The influence of fatty acids on model cholesterol/phospholipid membranes   总被引:1,自引:0,他引:1  
The aim of this work was to verify the influence of the saturated (SFA) (stearic acid) and the unsaturated (UFA) (oleic and alpha-linolenic) fatty acids on model cholesterol/phospholipid membranes. The experiments were based on the Langmuir monolayer technique. Cholesterol and phospholipid were mixed in the molar ratio that corresponds to the proportion of these lipids in the majority of natural human membranes. Into the binary cholesterol/phospholipid monolayers, various amounts of fatty acids were incorporated. Our investigations were based on the analysis of the interactions between molecules in ternary (cholesterol/phospholipids/fatty acid) mixtures, however, also binary (cholesterol/fatty acid and phospholipids/fatty acid) mixed system were examined. It was concluded that the influence of the fatty acids on model cholesterol/phospholipid membrane is closely connected with the shape of the fatty acid molecule, resulting from the saturation degree of the hydrocarbon chain. It was found that the saturated fatty acid makes the model membrane more rigid, while the presence of unsaturated fatty acid increases its fluidity. The increasing amount of stearic acid gradually destabilizes model membrane, however, this effect is the weakest at low content of SFA in the mixed monolayer. Unsaturated fatty acids in a small proportion make the membrane thermodynamically more stable, while higher content of UFA decreases membrane stability. This explains low proportion of the free fatty acids to other lipids in natural membrane.  相似文献

Many weeds interfere with the productivity of wheat and other winter annual crops; however, one common weed Medicago denticulata, appears to have a beneficial effect. An experiment was conducted at Agricultural Experimental Farm of the Indian Statistical Institute, Giridih, Jharkhand to observe the effect of M. denticulata (a legume) on different crops yield of barley (Hordeum vulgare Sensu lato), linseed (Linum usitatissimum L.), Indian mustard [Brassica juncea (L.) Czernj. & Cosson] and wheat (Triticum aestivum L.). The commensalisms interaction was studied, in which one species benefits, while the other is unaffected, between the crops, M. denticulata and other weeds, to observe the role of medicago as biological control agent of other weeds for enhancement of crop productivity. Further, a statistical test was performed to establish the effect of M. denticulata and weed infestations based on one-step Markov structure of the covariance matrix of the biomass. This permitted development of a mathematical model of a crop–M. denticulata–other weeds system and their interactions through direct and indirect competition for resources. Crops yield decreased considerably due to the presence of other weeds, while it increased due to introduction of medicago and hence medicago acted as a controlling agent in a crop–M. denticulata–other weeds system. Our mathematical analysis supported the experimental findings. The model parameters are estimated through a nonlinear least square and stochastic differential equations. Moreover, some thresholds for the growth of medicago population came out from our analysis that may be used as a biological control threshold for the enhancement of crop productivity.  相似文献

Tamoxifen (TAM) is a non-steroidal antiestrogen drug, which is widely used to prevent and treat breast, liver, pancreas and brain cancers. The present work investigates, in detail, the concentration dependent behavior of TAM (varying from 1 mol% to 45 mol%) on membrane fluidity. The differential scanning calorimetry (DSC) studies showed that tamoxifen eliminates the pre-transition and decreases the main phase transition to lower temperatures. Using visible spectroscopy at 440 nm and Fourier transform infrared (FTIR) spectroscopy it was found that membrane dynamics decreases for 1 and 3 mol% tamoxifen in both the gel and liquid crystalline phases. Above these concentrations up to 18–24 mol%, it increases and reaches its maximum values. As tamoxifen concentration was further increased, the membrane dynamics is found to be gradually decreased, although TAM still has fluidifying effect in comparison to pure phospholipid membrane. These findings are important for the effective use of tamoxifen in the cancer therapy to eliminate its dose dependent side effects reported in the literature.  相似文献

The origin of variation in animal growth rate and body size is not well understood but central to ecological and evolutionary processes. We develop a relationship that predicts the change in relative body size variation within a cohort will be approximately equal to the relative change in mean per unit size growth rate, when only size-dependent factors affect growth. When modeling cohort growth, relative size variation decreased, remained unchanged, or increased, as a function of growth rate-size scaling relationships, in a predictable manner. We use the approximation to predict how environmental factors (e.g., resource level) affect body size variation, and verified these predictions numerically for a flexible growth model using a wide range of parameter values. We also explore and discuss the assumptions underlying the approximation. We find that factors that similarly affect mean growth rate may differently affect size variation, and competition may increase body size variation without changing size-independent relationships. We discuss implications of our results to the choice of growth equations used in models where body size variation is an important variable or output.  相似文献

Most quadrupeds walk with lateral sequence (LS) gaits, where hind limb touchdowns are followed by ipsilateral forelimb touchdowns. Primates, however, typically walk with diagonal sequence (DS) gaits, where hind limb touchdowns are followed by contralateral forelimb touchdowns. Because the use of DS gaits is nearly ubiquitous among primates, understanding gait selection in primates is critical to understanding primate locomotor evolution. The Support Polygon Model [Tomita, M., 1967. A study on the movement pattern of four limbs in walking. J. Anthropol. Soc. Nippon 75, 120-146; Rollinson, J., Martin, R.D., 1981. Comparative aspects of primate locomotion, with special reference to arboreal cercopithecines. Symp. Zool. Soc. Lond. 48, 377-427] argues that primates' use of DS gaits stems from a more caudal position of the whole-body center of mass (COM) relative to other mammals. We tested the predictions of the Support Polygon Model by examining the effects of natural and experimental variations in COM position on gait mechanics in two distantly related primates: fat-tailed dwarf lemurs (Cheirogaleus medius) and patas monkeys (Erythrocebus patas). Dwarf lemur experiments compared individuals with and without a greatly enlarged tail (a feature associated with torpor that can be expected to shift the COM caudally). During patas monkey experiments, we experimentally shifted the COM cranially with the use of a weighted belt (7-12% of body mass) positioned above the scapulae. Examination of limb kinematics revealed changes consistent with systematic deviations in COM position. Nevertheless, footfall patterns changed in a direction contrary to the predictions of the Support Polygon Model in the dwarf lemurs and did not change at all in the patas monkey. These results suggest that body mass distribution is unlikely to be the sole determinant of footfall pattern in primates and other mammals.  相似文献

Cone photoreceptors function under daylight conditions and are essential for color perception and vision with high temporal and spatial resolution. A remarkable feature of cones is that, unlike rods, they remain responsive in bright light. In rods, light triggers a decline in intracellular calcium, which exerts a well studied negative feedback on phototransduction that includes calcium-dependent inhibition of rhodopsin kinase (GRK1) by recoverin. Rods and cones share the same isoforms of recoverin and GRK1, and photoactivation also triggers a calcium decline in cones. However, the molecular mechanisms by which calcium exerts negative feedback on cone phototransduction through recoverin and GRK1 are not well understood. Here, we examined this question using mice expressing various levels of GRK1 or lacking recoverin. We show that although GRK1 is required for the timely inactivation of mouse cone photoresponse, gradually increasing its expression progressively delays the cone response recovery. This surprising result is in contrast with the known effect of increasing GRK1 expression in rods. Notably, the kinetics of cone responses converge and become independent of GRK1 levels for flashes activating more than ∼1% of cone pigment. Thus, mouse cone response recovery in bright light is independent of pigment phosphorylation and likely reflects the spontaneous decay of photoactivated visual pigment. We also find that recoverin potentiates the sensitivity of cones in dim light conditions but does not contribute to their capacity to function in bright light.  相似文献

We present the first in silico model of the weak binding actomyosin in the initial powerstroke state, representing the actin binding-induced major structural changes in myosin. First, we docked an actin trimer to prepowerstroke myosin then relaxed the complex by a 100-ns long unrestrained molecular dynamics. In the first few nanoseconds, actin binding induced an extra primed myosin state, i.e. the further priming of the myosin lever by 18° coupled to a further closure of switch 2 loop. We demonstrated that actin induces the extra primed state of myosin specifically through the actin N terminus-activation loop interaction. The applied in silico methodology was validated by forming rigor structures that perfectly fitted into an experimentally determined EM map of the rigor actomyosin. Our results unveiled the role of actin in the powerstroke by presenting that actin moves the myosin lever to the extra primed state that leads to the effective lever swing.  相似文献

在高功率毫米波辐照新鲜皮肤组织加热时,实验测得的表面温升比表面绝热Pennes模型计算温升低很多,提出对于含水量较大且易从皮肤表面蒸发的组织,生物传热的Pennes方程必须考虑水分蒸发失热的因素.我们将蒸发失热项加入Pennes传热方程中,并对这样的理论模型作了一些探讨,在长时间毫米波辐照加热时,表面温度最终会趋于饱和平衡.  相似文献

目的建立一种稳定可靠的内膜下血管成形术(subintimal angioplasty,SIA)的动物模型,进而探索术后血管壁有关组成成分的变化规律。方法以巴马猪为实验对象,一组(7只)先建立动脉硬化狭窄模型,另一组(9只)为正常动脉,在颈总动脉施行SIA,在术后不同时间观察造模段动脉的通畅率及病理变化等。结果手术操作成功率为93.8%(15/16)。SIA术后总的通畅率为53.3%(8/15)。病理组织学观察发现SIA术后动脉管壁有新内膜形成,中膜增厚,平滑肌细胞数量增多,较多泡沫细胞出现,胶原纤维及弹力纤维染色范围增大、染色密度增加。超微结构观察提示SIA术后中膜层平滑肌细胞的收缩型与合成型同时存在;细胞外基质中胶原纤维和弹力纤维含量异常丰富。结论①首次成功设计、构建内膜下血管成形术的动物模型。②内膜下血管成形术模型构建术后的病理组织学改变与腔内血管成形术后的变化基本一致。  相似文献
