The binding of tritium-labelled 7,12-dimethylbenz[a]anthracene (DMBA), benzo[a]pyrene (BP) and 3-methylcholanthrene (MCA) to DNA or polynucleotides in vitro was re-examined both in the presence and in the absence of rat liver or human placental microsomes.A high level of non-enzymatic binding was evident when thymus DNA was used as acceptor. This non-enzymatic binding made it difficult to determine the effect of microsomes, except in the case of BP when induced rat microsomes were used. Better results were obtained using polynucleotides: a definite microsome-dependent binding occurred between all the polynucleotides and all the hydrocarbons tested.No clear evidence of binding catalysed by microsomes from human placenta was found except in polynucleotide-BP interactions: further studies are required to completely evaluate the ability of such nucleic acid-microsomal system for testing in vitro possible oncogenic substances in animals and humans.  相似文献

人白细胞介素-3(humanInterleukin3,hIL-3)是一种造血前体细胞早期分化的关键调节因子。用PCR方法从人T淋巴细胞cDNA文库中扩增出0.44kb的DNA片段,并克隆入pUC19载体中。经DNA序列测定,确定0.44kb的PCR产物合完整的编码人白细胞介素-3成熟蛋白cDNA序列,并在信号肽与成熟蛋白编码序列之间通过突变引入了限制性内切酶位点和ATG起始密码。构建的PL启动子控制下的hIL-3cDNA表达质粒,转入大肠杆菌Tap106,经42℃热诱导,获得hIL-3的表达产物。SDS-PAGE电泳显示表达产物为15kd,约占细菌总蛋白的15%。表达产物经ELISA和Western-blot验证。hIL-3表达产物在细胞内形成包涵体,纯化包涵体,使产物纯度提高到70.8%,产物复性后,能明显促进hIL-3依赖细胞株生长,具有明显的生物活性。产物转移到PVDF膜后进行N端序列分析,N端16个氨基酸正确。  相似文献

细胞色素P450(P450,Ec1.14.14.1)是一种十分重要的催化氧化反应的酶。本文测定了12个对氨基苯甲酸酯同系物与P450相互作用而形成P450代谢中间体络合物的活性,用半经验分子轨道法MNDO-PM3计算得到了这些同系物的分子轨道指数,并用逐步多元回归分析法导出了活性与分子轨道指数及正辛醇/水分配系数的对数值(LogP)之间的定量结构与活性关系(QSAR)。结果表明:对氨基苯甲酸脂同系物形成P450代谢中间体络合物的活性与原子净电荷的绝对值之和(∑Q)和LogP均具有很好的抛物线型相务性,同时,LogP与∑Q之间也存在相当好的相关性。  相似文献

The intracellular free Ca2+ ion concentration ([Ca2+]i) was measured using fura-2 microspec-trofluorimetry in individual rat pancreatic β-cells prepared by enzymatic digestion and fluorescence-activated cell sorting. The mean basal concentration of [Ca2+]i in β-cells in the presence of 4.4 mM glucose and 1.8 mM Ca2+ was 112±1.6 nM (n=207). The action of acetylcholine (ACh) was concentration-dependent, and raising the concentration resulted in [Ca2+]i spikes of increasing amplitude and duration in some, but not all of the β-cells. In addition, the β-cells demonstrated variable sensitivity to ACh. The increases in [Ca2+]i were rapid, transient and were blocked by atropine at 10-6M. A brief exposure to 50 mM K+ resulted in a transient increase in [Ca2+]i similar to that induced by ACh, but resistant to atropine. A high concentration of ACh (100μL 10-4M or 10-3M) induced [Ca2+]i oscillations in 11 out of 57 β-cells in the presence of 4.4 mM glucose. Using calcium channel blockers and Ca2+ free medium, the source of the increase in [Ca2+]i was deduced to be from extracellular spaces. Changing the temperature from 22 to 37°C did not affect the action of ACh on [Ca2+]i. These data strongly suggest that ACh exerted a direct action on [Ca2+]i in normal rat pancreatic β-cells and support a role for Ca2+ as a second messenger in the action of ACh.  相似文献

Recent investigations have provided significant evidence that many mesodermally derived tissues contain stem cell-like precursors capable of being stimulated to undergo differentiation into a variety of cellular lineages. We have recently reported that primary myoblasts isolated from 23-month-old mice have an increased adipogenic potential when compared to their 8-month-old counterparts. To further characterize the degree of adipocyte differentiation in these myoblasts, we examined early and late markers of adipocyte differentiation. Within the first 24h of adipocyte differentiation, expression of p130 and p107, two members of the retinoblastoma tumor suppressor gene family, are regulated and this event is an important one early in adipogenesis. Consistent with the increased adipogenic potential of the older myoblasts and in contrast to the younger cells, the p130:p107 pattern of expression is very similar to that observed in adipogenesis where there is a transient increase in p107 expression accompanied by a decrease in p130 expression. Interestingly, while these older cells accumulated lipid and expressed genes associated with lipid metabolism, they failed to express adipsin and leptin, two well-established markers of terminal adipocyte differentiation. These results suggest that older myoblasts are capable of initiating and progressing through the adipogenic program to a point where they express genes associated with lipid metabolism, but do not reach a terminally differentiated state. This finding may have important metabolic implications in the aging population.  相似文献

Lipid peroxidation in Laternula elliptica was assessed by detecting lipid radicals by electronic paramagnetic resonance. The values were compared with data from the temperate mud clam Mya arenaria. Lipid radical content was higher in the Antarctic bivalve than in the temperate mud clam, even within the range of its habitat temperature. The rate of generation of lipid radicals was affected by the iron content in the samples. The iron content in individual samples of digestive glands in L. elliptica ranged from 3 to 6 nmol g−1 fresh weight (fwt) and in M. arenaria from 0.6 to 2.7 nmol g−1 fwt. Arrhenius plots, developed from the rates obtained in the presence of 25 μM iron, showed no significant differences between the activation energy calculated for digestive glands of L. elliptica and M. arenaria. The Fe3+ reduction rate in L. elliptica was higher than in M. arenaria (4.7±0.9 vs. 1.8±0.4 nmol mg−1 protein min−1, respectively). L. elliptica had a higher content of α-tocopherol and β-carotene than M. arenaria. Our data suggest that increased lipid radical content in the membranes of cold-adapted organisms could be related to iron content.  相似文献

Paneth cells (PCs) are located at the base of small intestinal crypts and secrete the α‐defensins, human α‐defensin 5 (HD‐5) and human α‐defensin 6 (HD‐6) in response to bacterial, cholinergic and other stimuli. The α‐defensins are broad‐spectrum microbicides that play critical roles in controlling gut microbiota and maintaining intestinal homeostasis. Inflammatory bowel disease, including ulcerative colitis and Crohn's disease (CD), is a complicated autoimmune disorder. The pathogenesis of CD involves genetic factors, environmental factors and microflora. Surprisingly, with regard to genetic factors, many susceptible genes and pathogenic pathways of CD, including nucleotide‐binding oligomerization domain 2 (NOD2), autophagy‐related 16‐like 1 (ATG16L1), immunity‐related guanosine triphosphatase family M (IRGM), wingless‐related integration site (Wnt), leucine‐rich repeat kinase 2 (LRRK2), histone deacetylases (HDACs), caspase‐8 (Casp8) and X‐box‐binding protein‐1 (XBP1), are relevant to PCs. As the underlying mechanisms are being unravelled, PCs are identified as the central element of CD pathogenesis, integrating factors among microbiota, intestinal epithelial barrier dysfunction and the immune system. In the present review, we demonstrate how these genes and pathways regulate CD pathogenesis via their action on PCs and what treatment modalities can be applied to deal with these PC‐mediated pathogenic processes.  相似文献

摘要 目的:研究肺炎支原体肺炎(mycoplasma pneumoniae pneumonia, MPP)患儿血清可溶性共刺激分子B7-H3(soluble co-stimulatory molecule B7-H3, sB7-H3)含量与细胞因子水平及病情严重程度的相关性。方法:收集2019年3月至2020年6月期间我院收治的MPP患儿共96例,根据患儿病情严重程度分为轻症MPP组和重症MPP组,另选取同期于我院体检中心体检的健康儿童50例作为对照组。收集所有受试者的一般资料、主要临床表现、临床指标及细胞因子水平,对各指标进行Pearson相关性分析和多元逐步回归分析。结果:与对照组相比,MPP组患儿的白细胞计数(white blood cell, WBC)、中性粒细胞计数(neutrophil, NE)、红细胞沉降率(erythrocyta sedimentation rate, ESR)、C反应蛋白(C-reactive protein,CRP)、sB7-H3、粒细胞-巨噬细胞集落刺激因子(granulocyte-macrophage colony stimulating factor, GM-CSF)、干扰素-γ(interferon-γ, IFN-γ)、白介素-10(interleukin-10,IL-10)和白介素-17A(interleukin-17A,IL-17A)均较高(P<0.05);与轻症MPP组患儿相比,重症MPP组患儿的WBC、NE、ESR、CRP、sB7-H3、GM-CSF、IFN-γ、IL-10和IL-17A均较高(P<0.05)。Pearson相关性分析结果表明,sB7-H3与WBC、NE、ESR、CRP、GM-CSF、IFN-γ、IL-10和IL-17A呈正相关(P<0.05)。多元线性回归分析显示,GM-CSF(β=0.103,P<0.001)、IFN-γ(β=0.121,P<0.001)、IL-10(β=0.026,P<0.001)和IL-17A(β=0.093,P<0.001)是sB7-H3的独立影响因素。结论:MPP患儿血清sB7-H3、GM-CSF、IFN-γ、IL-10和IL-17A与MPP的病情严重程度密切相关,且sB7-H3的表达水平与GM-CSF、IFN-γ、IL-10和IL-17A的水平呈正相关。  相似文献
