A Raman scattering study of the channel-forming polyene antibiotic nystatin, is reported in the solid state, in organic and aqueous solutions as well as in phospholipid and phospholipid-cholesterol multilayers. Measurements of the solid and solution spectra as a function of excitation wavelengths in the 459.7–514.5 nm range, and the phospholipid spectra as a function of temperature in the 10–60°C range, have also been made. The spectral features indicate a pre-resonance-enhanced Raman spectrum in which the CC and CC stretching modes of the polyene segment of nystatin are dominant. However, in contrast to previously published results on the nearly isostructural polyene antibiotic amphotericin B, a line at 1610 cm?1 assignable to the CO stretching mode is also observed to be strongly resonance enhanced. This is explained by a postulated ground-state conformation model in which a twisting of the molecule is facilitated by the break in the polyene chain. This allows the CO group at one end of the molecule to be aligned along the polyene unit at the other end, and the CC stretching vibration, which is strongly modulated by the polyene π → π1 excited state, to mix with the CO stretching vibration. The peak frequencies and intensities of the CC and CC stretching modes in nystatin, however, remain essentially unchanged compared with amphotericin B, indicating that the polyene units in nystatin remain planar and trans both in the ground and excited states.The intensity of the CO mode with respect to the CC stretching mode was observed to vary appreciably with nystatin environment, indicating a  相似文献

Bow and arrow technology spread across California between ~AD 250 and 1200, first appearing in the intermountain deserts of the Great Basin and later spreading to the coast. We critically evaluate the available data for the initial spread in bow and arrow technology and examine its societal effects on the well‐studied Northern Channel Islands off the coast of Southern California. The introduction of this technology to these islands between AD 650 and 900 appears to predate the appearance of hereditary inequality between AD 900 and 1300. We conclude, based on the available data, that this technology did not immediately trigger intergroup warfare. We argue that the introduction of the bow and arrow contributed to sociopolitical instabilities that were on the rise within the context of increasing population levels and unstable climatic conditions, which stimulated intergroup conflict and favored the development of hereditary inequality. Population aggregation and economic intensification did occur with the introduction of the bow and arrow. This observation is consistent with the hypothesis that social coercion via intra‐group “law enforcement” contributed to changes in societal scale that ultimately resulted in larger groups that were favored in inter‐group conflict. We argue that the interplay between intra‐group “law enforcement” and inter‐group warfare were both essential for the ultimate emergence of social inequality between AD 900 and 1300.  相似文献

A specimen of the Chameleon sand tilefish Hoplolatilus chlupatyi was collected off Mozambique during a 2014 FAO Marine Ecosystem Survey by the R/V Dr. Fridtjof Nansen. Three additional larval specimens of H. chlupatyi were collected in the Mozambique Channel in 2008. These four specimens represent the first records of H. chlupatyi from the Western Indian Ocean, previously known to occur exclusively in the Western Pacific Ocean. Furthermore, the occurrence of the Indian tilefish Hoplolatilus fronticinctus, originally described from Mauritius, is reported for South Africa and Mozambique based on newly collected specimens. Identification of these species is based on genetic and morphological characteristics. Genetic, meristic and morphometric data of H. chlupatyi and H. fronticinctus are provided along with illustrations.  相似文献

The Ca2+-selective tetrameric Transient Receptor Potential Vanilloid 6 (TRPV6) channel is an inwardly rectifying ion channel. The constitutive current endures Ca2+-induced inactivation as a result of the activation of phospholipase C followed depletion of phosphatidylinositol 4,5-bisphosphate, and calmodulin binding. Replacing a glycine residue within the cytosolic S4-S5 linker of the human TRPV6 protein, glycine 516, which is conserved in all TRP channel proteins, by a serine residue forces the channels into an open conformation thereby enhancing constitutive Ca2+ entry and preventing inactivation. Introduction of a second mutation (T621A) into TRPV6G516S reduces constitutive activity and partially rescues the TRPV6 function. According to the recently revealed crystal structure of the rat TRPV6 the T621 is adjacent to the distal end of the transmembrane segment 6 (S6) within a short linker between S6 and the helix formed by the TRP domain. These results indicate that the S4-S5 linker and the S6-TRP-domain linker are critical constituents of TRPV6 channel gating and that disturbance of their sequences foster constitutive Ca2+ entry.  相似文献

A model based on the solution of the electrostatic potential for a geometry of three dielectric regions associated with a gramicidin A channel (GA) is presented. The model includes a cylindrical dielectric layer to represent the peptide backbone and dipole rings to account for dipolar side chains. Image potential and dipolar contributions for different orientations and positions along the channel are analyzed. The conductance of GA and two analogues obtained by substituting the amino acid at position 1 are studied. The numerical simulation reproduces experimental results (Barrett et al. 1986, Biophys J 49, 673–686) and supports the idea that electrostatic dipole-ion interactions are of primary importance in gramicidin channel function. Phosphorylation-dependent events have been shown to modulate the activity of several members of the mammalian CLC Cl channel gene family, including the inward rectifier ClC-2. In the present study we investigated the regulation of rat ClC-2 expressed in the TSA-201 cell line (a transformed HEK293 cell line that stably expresses the SV40 T-antigen) by protein kinases. Protein kinase A activation phosphorylated ClC-2 in vivo, whereas stimulation of protein kinase C with phorbol 12-myristate 13-acetate did not. In vitro labeling studies confirmed that protein kinase A could directly phosphorylate ClC-2, and that protein kinase C and Ca2+/calmodulin-dependent protein kinase II did not. Nevertheless, protein kinase A-dependent phosphorylation of CLC-2 failed to regulate either the magnitude or the kinetics of the hyperpolarization-activated Cl currents. Considered together, we demonstrate that protein kinase A activation results in the phosphorylation of rat ClC-2 in vivo, but this event is independent of Cl channel activity. From boxcore and dredge samples taken along a depth transect across the Faroe Shetland Channel (FSC) in 1997 and 1999, molluscs were sorted and identified. On the basis of their numerical abundance, the stations could be clustered into three groups, reflecting the depth extension of the main water masses in the FSC. Between 400 and 500 m depth, both the number of taxa per sample and the number of specimens, were highest. Bivalves were mainly caught by the boxcorer and the dredge was best suited to collect the epifaunal gastropods. According to the literature, at mid-slope depths, resuspension is increased by the action of internal waves, which leads to the development of intermediate nepheloid layers. This increased concentration of suspended matter may serve as a food source of low quality. Likewise the low amounts of suspended matter with a relatively high quality of particulate organic matter, such as from ∼400 and ∼700 m may act similarly and explain the relative high abundance of filter feeders at these depths. However, in the various multivariate analyses we could not distinguish the fauna from these depths from that collected from other sites. We observed, however, a pattern in the depth distribution of feeding guilds across the slope of the FSC. Deposit feeders demonstrated a trend of decreasing numbers with depth. Filter-feeding taxa were found to have peaks at depths of approximately 350 m, between 500 and 600 m and between 700 and 800 m. This observation supports the idea that the fauna at the deep sites reflects the locally high cross-slope currents and the subsequent increased amounts or availability of food with a better quality, which seems to be related to resuspension events.  相似文献

Comparisons were undertaken to investigate cost‐effective methods of implementing the enzyme‐linked immunosorbent assay (ELISA) for sporozoite determination in anophelines when large numbers require processing. Comparisons between ELISA plate reader and visual assessments were performed with Anopheles funestus and Anopheles gambiae s.l. (Diptera: Culicidae), as were comparisons between whole‐body mosquito samples, heads and thoraces, and abdomens alone. Rates obtained from pools of five or 10 mosquitoes were compared with those for individual mosquitoes, as were rates obtained using different sampling methods. A total of 41 792 An. funestus and 9431 An. gambiae s.l. collected in light traps, and 22 323 An. funestus and 6860 An. gambiae s.l. from exit collections were analysed. Visual assessments gave results similar to those of machine readings. Sporozoite rates were similar in both species, as were rates by collection method. The use of whole mosquitoes increased estimates of infection rate by 0.6%. Pool size did not affect infection rates of An. gambiae s.l., but rates were higher among individually tested An. funestus than among those tested in pools. For large‐scale surveys, the use of whole mosquitoes in pools of 10 mosquitoes, with correction for overestimation, and the noting of results according to a simple three‐stage visual assessment of positivity is the most cost‐effective approach and is sufficient to obtain reliable data for comparative purposes.  相似文献
