We investigated the capacity of Mycoplasma arthritidis mitogen (MAM) to induce (a) expression of the inducible enzymes cyclo-oxygenase (COX-2) and nitric oxide synthase (iNOS), (b) production of prostaglandin E2 (PGE2) and nitric oxide (NO), and (c) involvement of platelet-activating factor (PAF) in the MAM-induced activation pathway. Resident peritoneal cells from C3H/HePas mice were incubated with MAM in the presence or absence of a PAF-antagonist (WEB2170) or COX-2 inhibitors (nimesulide or NS398). Enzyme expression was evaluated by immunoblotting, PGE2 by EIA, and NO by Griess reaction. Following MAM-stimulation of peritoneal cells, expression of COX-2 was detected at 3 h (peak levels at 12 h) and of iNOS at 6 h (peak levels at 20 h). PGE2 increased till 20 h, decreasing thereafter, whereas NO increased with time. WEB2170 (5 x 10(-5) M) treatment caused 44% inhibition of NO output and reduced iNOS expression (48% at the peak of expression). Concomitant treatment with WEB2170 and nimesulide (10(-5) M) reversed these inhibitory effects. WEB2170 reduced COX-2 expression (43% at the peak of expression) and prevented the decline in PGE2 levels after 20 h. These results suggest the involvement of PAF in the signaling pathway triggered by MAM that leads to expression of iNOS and COX-2, and show that PAF regulates the production of NO, possibly by controlling levels of PGE2.  相似文献

The crystal structures of the Vbeta17+ beta chains of two human T cell receptors (TCRs), originally derived from the synovial fluid (SF4) and tissue (C5-1) of a patient with rheumatoid arthritis (RA), have been determined in native (SF4) and mutant (C5-1(F104-->Y/C187-->S)) forms, respectively. These TCR beta chains form homo-dimers in solution and in crystals. Structural comparison reveals that the main-chain conformations in the CDR regions of the C5-1 and SF4 Vbeta17 closely resemble those of a Vbeta17 JM22 in a bound form; however, the CDR3 region shows different conformations among these three Vbeta17 structures. At the side-chain level, conformational differences were observed at the CDR2 regions between our two ligand-free forms and the bound JM22 form. Other significant differences were observed at the Vbeta regions 8-12, 40-44, and 82-88 between C5-1/SF4 and JM22 Vbeta17, implying that there is considerable variability in the structures of very similar beta chains. Structural alignments also reveal a considerable variation in the Vbeta-Cbeta associations, and this may affect ligand recognition. The crystal structures also provide insights into the structure basis of T cell recognition of Mycoplasma arthritidis mitogen (MAM), a superantigen that may be implicated in the development of human RA. Structural comparisons of the Vbeta domains of known TCR structures indicate that there are significant similarities among Vbeta regions that are MAM-reactive, whereas there appear to be significant structural differences among those Vbeta regions that lack MAM-reactivity. It further reveals that CDR2 and framework region (FR) 3 are likely to account for the binding of TCR to MAM.  相似文献

目的:研究支原体肺炎(MP)患者血清肺表面活性蛋白-A(SP-A)、肺表面活性蛋白-D(SP-D)与超敏C反应蛋白(hs-CRP)的变化趋势及临床意义。方法:选择从2015年9月到2016年9月在我院治疗的MP患者56例纳入本次研究,将其记为观察组,给予常规治疗后,有效者49例,无效者7例。另选同期在我院进行健康体检的志愿者55例作为对照组,对比两组血清SP-A、SP-D及hs-CRP的变化趋势,比较观察组不同治疗结果患者的血清SP-A、SP-D及hs-CRP水平,并分析患者的血清SP-A、SP-D与hs-CRP的相关性。结果:治疗后3d及治疗后7d观察组的血清SP-A、SP-D及hs-CRP水平较治疗前依次降低,但均高于对照组(P0.05),在治疗后14d血清SP-A、SP-D及hs-CRP水平较治疗前降低(P0.05),但与对照组比较差异无统计学意义(P0.05)。治疗后14 d有效组血清SP-A、SP-D及hs-CRP水平均明显低于无效组,差异有统计学意义(P0.05)。根据Spearman相关性分析结果显示,MP患者的血清SP-A、SP-D与hs-CRP均呈正相关(r=0.713、0.699,P=0.000、0.000)。结论:MP患者的血清SP-A、SP-D及hs-CRP水平可伴随治疗过程的进行而逐渐回落,最终基本恢复至正常范围,且血清SP-A、SP-D水平与hs-CRP水平呈正相关,临床治疗时可针对上述3种指标进行监测以判断患者病情及治疗效果。  相似文献

根据支原体16s rDNA序列,选择RemyTeyssou设计的三条寡核苷酸链,组成两套引物:P_(1-2a)能检测出细胞培养中常见的各种支原体,P_(1-2b)能检出无胆甾原体。反应可检出体系中10CFV的菌体。此法先用于对实验室人为污染支原体Vero细胞的检测,后与DNA 染色法和培养法比较,检测了49份生物样品,其中24份传代细胞,PCR检测的阳性率为58%,DNA染色法为42%,培养法为33%;三者的灵敏性比较,PCR可检出10~(-3)稀释度的阳性样品,高于其他两种方法。此PCR方法快速、灵敏、特异,适用于细胞培养中支原体污染的检测。  相似文献

Abstract Mycoplasmas are cell wall-less organisms that require membrane precursors for growth. Activities involved in the acquisition of these materials have been hypothesized as mycoplasmal virulence factors because of the effects these activities might have on host cells. Twenty-nine species or strains of mycoplasmas were examined for membrane-associated hemolysis activity similar to that previously identified in Mycoplasma pulmonis . Membrane-associated hemolytic activity was found in most mycoplasma species, but the amount of activity varied between and within the species. All of the arginine-utilizing mycoplasmal species, one M. pulmonis strain, one Acholeplasma species, and the intracellular human pathogens M. penetrans and M. fermentans ssp. incognitus were devoid of activity. The wide distribution of the membrane-associated hemolysis activity suggests that it may be important to the survival of the organism.  相似文献

目的:探讨莫西沙星治疗老年支原体肺炎患者的临床疗效。方法:收集我院收治的老年支原体肺炎患者80例,年龄60~79岁,随机分为对照组和实验组,每组各40例,两组患者均给予相应常规对症治疗,对照组患者给予红霉素治疗;实验组患者给予莫西沙星治疗。治疗结束后,比较两组患者的退热时间、住院时间、白细胞计数、C反应蛋白水平以及患者的临床总有效率。结果:与治疗前相比,两组患者治疗后的白细胞计数、C反应蛋白水平均显著下降(P0.05),且实验组患者退热时间以及住院时间较对照组显著缩短(P0.05),白细胞计数、C反应蛋白水平明显低于对照组(P0.05),而临床治疗总显著高于对照组(P0.05)。结论:莫西沙星能显著提高老年支原体肺炎患者的临床疗效。  相似文献

Building structural models of entire cells has been a long-standing cross-discipline challenge for the research community, as it requires an unprecedented level of integration between multiple sources of biological data and enhanced methods for computational modeling and visualization. Here, we present the first 3D structural models of an entire Mycoplasma genitalium (MG) cell, built using the CellPACK suite of computational modeling tools. Our model recapitulates the data described in recent whole-cell system biology simulations and provides a structural representation for all MG proteins, DNA and RNA molecules, obtained by combining experimental and homology-modeled structures and lattice-based models of the genome. We establish a framework for gathering, curating and evaluating these structures, exposing current weaknesses of modeling methods and the boundaries of MG structural knowledge, and visualization methods to explore functional characteristics of the genome and proteome. We compare two approaches for data gathering, a manually-curated workflow and an automated workflow that uses homologous structures, both of which are appropriate for the analysis of mesoscale properties such as crowding and volume occupancy. Analysis of model quality provides estimates of the regularization that will be required when these models are used as starting points for atomic molecular dynamics simulations.  相似文献

Cervicitis is predominantly caused by Neisseria gonorrhoeae and Chlamydia trachomatis, which accounts for almost half of all the cases of cervicitis. The role of newer organisms like Mycoplasma genitalium and Ureaplasma sp. and association of bacterial load with cervicitis are also not well established. So the study aimed to determine the relative frequency of these organisms and their load in association with cervicitis cases from north India. A case–control study involving 300 women was conducted using quantitative real-time PCR from endocervical swabs for identification of organisms and quantification of bacterial load. Among 150 cervicitis cases, C. trachomatis, N. gonorrhoeae, M. genitalium and Ureaplasma parvum were detected in 5 (3·3%), 10 (6·6%), 37(24·6%) and 47 (31·3%) respectively. Old age (<0·001, chi-squared test) and irregular menstrual cycles (<0·001, chi-squared test) were significantly associated with cervicitis. M genitalium was the only organism to be associated significantly with cervicitis with regard to age (<0·031) and symptoms like discharge (P < 0·033, chi-squared test) and dysuria (P < 0·044, chi-squared test) in multivariate analysis. Our finding suggests that the bacterial load of these organisms is not significantly associated with cervicitis. However, we found significant association of M. genitalium infection with clinical characteristics of cervicitis cases.  相似文献

Heat‐shock proteins (HSP) are molecular chaperones that play key roles in the maintenance of cellular homeostasis under variable environmental conditions. Although HSP are frequently used in studies of wild vertebrates as indicators of stress, no one to date has assessed responses of HSP60, HSP70, and HSP90 in the same species to different environmental stressors. We studied changes in the circulating concentrations of HSP60, HSP70, and HSP90 in wild‐caught House Finches (Haemorhous mexicanus) in response to multiple and sequential stressful environments, including high temperatures, transportation, and pathogen exposure. House Finches sampled during a period of low‐environmental stress with moderate ambient temperatures had low levels of HSP60 and modest levels of HSP70 and HSP90 compared to birds sampled during a presumably more stressful period with high temperatures. After exposure to high‐ambient temperatures, transportation in a vehicle, and exposure to Mycoplasma gallisepticum, captive finches were found to have increasingly higher levels of HSP60. HSP70 tended to rise in response to each stressor, but to drop in the weeks between stress challenges. HSP90 levels increased significantly only in response to pathogen challenge. Our observations suggest that HSP60 and HSP70 are indices of a range of stressors in House Finches, whereas HSP90 primarily reflects health state.  相似文献
