Microcystis sp., especially in its colonial form, is a common dominant species during cyanobacterial blooms in many iron‐deficient water bodies. It is still not entirely clear, however, how the colonial forms of Microcystis acclimate to iron‐deficient habitats, and the responses of unicellular and colonial forms to iron‐replete and iron‐deficient conditions were examined here. Growth rates and levels of photosynthetic pigments declined to a greater extent in cultures of unicellular Microcystis than in cultures of the colonial form in response to decreasing iron concentrations, resulting in the impaired photosynthetic performance of unicellular Microcystis as compared to colonial forms as measured by variable fluorescence and photosynthetic oxygen evolution. These results indicate that the light‐harvesting ability and photosynthetic capacity of colonial Microcystis was less affected by iron deficiency than the unicellular form. The carotenoid contents and nonphotochemical quenching of colonial Microcystis were less reduced than those of the unicellular form under decreasing iron concentrations, indicating that the colonial morphology enhanced photoprotection and acclimation to iron‐deficient conditions. Furthermore, large amounts of iron were detected in the capsular polysaccharides (CPS) of the colonies, and more iron was found to be attached to the colonial Microcystis CPS under decreasing iron conditions as compared to unicellular cultures. These results demonstrated that colonial Microcystis can acclimate to iron deficiencies better than the unicellular form, and that CPS plays an important role in their acclimation advantage in iron‐deficient waters. 相似文献

In temperate latitudes, toxic cyanobacteria blooms often occur in eutrophied ecosystems during warm months. Many common bloom-forming cyanobacteria have toxic and non-toxic strains which co-occur and are visually indistinguishable but can be quantified molecularly. Toxic Microcystis cells possess a suite of microcystin synthesis genes (mcyA–mcyJ), while non-toxic strains do not. For this study, we assessed the temporal dynamics of toxic and non-toxic strains of Microcystis by quantifying the microcystin synthetase gene (mcyD) and the small subunit ribosomal RNA gene, 16S (an indicator of total Microcystis), from samples collected from four lakes across the Northeast US over a two-year period. Nutrient concentrations and water quality were measured and experiments were conducted which examined the effects of elevated levels of temperatures (+4 °C), nitrogen, and phosphorus on the growth rates of toxic and non-toxic strains of Microcystis. During the study, toxic Microcystis cells comprised between 12% and 100% of the total Microcystis population in Lake Ronkonkoma, NY, and between 0.01% and 6% in three other systems. In all lakes, molecular quantification of toxic (mcyD-possessing) Microcystis was a better predictor of in situ microcystin levels than total cyanobacteria, total Microcystis, chlorophyll a, or other factors, being significantly correlated with the toxin in every lake studied. Experimentally enhanced temperatures yielded significantly increased growth rates of toxic Microcystis in 83% of experiments conducted, but did so for non-toxic Microcystis in only 33% of experiments, suggesting that elevated temperatures yield more toxic Microcystis cells and/or cells with more mcyD copies per cell, with either scenario potentially yielding more toxic blooms. Furthermore, concurrent increases in temperature and P concentrations yielded the highest growth rates of toxic Microcystis cells in most experiments suggesting that future eutrophication and climatic warming may additively promote the growth of toxic, rather than non-toxic, populations of Microcystis, leading to blooms with higher microcystin content. 相似文献

Understanding the mechanisms underlying community assembly helps to define success and susceptibility to biological invasions. Here, we explored phytoplankton community assembly following niche and neutral paradigms and using a trait-based approach. Under the hypothesis that the morphology-based functional groups (MBFG) clusters species with similar niche, we analysed how trait-related differences in fitness influence dominance of an invasive species. This was based on literature review, field data and model simulations. We predict that invading species can be dominant if: 1) do not belong to the local MBFG but use unexploited areas of the niche, or 2) belong to the resident MBFG but exhibit a higher fitness due to a particular combination of traits. The invasive dinoflagellate Ceratium furcoides was used as the model species to evaluate these hypotheses, its morphological (e.g. volume) and physiological (e.g. growth rates) traits were compared with species from the same (V: photosynthetic flagellates) and different (VII: colonial cyanobacteria) MBFG. Fitness was estimated using models parametrized with MBFG rates (R*, ability to draw down phosphate) under different environmental conditions (i.e. flushing). Results contributed to support both hypotheses. First, the alternation of C. furcoides and cyanobacteria dominance was explained by the use of different niches. Secondly, species from MBFG V were dominant under similar environments. Within this group V C. furcoides showed higher fitness under low flushing and high predation, advantage provided by a distinctive combination of traits. The application of trait-based approaches to represent the niche and estimate fitness along environmental gradients was useful to evaluate community assembly and can be used to predict the dominance of microbial species invasions. 相似文献

Abstract A new sulfated, cyclic depsipeptide, called cyanopeptolin S, from Microcystis sp. was isolated from a water bloom in the Auensee/Leipzig (Germany). The depsipeptide had a relative molecular mass of 925 and contained l-arginine, l-threonine, l-isoleucine, N-methyl-l-phenylalanine, a l-glutamic acid-δ-aldehyde ring system and a sulfated d-configurated glyceric acid as a side chain. The structure was elucidated by means of two-dimensional 1 H and 13 C nuclear magnetic resonance spectroscopy, fast atom bombardment mass spectroscopy, Fourier transformed infrared spectroscopy and combined gas-liquid chromatography/mass spectrometry. 从滇池分离纯化了两种常见水华微囊藻即铜锈微囊藻和绿色微囊藻。在常规培养条件下,两种藻类在对数生长期的生长速率μ值分别为0.61和0.63;早期生长的抑制光强不大于100μmol m~(-2)s~(-1)。铜锈微囊藻主要产生3种微囊藻毒素:MYCST-RR,MYCST-YR和MYCST-LR,绿色微囊藻产生的主要微囊藻毒素为[Dha~7]-MYCST-RR,和[Dha~7]-MYCST-LR,另含有少量的[Dha~7]-MYCST-YR。在低光强15μE m~(-2)s~(-1)时,毒素含量每毫克干重细胞达到3.127μg微囊藻毒素,当光强达到100μE m~(-2)s~(-1)时,毒素含量降低到每毫克干重细胞1.971μg;光强对毒素形成的影响受到温度的调节,而温度对毒素形成的影响不大。探讨了两种微囊藻细胞在不同光照强度下叶绿素荧光比值Fv/Fm的变化,此比值的变化可以间接反映细胞受外界光照强度抑制程度。 相似文献

【目的】从水华蓝藻铜绿微囊藻(Microcystis aeruginosa PCC 7806)的细胞中分离纯化出高纯度且完整的气囊,并对气囊的结构组成蛋白进行鉴定。【方法】采用渗透冲击与溶菌酶处理相结合的方法,进行多次低速离心提纯气囊,纯化所得气囊用负染色透射电镜观察气囊的纯度、完整度和形态。将纯化得到的气囊溶解后进行SDS-PAGE电泳,电泳后的蛋白条带运用LC-MS质谱法完成鉴定。【结果】从气囊的电镜照片可以看出提纯后的气囊纯度高,完整性好。气囊是两端呈锥状的圆柱体状,各气囊的直径大小一致,约120 nm,但长度不同,从约500 nm到1 500 nm不等。SDS-PAGE电泳和质谱法鉴定出气囊的两种主要结构组成蛋白Gvp A和Gvp C。【结论】这些研究对后续揭示气囊的精细结构和深入研究水华蓝藻浮力调节机制具有重要的意义。 相似文献

The carbonic anhydrase (CA) activities were determined in three cyanobacterial species, namely Microcystis aeruginosa Kütz., Microcystis viridis (A.Br.) Lemm, and Microcystis wesenbergii (Kom.) Kom, which were dominant in a lake (Dianchi Lake) subject to major blooms. In more detailed experiments on M. aeruginosa, the effects of inorganic carbon, pH, temperature, nitrogen/phosphorus ratio, glucose, and light intensity on CA activity were also investigated. Because of the relatively alkaline pH value of the culture media for the optimum growth of algal cells, bicarbonate ions were the main form of exogenous inorganic carbon. The results showed that the CA activity of M. aeruginosa was influenced dramatically by the concentration of bicarbonate. Consequently, it was suggested that bicarbonate ions were the main form of exogenous inorganic carbon that M. aeruginosa could utilize. Cultures grown in the dark exhibited CA activity six times higher than that of cells cultured mixotrophically with the addition of glucose. Features of eutrophic water bodies promoted an increase in CA activity, and the resulting higher CA activity would accelerate the utilization of inorganic carbon and favor the growth and blooming of Microcystis spp. in eutrophic lakes. Although the experiments were carried out under controlled experimental conditions, they could provide some basic data that would prove useful for the control of cyanobacterial blooms in nature. 相似文献

In stratified lakes, dominance of the phytoplankton by cyanobacteria is largely the result of their buoyancy and depth regulation. Bloom-forming cyanobacteria regulate the gas vesicle and storage polymer contents of their cells in response to interactive environmental factors, especially light and nutrients. While research on the roles of nitrogen and phosphorus in cyanobacterial buoyancy regulation has reached a consensus, evaluations of the roles of carbon have remained open to dispute. We investigated the various effects of changes in carbon availability on cyanobacterial buoyancy with continuous cultures of Microcystis aeruginosa Kuetz. emend. Elenkin (1924), a notorious bloom-former. Although CO2 limitation of photosynthesis can promote buoyancy in the short term by preventing the collapse of turgor-sensitive gas vesicles and/or by limiting polysaccharide accumulation, we found that sustained carbon limitation restricts buoyancy regulation by limiting gas vesicle as well as polysaccharide synthesis. These results provide an explanation for the positive effects of bicarbonate enrichment on cyanobacterial nitrogen uptake and bloom formation in lake experiments and may help to explain the pattern of cyanobacterial dominance in phosphorus-enriched, low-carbon lakes. 相似文献

As a part of a whole-lake, long-term experiment in biomanipulation in. the hypertrophic Bautzen reservoir (G.D.R.), during three years (1984–1986) the dynamics of mouse-related LD 50 of Microcystis aeruginosa was compared with the biomass development of this blue-green and the grazing pressure exerted by Daphnia galeata. Since the three summer averages of the biomass of D. galeata revealed strong differences due to decreasing predation activity of fish from 1984 to 1986, the effects of different grazing pressure on Microcystis toxicity could be investigated under field conditions. Microcystis was nontoxic at the beginning of the growing season and developed high toxicity during its first strong biomass increase in summer in all three years. But this decrease of the LD 50 together with the first biomass increase of the season is found in quite different periods in different years (1984: August, 1985: July, 1986: June). It is obvious that the higher the mean effective filtration rate of D. galeata during summer is found the faster the toxicity of Microcystis is formed. If these observations are combined with findings of other authors, the conclusion can be drawn that the development of toxic Microcystis blooms seems to be promoted by a combination of five conditions: (1) Presence of a mixture of toxic and nontoxic Microcystis strains at the beginning of the growing season even if the portion of toxic strains is very low, (2) physical and chemical growth conditions which favour Microcystis over other phytoplankton, (3) high grazing pressure by zooplankton on edible food particles over a rather long period, (4) patchy distribution of the different Microcystis strains if nonselective filtrators such as Daphnia dominate the zooplankton, and (5) absence of defense mechanisms of Microcystis against grazing which are not coupled with toxicity (e.g. large colony size). These conclusions contribute to a better understanding of the possibilities and limits of in-lake eutrophication control by biomanipulation and emphasize the need to combine top-down and bottom-up control mechanisms in eutrophic and hypertrophic waters. 相似文献
