More than 90% of the Lesser White‐fronted Geese Anser erythropus in the Eastern Palearctic flyway population winter at East Dongting Lake, China. To explain this restricted distribution and to understand better the winter feeding ecology and habitat requirements of this poorly known species, we assessed their food availability, diet and energy budgets at this site through two winters. Lesser White‐fronted Geese maintained a positive energy budget when feeding on above‐ground green production of Eleocharis and Alopecurus in recessional grasslands in autumn and spring to accumulate fat stores. Such food was severely depleted by late November and showed no growth in mid‐winter. Geese fed on more extensive old‐growth Carex sedge meadows in mid‐winter where they were in energy deficit and depleted endogenous fat stores. Geese failed to accumulate autumn fat stores in one year when high water levels prevented the Geese from using recessional grassland feeding areas. Fat stores remained lower throughout that winter and Geese left for breeding areas later in spring than in the previous year, perhaps reflecting the need to gain threshold fat stores for migration. Sedge meadows are widespread at other Yangtze River floodplain wetlands, but recessional grasslands are rare and perhaps restricted to parts of East Dongting Lake, which would explain the highly localized distribution of Lesser White‐fronted Geese in China and their heavy use of these habitats at this site. Sympathetic management of water tables is essential to maintain the recessional grasslands in the best condition for Geese. Regular depletion of fat stores whilst grazing sedge meadows in mid‐winter also underlines the need to protect the species from unnecessary anthropogenic disturbances that enhance energy expenditure. The specialized diet of the Lesser White‐fronted Goose may explain its highly restricted winter distribution and global rarity.  相似文献

From August to December, thousands of Black‐necked Grebes Podiceps nigricollis concentrate during the flightless moult period in salt ponds in the Odiel Marshes, southern Spain, where they feed on the brine shrimp Artemia parthenogenetica. We predicted that because Black‐necked Grebes moulted in a food‐rich, predator‐free environment, there would be no net loss of body mass caused by the use of fat stored to meet energy needs during remigial feather replacement (as is the case for some other diving waterbirds). However, because the food resource disappears in winter, we predicted that grebes moulting later in the season would put on more body mass prior to moult because of the increasing risk of an Artemia population crash before the moult period is completed. Body mass determinations of thousands of birds captured during 2000–2010 showed that grebes in active wing‐moult showed greater mass with date of capture. Early‐moulting grebes were significantly lighter at all stages than late‐moulting birds. Grebes captured with new feathers post‐moult were significantly lighter than those in moult. This is the first study to support the hypothesis that individual waterbirds adopt different strategies in body mass accumulation according to timing of moult: early‐season grebes were able to acquire an excess of energy over expenditure and accumulate fat stores while moulting. Delayed moulters acquired greater fat stores in advance of moult to contribute to energy expenditure for feather replacement and retained extra stores later, most likely as a bet hedge against the increasing probability of failing food supply and higher thermoregulatory demands late in the season. An alternative hypothesis, that mass change is affected by a trophically transmitted cestode using brine shrimps as an intermediate host and Black‐necked Grebes as final host, was not supported by the data.  相似文献

Microorganisms dwelling in sediments have a crucial role in biogeochemical cycles and are expected to have a strong influence on the cycle of arsenic, a metalloid responsible for severe water pollution and presenting major health risks for human populations. We present here a metagenomic study of the sediment from two harbours on the Mediterranean French coast, l'Estaque and St Mandrier. The first site is highly polluted with arsenic and heavy metals, while the arsenic concentration in the second site is below toxicity levels. The goal of this study was to elucidate the potential impact of the microbial community on the chemical parameters observed in complementary geochemical studies performed on the same sites. The metagenomic sequences, along with those from four publicly available metagenomes used as control data sets, were analysed with the RAMMCAP workflow. The resulting functional profiles were compared to determine the over‐represented Gene Ontology categories in the metagenomes of interest. Categories related to arsenic resistance and dissimilatory sulphate reduction were over‐represented in l'Estaque. More importantly, despite very similar profiles, the identification of specific sequence markers for sulphate‐reducing bacteria and sulphur‐oxidizing bacteria showed that sulphate reduction was significantly more associated with l'Estaque than with St Mandrier. We propose that biotic sulphate reduction, arsenate reduction and fermentation may together explain the higher mobility of arsenic observed in l'Estaque in previous physico‐chemical studies of this site. This study also demonstrates that it is possible to draw sound conclusions from comparing complex and similar unassembled metagenomes at the functional level, even with very low sequence coverage.  相似文献

A convenient synthetic route and the characterization of complexes trans-[PtCl2(L)(PPh3)] (L = Et2NH (2), (PhCH2)2NH (3), (HOCH2CH2)2NH) (4) are reported. The antiproliferative activity was evaluated on three human tumor cell lines. The investigation on the mechanism of action highlighted for the most active complex 4 the capacity to affect mitochondrial functions. In particular, both the induction of the mitochondrial permeability transition phenomenon and an aspecific membrane damage occurred, depending on concentration.  相似文献

Monocarboxylate transporters MCT1-MCT4 require basigin (CD147) or embigin (gp70), ancillary proteins with a glutamate residue in their single transmembrane (TM) domain, for plasma membrane (PM) expression and activity. Here we use site-directed mutagenesis and expression in COS cells or Xenopus oocytes to investigate whether this glutamate (Glu218 in basigin) may charge-pair with a positively charged TM-residue of MCT1. Such residues were predicted using a new molecular model of MCT1 based upon the published structure of the E. coli glycerol-3-phosphate transporter. No evidence was obtained for Arg306 (TM 8) of MCT1 and Glu218 of basigin forming a charge-pair; indeed E218Q-basigin could replace WT-basigin, although E218R-basigin was inactive. No PM expression of R306E-MCT1 or D302R-MCT1 was observed but D302R/R306D-MCT1 reached the PM, as did R306K-MCT1. However, both were catalytically inactive suggesting that Arg306 and Asp302 form a charge-pair in either orientation, but their precise geometry is essential for catalytic activity. Mutation of Arg86 to Glu or Gln within TM3 of MCT1 had no effect on plasma membrane expression or activity of MCT1. However, unlike WT-MCT1, these mutants enabled expression of E218R-basigin at the plasma membrane of COS cells. We propose that TM3 of MCT1 lies alongside the TM of basigin with Arg86 adjacent to Glu218 of basigin. Only when both these residues are positively charged (E218R-basigin with WT-MCT1) is this interaction prevented; all other residue pairings at these positions may be accommodated by charge-pairing or stabilization of unionized residues through hydrogen bonding or local distortion of the helical structure.  相似文献

以一年生瑞典能源柳4个无性系(N2、N4、NC、NE)幼苗为材料,通过盆栽试验,在对照、轻度、中度、重度Hg2+胁迫条件下,测定了4个能源柳无性系幼苗生长过程中生理生化指标,分析其对Hg2+胁迫的生理响应及其耐受性.结果表明:(1)随着土壤中Hg2+浓度的上升,4个能源柳无性系叶片保护酶POD、SOD和CAT活性均呈先上升后下降的趋势,且各无性系之间存在差异,其中NC的POD和CAT活性在重度胁迫下仍高于对照.(2)各无性系叶片中渗透性调节物质可溶性蛋白和脯氨酸(Pro)含量均呈先上升后下降的趋势,并均在轻度或中度胁迫下达到峰值,以可溶性蛋白含量变化幅度更大,在NC和NE无性系中表现得更明显.(3)随着土壤中Hg2+胁迫强度的加剧,N2、N4和NE无性系根系活力都呈现连续下降趋势,而NC无性系根系活力在轻度胁迫条件下则略微上升,然后持续下降,并均在中度和重度胁迫下达到显著水平,而且4个能源柳无性系叶片MDA含量均表现为先下降后上升趋势.(4)在轻度Hg2+胁迫下,N4和NE叶绿素含量和Chl a/Chl b值呈下降趋势,而N2和NC则稍微上升;在中度和重度胁迫下,4个能源柳无性系叶绿素含量和Chl a/Chl b值迅速下降,但能源柳各无性系的下降速度存在差异,其中N4下降幅度最大,NC最小.(5)运用隶属函数模糊评判法综合评价表明,各能源柳无性系对Hg2+的耐受性表现为:NC> NE> N4>N2.研究认为,在不同程度Hg2+胁迫条件下,4个瑞典能源柳无性系通过提高自身保护酶活性、抗氧化物质GSH含量以及有机渗透物质含量来有效缓解胁迫伤害,从而对Hg2+表现出一定的耐性,并以无性系NC的耐受性较好.  相似文献

1-MCP和CO2对‘南果梨’冷藏后货架期能量代谢特性的影响   总被引:2,自引:0,他引:2  
分别对‘南果梨’果实采用0.75μL/L 1-甲基环丙烯(1-MCP)在20℃熏蒸20h后装入保鲜袋(0.02mm)、用2%CO2充气果实包装袋(0.04mm)、以保鲜袋果实为对照,各处理组果实均置于(0±1)℃贮藏5个月后移至室温下(18℃±3℃),测定各处理果实在货架期间的褐变度以及果实线粒体蛋白含量、MDA含量、ATP含量,以明确1-MCP和CO2对南果梨冷藏后果实货架期的能量代谢特性。结果显示:(1)1-MCP和CO2处理可不同程度延缓南果梨冷藏后货架期果实果心褐变指数和褐变度,且1-MCP处理效果更好,但CO2处理在货架后期反而使果实褐变度较对照提高。(2)1-MCP和CO2处理可有效抑制果实MDA含量增加,延缓细胞膜透性的升高,保持细胞完整性。(3)1-MCP处理有利于提高货架前期果实中线粒体蛋白质含量,能够在货架后期保持较高的ATP含量和能荷水平,而CO2处理在货架前期果实内含有较高水平的ATP,促进了果实内的能荷水平。研究发现,1-MCP和CO2处理均可以通过影响南果梨果实的能量代谢特性从而影响果实的成熟衰老进程,且1-MCP处理可以抑制货架前期的ATP含量和能量供应,有利于保持细胞膜完整性,抑制果心褐变的发生,延缓果实成熟衰老进程;而2%CO2处理与对照相比对果实能量代谢特性的影响不大。  相似文献

The concomitant production of formic acid and pterin compounds from guanosine-5′-triphosphate (GTP) has been found in cell-free extracts of Serratia indica. Among the pterin compounds, l-threo-neopterin–the major Crithidia factor in S. indica–, a cyclic phosphate of neopterin (cNP), d-erythro-neopterin and 6-hydroxymethyl pterin were detected and isolated. Formate-14C elimination from GTP-8-14C was quantitatively distributed in the ethyl acetate layer in the ehyl acetate-hydrochloric acid partition system. Carbon 8 of GTP was released as formic acid. Enzymatic production of formate and cNP was linear for 2 hr at 37°C. Formate production was proportional to the enzyme concentration. The optimum pH for formate elimination was observed around pH 8.6. Optimum temperature for the production of formate and cNP was 50°C. The apparent Km value of GTP for formate production was 6.2×10?bm. Formate eliminating activity was activated by disodium phosphate but was inhibited by Mg2+ or AMP. Incorporation of GTP-U-14C into pterin compounds was also regulated with disodium phosphate. Effective incorporation into cNP and d-erythro-neopterin occurred in the presence of phosphate. When phosphate was omitted from the system, however, effective incorporation into 6-hydroxymethyl pterin was observed. The biosynthetic process of the Crithidia factors, i.e. l-threo-neopterin and cNP, from GTP in S. indica is also discussed.  相似文献
