依照2005年版《中国药典》和2003年版《国家药品标准》,考察注射用胸腺肽制剂的热稳定性。将注射用胸腺肽制剂成品3批样品,于温度60℃±2℃、水浴自然光条件下,放置10h,多时段取样,观察其外观、多肽含量、酸碱度、可见异物和生物学活性。结果表明三批供试品的的各项指标均符合国家药品标准规定。注射用胸腺肽具有良好的热稳定性。  相似文献

A flow-injection method is described for the determination of carbaryl and carbofuran. It was found that a strong chemiluminescence (CL) signal was generated when these pesticides were mixed with Na(2)SO(3) and KMnO(4) in acidic medium. Under the optimum experimental conditions, the enhanced CL intensity was linear, with the concentrations in the range 0.1-2.0 microg/mL (r(2) = 0.9996 and 0.9993, n = 6) with relative standard deviation (n = 4) in the range 1.0-2.3%. The limits of detection (3sigma blank) were 10 and 50 ng/mL, respectively, with a sample throughput of 180/h. The proposed method was applied to determine carbaryl and carbofuran in freshwaters with satisfactory results. Most metal and non-metal ions and some pesticides, such as carbophenothion and aldicarb, do not interfere with the determination. Dinoseb, diazinon and malathion calibration graphs (in the range 0.2-2.0 microg/mL, r(2) = 0.9966-0.9988, n = 6) were also established with relative standard deviations (n = 4) in the range 1.2-2.0% with limits of detection (3sigma blank) in the range 100-300 ng/mL.  相似文献

An amperometric l-lactate biosensor was fabricated by confining lactate oxidase in a Prussian Blue-modified electrode with a Nafion membrane. The detector was assembled in a flow injection apparatus and operated at -0.1 V. Conditions for optimal electrode response were determined by investigating the influence of the amount of immobilized enzyme, the sample volume, and the flow rate. At the established operational conditions, the biosensor exhibited negligible response from interfering species usually present in biological fluids. The stability of the biosensor was also investigated, and its sensitivity was maintained unchanged at certain experimental conditions. l-Lactate was determined in blood samples, and the influence of physical exercise on the results was clearly evidenced, demonstrating that the proposed amperometric detector is suitable for monitoring changes in the l-lactate levels in biological fluids.  相似文献

Hypermetabolism and anorexia are significant problems associated with major burn trauma. Recent studies have implicated hypothalamic peptides and receptors of the corticotropin releasing factor (CRF) family as putative mediators of burn-induced hypermetabolism. Increased neuronal activity at the CRF type 2 receptor (CRF R-2) appeared particularly involved in the expression of elevated resting energy expenditure (REE) following major burn trauma. In the present study we continued these investigations of CRF R-2 mediation of burn-induced hypermetabolism, demonstrating that 3rd ventricle injection of CRF R-2 antisense oligodeoxynucleotide (ODN) normalized REE in burned rats. Similar treatments with CRF or CRF R-1 antisense ODNs had no significant effect in burned rats. In addition, 3rd ventricle injection of the selective CRF R-2 antagonist, antisauvagine-30, also reduced REE significantly in burned rats, while similar treatment with the selective CRF R-1 antagonist, antalarmin, was without effect. To determine which endogenous peptide was altered following burn we measured hypothalamic levels of urocortin (UCN) and CRF 15 days after burn injury, finding UCN was significantly elevated by nearly 3-fold, while CRF level tended to be decreased. We also assessed hypothalamic mRNA peptide and receptor expression by real-time PCR 7, 14, and 21 days post-burn, observing decreased CRF expression 7 and 21 days post-burn, decreased UCN-2 expression 7 days post-burn, and no significant alteration in UCN-1 at any time point. However, CRF R-2 mRNA was elevated at each post-burn time point. These results continue to suggest that increased neuronal activity is integrally involved in the mediation of burn-induced hypermetabolism, and that one of the UCN peptides may be the endogenous ligand affecting this receptor.  相似文献

摘要 目的:观察艾迪注射液联合XELOX方案(卡培他滨+奥沙利铂)对晚期结直肠癌患者肿瘤标志物、血管生成和外周血辅助性T细胞17(Th17)/调节性T细胞(Treg)平衡的影响。方法:选择我院2018年2月~2019年8月期间收治的98例经内镜活检证实的晚期结直肠癌患者,根据随机数字表法分为对照组(XELOX方案化疗,49例)和研究组(艾迪注射液联合XELOX化疗方案治疗,49例)。观察两组患者近、远期疗效,对比两组治疗前后的肿瘤标志物、血管生成和Th17/Treg平衡相关指标水平,记录两组用药安全性。结果:与对照组相比,研究组的客观缓解率、疾病控制率明显更高(P<0.05);研究组的总生存率、无进展生存率高于对照组,但两组间差异无统计学意义(P>0.05)。治疗后,两组癌胚抗原(CEA)、糖链抗原125(CA125)、糖链抗原242(CA242)水平均下降,且研究组低于对照组同期(P<0.05)。治疗后,两组血管内皮生长因子(VEGF)、肿瘤微血管密度(MVD)水平均下降,且研究组低于对照组同期(P<0.05)。治疗后,两组Th17比例、Th17/Treg比值均升高,Treg比例均下降(P<0.05);研究组治疗后Th17比例、Th17/Treg比值高于对照组同期,Treg比例低于对照组同期(P<0.05)。两组不良反应总发生率对比未见明显差异(P>0.05)。结论:艾迪注射液联合XELOX方案治疗晚期结直肠癌的疗效确切,可能与改善Th17/Treg平衡、抑制血管生成有关。  相似文献

关节腔内注射细胞制剂是治疗骨关节炎(OA)的新趋势,且展现了良好的临床效果、安全性及可行性。细胞制剂有多种类型,包括血小板、干细胞和单个核细胞(MNCs)。血小板制剂倾向于基层医疗机构应用和早期OA患者的治疗;MNCs较适用于不具备干细胞培养条件的医疗机构和中期OA患者应用;而干细胞制剂较适合高级别医疗机构和晚期OA患者应用。虽然腔内注射细胞产品治疗OA进行了大量临床研究,但亦有不足;因此制定腔内注射治疗OA不同细胞制剂的质量控制标准、临床应用标准和进行大样本多中心随机对照临床试验势在必行。探索外泌体、外周血MNCs治疗OA的临床研究及其分子机制,是研究的新切入点。  相似文献

The eye is a small and enclosed organ which makes it an ideal target for gene therapy. Recently various strategies have been applied to gene therapy in retinopathies using non-viral and viral gene delivery to the retina and retinal pigment epithelium (RPE). Subretinal injection is the best approach to deliver viral vectors directly to RPE cells. Before the clinical trial of a gene therapy, it is inevitable to validate the efficacy of the therapy in animal models of various retinopathies. Thus, subretinal injection in mice becomes a fundamental technique for an ocular gene therapy. In this protocol, we provide the easy and replicable technique for subretinal injection of viral vectors to experimental mice. This technique is modified from the intravitreal injection, which is widely used technique in ophthalmology clinics. The representative results of RPE/choroid/scleral complex flat-mount will help to understand the efficacy of this technique and adjust the volume and titer of viral vectors for the extent of gene transduction.  相似文献

目的:探讨丹红注射液对急性冠脉综合征患者血浆脑钠肽水平及心率变异性的影响。方法:选取我院收治的急性冠脉综合征患者72例,并将其随机分为实验组和对照组。对照组36例,予以阿司匹林肠溶片100 mg/日一次口服、低分子肝素钠200 IU/kg体重,每日一次皮下注射及普萘洛尔20 mg/日3次口服等常规治疗,2周为一个疗程,治疗两个疗程;实验组36例,在常规治疗的基础上加用丹红注射液,30 m L,加入5%葡萄糖注射液250 m L滴注,每日二次,2周为一个疗程,治疗两个疗程。治疗结束后,比较治疗前后两组患者的血浆脑钠肽水平及心率变异性中SDNN、SDANN、RMSSD、PNN50%变化。结果:1治疗后,两组患者血浆脑钠肽水平均有所改善,且实验组血浆脑钠肽(96.72±19.43 ng/L)较对照组血浆脑钠肽(147.53±26.89 ng/L)明显下降,差异有统计学意义(P0.05);2治疗后,两组患者心率变异性中SDNN、SDANN、RMSSD、PNN50%各指标均有所改善,且实验组改善情况明显优于对照组,差异有统计学意义(P0.05)。结论:丹红注射液能够明显降低急性冠脉综合征患者的血浆脑钠肽水平,并显著改善心率变异性各项参数,从而可能有助于改善记性冠脉综合征患者的预后。  相似文献

目的:探讨丹参酮IIA 磺酸钠注射液辅助治疗对突发性耳聋患者血液流变学及临床疗效的影响。方法:选取我院收治的突发 性耳聋患者78 例,并将其随机分为实验组和对照组,每组39 例。对照组给予三磷酸腺苷二钠、辅酶A、复合维生素、双密达莫片 及高压氧治疗,而实验组在对照组的基础上给予丹参酮IIA 磺酸钠注射液治复合组治疗。观察和比较两组患者治疗后的听力恢复 等级分布以及治疗前后的电测听情况和各项血液流变学指标的变化情况。结果:治疗后,与对照组比较,实验组听力恢复Ⅰ级患 者的比例更高,电听力平均值均显著降低,各项血液流变学指标均明显改善,差异均有统计学意义(P<0.05)。结论:丹参酮IIA 磺酸 钠注射液辅助治疗能够显著提高突发性耳聋的临床疗效,这可能其改善患者的血液流变学有关  相似文献

目的:探究黄芪注射液对慢性心力衰竭患者外周血清B型脑利钠肽水平的影响。方法:选择2013年12月-2015年1月我院收治的慢性心力衰竭住院患者334例,按用药不同分为实验组和对照组。对照组患者采取常规药物口服治疗,实验组患者应用黄芪注射液静脉滴注。观察并比较两组患者治疗前后外周血B型脑利钠肽水平的变化情况。结果:治疗后,两组患者外周血B型脑利钠肽水平均明显下降,且实验组下降更明显,实验组Ⅰ级、Ⅱ级患者比例较对照组显著升高,差异均具有统计学意义(P0.05)。结论:黄芪注射液能够降低慢性心力衰竭患者的外周血B型脑利钠肽水平,改善其心功能,值得临床推广应用。  相似文献
