Carbachol (CCh), a muscarinic agonist that elicits the formation of inositol trisphosphate (IP3) and diacylglycerol (DG), induces a calcium-dependent [3H]norepinephrine ([3H]NE) release [IC50 = (2.7 +/- 0.5) X 10(-4) M] in rat brain slices. Similarly, other muscarinic agonists evoke [3H]NE release which is specifically inhibited by muscarinic antagonists such as 3-quinuclidinyl benzilate, atropine, and N-methyl-4-piperidyl benzilate. The atropine-sensitive evoked release is effectively inhibited by neomycin (IC50 = 50 microM), a phospholipase C inhibitor that interferes with IP3-dependent cellular processes. In addition, polymyxin B, a rather selective inhibitor of protein kinase C (PK-C), abolishes the agonist-mediated release with a half-maximal effective concentration of 0.53 microM (750 ng/ml). These results have a significant implication for the mechanism by which agonists generating IP3 and DG act as inducers of neurotransmitter release in the CNS. However, since both neomycin and polymyxin B act also as N-calcium-channel blockers, other possible mechanisms are discussed. The CCh-induced release suggests that in the CNS an agonist-receptor interaction leads to a calcium-dependent neurotransmitter release, most likely via promoting the IP3/DG as second messengers followed by activation of PK-C. 相似文献

Maitotoxin-Evoked γ-Aminobutyric Acid Release Is Due Not Only to the Opening of Calcium Channels 总被引:1,自引:1,他引:0
The effects of maitotoxin (MTX) on endogenous amino acid release were tested on highly purified striatal neurons differentiated in primary culture. MTX induced a large and concentration-dependent release of gamma-aminobutyric acid (GABA). This effect was abolished when experiments were performed in the absence of external Ca2+, and restored when Ca2+ ions were added after removing the MTX-containing Ca2+-free solution. MTX-induced amino acid release was not affected by 1 microM nifedipine and only slightly inhibited by 1 mM Co2+. MTX also induced a massive accumulation of 45Ca2+ in the neurons which, in contrast to the MTX-evoked GABA release, was totally blocked in the presence of 1 mM Co2+. Whereas 500 nM tetrodotoxin was without significant effect, MTX-evoked GABA release was dependent on the presence of external Na+ and sensitive to nipecotic acid, a GABA uptake inhibitor. It is concluded that, on striatal neurons, MTX induced Na+ influx only in the presence of external Ca2+. The increase in cytoplasmic Na+ ions then triggers the release of GABA. 相似文献

Summary Relationships between fine root growth, rates of litter decomposition and nutrient release were analysed in a mixed forest on Tierra Firme, a Tall Amazon Caatinga and a Low Bana on podsolized sands near San Carlos de Rio Negro. Fine root growth in the upper soil layers (root mat+10 cm upper soil) was considerably higher in the Tierra Firme forest (1117 g m-2 yr-1) than in tall Cattinga (120) and Bana (235). Fine root growth on top of the root mat was stimulated significantly by added N in Tall Caatinga and Low Bana forests, by P in Tierra Firme and Bana forests, and by Ca only in the Tierra Firme forest. Rate of fine root growth in Tierra Firme forest on fresh litter is strongly correlated with the Mg and Ca content of litter. Rate of litter decomposition was inversely related to % lignin and the lignin/N ratio of litter. Litter contact with the dense root mat of the Tierra Firme increased rates of disappearance for biomass, Ca and Mg as compared with litter permanently separated or lifted weekly from the root mat to avoid root attachment. Nitrogen concentration of decomposing litter increased in all forests, net N released being observed only in Caryocar glabrum and Aspidosperma megalocarpum of the Tierra Firme forest after one year of exposure. Results emphasize the differences in limiting nutrients in amazonian forest ecosystems on contrasting soil types: Tierra Firme forests are particularly limited by Ca and Mg, while Caatinga and Bana forests are limited mainly by N availability. 相似文献

Calcium, magnesium and potassium dynamics in decomposing litter of three tree species were measured over a two-year period. The speices studied were flowering dogwood (Cornus florida), red maple (Acer rubrum) and chestnut oak (Quercus prinus). The order of decomposition was:C. florida>A. rubrum>Q. prinus.Calcium concentrations increased following any initial leaching losses. However, there were net releases of Ca from all three litter types since mass loss exceeded the increases in concentration. Net release of Ca by the end of two years from all three species combined was 42% of initial inputs in litterfall. Magnesium concentrations increased in the second year, following decreases due to leaching during the first year inC. florida andA. rubrum litter. Net release of Mg by the end of two years was 58% of initial inputs. Potassium concentrations decreased rapidly and continued to decline throughout the study. Net release of K by the end of two years was 91% of initial inputs.These data on cation dynamics, and similar data on N, S and P dynamics from a previous study, were combined with annual litterfall data to estimate the release of selected nutrients from foliar litter of these tree species at the end of one and two years of decomposition. Fine details of the infradian O2 consumption cycles that characterize pupal diapause in flesh flies have been monitored by a newly designed microrespirographic method coupled with an electronically regulated O2 generator. During the 4-5 days between the peaks of elevated O2 consumption, the diapausing pupae maintained a very low and fairly constant respiratory rate (13 microl O2 x g-1.h-1). During the intercalated peaks of increased respiratory metabolism, which lasted an average of 33.6 h to 24-27 degrees C, the average maximum rate of O2 consumption was 86.9 microl.g-1.h-1, a value of 6.7 times higher than the interpeak values. The respiratory peaks started abruptly in some cases while the decline was consistently gradual. During the periods between the peaks there were no discontinuous bursts of CO2 release, a feature common to diapause in many other insects. Diapause was characteristically terminated during a peak of the O2 consumption cycle. At diapause termination O2 consumption remained at the maximum values of the peak for many hours and then gradually increased to levels characteristic of nondiapause development. 相似文献

Dicotyledonous plants subjected to Fe-deficiency stress can decrease pH in the rhizosphere by proton excretion and reduce ferric iron by an activated reduction system in the plasma membranes of the root or by reductants released from the roots. The efficiency by which these plants take up Fe may strongly depend on their cation-anion balance. This study presents results of two experiments conducted to evaluate the effect of K, growth stage and cultivar on ionic balance and Fe acquisition of peanut (Arachis hypogaea L.) plants.Potassium applications to the high calcareous soil (30.3% CaCO3) favoured proton release, but did not ameliorate plant Fe acquisition. At the earliest stages of plant growth, anion uptake exceeded cation uptake due to intensive N uptake. With time, a shift in the ionic balance was observed as a result of predominant cation uptake. It appears that the relationship between H/OH-ion release and Fe nutrition of peanut plants is actually a complex phenomenon under soil conditions and depends on some soil parameters, such as CaCO3 content. Even by enhanced H-ion release Fe nutrition of plants can be impaired if soil CaCO3 is too high. 相似文献

Summary In rat pancreatic islets, a rise in extracellular D-glucose concentration is known to cause a greater increase in the oxidation of D-[6-14C]glucose than utilization of D-[5-3H]glucose. In the present study, such a preferential stimulation of acetyl residue oxidation relative to glycolytic flux was mimicked by nutrient secretagogues such as 2-aminobicyclo[2,2,1]heptane-2-carboxylate, 3-phenylpyruvate, L-leucine, 2-ketoisocaproate, D-fructose and ketone bodies. The preferential stimulation of D-[6-14C]glucose oxidation by these nutrients was observed at all hexose concentrations (0.5, 6.0 and 16.7 mM), coincided with an unaltered rate of D-[3,4-14C]glucose oxidation, was impaired in the absence of extracellular Ca2+, and failed to be affected by NH4
+. Although the ratio between D-[6-14C]glucose oxidation and, D-[5-3H]glucose utilization in islets exposed to other nutrient secretagogues could be affected by factors such as isotopic dilution and mitochondrial redox state, the present data afford strong support to the view that the preferential stimulation of oxidative events in the Krebs cycle of nutrient-stimulated islets is linked to the activation of key mitochondrial dehydrogenases, e.g. 2-ketoglutarate dehydrogenase. The latter activation might result from the mitochondrial accumulation of Ca2+, as attributable not solely to stimulation of Ca2+ inflow into the islet cells but also to an increase in ATP availability. 相似文献

Summary The giant mucous cells in the skin of the terrestrial banana slug Ariolimax columbianus secret intact granules containing mucins. Electron microscopy, after ultrarapid freezing and freeze-substitution in osmium, shows that the secreted granules are bounded by two distinct membranes, presumably derived from the Golgi apparatus and the plasmalemma. Relatively stable, intact granules can be obtained in great quantity in our in vitro system. Rapid lysis of the granules was induced by adenosine triphosphate. At much higher concentrations, adenosine diphosphate and 5-adenylimido-diphosphate also caused lysis. Other nucleotides and related compounds, as well as 1,4,5-inositol triphosphate and molluscan neurotransmitters and neuropeptides, had no effect on the granules. The stability of secreted granules varied with the ionic composition of the isosmotic medium in which they were suspended. When the predominant cation in the medium was potassium, and calcium was also present, granules lysed if exposed to shear stress (stirring of the suspension). This did not occur if sodium was the major cation present. None of the other ions in the suspension media had detectable effects on the stability of the granules. 相似文献

We have previously described a preferential reduction in the secretory response to nutrient secretagogues in pancreatic mouse
islets maintained in culture after in vitro exposure to streptozotocin (SZ). This reduction was associated with an impaired
substrate metabolism at the mitochondrial level. To further clarify this issue, mouse pancreatic islets were exposed in vitro
to 2.2 mM SZ for 30 min. At 4 h after SZ treatment ultrastructural changes were apparent in the endoplasmic reticulum and
Golgi areas of the B-cells. However, 2 and 6 days following SZ exposure the B-cells appeared well preserved, except for a
marked decrease in the number of insulin-containing secretory granules. A morphometric analysis of the B-cells 6 days after
SZ exposure showed a normal B-cell size and a normal volume fraction of B-cell mitochondria. However, there was a decrease
in total islet size and a 13% decrease in the volume fraction of B-cells in the islets. These mouse islets exhibited a decreased
content of the mitochondrial DNA-encoded cytochrome b mRNA, as evaluated by dot-blot analysis. As a whole, the data obtained
indicate that SZ treatment does not induce a decrease in the number of mitochondria or long-lasting ultrastructural damage
to this organelle. However, there is a clear decrease in the cytochrome b mRNA, suggesting that SZ can induce damage to the

The release of endogenous acetylcholine and dopamine and the appearance of their metabolites, choline and dihydroxyphenylacetic acid (DOPAC), from neostriatal slices prepared from Fischer 344 rats was examined under various experimental conditions. There was a dose-dependent increase in the amount of neurotransmitter or metabolite as the medium potassium concentration was increased from 5 to 50 mM. Over an eight minute period in Krebs Ringer bicarbonate buffer containing 25 mM potassium, the rate of release of acetylcholine was 6 to 13 times greater than that of dopamine. The dopamine endogenous to the slice preparation appeared to have little effect on the release of endogenous acetylcholine since manipulations that significantly altered dopamine release (depletion with 6-hydroxydopamine or uptake inhibition with nomifensine) had minimal effects on the cholinergic neurons. In contrast, increasing the endogenous acetylcholine in the preparation by inhibiting acetylcholinesterase resulted in a 1.2 to 12 fold increase in dopamine release depending upon the incubation time and the potassium concentration. These studies indicate that within the neostriatal slices there is minimal influence of the endogenous dopamine on the cholinergic neurons, whereas the extracellular acetylcholine can influence dopamine release when its concentration is increased by inhibition of acetylcholinesterase. 相似文献
