目的通过克隆分析中国地鼠16S基因的部分序列,对中国地鼠16S基因的结构和功能进行初步探索和揭示。方法从GenBank中已报道的啮齿动物16S基因保守区设计一对引物,进行PCR扩增,测序。用Blastn与GenBank中七种啮齿类动物的16S基因进行序列比较,分析其碱基组成及变异情况,并用邻接法(NJ)、非加权组平均法(UPGMA)构建分子系统树,在分子水平上探讨中国地鼠和其他啮齿类动物的进化关系,对中国地鼠的种属地位进行了进一步验证。结果获得了中国地鼠线粒体16S基因的部分序列,其碱基组成A、T、C、G分别为40.5%、24.5%、18.7%、16.3%,与其他七种啮齿类动物的碱基含量相比,各碱基含量基本相似。NJ进化树表明,中国地鼠、金黄地鼠与欧洲仓鼠先聚为一支,小鼠与大鼠先聚为一支,东方田鼠、台湾田鼠与东欧田鼠先聚为一支。结论中国地鼠和金黄地鼠的亲缘关系最近,与传统的分类地位基本吻合。  相似文献

In situ hybridization and northern/slot blot analyses were used to quantify the expression of calcyclin (2A9, 5B10), osteopontin (opn, secreted phosphoprotein, 2ar) and calmodulin mRNAs in mouse tissues that support pregnancy. High-to-moderate levels of the mRNAs of all three genes were detected at discrete locations in the uterus, decidua and placenta as a function of gestation time. Calmodulin expression was constant in these tissues; calcyclin mRNA was high during early pregnancy and declined after day 8-9 of gestation; and opn mRNA was undetectable before day 7, with maximal levels on days 9-12 in each of these tissues. Calcyclin, but not opn, expression was also observed in the chorioamnion after day 12. Calcyclin was expressed throughout the decidua on day 8 but became restricted to the primary (antimesometrial) decidual zone and decidua lateralis on day 9, and the decidua capsularis after day 9. By contrast, opn mRNA was localized on day 9 to the mesometrial triangle, which contains a large population of granulated metrial gland cells, and to the decidua basalis. These two genes may serve as markers for the two types of decidual tissue. We suggest that one function of OPN, which may be an indicator of cells in the decidua that have a bone marrow genealogy, is to mediate the flux of calcium from the maternal circulation to the developing embryo.  相似文献

Previous histological studies showed that in addition to a sinus node, an atrioventricular (AV) node, an AV bundle, left and right bundle branches, birds also possess a right AV‐Purkinje ring that is located in the atrial sheet of the right muscular AV‐valve along all its base length. The functionality of the AV‐Purkinje ring is unknown. In this work, we studied the topology of pacemaker myocytes in the atrial side of the isolated chicken spontaneously contracting right muscular AV‐valve using the method of microelectrode mapping of action potentials. We show that AV‐cells having the ability to show pacemaking reside in the right muscular AV‐valve. Pacemaker action potentials were exclusively recorded close to the base of the valve along its whole length from dorsal to the ventral attachment to the interventricular septum. These action potentials have much slower rate of depolarization, lower amplitude, and higher diastolic depolarization than action potentials of Purkinje (conducting) cells. 本文运用常规染色和Bernhard染色方法对切片标本中小鼠粗线期精母细胞联会复合体(SC)的超微结构和电镜细胞化学特点进行了研究。经常规染色后,可见SC由侧生组分(LE)、中央组分(CE)和L-C纤维组成;SC宽约210nm,LE宽约60nm,中央间隔区宽约90nm。在Bernhard染色标本中,SC的LE、CE和L-C纤维着色较深,说明其中含有RNA;SC各结构组分的宽度和形态特点与常规染色标本中的基本一致。本文讨论了SC中存在有RNA等问题。  相似文献

向非鸣禽家鸽(Columba livia domesticus)和鸣禽黄雀(Carduelis spinus)、黄喉鹀(Emberiza, elegans)耳蜗内注射 HRP作顺行追踪表明,耳蜗纤维组成第八脑神经的听支( N Ⅷ)后,分别投射至延髓的角状核(NA)和巨细胞核(NM),由 NA及 NM两个亚核组成耳蜗核(nucleus cochlea),它是听觉的上行通路中的第一级中继站,延髓层状核并不接受耳蜗纤维的直接投射。家鸽与黄雀、黄喉鹀之间的NA、NM及NⅧ在形态和分布上都有较明显的差别。  相似文献

A general assumption in quantitative genetics is the existence of an intermediate phenotype with higher mean individual fitness in the average environment than more extreme phenotypes. Here, we investigate the evolvability and presence of such a phenotype in wild bird populations from an eleven‐year experiment with four years of artificial selection for long and short tarsus length, a proxy for body size. The experiment resulted in strong selection in the imposed directions. However, artificial selection was counteracted by reduced production of recruits in offspring of artificially selected parents. This resulted in weak natural selection against extreme trait values. Significant responses to artificial selection were observed at both the phenotypic and genetic level, followed by a significant return toward preexperimental means. During artificial selection, the annual observed phenotypic response closely followed the predicted response from quantitative genetic theory ( = 0.96, = 0.56). The rapid return to preexperimental means was induced by three interacting mechanisms: selection for an intermediate phenotype, immigration, and recombination between selected and unselected individuals. The results of this study demonstrates the evolvability of phenotypes and that selection may favor an intermediate phenotype in wild populations.  相似文献

Accurate, quantitative characterization of complex shapes is recognized as a key methodological challenge in biology. Recent development of automated three‐dimensional geometric morphometric protocols (auto3dgm) provides a promising set of tools to help address this challenge. While auto3dgm has been shown to be useful in characterizing variation across clades of morphologically very distinct mammals, it has not been adequately tested in more problematic cases where pseudolandmark placement error potentially confounds interpretation of true shape variation. Here, we tested the sensitivity of auto3dgm to the degree of variation and various parameterization settings using a simulation and three microCT datasets that characterize mammal tooth crown morphology as biological examples. The microCT datasets vary in degree of apparent morphological differentiation, with two that include grossly similar morphospecies and one that includes two laboratory strains of a single species. Resulting alignments are highly sensitive to the number of pseudolandmarks used to quantify shapes. The degree to which the surfaces were downsampled and the apparent degree of morphological differentiation across the dataset also influenced alignment repeatability. We show that previous critiques of auto3dgm were based on poorly parameterized alignments and suggest that sample‐specific sensitivity analyses should be added to any research protocol including auto3dgm. Auto3dgm is a useful tool for studying samples when pseudolandmark placement error is small relative to the true differences between specimens. This method therefore represents a promising avenue forward in morphometric studies at a wide range of scales, from samples that differ by a single genetic locus to samples that represent multiple phylogenetically diverse clades.  相似文献

Commensal house mice have spread from their probable origin in north India, differentiating into a number of forms described variously as species, semispecies or subspecies. The different taxa can breed together and exchange genes but they retain their distinctiveness (although Mus ( musculus ) molossinus of Japan seems to be the result of a complete fusion between M.  ( m .)  musculus and M.  ( m .)  castaneus ). The most widespread form is M.  ( m. )  domesticus , which has successfully colonized every continent as a commensal, albeit with varying contributions from other Mus genomes. It has also been domesticated as the laboratory mouse. This means that the same genome is exposed to a wide variety of environments and gives tremendous opportunities for exploring the operation of different evolutionary mechanisms. Mice have accompanied evolutionary understanding from early Darwinian days – confirming Mendelian ratios and showing they applied in mammals, providing data on rates of evolution, and representing examples of dominance modification, differential survival, competition and other indicators of a struggle for existence. However, they have an unfulfilled potential to drive as well as to illuminate evolutionary theory – by revealing more about, for example, the interactions between gene flow and social determinants, constraints on introgression and physiological adjustments. This potential can be explored through our ever-deepening knowledge of the genome and molecular mechanisms, and by the application of new techniques, but its most effective agent will always be the cross-disciplinary synergy of visionary scientists like Julian Huxley, Charles Elton and Louis Thaler.  © 2005 The Linnean Society of London, Biological Journal of the Linnean Society , 2005, 84 , 335–347.  相似文献
