烟草抗CMV突变体的抗病性分析 总被引:2,自引:0,他引:2
在突变体植株“Ea201主、Ea201侧、Aa201侧”的5~6叶期接种CMV汁液,并按优选法去除病症最严重的病株,6周后将剩余植株分别移栽到田间露地和防虫网内,到盛花期分别统计植株的感病情况。其结果为:在田间露地和防虫网内,①感病的对照品种的植株高度只及健康的对照植株高度的30%和40%,突变体的则可达到70%和80%;②对照品种的植株感CMV的病情指数可达90%和50%,突变体的则只有30%和3%;③对照品种植株感TMV的病情指数可达60%和30%,突变体植株感TMV的病情指数只有30%和10%;④突变体植株群体中有症状回复现象,同时其育性不受任何影响。从而表明所筛选到的烟草突变体“Ea201主、Ea201侧、Aa201侧”对CMV、TMV的相对抗性较好,具有较高的抗病性。 相似文献

本研究构建丙型肝炎病毒(HCV)糖蛋白E2的N-糖基化位点定点突变体。采用高保真性的Pfx DNA聚合酶,设计两对引物,分别引入两个突变位点,通过PCR体外定点突变,使E2第535、583位核苷酸由A突变为T,从而使AAC编码的天冬酰氨突变为TAC编码的酪氨酸,使得N-糖苷化位点NNT、NST突变为YNT、YST.结果得到两个单位点以及一个双位点突变体,并将突变型E2连接到真核表达载体peDNA3.1(-)/Myc—HisB上。成功获得的3个HCVE2糖蛋白糖基化位点定点突变体,为进一步进行HCVE2糖蛋白糖基化位点与分子伴侣之间的相互关系以及突变体对机体的免疫功能的影响的研究奠定了基础。 相似文献

研究了一个在田间发现的水稻叶绿素b含量低的突变体叶片的光合功能衰退。和野生型相比,突变体的光合功能在叶片一生中较稳定;超氧阴离子和H2O2含量低,但活性氧清除系统的SOD和CAT酶活性差异不显著。由于突变体叶片中还原态辅酶Ⅱ和氧化态辅酶Ⅱ的比值低于野生型,因而认为其光合电子传递速率和羧化反应消耗电子速率之间比野生型更加平衡,是其光合功能较野生型稳定的原因。 相似文献

The first complete mitochondrial genome of dobsonfly Protohermes concolorus Yang et Yang, 1988 (Megaloptera: Corydalidae) was sequenced in this study. The genome was a circular molecule of 15,851 bp
containing the typical 37 genes that arranged in the same order as that of the putative ancestor of hexapods. Sequences overlaps
were observed between several neighbor genes, which made the genome relatively compact. The tRNA-Ser (GCT) could not be folded
into typical secondary structure because its DHU arm was replaced with a simple loop. Six of the 13 protein genes were terminated
with a single T adjacent to a downstream tRNA gene in the same strand. The variation of GC content caused the different nucleotide
substitution patterns of the protein genes. The genome was AT-biased with a total A + T content of 75.83% which was also demonstrated
by the codon usage. The control region was the most AT-rich region with a sub-region of even higher A + T content. Protein
genes of two strands presented opposite CG-skew trends which was also reflected by the codon usage. For most of the amino
acids, the protein coding sequences did not prefer to use the cognate codons of corresponding tRNAs and the codon usage of
the protein genes was not random. The variation of nucleotide substitution patterns of protein genes was significantly correlated
with the GC content. The phylogenetic analyses based on all the 13 protein genes showed that Megaloptera was the sister group
of other holometabolous insects except Coleoptera. 相似文献

Hypocrealean arthropod pathogenic fungi have profound impact on the regulation of agricultural and medical pests. However, until now the genetic and phylogenetic relationships among species have not been clarified, such studies could clarify host specificity relationships and define species boundaries. Our purpose was to compare the sequences of the mitochondrial SSU rDNA fragments from several mitosporic entomopathogenic Hypocreales to infer relationships among them and to evaluate the possibility to use these sequences as species diagnostic tool in addition to the more commonly studied sequences of nuclear SSU rDNA. The SSU mt-rDNA proved to be useful to help in differentiation of species inside several genera. Clusters obtained with Parsimony, Bayesian, and Maximum Likelihood analyses were congruent with a new classification of the Clavicipitaceae (Sung et al. Stud Mycol. 2007;57:5-59) in which the anamorphic genera Nomuraea and Metarhizium species remain in the Clavicipitaceae and Isaria species sequenced here are assigned to the family Cordycipitaceae. Mitochondrial genomic information indicates the same general pattern of relationships demonstrated by nuclear gene sequences. 相似文献

Gluconacetobacter diazotrophicus is a plant-growth-promoting bacterium, which is able to colonize sugarcane and other plant species of economic importance.
The potentially beneficial effects promoted by this bacterium on plants are nitrogen-fixation, production of phythormones,
action against pathogens and mineral nutrient solubilization. In this study, the molecular mechanisms associated with phosphorus
and zinc solubilization were analyzed. A transposon mutant library was constructed and screened to select for mutants defective
for phosphorous [Ca5(PO4)3OH] and zinc (ZnO) solubilization. A total of five mutants were identified in each screen. Both screenings, performed independently,
allowed to select the same mutants. The interrupted gene in each mutant was identified by sequencing and the results demonstrate
that the production of gluconic acid is a required pathway for solubilization of such nutrients in G. diazotrophicus.

The mechanism by which we age has sparked a huge number of theories, and is an area of intense debate. As the elderly population rises, the importance of elucidating these mechanisms is becoming more apparent as age is the single biggest risk factor for a number of diseases such as cancer, diabetes and neurodegenerative disease. Mitochondrial DNA (MtDNA) mutations have been shown to accumulate in cells and tissues during the ageing process; however the question as to whether these mutations have a causal role in the ageing process remains an area of uncertainty. Here we review the current literature, and discuss the evidence for and against a causal role of mtDNA mutations in ageing and in the pathogenesis of age-related disease. 相似文献

Background
Plagiochin E (PLE) is an antifungal macrocyclic bis(bibenzyl) isolated from liverwort Marchantia polymorpha L. Its antifungal mechanism is unknown. To elucidate the mechanism of action, its effect on mitochondria function in Candida albicans was studied.Methods
We assayed the mitochondrial membrane potential (mtΔψ) using rhodamine 123, measured ATP level in mitochondria by HPLC, and detected the activities of mitochondrial F0F1-ATPase and dehydrogenases. Besides, the mitochondrial dysfunction-induced reactive oxygen species (ROS) production was determined by a fluorometric assay, and the effects of antioxidant L-cysteine on PLE-induced ROS production and the antifungal effect of PLE on C. albicans were also investigated.Results
Exposure to PLE resulted in an elevation of mtΔψ, and a decrease of ATP level in mitochondria. The ATP depletion owed to PLE-induced enhancement of mitochondrial F0F1-ATPase and inhibition of the mitochondrial dehydrogenases. These dysfunctions of mitochondria caused ROS accumulation in C. albicans, and this increase in the level of ROS production and PLE-induced decrease in cell viability were prevented by addition of L-cysteine, indicating that ROS was an important mediator of the antifungal action of PLE.Conclusions
PLE exerts its antifungal activity through mitochondrial dysfunction-induced ROS accumulation in C. albicans.General significance
The effect of PLE on the mitochondria function in C. albicans was assayed for the first time. These results would conduce to elucidate its underlying antifungal mechanism. 相似文献999.

Background
We examined the expression of the mitochondrial selenoenzyme TrxR2 in the endothelial cell line EAhy926 under conditions known to modify its cytoplasmic counterpart TrxR1.Methods
Cells were cultured with varying concentrations of selenite, sulforaphane or the Ca2+ ionophore A23187 for 72-h, prior to assay of TrxR concentration and activity. Further cultures underwent prolonged (7-day) Se-depletion before selenoprotein measurement.Results
In Se-deficient cultures, neither Se, A23187 or sulforaphane affected TrxR2 concentration, while these treatments induced TrxR1 concentration (p < 0.05). When co-incubated, optimal concentrations of Se (40 nM) and sulforaphane (4 μM) only modestly increased TrxR2 protein (∼ 1.3-fold), compared with TrxR1 (∼ 4-fold). In Se-deficient cells, TrxR activity was unaffected by sulforaphane or A23187. Prolonged Se-depletion caused a comparatively small reduction in TrxR2 (66% TrxR2 retained) against TrxR1 and glutathione peroxidase-1 activity (38% and 17% retained, respectively).Conclusions
The relative resistance of TrxR2 to Se-deprivation and induction by sulforaphane and A23187 suggests TrxR2 lies near the top of the selenoprotein hierarchy in EAhy926 cells and exhibits near maximum expression under a range of culture conditions. In Se deficiency an inactive (possibly truncated) TrxR1 is produced in response to stimulus by sulforaphane and A23187.General significance
