In the temperate forests of the southern Andes, Nothofagus pumilio, the dominant species of the most extensive forest type, experiences severe defoliation caused by caterpillars of the Ormiscodes genus (Lepidoptera: Saturniidae). This study uses tree rings to reconstruct the history of Ormiscodes outbreaks for the 1850–2005 period and examines relationships between outbreaks and climate variability. We used local climate records to compare outbreak–climate relationships in the northern Patagonian Andes (c. 41°S) and the cooler southern Patagonian Andes (c. 49°S). We also examined relationships between outbreak events and regional climate variability driven by variability in the Southern Annular Mode (SAM) and the El Niño‐Southern Oscillation. Although relationships between Ormiscodes outbreaks and climate proved to be complex, in northern Patagonia defoliation events are associated with drier and warmer than average growing seasons. Warming and drying trends in Patagonia during the latter part of the 20th century have been linked to a positive trend in SAM. During the post‐1976 period of accelerated warming in Patagonia, widespread defoliation outbreaks have occurred in both northern and southern Patagonia but the increase in frequency of events has been greater in the south. In southern Patagonia the increases in frequency of outbreaks in the late 20th century appear to be unprecedented over the c. 150 year tree‐ring record of reconstructed outbreaks. These results are consistent with the greater magnitude of recent warming in southern Patagonia, and suggest that under predicted warmer and drier climates in the 21st century, defoliator outbreaks may continue to increase in frequency. This study is the first systematic reconstruction of past insect outbreaks in South America and provides a preliminary understanding of how climate variability affects defoliator outbreaks in Patagonian Nothofagus forests. 相似文献

In the present article we examine the antiplasmodial activities of novel quinolone derivatives bearing extended alkyl or alkoxy side chains terminated by a trifluoromethyl group. In the series under investigation, the IC50 values ranged from 1.2 to approximately 30 nM against chloroquine-sensitive and multidrug-resistant Plasmodium falciparum strains. Modest to significant cross-resistance was noted in evaluation of these haloalkyl- and haloalkoxyquinolones for activity against the atovaquone-resistant clinical isolate Tm90-C2B, indicating that a primary target for some of these compounds is the parasite cytochrome bc1 complex. Additional evidence to support this biochemical mechanism includes the use of oxygen biosensor plate technology to show that the quinolone derivatives block oxygen consumption by parasitized red blood cells in a fashion similar to atovaquone in side-by-side experiments. Atovaquone is extremely potent and is the only drug in clinical use that targets the Plasmodium bc1 complex, but rapid emergence of resistance to it in both mono- and combination therapy is evident and therefore additional drugs are needed to target the cytochrome bc1 complex which are active against atovaquone-resistant parasites. Our study of a number of halogenated alkyl and alkoxy 4(1H)-quinolones highlights the potential for development of "endochin-like quinolones" (ELQ), bearing an extended trifluoroalkyl moiety at the 3-position, that exhibit selective antiplasmodial effects in the low nanomolar range and inhibitory activity against chloroquine and atovaquone-resistant parasites. Further studies of halogenated alkyl- and alkoxy-quinolones may lead to the development of safe and effective therapeutics for use in treatment or prevention of malaria and other parasitic diseases. 相似文献

Recent arguments for conventional fisheries management approaches (CFMAs) and against no-take marine protected areas (NTMPAs)
are reviewed, i.e. CFMAs are more effective, density-dependent factors will lead to reduced fish stock production in and around
NTMPAs, rights-based approaches in combination with CFMAs will be more effective, and natural refuges from fishing already
exist. It is concluded that these are largely valid but only from a fisheries management perspective. The arguments of proponents
of NTMPAs and those of proponents of CFMAs are considered as contrasting storylines, the divergences between which are based
on two key factors: different objectives and different science. In relation to different objectives, it is concluded that
the arguments against NTMPAs based on their lack of fisheries management benefits must be considered as only applying to the
secondary resource conservation objectives of such designations and not to the primary marine biodiversity conservation objectives.
On this basis it is argued that it is counter-productive for NTMPAs to be ‘sold’ on a win–win basis, including their potential
to deliver fisheries management benefits, as this detracts from their marine biodiversity conservation objectives and leaves
such calls open to arguments that CFMAs are better able to deliver fisheries management objectives. In relation to different
science, it is concluded that criticisms of NTMPAs and support for CFMAs implicitly resist the shift from Mode 1 (reductive,
intra-disciplinary) to Mode 2 (holistic, trans-disciplinary) science that is inherent in calls for NTMPAs as part of an ecosystem
approach. Mode 2 science attempts to accommodate both uncertainty and wider societal values and preferences, and it is argued
that arguments for NTMPAs should be more explicitly focussed on this potential. It is difficult, if not impossible and inappropriate,
to extend the reductive approach inherent in CFMA analyses to encompass the broader ethical and scientific concerns for the
health of marine ecosystems and their component populations and habitats that arguments for NTMPAs reflect. NTMPA proponents
might focus on stressing that arguments against such designations and in favour of CFMAs do not encompass such valid concerns,
therefore they tell only half of the story.

Diversity and functional significance of C-terminal sequences have been appreciated from studies of PDZ and PEX domains. Signaling 14-3-3 protein signaling by recognizing phosphorylated peptides plays a critical role in a variety of biological processes, including oncogenesis. The preferential binding of 14-3-3 to phosphorylated C-terminal sequences, mode III, provides a means of regulated binding and considerably expands the substrate repertoire of 14-3-3 interaction partners. 相似文献

Background
The tip of an optical fiber has been considered an attractive platform in Biology. The simple cleaved end of an optical fiber can be machined, patterned and/or functionalized, acquiring unique properties enabling the exploitation of novel optical phenomena. Prompted by the constant need to measure and manipulate nanoparticles, the invention of the Scanning Near-field Optical Microscopy (SNOM) triggered the optimization and development of novel fiber tip microfabrication methods. In fact, the fiber tip was soon considered a key element in SNOM by confining light to sufficiently small extensions, challenging the diffraction limit. As result and in consequence of the newly proposed “Lab On Tip” concept, several geometries of fiber tips were applied in three main fields: imaging (in Microscopy/Spectroscopy), biosensors and micromanipulation (Optical Fiber Tweezers, OFTs). These are able to exert forces on microparticles, trap and manipulate them for relevant applications, as biomolecules mechanical study or protein aggregates unfolding.Scope of review
This review presents an overview of the main achievements, most impactful studies and limitations of fiber tip-based configurations within the above three fields, along the past 10?years.Major conclusions
OFTs could be in future a valuable tool for studying several cellular phenomena such as neurodegeneration caused by abnormal protein fibrils or manipulating organelles within cells. This could contribute to understand the mechanisms of some diseases or biophenomena, as the axonal growth in neurons.General significance

Three diploid taxa – Hieracium transylvanicum (subgenus Hieracium), Hieracium caespitosum subsp. brevipilum and Hieracium pavichii (subgenus Pilosella) – from five natural Bulgarian populations were investigated embryologically. The peculiarities of the male and female gametophytes, embryo- and endospermogenesis were established in each species. The results suggest that the species propagate sexually as expected from their diploid chromosome number. However, some forms of apomixis have also been observed, e.g. somatic apospory and integumental embryony in H. pavichii. The presence of apomixis in a diploid taxon shows that polyploidy is not an obligatory prerequisite for apomixis and the two phenomena are independent. The embryological plasticity detected in H. pavichii and H. transylvanicum suggests they may have higher opportunities for adaptation and successful reproduction. 相似文献

An investigation was undertaken to understand the mechanism(s) by which chitosan exerts its antifungal effects against the wood-degrading fungi Sphaeropsis sapinea and Trichoderma harzianum. Exposure to increasing concentrations of chitosan caused an increase in the amount of hydrogen peroxide accumulation in cultures of S. sapinea, which was accompanied by a decrease in superoxide formation. The same effect was not observed in T. harzianum. Potassium ion leakage was an early event for both test fungi, leakage being more pronounced for S. sapinea than T. harzianum for the first 5 min, particularly at higher concentrations of chitosan treatment. Fluorescence microscopy provided evidence that the effect of chitosan on fungal hyphae was mediated through alterations in the plasma membrane properties. Chitosan also severely affected fungal morphology. Increasing concentrations of chitosan induced excessive branching, vacuolation, and a reduction in hyphal diameter. Transmission electron microscopy, which showed more severe ultrastructural changes in S. sapinea hyphae from chitosan treatment as compared to T. harzianum, provided valuable complementary information. The data suggest that the plasma membrane may be the primary target of chitosan action, and that the two fungi differ in the extent to which they are affected. 相似文献

In this paper, the nitrogen atom was inserted into the anthracycline system of the isocryptolepine nucleus to obtain the “Aza”-type structure benzo[4,5]imidazo[1,2-c] quinazoline. A series of “Aza”-type derivatives were designed, synthesized and evaluated for their antifungal activity against six plant fungi in vitro. Among all derivatives, compounds A-0, B-1 and B-2 showed significant antifungal activity against B. cinerea with the EC50 values of 2.72 μg/mL, 5.90 μg/mL and 4.00 μg/mL, respectively. Compound A-2 had the highest activity against M. oryzae with the EC50 values of 8.81 μg/mL, and compound A-1 demonstrated the most control efficacy against R. solani (EC50, 6.27 μg/mL). Moreover, compound A-0 was selected to investigate the in vivo tests against B. cinerea and the results indicated that the preventative efficacy of it up to 72.80% at 100 μg/mL. Preliminary mechanism studies revealed that after treatment with A-0 at 5 µg/mL, the B. cinerea mycelia appeared curved, collapsed and the cell membrane integrity may be damaged. The reactive oxygen species production, mitochondrial membrane potential and nuclear morphometry of mycelia have been changed, and the membrane function and cell proliferation of mycelia were destroyed. Compounds A-0, A-1, B-1 and B-2 presented weaker toxicities against two cells lines than isocryptolepine. This study lays the foundation for the future development of isocryptolepine derivatives as environmentally friendly and safe agricultural fungicides. 相似文献

目的 探讨不同分娩方式对晚期早产儿肠道菌群定植的影响。方法 以胎龄(周)为340/7~366/7的15例晚期早产儿为研究对象,根据分娩方式分为自然分娩组(n=8)和剖宫产组(n=7)。收集早产儿出生后3 d、7 d、14 d的粪便标本,应用高通量测序技术对细菌16S rRNA可变区中的V4区进行测序,分析肠道菌群多样性及组成结构。结果 (1)自然分娩组晚期早产儿粪便标本菌群多样性指数逐渐上升,剖宫产组的多样性指数较平稳,两组相比差异无统计学意义;(2)45份粪便标本中共检测出10个菌门,均以变形菌门、厚壁菌门、放线菌门和拟杆菌门为优势菌门,两组晚期早产儿生后变形菌门、拟杆菌门所占比例逐渐降低,厚壁菌门、放线菌门呈增多趋势。两组相比,剖宫产组7 d、14 d时拟杆菌门的相对丰度显著低于自然分娩组(Z=‒2.896,P=0.004;Z=‒2.120,P=0.040),变形菌门相对丰度仅在7 d时显著高于自然分娩组(Z=‒2.190,P=0.030);(3)两组研究对象中,除自然分娩组14 d时以双歧杆菌属为优势菌属外,余下均以肠杆菌属为优势菌属。相比于自然分娩组,在7 d时剖宫产组拟杆菌属所占比例显著降低(Z=‒2.806,P=0.005),肠杆菌属所占比例显著升高(Z=‒2.199,P=0.030)。结论 剖宫产能显著影响婴儿早期肠道菌群的定植,降低肠道中早期拟杆菌的水平。 相似文献
