Viral particles of human severe acute respiratory syndrome coronavirus (SARS CoV) consist of three virion structural proteins, including spike protein, membrane protein, and envelope protein. In this report, virus-like particles were assembled in insect cells by the co-infection with recombinant baculoviruses, which separately express one of these three virion proteins. We found that the membrane and envelope proteins are sufficient for the efficient formation of virus-like particles and could be visualized by electron microscopy. Sucrose gradient purification followed by Western blot analysis and immunogold labeling showed that the spike protein could be incorporated into the virus like particle also. The construction of engineered virus-like particles bearing resemblance to the authentic one is an important step towards the development of an effective vaccine against infection of SARS CoV. 相似文献

The severe acute respiratory syndrome (SARS) has been one of the most epidemic diseases threatening human health all over the world. Based on clinical studies, SARS-CoV (the SARS-associated coronavirus), a novel coronavirus, is reported as the pathogen responsible for the disease. To date, no effective and specific therapeutic method can be used to treat patients suffering from SARS-CoV infection. RNA interference (RNAi) is a process by which the introduced small interfering RNA (siRNA) could cause the degradation of mRNA with identical sequence specificity. The RNAi methodology has been used as a tool to silence genes in cultured cells and in animals. Recently, this technique was employed in anti-virus infections in human immunodeficiency virus and hepatitis C/B virus. In this study, RNAi technology has been applied to explore the possibility for prevention of SARS-CoV infection. We constructed specific siRNAs targeting the S gene in SARS-CoV. We demonstrated that the siRNAs could effectively and specifically inhibit gene expression of Spike protein in SARS-CoV-infected cells. Lyophilized aqueous extract of Maytenus ilicifolia leaves (LAEMIL) is commonly used in Brazilian folk medicine in the treatment of dyspepsia as well as gastric ulcers. We have investigated the effect and the possible mechanism of action of the LAEMIL on acid secretion in isolated frog gastric mucosa incubated in an Ussing chamber. It was observed that LAEMIL (7–28 mg%) as well as cimetidine (125–4,000 M), a well-known histamine H2 receptor antagonist, decreased basal acid secretion in a concentration-dependent manner. Similarly to cimetidine (190 M), LAEMIL (21 mg%) also inhibited gastric acid secretion induced by increasing concentrations of histamine (50–800 M). The EC50 values for histamine alone and histamine in the presence of LAEMIL or cimetidine were 94.6 M (71.1–125.9 M), 244.9 M (209.4–286.4 M) and 142.2 M (23.6–855.0 M), respectively. LAEMIL, histamine and cimetidine were effective on acid secretion only when added to the serosal surface of the mucosa. Furthermore, simultaneous addition of LAEMIL and cimetidine at concentrations, per se, ineffective, caused a 16% reduction in the basal acid secretion [from 8.3±0.3 to 6.9±0.2 Eq g–1 (15 min)–1, n=4]. Although effects such as inhibition of histamine biosynthesis and/or histamine release can not be ruled out, our data suggest that LAEMIL, like cimetidine, reduces acid secretion in the isolated frog gastric mucosa by antagonising histamine H2 receptors.Abbreviations LAEMIL Lyophilized aqueous extract of Maytenus ilicifolia leaves - Hist Histamine - Cim Cimetidine 相似文献

实验选用健康成年SD大鼠,观察电刺激面神经核对前包钦格复合体(pre-—Boetzinger complex,PBC)呼吸神经元(RNs)放电活动的影响,并观察微电泳6-氰基-7-硝基喹喔啉-2,3-二酮(CNQX)、荷包牡丹碱(BIC)、士的宁(Stry)和阿托品(Atr)对电刺激面神经核引起的PBCRNs放电变化的拮抗效应,以进一步探讨面神经核是否参与呼吸调节及其可能的神经机制。在12只面运动神经元逆行溃变大鼠同侧PBC内共记录到各类RNs116个,电刺激溃变侧面神经核时,前吸气(Pre-I)神经元(24/26个)和吸气(I)神经元(30/35个)主要表现为兴奋,呼气(E)神经元(20/22个)和吸气-呼气(I-E)跨时相神经元(28/33个)表现为抑制。CNQx可完全或部分拮抗电刺激面神经核对Pre-I(18/24)和I(23/27)神经元的兴奋效应;Stry可拮抗电刺激面神经核对Pre-I(12/18)和I(14/23)神经元的瞬时抑制效应以及对I-E(20/28)和E(9/16)神经元的抑制效应;BIC可拮抗电刺激面神经核对I—E(22/25)和E(9/9)神经元的抑制效应;微电泳Atr对各类RNs的放电变化无明显作用。这些结果表明,面神经核非运动神经元可能通过向PBC的纤维投射,以Glu、GABA和Gly为神经递质或调质,调节PBC RNs的活动,从而参与对呼吸运动的调节。 相似文献

目的:探讨双水平气道正压通气(Bi-PAP)联合舒利迭对慢性阻塞性肺疾病(COPD)合并Ⅱ型呼吸衰竭患者呼吸衰竭症状、免疫功能以及生活质量的影响。方法:按照随机数字表法将2012年6月至2016年12月间我院收治的157例COPD合并Ⅱ型呼吸衰竭患者分为观察组(n=79)和对照组(n=78),对照组患者在平喘、解痉、化痰、抗感染等常规治疗同时联合Bi-PAP治疗,观察组患者在对照组治疗方案的基础上给予舒利迭治疗。比较两组患者治疗前后呼吸衰竭症状、免疫功能以及生活质量改善情况。结果:观察组治疗后有效率为89.87%(71/79),显著高于对照组的73.08%(57/78)(P0.05)。治疗后两组患者的呼吸、心率、动脉二氧化碳分压较治疗前降低,动脉氧分压较治疗前升高,且观察组患者呼吸、心率、动脉二氧化碳分压低于对照组,动脉氧分压高于对照组(均P0.05)。治疗后两组患者血清免疫球蛋白M(IgM)、免疫球蛋白G(IgG)以及免疫球蛋白A(IgA)水平均较治疗前明显升高,且观察组患者治疗后的IgM、IgA、IgG水平高于对照组(均P0.05)。随着时间的推移,两组患者治疗后的圣·乔治医院呼吸问卷(SGRQ)评分逐渐降低,且观察组治疗后3个月、6个月的SGRQ评分均低于对照组(均P0.05)。结论:Bi-PAP联合舒利迭能明显改善COPD合并Ⅱ型呼吸衰竭患者呼吸衰竭症状,提高机体免疫力和患者的生活质量,临床推广应用价值高。 相似文献

Porcine reproductive and respiratory syndrome (PRRS) continues to be one of the most important swine diseases worldwide. Interferon-γ (IFNγ)-mediated type Ⅰ cell-mediated immune response plays an important role in protection from, and clearance of, PRRS virus (PRRSV). Several lymphocyte subsets including T-helper, CTLs, Th/memory cells, and γδ T lymphocytes were previously reported to produce IFNγ during PRRSV infection. However, the proportion and phenotypic characterization of these IFNγ-secreting lymphocytes have not been explored. In this study, IFNγ producted by different lymphocyte subsets was assessed by multi-color flow cytometry after vaccination with PRRSV modified live vaccine (PRRSV-MLV) and challenge with homogeneous or heterogeneous PRRSV. The results showed that T-helper cells were the major IFNγ-secreting cell population after PRRSV-MLV vaccination and PRRSV challenge. Additionally, the proportion of IFNγ producing Th/memory cells and γδ T cells increased after PRRSV challenge. This difference was accounted for an enhanced ability to produce IFNγ in Th/memory cells and an enlarged quantity of γδ T cells. The results presented here could contribute to our understanding of the roles of IFNγ in protective immunity against PRRSV infection and may be useful for assessment of cell-mediated immunity in vaccine tests. 相似文献

Technovit 7200 VLC is an acrylic resin formulated for embedding undecalcified hard tissues which are prepared for light microscopy according to a cutting-grinding technique. To employ this resin for embedding and cutting soft tissues by ultramicrotomy, we carried out a qualitative study on biopsies of canine gingival mucosa using light and transmission electron microscopy. For a critical evaluation of this resin, some biopsies were embedded in Agar 100, an epoxy resin widely used in morphological studies. At the light microscopic level the samples embedded in Technovit 7200 VLC showed good morphology and excellent toluidine blue staining of different cell types and extracellular matrix. At the ultrastrueturallevel, nuclei, cytoplasmic organelles, collagen fibrils and ground substance appeared well preserved and showed high electron density. The acrylic resin was stable under the electron beam and its degree of shrinkage appeared to be very low. We conclude that Technovit 7200 VLC can be employed for ultramicrotomy for both light and electron microscopic investigation of soft tissues. 相似文献

Norathyriol, aglycone of a xanthone C-glycoside mangiferin isolated from Tripterospermum lanceolatum, concentration dependently inhibited the formylmethionyl-leucyl-phenylalanine (fMLP)-induced superoxide anion (O2˙−) generation and O2 consumption in rat neutrophils. In cell-free oxygen radical generating system, norathyriol inhibited the O2˙− generation during dihydroxyfumaric acid (DHF) autoxidation and in hypoxanthine-xanthine oxidase system. fMLP-induced transient elevation of [Ca2+]i and the formation of inositol trisphosphate (IP3) were significantly inhibited by norathyriol (30 μM) (about 30 and 46% inhibition, respectively). Norathyriol concentration dependently suppressed the neutrophil cytosolic phospholipase C (PLC). In contrast with the marked attenuation of fMLP-induced protein tyrosine phosphorylation (about 70% inhibition at 10 μM norathyriol), norathyriol only slightly modulated the phospholipase D (PLD) activity as determined by the formation of phosphatidic acid (PA) and, in the presence of ethanol, phosphatidylethanol (PEt). Norathyriol did not modulate the intracellular cyclic AMP level. In the presence of NADPH, the phorbol 12-myristate 13-acetate (PMA)-activated particulate NADPH oxidase activity was suppressed by norathyriol in a concentration-dependent manner and the inhibition was noncompetitive with respect to NADPH. Norathyriol inhibited the iodonitrotetrazolium violet (INT) reduction in arachidonic acid (AA)-activated cell-free NADPH oxidase system at the same concentration range as those used in the suppression of PMA-activated particulate NADPH oxidase activity. Taken together, these results suggest that the scavenging ability of norathyriol contributes to the reduction of generated O2˙−, however, the inhibition of O2˙− generation from neutrophils by norathyriol is attributed to the blockade of PLC pathway, the attenuation of protein tyrosine phosphorylation, and to the suppression of NADPH oxidase through the interruption of electrons transport. 相似文献

A vigorous and transient increase of O2 uptake associated with a simultaneous release of CO2 was elicited in Egeria densa and in Potamogeton crispus leaves by treatment with N-ethylmaleimide (NEM) and by other -SH group reagents (iodoacetate, p-(chloromercuri)benzenesulfonate (p-CMBS), Ag+, Hg2+, Cu2+). The NEM-induced respiratory burst was apparent even in the absence of photosynthesis (darkness, or presence of DCMU) as well as in the presence of the respiration inhibitors cyanide and propyl gallate or SHAM, separate or in combination. In contrast, a complete suppression of the respiratory burst was induced by diphenylene iodonium and by quinacrine, inhibitors of the plasma membrane NADPH oxidase activated in the pathogen-elicited oxidative burst in granulocytes and in plants. The respiratory burst induced by NEM and by the uncoupler CCCP were additive. The intensity of the respiratory burst was markedly decreased by increasing the pH of the medium from 5 to 8, and partially decreased by the presence of K+ in the medium. Azide inhibited the burst (as well as basal respiration) at pH 6.5 but not at pH 5. The stimulation of QO2 by SH reagents was associated with an early, pronounced membrane depolarization together with a rapid increase of the release into the medium of K+ and other electrolytes, and with a rapid decrease of the intracellular ATP, ADP and G6 P contents. The possible relationships between this SH reagent-induced respiratory burst and the associated effects on Em and electrolyte leakage are discussed. 相似文献
