Flow cytometric investigation of heterogeneous copper-sensitivity in asynchronously grown Saccharomyces cerevisiae 总被引:8,自引:0,他引:8
The variable stress-sensitivity of individual cells within pure cultures is widely noted but generally unexplained. Here, factors determining the heterogeneous susceptibility to copper toxicity in Saccharomyces cerevisiae were examined with a rapid non-perturbing approach based on flow cytometry. By determination of the DNA content (with propidium iodide) in cell fractions gated by forward angle light scatter (an indicator of the cell volume), it was shown that forward angle light scatter measurements gave an approximation of the cell cycle stage. Thus, our observation that cells in different forward angle light scatter fractions displayed differing Cu-sensitivities indicated that heterogeneous Cu-sensitivity is a function of the cell cycle stage. Furthermore, cells sorted by their Cu-sensitivity and-resistance and subsequently analyzed for DNA content were found predominantly to occupy G1/S and G2/M cell cycle stages, respectively. The oxidant-sensitive probe 2',7'-dichlorodihydrofluorescein diacetate was used to show that the Cu-sensitivity of G2/M phase S. cerevisiae was correlated with greater levels of pre-existing reactive oxygen species in these cells. The results indicate that differential Cu-sensitivity in a S. cerevisiae culture is linked to the cell cycle stage and this link may be determined partly by cell cycle-dependent fluctuations in basal reactive oxygen species generation. 相似文献

A novel aqueous solution-ionic liquid biphasic catalytic system was established for the oxidative degradation of chitosan under mild conditions. In this process, the environmentally acceptable and inexpensive molecular oxygen was first used as oxidant, the metal phthalocyanine was immobilized in ionic liquid as catalyst, and the aqueous solution as medium carried the reactants and the products. Under vigorous stirring and heating, the reactants fully contacted the catalysts in the emulsion and chitosan efficiently degraded into water-soluble materials. At the end of the reaction, the catalytic system could be easily separated by simple decantation and could also be reused in subsequent runs without apparent change in activity. These characters are in favor of the elimination of pollution and the reduction of the economic cost in the large-scale production of the water-soluble chitosan derivatives in chemical industry. 相似文献

In this study 80 wine strains of Saccharomyces cerevisiae were characterized for the production of acetic acid. A significant variability in the production levels was determined among the strains, which produced from a few mg/l to more than 1 g/l. Fifteen strains, differing in acetic acid production, were tested in fermentation of grape musts of different varieties (Aglianico Basilicata, Aglianico Apulia, Cannonau, Bombino nero, Nero d'Avola, Vermentino, Fiano). The results emphasized a great strain variability, but the best strain behaviour was strictly related to the must composition, which is a determinant factor on the expression of the best strain potentiality. Therefore, this study, confirming the high/low production of acetic acid as a strain characteristic, demonstrated also that the inoculated fermentation becomes more advantageous when the starter culture is chosen in relation to the interaction of yeast strain/vine variety. 相似文献

The structures of new compounds containing Zn(II) ions and bpp (1,3-bis(4-pyridyl)propane) ligands have been determined. The coordinating halides (Br− or Cl−) produce one-dimensional compounds 6 and 7, and intra- and inter-chain CH?X (X = Br or I) interactions play roles for building crystal structures with the flexible bpp ligands. The non-coordinating anions do not produce hydroxyl bridged zinc cations or polymeric compounds, and produce only a monomeric complex 4 containing four bpp ligands and two water ligands. Previously reported polymeric compounds 1 and 2 containing hydroxyl-bridged zinc cations [Zn2OH] were found to carry out the catalytic transesterification of a range of esters with methanol at room temperature under the mild conditions, whereas the rest of compounds did not catalyze the transesterification reactions at all. Separating individual compounds by HPLC represents an effective method for the detection and quantification of phenolic compounds and has been widely utilised. However, phlorotannins are commonly quantified using colorimetric methods, as the total amount of the whole compound group. In the present paper the separation of a set of individual soluble phlorotannins from the phenolic crude extract of Fucus vesiculosus was achieved by HPLC with UV photodiode array detection. Different gradient programs for reversed- and normal-phase HPLC methods were developed and tested. Normal-phase (NP) conditions with a silica stationary phase and a mobile phase with a linear gradient of increasing polarity were found to separate 16 individual components of the phenolic extract. The suitability of the NP-HPLC method for mass spectrometric application was preliminarily tested. Sample preparation was found to be a critical step in the analysis owing to the rapid oxidation of phlorotannins; ascorbic acid was used as an antioxidant. 相似文献

Effector studies with two isoenzymes (I and IV) of glucose-6-phosphate dehydrogenase (G6PDH) from tobacco suspension culture WR-132 revealed that chlorogenic acid, at 0.4 mM, inhibited both isoenzymes almost 100%, with the inhibition decreasing as the concentration of the acid was reduced. At 0.3 and 0.4 mM, the coumarin glucosides scopolin and esculin were inhibitory, whereas their aglucones scopoletin and esculetin were less inhibitory, and at low concentrations of glucose-6-phosphate (G6P), the latter two were actually stimulatory for G6PDH I. Of the possible effectors studied, only scopoletin and esculetin exhibited a significant activation of G6PDH I under these conditions. However, with G6PDH IV these two effectors do not show the same marked activation at the low G6P concentrations. The phenolic acids, caffeic and ferulic, were less inhibitory than the coumarins tested. The activation of G6PDH I by scopoletin, a compound which accumulates in tobacco under certain stress conditions, gives a possible clue as to the resulting enhanced activity of the hexose monophosphate pathway that has been reported for some plants subjected to stress conditions. 相似文献

Photosynthates stimulate the UV-B induced fungal anthraquinone synthesis in the foliose lichen Xanthoria parietina 总被引:1,自引:0,他引:1
Synthesis of the cortical anthraquinone pigment parietin (= physcion) was studied in acetone‐rinsed, parietin‐free Xanthoria parietina thalli. UV‐B induced the synthesis, which increased linearly with UV‐B (log‐transformed) to the highest applied UV‐B level (1.8 W m?2). At natural UV‐B levels (0.75 W m?2), parietin resynthesis occurred at a constant pace (106 mg m?2 d?1) during a 14‐d period at 220 µmol m?2 s?1 PAR. Under these conditions, 56% of the natural parietin content prior to extraction was resumed, accounting for 10% of total net carbon gain. In the presence of UV‐B, the remaining results were consistent with the hypothesis assuming that photosynthates regulate the pace at which parietin is synthesized by the mycobiont. Resynthesis was rapid when photosynthesis was activated by light, or when certain carbohydrates were added exogenously. Additions of ribitol, the carbohydrate delivered from the photobiont, increased the parietin resynthesis substantially. Mannitol, the main fungal polyol, was significantly less effective. Furthermore, parietin resynthesis in X. parietina was depressed at high and low hydration when net photosynthesis is depressed. Therefore, the photobiont regulates the parietin resynthesis pace in its mycobiont partner by the delivery of photosynthates. In conclusion, both lichen bionts play important roles in the synthesis of parietin, which probably acts as a PAR‐ rather than a UV‐B‐screen. 相似文献
