Active in both binding and biological assays, morphiceptin (NH2 Tyr-Pro-Phe-Pro-CONH2), a potent opioid peptide derivative of β-casamorphine, binds specifically and selectively to mu or morphine-type receptors with little affinity for delta sites. Displacement studies of a variety of 3H-labeled opiates and enkephalins show biphasic curves. Naloxazone, which blocks irreversibly and selectively high affinity opiate and enkephalin binding, abolishes morphiceptin's inhibition of binding at low concentrations, suggesting that the high affinity binding of enkephalins and opiates represents a mu or morphine-type receptor. Unlike the reversible antagonist naloxone, naloxazone treatment invivo inhibits for over 24 hours the analgesic activity of morphiceptin like it inhibits morphine, β-endorphin and enkephalin analgesia. Together, these studies imply that opiates and enkephalins bind with highest affinity to a mu receptor which mediates their analgesic activity. The 3H-D-ala2-D-leu5-enkephalin binding remaining after naloxazone treatment, representing a lower affinity site (KD 4 nM), is quite insensitive to morphiceptin inhibition and has the characteristics of a delta receptor. However, the 3H-dihydromorphine binding present after naloxazone treatment, which also represents a lower affinity site (KD 6 nM), is far more sensitive to both morphine and morphiceptin and may represent a second morphine-like, or mu, receptor.  相似文献

ADP and Pi-loaded membrane vesicles from l-malate-grown Bacillus alcalophilus synthesized ATP upon energization with ascorbateN,N,N′,N′-tetramethyl-p-phenylenediamine. ATP synthesis occurred over a range of external pH from 6.0 to 11.0, under conditions in which the total protonmotive force Δ\?gmH+ was as low as ?30 mV. The phosphate potentials (ΔGp) were calculated to be 11 and 12 kcal/mol at pH 10.5 and 9.0, respectively, whereas the Δ\?gmH+ values in vesicles at these two pH values were quite different (?40 ± 20 mV at pH 10.5 and ?125 ± 20 mV at pH 9.0). ATP synthesis was inhibited by KCN, gramicidin, and by N,N′-dicyclohexylcarbodiimide. Inward translocation of protons, concomitant with ATP synthesis, was demonstrated using direct pH monitoring and fluorescence methods. No dependence upon the presence of Na+ or K+ was found. Thus, ATP synthesis in B. alcalophilus appears to involve a proton-translocating ATPase which functions at low Δ\?gmH+.  相似文献

Superoxide dismutase, a scavenger of O?2. does not affect the rate of ethanol oxidation in a reconstituted system containing purified cytochrome P-450, NADPH-cytochrome c reductase, and dilauroyl l-3-phosphatidyl choline. The same concentration of Superoxide dismutase (50 μg/ml) completely abolishes the oxidation of epinephrine in this reconstituted system and ethanol oxidation by the xanthine-xanthine oxidase. Ethanol is not oxidized by the reconstituted system when NADPH is replaced by H2O2 but the addition of H2O2 to this sytem containing NADPH accelerates ethanol oxidation. This increase is abolished by the addition of Superoxide dismutase. Hydroxyl radical scavengers (50 mm dimethylsulfoxide, 100 mm benzoate, 100 mm mannitol, 20 mm thiourea) diminish the oxidation of ethanol in the reconstituted system by 48 to 76%. Thus hydroxyl radical may participate in the activity of reconstituted ethanol-oxidizing system, whereas Superoxide is not involved.  相似文献

European pear psylla Cacopsylla pyri L. (Hemiptera Psyllidae) is one of the worst pests of pear (Pyrus communis L.) in Europe. We investigated probing behaviour in adults and nymphs of C. pyri by full EPG on a psylla‐resistant pear selection, NY 10353. Concerning stylet probing behaviour on the plant surface, the results showed no significant differences between the resistant selection and the susceptible cultivar Bartlett, and no differences were also detected for epidermis and mesophyll resistance in the same conditions. For mesophyll/phloem, no differences were found in adults. However, in nymphs, weak resistance factors (longer stylet penetration and mesophyll salivation) were detected in the resistant selection. In phloem, EPG data indicate strong resistance factors in NY 10353, especially for nymphs and summer‐form adults (longer time before the first phloem ingestion and a lower duration of each phloem ingestion event). No prolonged (>10 min) phloem ingestion was performed by nymphs and adults in the resistant selection. The results support the hypothesis that NY 10353 resistance factors are located in the phloem sap and cause high C. pyri nymph mortality: this could be useful as a basis for further investigations of resistance mechanisms at the metabolic, chemical and genetic levels.  相似文献

Physiological concentrations (10(-10) M) of L-thyroxine and triiodo-L-thyronine were found in vitro to enhance Ca2+-ATPase activity in reticulocyte-enriched red cell membranes from female rabbits and to inhibit this enzyme in the male reticulocyte. Cross-incubation experiments with reticulocyte-enriched red cells and plasma from the opposite sex demonstrated that this sex-specific membrane response to thyroid hormone was transferable by plasma. Similar experiments with intact reticulocytes exposed to physiological concentrations (10(-11) M) of testosterone and 17 beta-estradiol indicated that the plasma factors were the sex steroids. That is, incubation in vitro with testosterone converted female-source reticulocytes to male-type responsiveness to thyroid hormone (inhibition of Ca2+-ATPase activity); incubation with estradiol converted male-source reticulocyte-enriched red cells to female-type responsiveness (stimulation by iodothyronines of membrane Ca2+-ATPase activity). Similar results were obtained when reticulocyte ghosts were incubated with testosterone and 17 beta-estradiol prior to determination of membrane enzyme activity. Etiocholanolone (5 beta-androstan-3 alpha-ol-17-one) and testosterone were equipotent, but 5 alpha-dihydrotestosterone had little activity in this system. Estrone and estradiol were equipotent, but estriol had no permissive effect on the stimulation by iodothyronine of reticulocyte membrane Ca2+-ATPase activity. Expression of thyroid hormone action in vitro on Ca2+-ATPase activity in the rabbit reticulocyte is determined at the membrane level by testosterone and estrogen. The structure-activity relationships of the sex steroids for this membrane action are different than those reported for nuclear actions of the steroids.  相似文献

Ultrasound technology was used to measure live animal meat traits instead of true carcass meat traits for beef production and cattle breeding by an increasing number of institutions. In this study, we analyzed the association between genetic polymorphisms of proopiomelanocortin (POMC) and ultrasound measurement traits in Chinese cattle. Using direct DNA sequencing in 322 individuals of 7 different cattle subpopulation, 7 SNPs were identified for genotyping within 790 bp region of intron 2 and exon 3 of POMC. 6586 T>G in intron 2 and 6769 C>T and 7216 C>T in exon 3 were significantly associated with ultrasound backfat thickness (UBF) (P < 0.05) and ultrasound loin muscle area (ULMA) (P < 0.01) in the total population; 6694 C>T, 6706 T>C, 6796 C>T and 6810 C>T in exon 3 were significantly associated with ULMA (P < 0.0001) in the total population. These results clearly suggest that these SNPs of POMC be benefit for selection of individuals with good quality meat in Chinese cattle breeding program. Following validation in other populations and breeds, these markers could be incorporated into breeding programs to increase the rate of improvement in carcass and meat quality traits.  相似文献

为了研究铜绿假单胞菌Pseudomonas aeruginosa NY3胞外分泌物对烃类降解的作用,对胞外分泌物影响烃类降解的效率进行了研究。研究发现,NY3菌以LB培养基和以烃类为唯一碳源的无机盐培养基培养得到的种子液,离心并洗涤菌体后,菌细胞代谢烃类效率明显下降。分别利用盐析和溶剂萃取法,提取种子液上清液中胞外大分子和小分子,且投加在NY3菌(离心后的菌细胞)降解十四烷的无机盐培养基中,发现24 h内外加胞外大分子化合物,使NY3菌对十四烷去除率可提高约14%,而投加小分子化合物使NY3菌24 h内对十四烷去除率提高约6%。利用SDS-page聚丙烯酰氨凝胶电泳分离胞外大分子,结果发现主要是该菌胞外所分泌的蛋白或多肽类,主要有3个条带,它们的分子量约在35~55 k D范围内,其对烃降解促进作用机理有待进一步验证。利用飞行质谱(LCMS-IT-TOF)鉴定胞外小分子分泌物,结果表明在本研究条件下,它们均为氧化还原反应活性物,可以加快反应体系中电子传递速度,促进底物的氧化还原反应。  相似文献

Living mitotic HeLa cells were microinjected with sodium fluorescein, fluorescein isothiocyanate (FITC)-labelled IgG, and Lucifer Yellow-CH to determine if the intracellular bridge and midbody forms a barrier to the migration of these molecules. All three fluorescent molecules were found to pass across the intercellular bridge. The post-mitotic cells maintained a molecular patency across the intercellular bridge that persisted for considerable lengths of time, extending into the next cell cycle.  相似文献

The lower six miles of the tidal portion of the Passaic River (Study Area) has long been heavily industrialized. The objectives of this study were to: quantify the present extent and magnitude of metals contamination in surface sediments in the Study Area, evaluate the contamination in the Study Area relative to a reference area and surrounding regional waterways, assess the potential for adverse effects to aquatic organisms, and identify spatial gradients in concentrations that may indicate potential point-sources of metals. We also examined the role of natural sediment characteristics in metals concentration variability. Study Area sediments were generally enriched in barium, cadmium, chromium, copper, lead, manganese, mercury, silver, selenium, and zinc relative to the reference area. Compared to available sediment quality benchmarks, the only metals in the Study Area presently at average concentrations sufficiently high to warrant concern about potential aquatic toxicity are lead, mercury, and zinc. Compared to the rest of the NY/NJ Harbor Estuary, the Study Area generally contains elevated levels of lead, manganese, silver, and zinc. Variability in metals concentrations can not be accounted for by TOC or percent fines. However, a substantial proportion of the spatial variability in a number of metals can be explained by normalization to either aluminum or iron.  相似文献
