Cutinase enzymes from fungi have found diverse applications in industry. However, most of the available literature on cutinase
production is related to the cultivation of genetically engineered bacteria or yeast cells. In the present study, we use mixture
design experiments to evaluate the influence of six nutrient elements on production of cutinase from the fungus Colletotrichum lindemuthianum. The nutritional elements were starch, glucose, ammonium sulfate, yeast extract, magnesium sulfate, and potassium phosphate.
In the experimental design, we imposed the constraints that exactly one factor must be omitted in each set of experiments
and no factor can account for more than one third of the mixture. Thirty different sets of experiments were designed. Results
obtained showed that while starch is found to have negative influence on the production of the enzyme, yeast extract and potassium
phosphate have a strong positive influence. Magnesium sulfate, ammonium sulfate, and glucose have low positive influence on
the enzyme production. Contour plots have also been created to obtain information concerning the interaction effects of the

We develop and characterize a disordered polymer optical fiber that uses transverse Anderson localization as a novel waveguiding mechanism. The developed polymer optical fiber is composed of 80,000 strands of poly (methyl methacrylate) (PMMA) and polystyrene (PS) that are randomly mixed and drawn into a square cross section optical fiber with a side width of 250 μm. Initially, each strand is 200 μm in diameter and 8-inches long. During the mixing process of the original fiber strands, the fibers cross over each other; however, a large draw ratio guarantees that the refractive index profile is invariant along the length of the fiber for several tens of centimeters. The large refractive index difference of 0.1 between the disordered sites results in a small localized beam radius that is comparable to the beam radius of conventional optical fibers. The input light is launched from a standard single mode optical fiber using the butt-coupling method and the near-field output beam from the disordered fiber is imaged using a 40X objective and a CCD camera. The output beam diameter agrees well with the expected results from the numerical simulations. The disordered optical fiber presented in this work is the first device-level implementation of 2D Anderson localization, and can potentially be used for image transport and short-haul optical communication systems. 相似文献

The use of cruciferous plant residues to reduce the amount of sclerotia of Sclerotium cepivorum and chlamydospores of Fusarium oxysporum f. sp. lycopersici in soil was investigated. Air‐dried and crushed mustard (Brassica juncea) added to the soil effectively reduced the viability of fungal propagules. Consequently, the reduction of white rot of onion, caused by S. cepivorum and wild of tomato caused by F. oxysporum was observed. The addition of rapeseed (Brassica. napus cv. Bolko and B. napus cv. Gorczanski) residues to soil also resulted in a significant decrease of number of S. cepivorum sclerotia but the effect on F. oxysporum chlamydospores was variable. Introduction of the plant material increased the total number of bacteria, spore‐forming bacteria, fluorescent pseudomonads, actinomycetes, and fungi in soil. One year after the soil amendment, the amount of sporeforming bacteria in treatments with cruciferous residues was higher as compared to the control soil without plant residues. The possible contribution of the decomposition of plant residues and soil micro‐organisms to the reduction of the pathogens population is discussed. 相似文献

In an outside pot experiment, dry pig manure processed on pine sawdust litter and fermented for seven days by house fly larvae (fermented manure), and pine sawdust applied alone, and in combination with a spring application of inorganic nitrogen fertilizer were used to determine their effects on plant parasitic and free-living soil nematodes on sugar beets (cv. Antek). Non amended soil was used as a control. All treatments with fermented pig manure and sawdust with nitrogen fertilizer decreased number of plant parasitic nematodes and also root-fungal feeding nematodes compared to the untreated control. Sawdust applied alone had no effect on plant parasitic and root-fungal feeding nematode suppression. Free-living nematodes which were mainly bacteriovores and fungivores were significantly more abundant in soil amended with fermented pig manure, while the sawdust had no effect on these nematodes. The effect of all tested treatments on omnivores-predators was rather random, and in general, the number of these nematodes decreased after soil amendment applications compared to the untreated control. 相似文献

The composition and formulation of an artificial medium and handling techniques are described for rearing the chalcid insect endoparasite, Brachymeria lasus (Walker) from the egg to adult stage under in vitro conditions. The medium was devoid of host materials, prepared aseptically, and contained 15% () albumin, 1% amino acids, 4% glucose, and approximately 2.5% lipids, 0.3% inorganic salts, 0.01% B vitamins, and 0.2% lipogenic growth factors. Inflammation of the middle ear cavity (otitis media) and the abnormal deposition of bone at the otic capsule are common causes of conductive hearing impairment in children and adults. Although a host of environmental factors can contribute to these conditions, a genetic predisposition has an important role as well. Here, we analyze the Tail-short (Ts) mouse, which harbors a spontaneous semi-dominant mutation that causes skeletal defects and hearing loss. By genetic means, we show that the Ts phenotypes arise from an 18-kb deletion/insertion of the Rpl38 gene, encoding a ribosomal protein of the large subunit. We show that Ts mutants exhibit significantly elevated auditory-brain stem response thresholds and reduced distortion-product otoacoustic emissions, in the presence of normal endocochlear potentials and typical inner ear histology suggestive of a conductive hearing impairment. We locate the cause of the hearing impairment to the middle ear, demonstrating over-ossification at the round window ridge, ectopic deposition of cholesterol crystals in the middle ear cavity, enlarged Eustachian tube, and chronic otitis media with effusion all beginning at around 3 weeks after birth. Using specific antisera, we demonstrate that Rpl38 is an ~8-kDa protein that is predominantly expressed in mature erythrocytes. Finally, using an Rpl38 cDNA transgene, we rescue the Ts phenotypes. Together, these data present a previously uncharacterized combination of interrelated middle ear pathologies and suggest Rpl38 deficiency as a model to dissect the causative relationships between neo-ossification, cholesterol crystal deposition, and Eustachian tubes in the etiology of otitis media. 相似文献

Conservation biological control is the modification of the environment or existing practices to protect and enhance antagonistic organisms to reduce damage from pests. This approach to biological control has received insufficient attention compared with inundative applications of microbial antagonists to control nematodes. This review provides examples of how production practices can enhance or diminish biological control of plant-parasitic nematodes and other soilborne pests. Antagonists of nematodes can be enhanced by providing supplementary food sources such as occurs when organic amendments are applied to soil. However, some organic amendments (e.g., manures and plants containing allelopathic compounds) can also be detrimental to nematode antagonists. Plant species and genotype can strongly influence the outcome of biological control. For instance, the susceptibility of the plant to the nematode can determine the effectiveness of control; good hosts will require greater levels of suppression than poor hosts. Plant genotype can also influence the degree of rhizosphere colonization and antibiotic production by antagonists, as well the expression of induced resistance by plants. Production practices such as crop rotation, fallow periods, tillage, and pesticide applications can directly disrupt populations of antagonistic organisms. These practices can also indirectly affect antagonists by reducing their primary nematode host. One of the challenges of conservation biological control is that practices intended to protect or enhance suppression of nematodes may not be effective in all field sites because they are dependent on indigenous antagonists. Ultimately, indicators will need to be identified, such as the presence of particular antagonists, which can guide decisions on where it is practical to use conservation biological control. Antagonists can also be applied to field sites in conjunction with conservation practices to improve the consistency, efficacy, and duration of biological control. In future research, greater use should be made of bioassays that measure nematode suppression because changes in abundance of particular antagonists may not affect biological control of plant parasites. 相似文献

Pot and field experiments were conducted to investigate the effects of soil amendments (cow manure, rice straw, zeolite, dicalcium phosphate) on the growth and metal uptake (Cd, Zn) of maize (Zea mays) grown in Cd/Zn contaminated soil. The addition of cow manure and rice straw significantly increased the dry biomass, shoot and root length, and grain yield of maize when compared with the control. In pot study, cow manure, rice straw, and dicalcium phosphate all proved effective in reducing Cd and Zn concentrations in shoots and roots. Cd and Zn concentrations in the grains of maize grown in field study plots with cow manure and dicalcium phosphate amendments to highly contaminated soil (Cd 36.5 mg kg?1 and Zn 1520.8 mg kg?1) conformed to acceptable standards for animal feed. Additionally both cow manure and dicalcium phosphate amendments resulted in the significant decrease of Cd and Zn concentrations in shoots of maize. 相似文献

The abandoned chromite-asbestos mines are located in the Roro hills, West Singhbhum, Jharkhand, India, where mining operation ceased in 1983, and since then these mines are causing environmental pollution. The present study was planned to phytoremediate these metalloid and metal contaminated mine waste by using two aromatic grasses, Cymbopogon citratus and Chrysopogon zizanioides by applying different proportions of amendments (chicken manure, farmyard manure and garden soil). Mine waste has neutral pH, low electrical conductivity and organic carbon with higher concentration of total metals (Cr and Ni) as compared to soil. Application of manures resulted significant improvements of mine waste characteristics and plant growth, reduction in the availability of total extractable toxic metals (Cr, Ni) and increase in Mn, Zn and Cu concentration in the substrate. The maximum growth and biomass production for C. citratus and C. zizanioides were found in T-IV combination comprising of mine waste (90%), chicken manure (2.5%), farmyard manure (2.5%) and garden soil (5%). Addition of T-IV combination also resulted in low Cr and Ni accumulation in roots and reduction in translocation to shoots. Study indicates that C. citratus and C. zizanioides can be used for phytostabilization of abandoned chromite-asbestos mine waste with amendments. 相似文献
