Summary Using an automated cell analyzer system, the effect of hepatocyte growth factor/scatter factor (HGF/SF), epidermal growth
factor (EGF), basic fibroblast growth factor (bFGF), endothelial acidic fibroblast growth factor (a-FGF), platelet derived
growth factor (PDGF), and recombinant human insulinlike growth factor (IGF) on the motility and morphology of Madin-Darby
canine kidney (MDCK), rat hepatomas, C2, and H5–6 and murine mammary carcinoma (EMT-6) cells was investigated. Treatment of MDCK cells with HGF/SF, bFGF, EGF, and a-FGF resulted
in an increase in average cell velocity and in the fraction of moving cells. Cells treated with the PDGF and IGF did not show
significant alterations in velocity. MDCK cells treated with each growth factor were classified into groups of “fast” and
“slow” moving cells based on their average velocities, and the average morphologic features of the two groups were quantitated.
Fast-moving cells had larger average area, circularity, and flatness as compared to slow-moving cells. Factors that stimulated
cell movement also induced alterations in cell morphologic parameters including spreading, flatness, area, and circularity.
HGF/SF also scattered and stimulated motility of C2 and H5–6 hepatoma cells. In contrast to MDCK cells, there was no significant difference between the morphology of the fast moving
and slow moving C2 and H5–6 cells. These studies suggest that growth factor cytokines have specific effects on motility of normal and tumor cells. 相似文献

Monthly trends shown by gonadosomatic indices, the prevalence of the different gonadal stages, and the size distribution of the oocytes, indicate that the large marine and commercially important plotosid Cnidoglanis macrocephalus spawns in Wilson Inlet between October and January. The conclusion that spawning occurs within this seasonally closed estuary was confirmed by the presence of males in large nests and by the capture of newly-hatched, yolk sac larvae from one of those nests. The fact that C. macrocephalus, which is also widely distributed in coastal marine waters throughout much of southern Australia, can spawn within Wilson Inlet would be of particular value to this species in those periods when closure of the estuary would preclude a seawards spawning migration. Sexual maturity is size dependent, with spawning rarely occurring before fish have reached a total length of 425 mm. Sexual maturity was attained by a few fish at the end of their second year, by several at the end of their third year and by most, if not all fish, at the end of their fourth year. Comparisons with data for the more northern and permanently open Swan Estuary indicate that C. macrocephalus also spawns within that system and that the spawning time of this species is related to water temperature. The adult male guards the larvae under its pelvic fins in burrows. The larvae increased in total length from 29 mm just after hatching to 43 mm in the 17–18 days after capture, during which time their yolk sac was resorbed. Details are given of the morphology, morphometrics, meristics and pigmentation of larval C. macrocephalus. In comparison with the larvae of three other plotosid genera, the larva of C. macrocephalus is far larger in size and more developed at hatching and takes a shorter time to transform into a juvenile. 相似文献

Meloidogyne sasseri n. sp. is described and illustrated from American beachgrass (Ammophila breviliffulata) originally collected from Henlopen State Park and Fenwick Island near the Maryland state line in Delaware, United States (6). Its relationship to M. graminis, M. spartinae, and M. californiensis is discussed. Primary distinctive characters of the female perineal pattern were a high to rounded arch with shoulders, widely spaced lateral lines interrupting transverse striations, a sunken vulva and anus, and coarse broken striae around the anal area. Second-stage juvenile body length was 554 μm (470-550), stylet length 14 μm (13-14.5), tail length 93 μm (83-115), tapering to a finely rounded terminus. Male stylet length 20 μm (19-21.5), spicule length 33 μm (30-36). Scanning electron microscope observations provided additional details of perineal patterns and face views of the female, male, and J2 head. Wheat, rice, oat, Ammophila sp., Panicum sp., bermudagrass, zoysiagrass and St. Augustinegrass were tested as hosts. Distribution of the species was the coasts of Delaware and Maryland. The common name "beachgrass root-knot" is proposed for M. sasseri n. sp. 相似文献

Three new benthic dinoflagellate species, Prorocentrum belizeanum, Prorocentrum elegans, and Prorocentrum caribbaeum, from mangrove floating detritus are described from scanning electron micrographs. Species were identified based on shape, size, surface micromorphology, ornamentation of thecal plates, and architecture of the periflagellar area and intercalary band. Cells of P. belizeanum are round to slightly oval with a cell size of 55–60 μm long and 50–55 μm wide. Areolae are round and numerous (853–1024 per valve) and range from 0.66 to 0.83 μm in size. The periflagellar area of P. belizeanum is a broad V-shaped depression; it accommodates a flagellar and an auxiliary pore and a flared, curved apical collar. The intercalary band of P. belizeanum is horizontally striated. Prorocentrum elegans is a small species 15–20 μm long and 10–14 μm wide, with an ovate cell shape. The thecal surface is smooth. Two sizes of valve pores were recognized: large, round pores (20–22 per valve) arranged in a distinct pattern and smaller pores situated in an array along the intercalary band. The periflagellar area is V-shaped; it accommodates an uneven sized flagellar pore, an auxiliary pore, and an angled protuberant flagellar plate. The intercalary band is transversely striated. It is a bloom-forming species. Prorocentrum caribbaeum cells are heart-shaped with a rounded anterior end and a pointed posterior end. Cells range from 40 to 45 μm long and 30 to 35 μm wide. Thecal surface has two different-sized pores: large, round pores (145–203 per valve) arranged perpendicularly from the posterior margins, and small, round pores unevenly distributed on the thecal surface. The periflagellar area is ornate. It is V-shaped with a curved apical collar located next to the auxiliary pore; a smaller protuberant apical plate is adjacent to the flagellar pore. DNA containing structures (nucleoids) were visualized by 4′, 6-diamidino-2-phenylindole (DAPI) fluorescent staining in two groups of cyanophytes (59 filamentous oscillatorialean species and 12 coccal Synechococcus-like organisms) to test the possibility of using nucleoid morphology in cyanophyte taxonomy. The morphology of nucleoids (size, shape, and structure) in oscillatorialean species is specific for individual families. The morphology of the nucleoid in Synechococcus-like species agrees from the proposed separation of the genus Cyanothece from Synechococcus. A much different nucleoid morphology in three species of Cyanothece suggests that these species should be separated into a new genus. On the basis of other characters, the species could be returned to the genus Cyanobacterium. My results indicate that the morphology of nucleoids is a valuable character in the classification of the cyanophytes examined; thus, it is a prospective feature that could be used in the taxonomy of other groups of cyanophytes. Additionally, DAPI staining is not a complicated procedure. The new character is easy to see in samples taken from nature, both living and preserved. 相似文献

Function and biological role of morphological specialization in desmognathine salamanders are analysed in the light of studies of feeding in Leurognatthus marmoratus. Nine morphological features uniquely characterize the Desmognathinae as compared to its sister group, the Plethodontinae, and other salamanders: (1) heavily ossified and strongly articulated skull and mandible; (2) flat, wedgelike head profile; (3) stalked occipital condyles; (4) modified atlas; (5) modified anterior trunk vertebrae; (6) atlanto-mandibular ligaments; (7) enlarged dorsal spinal muscles; (8) enlarged quadrato-pectoralis muscles; and (9) hind limbs relatively larger than forelimbs. Dorsoventral head mobility is increased at the atlanto-occipital joint by the stalked occipital condyles which simultaneously increase the mechanical advantage of the hypertrophied axial muscles that cross the joint. During head depression the atlanto-mandibular ligaments are placed in tension. Force generated by the quadrato-pectoralis muscles is transmitted directly to the mandible, creating a powerful bite with the jaws in full occlusion. Desmognathines use an efficient static pressure system for subduing and/or killing prey items held in the jaws, not a kinetic-inertial mechanism, as previously suggested. Leurognathus exhibits a behaviour ('head-tucking') unique to desmognathines that is consistent with the static-pressure hypothesis. Several desmognathine features (1, 2, 5, 7, 9) are not explicable as adaptations for feeding; these function as locomotory specializations for burrowing, especially for wedging under rocks within and alongside streams. Desmognathines use head-tucking during such wedging and burrowing movements, thus locomotory specializations act in concert with the feeding specializations. We suggest that origin of the atlanto-mandibular ligaments can be considered a 'key innovation' in that it allowed the secondary invasion of stream habitats by adults of ancestral desmognathines. 相似文献

The frequency and form of the middle trigonid crest (MTC) in lower permanent molars is reported for 1,131 dental casts of Bushman (San), Bantu, Solomons, Hawaiians, Pima, Eskimo, Navajo, Chinese, and American whites. The MTC occurs most often on the first molar. We found very little intra-trait variation, so observations were scored on a present-absent basis. The MTC is most frequent in the African samples and rare in those of the other populations. Many organisms show distinct morphological types. We argue that the evolution of these alternate morphologies depends upon both fitness differences between morphs within each sex and the genetic correlation between sexes. In this paper, we examine the evolution of alternate morphologies using wing dimorphism in insects as a model system. Many insect species are wing dimorphic, one morph having wings and being capable of flight, the other lacking functional wings. While there is a well established trade-off in females between macroptery and reproduction, there are few data on the possible costs in males. We examine trade-offs between macroptery and life-history traits in male sand crickets, Gryllus firmus, and estimate the genetic correlation of wing dimorphism between the sexes. Macropterous males develop faster than micropterous males and are either larger or the same size depending upon rearing conditions. There is no difference in absolute or relative testis size at eclosion or 7 d thereafter. Finally, there is no difference between macropterous and micropterous males in relative success at siring offspring. Thus, with respect to the above traits, there are no costs associated with being winged in male G. firmus. It is possible that there may be a trade-off between calling rate and macroptery. A comparison of the relative frequency of macroptery between males and female across different orders of insects supports this hypothesis. The genetic correlation of wing dimorphism between the sexes is high (r8 = 0.86), and hence the frequency of macroptery in males may be strongly influenced by selection acting on females. 相似文献

A new technique based on a fractal model has been developed for the quantification of the macroscopic morophology of mycelia. The morphological structuring is treated as a fractal object, and the fractal dimension, determined by an ultrasonic scattering procedure developed for the purpose, serves as a quantitative morphological index. Experimental observations reported earlier and simulations of mycelial growth, carried out using a probabilistic-geometric growth model developed for the purpose, both validate the applicability of the fractal model. In experiments with three different species, the fractal dimensions of pelletous structures were found to be in the range 1.45-2.0 and those of filamentous structures were in the range 1.9-2.7, with values around 2.0 representing mixed morphologies. Fractal dimensions calculated from simulated mycelia are in rough agreement with these ranges. The fractal dimension is also found to be relatively insensitive to the biomass concentration, as seen by dilution of the original broths. The relation between morphology and filtration properties of the broths has also been studied. The fractal dimension shows a strong correlation with the index of cake compressibility and with the Kozeny constant, two filtration parameters that are known to be morphology dependent. This technique could thus be used to develop correlations between the morphology, represented by the fractal dimension, and important morphology-dependent process variables.
