upon another. 相似文献

Reliable information about soil conservation capacities of different natural ecosystems is an important reference for the design of targeted erosion and sediment control strategies. The objective of this paper is to quantify the soil conservation capacities of different natural ecosystems that can represent different climatic zones. The 137Cs technique has been used to estimate soil redistribution rates in different natural ecosystems over the past 40 years in Wolong Nature Reserve. The reserve, transiting from the Chengdu plain to the Qinghai-Tibet plateau, maintains rich ecosystems from subtropical to frigid.The net soil erosion rates of 5 selected ecosystems that represent a warm coniferous-broadleaf-mixed forest, a cold-resistant deciduous taiga forest, a cold-resistant shrub, an evergreen cold-resistant taiga servation capacities are reversed in order. The reference inventories for 137Cs in different ecosystems velop effective erosion and sediment strategies in areas with similar climates should consider natural ecosystem types. 相似文献
