AIMS: The life cycle of the dimorphic fungus Mucor circinelloides was studied in a temperature-controlled flow-through cell, which constitutes an ideal tool when following the development of individual cells, with a view to understanding the growth and differentiation processes occurring in and between the different morphological forms of the organism. METHODS AND RESULTS: Mycelial growth and the transformation of hyphae into chains of arthrospores were characterized by image analysis techniques and described quantitatively. The influence of the nature (glucose and xylose) and concentration of the carbon source on specific growth rate and hyphal growth unit length were studied. The organism branched more profusely on xylose than on glucose while the specific growth rates determined were rather similar. Methods were developed to study the yeast-like growth phase of M. circinelloides in the flow-through cell, and combined with fluorescent microscopy which allowed new insights to bud formation. Additionally, numbers and distribution of nuclei in arthrospores, hyphae and yeasts were studied. CONCLUSIONS: The results give essential information on the morphological development of the organism. SIGNIFICANCE AND IMPACT OF STUDY: Development of any industrial process utilizing this organism will be dependent on the information obtained here for effective process optimization.  相似文献

The sesquiterpenes cadina-4,10(15)-dien-3-one (1) and aromadendr-1(10)-en-9-one (squamulosone) (14) along with the triterpenoid methyl ursolate (21) were incubated with the fungus Mucor plumbeus ATCC 4740. Substrates 1, 14 and ursolic acid (20) were isolated from the plant Hyptis verticillata in large quantities. M. plumbeus hydroxylated 1 at C-12 and C-14. When the iron content of the medium was reduced, however, hydroxylation at these positions was also accompanied by epoxidation of the exocyclic double bond. In total nine new oxygenated cadinanes have been obtained. Sesquiterpene 14 was converted to the novel 2alpha,13-dihydroxy derivative along with four other metabolites. Methyl ursolate (21) was transformed to a new compound, methyl 3beta,7beta,21beta-trihydroxyursa-9(11),12-dien-28-oate (22). Two other triterpenoids, 3beta,28-dihydroxyurs-12-ene (uvaol) (23) and 3beta,28-bis(dimethylcarbamoxy)urs-12-ene (24) were not transformed by the micro-organism, however.  相似文献

Using 5-fluoroorotic acid (5-FOA) as a positive selection system we isolated mutants of Mucor circinelloides altered in the pyrimidine biosynthetic pathway. These mutants were found to be deficient either in orotidine-5′-monophosphate decarboxylase (OMPdecase), or in orotate phosphoribosyltransferase (OPRTase) activity. Complementation tests among mutants lacking OPRTase activity classified them into three groups, thus suggesting the possibility of interallelic complementation. To investigate this hypothesis a cDNA clone corresponding to the OPRTase-encoding gene of M. circinelloides was isolated by direct complementation of E. coli. The genomic copy transformed to prototrophy one member of each of the three classes of OPRTase-deficient mutants. We therefore concluded that they were all altered at the same locus, the pyrF locus. The corresponding alleles were cloned and sequenced. Comparisons of the amino acid sequence of M. circinelloides OPRTase with those of E. coli and S. typhimurium revealed a high degree of similarity in secondary and tertiary structure. As the two bacterial enzymes exist as dimers, a homodimeric quaternary structure of the M. circinelloides mature protein can be assumed. This would also explain the interallelic complementation between some pyrF mutants. The mutations found could affect either the active site or the structure of the dimer interface of the OPRTase. The kinetics of the synthesis of triolein catalyzed by immobilized Mucor miehei lipase were studied. Equilibrium constants for the synthesis of mono-, di-, and triolein were calculated from the equilibrium compositions for different initial ratios of glycerol and oleic acid by means of multiresponse regression. The 1,3-specific lipase can catalyze the synthesis of triolein because the ester enzymatically formed with the primary alcohol isomerizes, through acyl migration, to an ester on the secondary hydroxyl. The freed primary hydroxyl may then undergo further enzymatic conversion. The rates of isomerization depend on the concentration of oleic acid. (c) 1993 Wiley & Sons, Inc.  相似文献

The distribution of F-actin microfilaments and microtubules was analyzed in germinating sporangiospores of Mucor rouxii by labeling with rhodamine-tagged phalloidin and by immunofluorescence microscopy. The transition from isodiametrical to apical growth was accompanied by a switch from uniform distribution of F-actin patches to a polarized accumulation of F-actin material at the germ tube tips. Immunoblotting of cell-free extracts of M. rouxii with a monoclonal anti-porcine -tubulin antibody (TU-01) disclosed two discrete bands of -tubulin suggesting the existence of two -tubulin genes in this fungus. Immunofluorescence microscopy of germinating cells stained with the same antibody revealed an elaborate network of cytoplasmic microtubules that persisted during the entire germination process and extended into the apex of the germ tube. The effect of the nucleoside-peptide antibiotics nikkomycin Z, nikkomycin X, and polyoxin A was tested on chitosomal chitin synthetase from yeast cells of the dimorphic fungus Mucor rouxii. The K i was 0.6 M for polyoxin A and 0.5 M for nikkomycin X; nikkomycin Z was slightly less inhibitory (K i=3.5M). Whereas the minimum inhibitory concentrations of the nikkomycins for growth and germination were quite low (about 1M, or lower), polyoxin A displayed no antifungal activity against yeast cells and sporangiospores of the test organism, even when present in high concentrations. 目的 总结与探讨中国大陆地区近25 a肺接合菌病的流行病学特点及诊疗情况.方法 检索分析重庆维普、CNKI系列数据库、万方数据库、中国医师协会全文期刊库及Pubmed等数据库中1988年以来的中国大陆地区肺接合菌病的相关文献资料.结果 共检索文献73篇,病例102例.患者的平均发病年龄为48.4岁,男女比例为2.46∶1,总死亡率为40.82%.我国肺接合菌病多见于糖尿病、HIV感染、病毒性肝炎、长期使用糖皮质激素和(或)免疫抑制剂等伴基础疾病的人群.其临床及影像学检查无特异性,诊断主要依赖于真菌学及组织病理学检查.治疗方法首选两性霉素B,联合手术治疗较之单独抗真菌药物疗效更好.结论 近年来中国大陆地区肺接合菌病发病率呈上升趋势.对高度怀疑肺接合菌感染的患者,宜尽早启动抗真菌治疗,及时联合病灶切除者疗效更佳.  相似文献

After Screening 100 micro-organisms to detect intracellular myrosinase, only Aspergillus niger produced myrosinase.

Enzyme production was induced by the addition of ten percent of a mustard extract* to the culture medium. The enzyme was produced in considerable amounts on the first and second day of cultivation. L-Ascorbic acid was an excellent carbon source.

The enzyme was unstable but was stabilized by coexistence with 2-mercaptoethanol (10?2 M) and ascorbic acid (10?3 M).  相似文献

Microorganisms that accumulated the eicosapentaenoic acid (EPA)-enriched triacylglycerol (TG), were screened for using yeast-malt medium containing 1% free EPA. The best strain was identified as Mucor hiemalis HA-30. The optimum culture conditions for the accumulation of EPA-enriched TG were : 3% soluble starch, 0.5% polypeptone, 0.3% yeast extract, 0.5% free EPA, and pH 6.0 at 25°C. After the cultivation, 1.77 mg/ml of the TG with EPA purity of 79% was obtained. The EPA content in TG increased in conjunction with the EPA content of the supplemented free fatty acids or ethyl esters. Free EPA were more efficiently incorporated than the ethyl esters. Trieicosapentaenoyl glycerol (EPA, EPA, EPA) accounted for 73% of total TGs.  相似文献

alpha-chymotrypsin (CT) and lipase (LP) were immobilized in hierarchically-ordered mesocellular mesoporous silica (HMMS) in a simple but effective way for the enzyme stabilization, which was achieved by the enzyme adsorption followed by glutaraldehyde (GA) crosslinking. This resulted in the formation of nanometer scale crosslinked enzyme aggregates (CLEAs) entrapped in the mesocellular pores of HMMS (37 nm), which did not leach out of HMMS through narrow mesoporous channels (13 nm). CLEA of alpha-chymotrypsin (CLEA-CT) in HMMS showed a high enzyme loading capacity and significantly increased enzyme stability. No activity decrease of CLEA-CT was observed for 2 weeks under even rigorously shaking condition, while adsorbed CT in HMMS and free CT showed a rapid inactivation due to the enzyme leaching and presumably autolysis, respectively. With the CLEA-CT in HMMS, however, there was no tryptic digestion observed suggesting that the CLEA-CT is not susceptible to autolysis. Moreover, CLEA of lipase (CLEA-LP) in HMMS retained 30% specific activity of free lipase with greatly enhanced stability. This work demonstrates that HMMS can be efficiently employed as host materials for enzyme immobilization leading to highly enhanced stability of the immobilized enzymes with high enzyme loading and activity.  相似文献
