A total of 90 cows from three commercial farms were used to evaluate the relationship between subclinical mastitis and clinical mastitis and thermal nociceptive threshold. Milk strips from all udder quarters were tested for clinical mastitis with visual inspection of milk and udder alterations and for subclinical mastitis using California Mastitis Test. Milk yield was recorded, milk was sampled and further analyzed for somatic cells count (SCC). Cows were considered healthy when SCC<200 000 cells/ml and no visual alterations in milk and/or udder, with mild subclinical mastitis when SCC>200 000 cells/ml and no visual alterations in milk and/or udder, with moderate subclinical mastitis when SCC>500 000 cells/ml and no visual alterations in milk and/or udder and with clinical mastitis when visual alterations in milk and/or udder were detected. Nociceptive threshold was evaluated with the thermal threshold meter apparatus applied to the rear legs. Thermal threshold (TT) decreased when we compared healthy cows with cows presenting clinical mastitis and tended to decrease when we compare healthy cows with those with moderate subclinical mastitis. TT was lower at the ipsilateral rear leg compared with the contralateral leg to the infected mammary gland. TT linearly decreases as log10SCC increased and it showed sharp decrease as log10SCC exceed the value of 6.4. Increase in one unit of log10SCC increased the odds of low thermal threshold (lower than 55.8°C). Subclinical mastitis might be a welfare issue as it tended to decrease nociceptive thermal threshold.  相似文献

氮敏感性鉴定与评价是培育耐低氮与氮高效利用玉米品种的重要的研究内容。本研究采用前期建立的快速无损伤测定SPAD值分析相对叶绿素含量的模型,于低氮与正常供氮大田试验处理下,对我国玉米育种与生产上重要的 189 份玉米自交系开展了低氮与正常供氮条件下不同时期与不同部位叶片的相对叶绿素含量分析。方差分析结果表明,低氮与正常供氮条件下,叶绿素含量均存在极显著的基因型与环境差异,基因型差异是氮敏感性差异的重要原因之一。开花期和开花后10天玉米主要功能叶穗三叶(穗位上第1叶、穗位叶、穗位下第1叶)叶绿素含量之间具有极显著正相关;大喇叭口期玉米全展叶叶绿素含量与各时期各部位叶绿素含量之间呈现极显著中度相关。将正常供氮条件下的性状值与低氮胁迫下性状值的差值占正常供氮条件下性状值的百分比定义为氮敏感性。189份玉米自交系氮平均氮敏感指数变幅为23.86%- 36.00%,表现高度耐低氮的材料有合344和昌7-2等40份自交系,高度敏感的材料有CML206和CA375等40份自交系。本研究提供了一种快速高效的种质资源氮敏感性鉴定与评价方法,并为玉米耐低氮与氮高效育种的遗传育种提供了重要分析结果与基础数据。  相似文献

The wall-yielding properties of cell walls were examined using frozen-thawed and pressed segments (FTPs) obtained from the elongation zones of cucumber hypocotyls with a newly developed programmable creep meter. The rate of wall extension characteristically changed depending on both tension and pH. By treatment of the FTPs with acid, the yield tension (y) was shifted downward and the extensibility (phi) was increased. However, the downward shift of y was greatly suppressed and the increase in phi was partly inhibited in boiled FTPs. The boiled FTPs reconstituted with expansin fully recovered the acid-induced downward y shift as well as the increase in phi. Even under the tension below y, wall extension took place pH dependently. Such extension was markedly slower (low-rate extension) than that under the tension above y (high-rate extension). At a higher concentration (8 M), urea markedly inhibited the creep ascribable to the inhibition of the acid-induced downward y shift and increase in phi. Moderate concentrations (2 M) of urea promoted wall creep pH dependently. The promotion was equivalent to a 0.5 decrease in pH. The promotion of creep by 2 M urea was observed in boiled FTPs reconstituted with expansin but not in boiled FTPs. These findings indicated that the acid-facilitated creep was controlled by y as well as in cucumber cell walls. However, y and phi might be inseparable and mutually related parameters because the curve of the stress extension rate (SER) showed a gradual change from the low-rate extension to the high-rate extension. Expansin played a role in pH-dependent regulation of both y and phi. The physiological meaning of the pH-dependent regulation of wall creep under different creep tensions is also discussed with reference to a performance chart obtained from the SER curves.  相似文献

The ability of the SPAD-502 chlorophyll meter to quantify chlorophyll amounts in ozone-affected leaves of cutleaf coneflower (Rudbeckia laciniata var. digitata) was assessed in this study. When relatively uninjured leaves were measured (percent leaf area affected by stipple less than 6%), SPAD meter readings were linearly related to total chlorophyll with an adjusted R 2 of 0.84. However, when leaves with foliar injury (characterized as a purple to brownish stipple on the upper leaf surface affecting more than 6% of the leaf area) were added, likelihood ratio tests showed that it was no longer possible to use the same equation to obtain chlorophyll estimations for both classes of leaves. Either an equation with a common slope or a common intercept was necessary. We suspect several factors are involved in altering the calibration of the SPAD meter for measuring chlorophyll amounts in visibly ozone-injured leaves, with the most likely being changes in either light absorption or scattering resulting from tissue necrosis.  相似文献

The portable chlorophyll meter (SPAD-502) has been successfully used for a rapid and direct estimation of total chlorophyll content (TCHL) in the leaves of some crops. In this work, SPAD-502 meter readings and TCHL concentration were compared for the leaves of Amaranthus vlitus L., a common weed. SPAD readings were linearly and positively correlated to TCHL concentration in the leaves. A linear correlation was also shown between SPAD-502 readings and some physiological parameters of the leaves, such as photosynthesis, transpiration, and stomatal conductance.  相似文献

We examined the therapeutic effect of a 1% cream preparation of NND-502, a novel topical antifungal agent, in a guinea pig tinea pedis model produced by infecting the plantar skin of guinea pigs with Trichophyton mentagrophytes. Animals developing tinea pedis were divided into two groups: an untreated control group and a treated group. In the latter group, after confirming infection had been established, the infected animals were topically treated with the NND-502 cream once daily for one week. The animals were reared in a clean environment free from exposure to exogenous dermatophytes. At one week (5 weeks post-infection), 6 weeks (10 weeks post-infection) and 16 weeks (20 weeks post-infection) after completion of the treatment, plantar skin samples were taken from a certain number of both groups of animals. The results demonstrated that all of the animals in the untreated control group and none of those in the treated group were culture-positive in this animal model of tinea pedis. The topical treatment with NND-502 achieved a mycological cure. Thus NND-502 can be considered a promising candidate as a new anti-dermatophytic agent for topical use.  相似文献

An improved reverse-phase high-performance liquid chromatographic method (RP-HPLC) for the determination of a novel iron chelator CP502 (1,6-dimethyl-3-hydroxy-4-(1H)-pyridinone-2-carboxy-(N-methyl)-amide hydrochloride) in rat plasma, urine and feces was developed and validated. The separation was performed on a polymeric column using a mobile phase composed of 1mM ethylenediaminetetra-acetic acid disodium salt (EDTA), acetonitrile, methanol and methylene chloride. Separation of CP502 from plasma, urine or feces endogenous compounds was achieved by gradient elution. Retention times of CP502 and its major metabolite (glucuronide) were about 13 and 4 min, respectively. The method was validated in terms of limit of detection (LOD), limit of quantification (LOQ), selectivity (endogenous from plasma, urine or feces), linearity, extraction recovery, robustness (column selection, mobile phase composition, detection mode, internal standard (IS) selection, analyte stability), day-to-day reproducibility and system suitability (repeatability, peak symmetry and resolution). The method is applicable to bioavailability and pharmacokinetic studies of CP502 in rats.  相似文献

The practicality of the portable, non-destructive type nitrogen meter (Agriexpert PPW-3000) was tested on ten forest species. Also investigated was the potential relationship between leaf nitrogen and chlorophyll (Chl) contents and the readings taken with the PPW-3000 and a Chl meter (SPAD-502). There was a significantly positive correlation between the readings of PPW-3000 and N content in the same leaves, whereas the correlation between leaf Chl content and the PPW-3000 values was less positive. Similarly there was a significant positive correlation between actual Chl content and the SPAD-502 readings and the less positive correlation between actual N content and the SPAD-502 readings. Thus using both the PPW-3000 and SPAD-502 enables to determine leaf N and Chl contents simply and non-destructively in the field.  相似文献

Use of leaf meters to provide an instantaneous assessment of leaf chlorophyll has become common, but calibration of meter output into direct units of leaf chlorophyll concentration has been difficult and an understanding of the relationship between these two parameters has remained elusive. We examined the correlation of soybean (Glycine max) and maize (Zea mays L.) leaf chlorophyll concentration, as measured by organic extraction and spectrophotometric analysis, with output (M) of the Minolta SPAD-502 leaf chlorophyll meter. The relationship is non-linear and can be described by the equation chlorophyll (mol m–2)=10(M0.265), r 2=0.94. Use of such an exponential equation is theoretically justified and forces a more appropriate fit to a limited data set than polynomial equations. The exact relationship will vary from meter to meter, but will be similar and can be readily determined by empirical methods. The ability to rapidly determine leaf chlorophyll concentrations by use of the calibration method reported herein should be useful in studies on photosynthesis and crop physiology.Abbreviations Chl- chlorophyll - M- SPAD-502 meter value  相似文献
