In this work, a novel thiol aromatic aldehyde was synthesized. It can be used as a substrate to directly immobilize antibodies on a gold electrode, for which no additional chemical cross-linker is required. It was also applied as a linker to prepare Fe3O4@Au/PAMAM/Ab2–horseradish peroxidase bioconjugates, which introduced multiple enzymes onto a sensing interface owing to the high surface-to-volume ratio of Fe3O4@Au nanoparticles and many functional groups of the poly(amidoamine) dendrimer (PAMAM). The introduced multiple enzymes greatly improved the detection signal. Under optimal conditions, the proposed electrochemical immunosensor exhibited desirable performance for detection of IgG in the range 0.005–50 ng ml−1 with a detection limit of 3 pg ml−1 based on a signal-to-noise ratio of 3. It has great potential application in the area of clinical analysis.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are known to be toxic to living organisms and have been identified as carcinogenic. In this study, a pathway of surfactant flushing, chemical oxidation, and biological treatment is proposed to remediate the soils polluted with the hydrophobic PAHs. Different surfactants such as Tween 80, Brij 35, sodium dodecyl sulfate (SDS), and polyethylene glycol (PEG) 6000 were tested in order to increase the PAH solubilization from the soil matrix. The maximum desorption efficiency of naphthalene and anthracene were found to be 56.5% and 59%, respectively, when Brij and SDS were used. The soluble PAH in the aqueous phase was amended with sodium thiosulfate (3%) to oxidize the PAH into a more bioavailable form. The chemical oxidation with subsequent biodegradation by Pseudomonas aeruginosa exhibited the relatively high PAH degradation rate (1.24 times higher) when compared with chemical oxidation alone. These results display the efficiency of chemical pretreatment of PAH-contaminated soil for improved bioremediation.  相似文献

AIMS: This paper investigates the diversity of polycyclic aromatic hydrocarbon (PAH)-degrading mycobacterium isolates from three different sites within United States: Montana, Texas and Indiana. METHODS AND RESULTS: All five mycobacterium isolates differed in chromosomal restriction enzyme-fragmentation patterns; three isolates possessed linear plasmids. The DNA sequence between the murA and rRNA genes were divergent but the sequence upstream of nidBA genes, encoding a dioxygenase involved in pyrene oxidation, was more highly conserved. Long-chain fatty acid analysis showed most similarity between three isolates from the same Montana site. All isolates were sensitive to rifampicin and isoniazid, used in tuberculosis treatment, and to syringopeptins, produced by plant-associated pseudomonads. Biofilm growth was least for isolate MCS that grew on plate medium as rough-edged colonies. The patterns of substrate utilization in Biolog plates showed clustering of the Montana isolates compared with Mycobacterium vanbaalenii and Mycobacterium gilvum. CONCLUSION: The five PAH-degrading mycobacterium isolates studied differ in genetic and biochemical properties. SIGNIFICANCE AND IMPACT OF THE STUDY: Different properties with respect to antibiotic susceptibility, substrate utilization and biofilm formation could influence the survival in soil of the microbe and their suitability for use in bioaugmentation.  相似文献

Bioremediation represents one of the most cost-effective technologies for treatment of petroleum hydrocarbons in contaminated surface soils. A major concern for regulatory agencies when evaluating bioremediation is how to determine acceptable levels for residual organics in soil. Although guidelines have been developed for some organics in soil, limited information is available to define acceptable levels of the metabolites of biological degradation. The products of oxidative degradation are likely to be more water soluble and may also be more toxic. The purpose of the current study was to monitor changes in compound concentration and genotoxicity in soils undergoing bioremediation. The site selected for this study was a former wood-preserving site in the northwestern United States. Soil samples were collected over a 4-year period from two 6075-m2 land treatment units. Conditions for biodegradation were enhanced by the addition of water and nutrients, as well as by frequent tilling to add oxygen. Due to the location of the facility, the temperature was conducive to a more rapid rate of biodegradation for approximately 6 months per year. Soil samples were collected using a grid system and solvent extracted. Polycyclic aromatic hydrocarbons were quantified in soil extracts using gas chromatography-mass spectrometry (GC/MS), and genotoxicity measured using the Salmonella/microsome assay. After 2 years of treatment, concentrations of total and carcinogenic polycyclic aromatic hydrocarbons (PAHs) were reduced to approximately 10% the concentration in the untreated soil. However, the mean weighted activity of the untreated soil was 293 net revertants per g soil, whereas the extracts of soil collected after 2 years induced a mean weighted activity of 325 net revertants per g soil. Thus, although biodegradation clearly reduced the concentration of total and carcinogenic PAHs in the surface soils, the results from the genotoxicity bioassay indicate that there was a lag in the reduction of mutagenicity in treated soils.  相似文献

An open field experiment was established in 2014 in Budapest (BP) and Poznań (PZ), which are characterized by different soil and weather conditions. The purpose was to study the intraspecific responses of Melissa officinalis L. (lemon balm -Lb) and Thymus vulgaris (thyme -T) to varying locations. Five accessions of lemon balm (‘Lorelei’, ‘Lemona’, ‘Soroksár’, ‘Quedlinburger Niederliegende’, ‘Gold Leaf’) and four of thyme (‘Sloneczko’, French Summer’, ‘Varico 3′, ‘Standard Winter’) were tested for yield and chemical characteristics.Biomass of both species was significantly higher in PZ, than in BP (291 and 107 g/plant for Lb and 105 and 53 g/plant for T in PZ and BP, respectively). In Lb, the environmental circumstances of the actual vegetation period of BP location were more favourable for the accumulation of volatiles (+76%), while the level of phenolic compounds was higher in PZ (up to +53% for rosmarinic acid). In T, each chemical parameter increased (up to +27% both for volatiles and total flavonoid content) in PZ, where lower temperatures and less precipitation prevailed.In most traits a significant interaction of genotype and growing location was established, which was more characteristic in T. The results show that breeding of specific cultivars for a given location might be of primary importance.  相似文献

We report the hierarchical supramolecular organization of metallosupramolecular homochiral complexes 1 ‐Λ‐(S,S,S,S)‐M2+/ 1 ‐?‐(R,R,R,R)‐M2+ and 2 ‐ Λ‐(S,S,S,S)‐M2+/ 2 ‐?‐ (R,R,R,R)‐M2+ of M2+ = Co2+, Fe2+, Zn2+ metal ions with chiral pseudo‐terpyridine‐type ligands: 1‐ (S,S) or 1‐ (R,R) = 2,6‐bis (naphthyl ethylimine)pyridine and 2‐ (S,S) or 2‐ (R,R) = 2,6‐bis (phenyl‐ethylimine)pyridine. Circular dichroism measurements in solution were used to confirm the enantiomeric nature of all twelve complexes. For crystal structures of 1 ‐ Λ‐ (S,S,S,S)‐M2+ or 1 ‐?‐ (R,R,R,R)‐M2+ complexes, absolute configurations {? (or P), Λ (or M)} were confirmed by refinement of the Flack parameter x: ?0.007 ≤ x ≤ 0.11 for the single crystals of 1 ‐Λ‐(S,S,S,S)‐M2+/ 1 ‐?‐ (R,R,R,R)‐M2+, 2 ‐ Λ‐ (S,S,S,S)‐Fe2+, and 2 ‐?‐ (R,R,R,R)‐Co2+.  相似文献

The capital city of Prague is one of the most polluted localities of the Czech Republic. Therefore, the effect of exposure to carcinogenic polycyclic aromatic hydrocarbons (c-PAHs) adsorbed onto respirable air particles (<2.5 μm) on chromosomal aberrations was studied in a group of policemen (males, aged 22–50 years) working in the downtown area of Prague and spending daily >8 h outdoors (N = 53). Age- and sex-matched healthy volunteers spending > 90% daily time indoors were chosen as controls (N = 52). Ambient air particles (PM10, PM2.5) and c-PAHs were monitored using versatile air pollution sampler (VAPS), and personal exposure was evaluated using personal samplers during working shift. Chromosomal aberrations were analyzed by conventional cytogenetic analysis and fluorescent in situ hybridization (FISH). Urinary cotinine plasma levels of vitamins A, E and C, folate, total cholesterol, HDL, LDL cholesterols and triglycerides were also analyzed as possible effect modifiers. Genotypes CYP1A1*2A, CYP1A1*2C, GSTM1, GSTP1, GSTT1, EPHX1, NAT2, hOGG1, XRCC1, XPD, p53 BstI, p53 MspI, MTHFR677, and MS2656 were determined by PCR-based RFLP assays. The following levels of air pollution were recorded during the study period (mean from HiVol sampling): PM10 62.6 μg/m3, c-PAHs 24.7 ng/m3, B[a]P 3.50 ng/m3. The conventional cytogenetic analysis did not reveal any differences between the group of policemen exposed to the ambient air pollution and the control group. The cytogenetic analysis by FISH analysis used the whole chromosome painting probes for chromosomes #1 and #4 (Cambio, UK). It detected a significant increase in all studied endpoints in the policemen compared to controls (% AB.C. = 0.33 ± 0.25 versus 0.24 ± 0.18, p < 0.05, FG/100 = 1.72 ± 1.57 versus 1.25 ± 1.11, p < 0.05, AB/1000 (aberrations/1000 cells) = 5.58 ± 4.62 versus 3.90 ± 3.06, p < 0.05). CYP1A1*2C (Ile/Ile), XPD 23 (Lys/Lys), and XPD 6 (CC) genotypes were associated with an increase of aberrant cells by conventional method. Factors associated with an increased level of translocations by FISH included age, smoking, B[a]P-like DNA adducts (corresponding to the exposure of c-PAHs), folate, polymorphisms of CYP1A1*2C, GSTP1, EPHX1, p53 MspI and MTHFR. Ambient air exposure to c-PAHs significantly increased FISH cytogenetic parameters in nonsmoking policemen. We may conclude that FISH indicates that the city policemen in Prague represent a group of increased genotoxic risk. This is the first study that has reported a relationship between DNA adducts (biomarker of exposure) and chromosomal aberrations by FISH (biomarker of effect).  相似文献

The exploitation of methoxylated aromatic monomers by Eubacterium limosum was restricted to the cleavage and consumption of the methoxyl substituents: the phenolic residues were not further attacked. Growth characteristics were similar to those previously described for this organism on methanol. Degradation of aromatics containing more than one methoxyl group occurred in a sequential manner and transient accumulation of intermediates (particularly methyl-gallate) took place, though the enzymic mechanism for this phenomenon remains obscure. Degradation of 3,4,5-trimethoxybenzoate necessitated the initial attack of the para-methoxyl group before those groups in meta positions could be metabolised.  相似文献
