Infection efficiency is the key issue for gene delivery using adenovirus vector and usually unsatisfactory. In this study, recombinant adenoviruses encoding recombinant human EPO were prepared using the Adeasy system, and injected into the mammary gland of goats via the teat canal. BAPTA was used to treat the mammary gland to facilitate adenoviruses infection compared with EGTA. Milk serum was collected from the infected mammary gland and characterized by ELISAs and Western blotting. Seventy-two lactating Chios ewes were used in two experiments to determine effects of supplemental dietary biotin on productivity and milk composition. The first experiment started after weaning on day 42 postpartum, and lasted 20 weeks, while the second started on week 24 postpartum and lasted 12 weeks. In both experiments, ewes were allocated, after equal distribution relative to milk yield, body weight, time of lambing, and lactation number (i.e., two or three), into three groups of 24 ewes each, and were accommodated in three floor pens/groups of eight ewes/treatment. Ewes were fed one of three diets varying in supplemental biotin (BIOTIN0, no supplemental biotin; BIOTIN3, 3 mg supplemental biotin/ewe/day; BIOTIN5, 5 mg supplemental biotin/ewe/day) in each experiment. Milk, fat, protein, lactose and ash yield, and milk fat content increased linearly (P<0.012) with increased dietary biotin in both studies. This study compared pancreatic tissue growth and functional changes during the first 3 postnatal days in piglets and rat pups. In piglets the absolute weight and the relative weight per unit body weight of the pancreas increased by 97 and 70%, respectively, while in rat pups the same parameters decreased by 33 and 48%, respectively, during this period. The specific activity of pancreatic amylase rose by 336% while that of trypsin, chymotrypsin and lipase remained at newborn level in piglets. In rat pups the specific activities of all enzymes measured declined by 61 to 92% during the first 3 postnatal days. The rate of postnatal pancreatic growth in the two species coincide with the levels of epidermal growth factor and insulin-like growth factors in maternal milk as reported in the literature, suggesting that milk-borne growth factors may stimulate pancreatic development in newborn animals. 相似文献

A total of 42,197 records of test-day milk yield (TDY) and 7654 lactations from 3854 individuals belonging to the Spanish Assaf (Assaf.E) breed were analysed using univariate and multivariate models to estimate genetic parameters affecting TDY, total milk yield standardised to 180 days (MY180) and lactation length (LL) in order to asses their possibilities of use in the selection scheme of the Assaf.E population. Estimates of h2 were, in general, on the lower limit of those usually reported for the three analysed traits. Estimates of heritability for MY180 varied from 0.131 to 0.177. Estimates of h2 for LL and TDY were consistent regardless the estimation model being of, roughly, 5% and 10%, respectively. The estimates for the permanent environmental effect (c2) of TDY were consistently the same (0.27–0.28) regardless the fitted model whilst they showed large differences for LL and MY180. Genetic correlations were always positive and high ranging from 0.792 for the pair LL-TDY to 0.999 for the pair MY180-TDY. Correlations between permanent environmental effects were even higher ranging from 0.932 for the pair LL-MY180 and 0.999 for the pair MY180-TDY. The advantages of using TDY as selection criterion in the Assaf.E improvement scheme are discussed. 相似文献

Summary Changes in mammary gland tubulin were studied immunocytochemically during transition from late pregnancy to lactation. Indirect immunofluorescence was used to localize tubulin in mammary glands from late pregnant, early lactating and peak lactating guinea pigs. Whole rabbit antiserum against guinea pig brain tubulin and affinity-purified antibody indicated increases in alveolar cell tubulin content from late pregnancy through peak lactation coincident with the development of lactation. Only alveolar cells displayed high, specific fluorescence or underwent a developmental increase. Tubulin was concentrated apically, in association with secretory structures. In a second study comparing mammary tissues from 18 days pregnant and 10 days lactating rats, EM immunogold was used with three commercial antitubulins ranging from a rabbit polyclonal antiserum against chick brain MTs to a monoclonal mouse anti-alpha tubulin. Gold particle counts indicated 2- to 5- fold tubulin increases in alveolar cells with lactation and development of an apicobasal (high apical) tubulin gradient. Variations among the three anti-tubulins is discussed. The results confirmed previous observations of whole gland tubulin increases based on colchicine binding assays and localized the site of the increase primarily in alveolar cells.Abbreviations EM
