The objective of this study was to examine the effects of nitrate on methane production, important fermentation characteristics, Fibrobacter succinogenes, Ruminococcus albus, Ruminococcus flavefaciens, total bacteria, and methanogens using in vitro ruminal cultures. Potential adaptation of the above microbes and persistency of nitrate to mitigate CH4 production were also evaluated. Methane production was reduced by 70% at 12 μmol ml−1 and nearly completely at ?24 μmol ml−1 nitrate. Production of volatile fatty acids (VFAs) was affected to different extents at different nitrate concentrations. Over a series of six consecutive cultures receiving 12 μmol ml−1nitrate, production of CH4 and VFA did not change significantly. R. albus and R. flavefaciens seemed to adapt to nitrate, while F. succinogenes and methanogens did not. Nitrate may be used in achieving persistent mitigation of CH4 production by ruminants. 相似文献

Mosquito larvae experience multiple environmental stressors that may modify how subsequent adults interact with pathogens. We evaluated the effect of larval rearing temperature and intraspecific larval competition on adult mosquito immunity and vector competence for Sindbis virus (SINV). Aedes aegypti larvae were reared at two intraspecific densities (150 and 300 larvae) at 20° C and 30° C and the adults were fed artificially on citrated bovine blood containing 10(5) plaque forming units of SINV. Expression of cecropin, defensin, and transferrin was also evaluated in one- and five-day-old female adults. There was a direct relationship between larval density and SINV infection and dissemination rates at low temperature (20° C) and an inverse relationship between larval density and SINV infection rate at high temperature (30° C). Cecropin was only expressed in five-day-old adults that were raised at high temperature as larvae and was 20-fold over-expressed at low compared to high density treatments. Defensin and transferrin were under-expressed in one-day-old adults and over-expressed in five-day-old adults in all competition-temperature combinations relative to low density treatments at 20° C. These findings suggest that interaction between biotic and abiotic conditions of the larval environment may alter adult mosquito immunity resulting in enhanced vector competence for arboviruses. 相似文献

The recent evidences of presence of subsurface oceans of liquid water and ice on Saturn's moons, and the possible presence and astrobiological importance of polycyclic aromatic hydrocarbons (PAHs) in these environments, provide strong motivation for the exploration of the prebiotic chemistry in ice and to test if PAHs could be experimentally synthesized in ice surfaces under atmospheres containing methane as carbon source. In this work, we present a new design for prebiotic‐chemistry experiments in ice matrix. Using this design, a mixture of products including PAHs, polar aromatic compounds, and hydrophilic acetylene‐based polymers was obtained. We propose that acetylene generation in a methane/nitrogen atmosphere and subsequent polymerization to PAHs and polyynes could be a favored pathway in the presence of water freeze–melt cycles. These results shed light on the processes involved in PAH synthesis in icy environments and on the physical factors that drive the different competing pathways in methane/nitrogen atmospheres. 相似文献

The symbiotic plasmid (pSym) DNA present in bacteroids of strain RCR1001 of Rhizobium leguminosarum biovar viceae has been compared qualitatively and quantitatively with that present in free living bacteria by hybridization experiments with appropriate probes. A decrease in the relative amount of pSym DNA was observed in bacteroids as compared to bacteria. No rearrangements of the symbiotically expressed pSym borne genes were detected in bacteroids. 相似文献

Abstract Salterns in the Bretagne (France), exhibiting different stages of salinity of 5% to 33% evolved up to 0.7 mmol methane per m2 per day. Methane concentrations of up to 0.4 mM were found. High methane evolution rates and increased methane concetration were restricted to basins of up to 7% and more than 15% salinity, and to the upper 10 cm of the sediments, where high sulfate concentrations (50 to 100 mM) occurred as well. Basins of 10–15% salinity exhibited only low methane evolution rates (less than 0.05 mmol methane per m2 per day) and low methane concentrations (less than 0.02 mM). Gas bubbles arising during times of increased photosynthetic activity from the microbial mats covering the sediments of the saltern basins contained up to 2% methane. Addition of methylated amines and methanol, but not of H2 /CO2 , formate or acetate, to sediments slurries from basins of up to 12% salinity resulted in a rapid enrichment of methanogenic populations. Enriched methanogenic bacteria did not grow at salinities exceeding 15% or temperatures exceeding 45°C, and showed characteristics similar to those documented for Methanococcus halophilus and strain SF1 (DSM 3243). No enrichments were obtained from basins of more than 20% salinity in spite of methane being produced and evolved from those basins. 相似文献
