Cultured L1210 lymphocytic leukemic cells resistant to cytosine arabinoside or Cytoxan were frozen under different conditions for up to 5 months and transplanted into recipient mice. Biochemical determinations including DNA, total RNA and drug resistance suggested that the more rapid, less expensive method of freezing mouse leukemic cells enables good retention of each parameter. Examination of cellular and subcellular RNA fingerprints indicated several alterations in the localizations f certain subspecies of RNA. Nonetheless, all cells retained their overall viability and the capacity to induce leukemia in CDF1 mice.  相似文献

Grazer-periphyton interactions were investigated in 11 laboratory streams holding a range of densities of three herbivore taxa during a 32-d experiment. Effects of grazers on algae were strongest with Dicosmoecus gilvipes caddisflies, intermediate with Juga silicula snails, and weakest with Baetis spp. mayflies. Algal standing crop, export, and gross primary production declined logarithmically with increasing grazer density. Algal turnover rate, however, increased with grazer abundance. At high densities of all grazers, responses in most algal parameters converged, suggesting that high grazing pressure, regardless of taxon, will similarly affect periphyton. Growth of both Dicosmoecus caddisflies and Juga snails was density-dependent, with the highest growth rates occurring at the lowest densities. Caddisflies displayed high growth rates but low efficiency in resource use. Snails had lower growth rates but were more efficient in resource use. The coexistence of Dicosmoecus and Juga, or other competing herbivores, in natural streams may be related to these fundamental differences in life history strategies.Department of Fisheries and Wildlife  相似文献

Rating scales for difficulty in capturing and holding mice were devised that proved to be easy to use and highly sensitive to differences among mouse strains on the A and B priority lists of the Mouse Phenome Project. The simplicity of the scales makes it feasible to rate wildness during behavioral test sessions without adding much to testing time or distracting the technician from the principal task at hand. Overall wildness and placidity ratings obtained by combining capture and hold ratings provide a good impression of the difficulties encountered while working with lab mice in the course of complex experiments. Ratings of 21 inbred strains during the course of 15 behavioral tests in two laboratories demonstrated that the SPRET/Ei, PERA/Ei, CAST/Ei and SWR/J strains were particularly difficult to handle. The NOD/LtJ strain posed no special challenge in the Edmonton laboratory but was very difficult to handle in the Portland lab. The rating scales should be useful for judging the difficulties in working with novel targeted or induced mutations in mice as well as effects of a variety of environmental treatments or drugs.  相似文献

The recent interest in natural killer (NK) cells in immunosurveillance and the ability of infection with certain organisms to modulate NK activity led us to examine the influence of Toxoplasma gondii infection on mouse NK cells. Infection of BALB/c mice with 5 × 103 virulent Toxoplasma intraperitoneally (ip) resulted in significantly enhanced NK activity in peritoneal exudate cells (PC) and in spleen cells (SC). Intravenous (iv) and subcutaneous (sc) challenge of BALB/c mice with Toxoplasma also resulted in enhanced natural killer (NK) activity in PC and SC. In BALB/c mice, as well as in other strains (A/J, C57BL/6, C3H/HeJ, CeH/HeN, [A/J × C3H]F1), peak augmentation of PC and SC NK activity was observed 3 days following ip Toxoplasma challenge. Administration of silica to mice abolished Toxoplasma-induced NK cytotoxicity. BALB/c mice chronically infected with Toxoplasma had significantly higher endogenous NK activity than did controls in PC but not in SC. Chronically infected BALB/c mice boosted with virulent Toxoplasma ip exhibited significantly enhanced NK activity in PC but not in SC. Thus, acute and chronic infection with Toxoplasma modulates NK activity in addition to macrophage activation and thereby provides a system that should facilitate study of the relative contribution of NK cells and activated macrophages in resistance to tumor growth and spread.  相似文献

Cutaneous leishmaniasis (CL) is gaining attention as a public health problem. We present two cases of CL imported from Syria and Venezuela in Japan. We diagnosed them as CL non-invasively by the direct boil loop-mediated isothermal amplification method and an innovative sequencing method using the MinION? sequencer. This report demonstrates that our procedure could be useful for the diagnosis of CL in both clinical and epidemiological settings.  相似文献

Preliminary gut analysis of a recent Great Lakes invader, the round goby, Neogobius melanostomus (7.0–8.4 cm), collected from the Detroit River, showed that they ate zebra mussels (58%), snails (6%), and other invertebrates (36%), including aquatic insects (Hexagenia), softshelled crayfish, and zooplankton. Because zebra mussels, Dreissena polymorpha, predominated as prey, we investigated the ability of round gobies to consume different size classes of zebra mussels. In laboratory experiments, we examined feeding preferences of three size classes of round gobies (5.5–6.9 cm; 7.0–8.4 cm; 8.5–10.3 cm standard length) on four different size classes of zebra mussels (6.0–9.9 mm, 10.0–12.9 mm, 13.0–15.9 mm, 16.0–18.9 mm). All sizes of round gobies ate zebra mussels < 10.0 mm. Only the largest size class of round gobies ate larger zebra mussels (10.0–12.9 mm) when all prey sizes were presented. The association between the total mass of zebra mussels available and the amount consumed by round gobies increased positively up to about 6.5 g of available mussels and then levelled off. Round gobies consumed an average of 1.0 g of mussels in 24 h. There was a significant positive relationship between gape size and standard length of round gobies. Although larger round gobies (over the size range of fish in our study) are able to consume larger zebra mussels, small mussels were preferred. Our findings suggest that the preference of small zebra mussels by round gobies has the potential to alter the size structure of zebra mussel populations. On the basis of the currently accepted model for the cell membrane structure, a physico-chemical model for mediated transport is developed and solved for the case of polar non-electrolyte migration through the cell membrane. The model considers the interstitial space defined by the transport protein subunits to be the migration pathway for polar solutes. A Langmuir-type adsorption equilibrium is assumed at the interfaces and a multicomponent diffusion mechanism of solute and water is postulated within the migration pathway, where the polar residues of the transport protein represent another component of the system. Membrane selectivity is governed by the adsorption constants, which are shown to affect strongly the kinetics of transport. Isosmotic transport and the volume change of the cell are important features incorporated in the model, which is shown to fulfill the peculiar properties of facilitated diffusion systems. It is concluded that the same type of pathway can be used for the transport of other polar solutes through existing or induced hydrophilic channels, for which a similar approach is suggested.  相似文献
